Young adult rabbits (2 to 3 kg.) were subjected to a single total body exposure of x-radiation ranging from 50 to 1200 r and tested frequently for the presence of Cx-reactive protein in their blood. It usually appeared in two phases separated by an interval of several days. The primary phase occurred 24 to 48 hours after irradiation in almost all (92 of 97) rabbits exposed to 500 r or more and in 4 of 16 exposed to 300 r or less. The secondary phase occurred during the 2nd week in many of the rabbits irradiated with 900 r or more and in a few irradiated with 700 r. Autopsy cultures failed to demonstrate the presence of infection in rabbits which died or were sacrificed during the primary phase. Bacterial infection was demonstrated, however, in almost all (21 of 24) rabbits autopsied and cultured during the secondary phase. After the disappearance of the primary phase in rabbits exposed to 700 or 900 r, the secondary phase could be elicited by initiating bacterial infection. Within 6 to 24 hours after intravenous inoculation of E. coli, Cx-reactive protein reappeared in the blood and persisted until death or termination of the experiment. Reappearance of the protein also followed the intravenous injection of killed E. coli but it disappeared again 1 to 2 days later. The results indicate that the primary phase is elicited by radiation injury per se and the secondary phase by bacterial infection.

Multiphase systems, such as sprays and aerosols, are characterized by the existence of a continuous primary phase and a disperse secondary phase. The interaction between the two phases and/or the chemical reactions can affect both composition and characteristic velocity of the primary and secondary phases, as well as the size distribution of the secondary phase. In order to describe these systems, the continuity, mass balance and momentum balance equations as well as additional equations for turbulence, must be solved. Nevertheless if there is the need to account for the evolution of the secondary phase because of continuous and discrete events the population balance equation must be solved. In this work two very efficient ways to cope with these issues will be presented. In particular the use of the quadrature method of moments coupled with the mixture multiphase model, and the multi-fluid model will be presented and discussed.

SummaryConditions affecting the responses of human platelets to epinephrine were examined. In platelet-rich plasma prepared from blood anticoagulated with hirudin or PPACK (D-pheny- lalanyl-L-prolyl-L-arginine chloromethyl ketone), epinephrine did not cause shape change or aggregation. In a Tyrode-albumin- apyrase solution containing a concentration of Ca2+ in the physiological range, and fibrinogen, epinephrine in concentrations as high as 40 μM did not induce platelet shape change, caused either no primary aggregation or very slight primary aggregation, and did not induce thromboxane formation, release of dense granule contents, or secondary aggregation. In contrast, in citrated platelet-rich plasma, epinephrine induced two phases of aggregation. This is not attributable to the generation of traces of thrombin since the same effects were evident when blood was taken into a combined citrate-hirudin anticoagulant or a combined citrate-PPACK anticoagulant. In a modified Tyrode-albu- min-apyrase solution containing approximately 20 μM Ca2+, 1 mM Mg2+, and fibrinogen, epinephrine induced extensive aggregation after a lag phase, but no primary phase was evident; thromboxane formation and release of dense granule contents accompanied the aggregation response. These responses were also observed when PPACK was included with the acid-citrate- dextrose anticoagulant, and in the washing and resuspending fluids. In the presence of aspirin or the thromboxane receptor blocker BM 13.177 a few small aggregates were detected by particle counting and by scanning electron microscopy; with the latter inhibitor, the platelets in the aggregates retained their disc shape; secondary aggregation and the responses associated with it did not occur. Thus thromboxane A2 formation is not necessary for the formation of these small aggregates, but is required for extensive aggregation and release. As with other weak agonists, the close platelet-to-platelet contact in the low Ca2+ medium appears to be necessary for full secondary aggregation. Omission of fibrinogen from the low Ca2+ medium prevented both primary and secondary aggregation in response to epinephrine. An antibody (10E5) to the glycoprotein Ilb/IIIa complex was completely inhibitory in the presence of fibrinogen. Thus the response of human platelets to epinephrine is influenced by the concentration of Ca2+ and the presence of fibrinogen in the medium in which they are suspended.

SummaryEarlier, we found that ε-aminocaproic acid (EACA) inhibited human platelet aggregation induced by adenosine diphosphate (ADP) and collagen, but not aggregation by arachidonic acid (AA). Since EACA is structurally similar to lysine, yet these two agents exhibit vast difference in their antifibrinolytic activities, we chose to study the effect of lysine on platelet aggregation. We used L-lysine-HCl in these studies because of its high solubility in aqueous solutions while causing no change in pH when added to human plasma. With lysine, we repeatedly found inhibition of ADP-, collagen- and ristocetin-induced aggregation, but potentiation of AA-induced aggregation. Both the inhibitory and potentiation effects were dose-dependent. Low doses of lysine inhibited the secondary phase of aggregation; high doses of it also inhibited the primary phase of aggregation. Potentiation of AA-induced aggregation was accompanied by increased release of serotonin and formation of malondialdehyde. These effects were not confined to human platelets; rat platelets were similarly affected. Platelets, exposed to lysine and then washed and resuspended in an artificial medium not containing lysine, remained hypersensitive to AA, but no longer showed decreased aggregation by collagen. Comparing the effects of lysine with equimolar concentrations of sucrose, EACA, and α-amino-n-butyric acid, we attribute the potent inhibitory effect of lysine to either the excess positive charge or H+ and C1− ions. The -NH2 group on the α-carbon on lysine appears to be the determining factor for the potentiation effect; the effect seems to be exerted on the cyclooxygenase level of AA metabolism. Lysine and other chemicals with platelet-affecting properties similar to lysine may be used as a tool for the study of the many aspects of a platelet aggregation reaction.

Sepsis is a serious bacterial infection. The main treatment is using antibiotics. However, the rate of antibiotic resistance is very high and this resistance is related to the outcome of treatment. Objectives: To evaluate the situation of antibiotic resistance of some isolated bacteria in sepsis patients treated at Hue Central Hospital; to evaluate the relationship of antibiotic resistance to the treatment results in patients with sepsis. Subjects and methods: prospective study of 60 sepsis patients diagnosed according to the criteria of the 3rd International Consensus-Sepsis 3 and its susceptibility patterns from April 2017 to August 2018. Results and Conclusions: The current agents of sepsis are mainly S. suis, Burkhoderiae spp. and E. coli. E. coli is resistant to cephalosporins 3rd, 4th generation and quinolone group is over 75%; resistance to imipenem 11.1%; the ESBL rate is 60%. S. suis resistant to ampicilline 11.1%; no resistance has been recorded to ceftriaxone and vancomycine. Resistance of Burkholderiae spp. to cefepime and amoxicillin/clavulanic acid was 42.9% and 55.6%, resistant to imipenem and meropenem is 20%, resistance to ceftazidime was not recorded. The deaths were mostly dued to E. coli and K. pneumoniae. The mortality for patients infected with antibiotic-resistant bacteria are higher than for sensitive groups. To determine the resistance pattern of bacterial pathogens involved in infections of the patients aged between 18-64 years, admitted in a ICU from a 1518-bed university-affiliated hospital. A retrospective study of bacterial pathogens was carried out on 351 patients aged between 18-64 years admitted to the ICU, from January to December 2017. In this study there were analysed 469 samples from 351 patients (18-64 years). A total of 566 bacterial isolates were obtained, of which 120 strains of Klebsiella spp. (35.39%%), followed by Nonfermenting Gram negative bacilli, other than Pseudomonas and Acinetobacter (NFB) (75- 22.12%), Acinetobacter spp. (53 - 15.63%), Pseudomonas aeruginosa and Proteus (51 - 15.04%), and Escherichia coli (49 - 14.45%). The most common isolates were from respiratory tract (394 isolates � 69.61%). High rates of MDR were found for Pseudomonas aeruginosa (64.70%), MRSA (62.65%) and Klebsiella spp. (53.33%), while almost all of the isolated NFB strains were MDR (97.33%). There was statistic difference between the drug resistance rate of Klebsiella and E. coli strains to ceftazidime and ceftriaxone (p[0.001), cefuroxime (p[0.01) and to cefepime (p[0.01). The study revealed an alarming pattern of antibiotic resistance in the majority of ICU isolates.

Manual transplanting, a pre-dominant practice in almost all the paddy growing areas in India, is laborious, burdensome, and has many expenses on raising, settling, and transplanting nursery. The transplanting process’s limitations motivated the replacement of conventional paddy transplanting methods. The study was divided into two phases. The first phase included laboratory testing of three levels of metering mechanisms, namely cell type (M1) with 10 cells grooved around a circular plate having a 13 cm diameter, inclined plate (M2) containing 24 U shaped cells provided on an 18 cm diameter plate, and fluted roller (M3) with 10 flutes on a 5 cm diameter shaft. The testing matrix included a missing index, multiple index, and seed damage with forward speeds (2.5, 3.0, and 3.5 km/h), and pre-germination levels of 24 h soaked (P1), 24 h pre-germinated (P2), and 36 h pre-germinated paddy seeds (P3)). The second phase included selecting the best combination obtained from the laboratory study and developing a new efficient planter for the puddled field. The inclined plate metering mechanism operating at 2.5 km/h for 24 h pre-germinated seeds was reported most efficient from the first phase. Therefore, a self-propelled 8-row planter equipped with an inclined plate metering mechanism having a row-to-row spacing of 22.5 cm was developed, fabricated, and evaluated in the puddled field. The designed planter was assessed on two different soils: sandy loom (ST1) and clay loom (ST2) and at two different hopper fill levels as ½ filled hopper (F1) and ¾ filled hopper (F2). The number of plants per square meter and hill-to-hill spacing was measured. The on-field evaluation revealed that the number of plants per square meter was non-significantly affected by the type of soil but was significantly affected by hopper fill.

Canine parvoviral enteritis (CPE) is a severe disease characterized by systemic inflammation and immunosuppression. The function of circulating phagocytes (neutrophils and monocytes) in affected dogs has not been fully investigated. We characterized the functional capacity of canine phagocytes in CPE by determining their oxidative burst and phagocytic activities using flow cytometry. Blood was collected from 28 dogs with CPE and 11 healthy, age-matched, control dogs. Oxidative burst activity was assessed by stimulating phagocytes with opsonized Escherichia coli or phorbol 12-myristate 13-acetate (PMA) and measuring the percentage of phagocytes producing reactive oxygen species and the magnitude of this production. Phagocytosis was measured by incubating phagocytes with opsonized E. coli and measuring the percentage of phagocytes containing E. coli and the number of bacteria per cell. Complete blood counts and serum C-reactive protein (CRP) concentrations were also determined. Serum CRP concentration was negatively and positively correlated with segmented and band neutrophil concentrations, respectively. Overall, no differences in phagocyte function were found between dogs with CPE and healthy control dogs. However, infected dogs with neutropenia or circulating band neutrophils had decreased PMA-stimulated oxidative burst activity compared to healthy controls. Additionally, CPE dogs with neutropenia or circulating band neutrophils had decreased PMA- and E. coli–stimulated oxidative burst activity and decreased phagocytosis of E. coli compared to CPE dogs without neutropenia or band neutrophils. We conclude that phagocytes have decreased oxidative burst and phagocytic activity in neutropenic CPE dogs and in CPE dogs with circulating band neutrophils.

Assessment of the pollution for the selected six heavy metals (Pb, Cu, Zn, Fe, Mn and Ni) in ten surface sediments sampled from Aha Lake in a dry season was made in the present investigation. Principal component analysis (PCA) was used to assess the sources of the heavy metals contamination and two components were extracted. Analysis of the lake characteristics and point source pollution revealed that the discharge of industrial effluent and coal mining wastewater were the possible sources of these heavy metals contamination. Based on the speciation characteristics of heavy metals in sediments, the method ratio of secondary phase to primary phase (RSP) was applied to evaluate the loadings and the bioavailability of these heavy metals. The RSP evaluation exhibited that Pb, Zn, Fe, Mn and Ni were mainly associated with Fe-Mn oxides besides residual phase, while Cu mainly existed in organic phase and residual phase. In summary, the potential risk posed to the lake caused by the heavy metals was high and descended in the order of Mn > Ni > Zn > Pb > Cu > Fe.

ABSTRACTIn line with recent reports of extended-spectrum beta-lactamases (ESBLs) inEscherichia coliisolates of highly virulent serotypes, such as O104:H4, we investigated the distribution of phylogroups (A, B1, B2, D) and virulence factor (VF)-encoding genes in 204 ESBL-producingE. coliisolates from diarrheic cattle. ESBL genes, VFs, and phylogroups were identified by PCR and a commercial DNA array (Alere, France). ESBL genes belonged mostly to the CTX-M-1 (65.7%) and CTX-M-9 (27.0%) groups, whereas those of the CTX-M-2 and TEM groups were much less represented (3.9% and 3.4%, respectively). One ESBL isolate wasstx1andeaepositive and belonged to a major enterohemorrhagicE. coli(EHEC) serotype (O111:H8). Two other isolates wereeaepositive butstxnegative; one of these had serotype O26:H11. ESBL isolates belonged mainly to phylogroup A (55.4%) and, to lesser extents, to phylogroups D (25.5%) and B1 (15.6%), whereas B2 strains were quasi-absent (1/204). The number of VFs was significantly higher in phylogroup B1 than in phylogroups A (P= 0.04) and D (P= 0.02). Almost all of the VFs detected were found in CTX-M-1 isolates, whereas only 64.3% and 33.3% of them were found in CTX-M-9 and CTX-M-2 isolates, respectively. These results indicated that the widespread dissemination of theblaCTX-Mgenes within theE. colipopulation from cattle still spared the subpopulation of EHEC/Shiga-toxigenicE. coli(STEC) isolates. In contrast to other reports on non-ESBL-producing isolates from domestic animals, B1 was not the main phylogroup identified. However, B1 was found to be the most virulent phylogroup, suggesting host-specific distribution of virulence determinants among phylogenetic groups.
