Effects of carnosine, ankaferd and silver sulfadiazine on an experimental burn model; roles of irisin and HSP70

Abstract In this study, the effects of carnosine, ankaferd and 1% silver sulfadiazine applied topically on second-degree burns were investigated and the roles of irisin and Heat shock protein 70 (HSP70) in this healing process were evaluated. Ninety male albino rats were used and divided into 5 groups. The groups classified as; control, burn, burn + carnosine (CAR), burn + ankaferd (ABS) and burn + silver sulfadiazine (SS). It was found that level of irisin increased in the first week and decreased in the second week in the burn and CAR groups. In the ABS and SS groups, the level of irisin was determined that started to increase in the first week and continued to increase in the second week. The level of HSP70 was found to increased in the first week in burn and CAR groups and decreased in the second week, but started to increase in the second week in ABS and SS groups. Both levels of irisin and HSP70 were observed to decreased in all treatment groups in the third week. In this study, it was shown that ankaferd and silver sülfadiazine treatments cause an increase in the irisin levels in the early period and a gradually increase in HSP70 levels in the later period in burns. The inflammatory response was observed to be limited in the early period in the ankaferd and sulfadiazin groups. It was concluded that these findings were effective in early wound healing in burns.

Our study aimed to determine the effects of losartan and PD123319 in ischemia–reperfusion (IR) injury in isolated perfused rat heart. The study used 40 male Wistar albino rats that were grouped as Control, IR, and IR treatment groups that received losartan (20 mg/kg), PD123319 (20 mg/kg), and losartan+PD123319. The hearts were attached to Langendorff isolated heart system by employing in situ cannulation method, and cardiodynamic parameters were recorded during the experiment. At the end of experiment, hearts were retained for biochemical analysis and all data were statistically evaluated. A partial recovery of cardiodynamic parameters was observed in all treatment groups. A significant increase in oxidative stress parameters were seen in the IR group, whereas all treatment groups exhibited lower increase. Furthermore, levels of all antioxidant parameters were significantly lower in the IR group, but higher in the treatment groups. Effects on all parameters were much more remarkable in the PD123319 group. Levels of angiotensin II and renin were increased (P < 0.001) with IR application and decreased (P < 0.001) with the treatment of both antagonists. In conclusion, treatment of losartan and PD123319 played a cardioprotective role against IR injury, PD123319 being more effective in this protection.

This study was conducted to prove whether ellagic acid can be used as a regulator of incision wound healing process in male albino rats as experimental animals. A total of 24 male healthy rats based on physical examination, aged at 3 months old and weight at 150-180 grams used in this study. Rats were divided into three groups and each group has 8 replications, group of control (P0) has no incision wound and no medication, treatment group (P1) has incision wound and treated with Betadine® and (P2) has incision wound and treated with 2,5 % ellagic acid. The variable that observed are histopathology changes (collagen, PMN, angiogenesis and fibrosis) from the male albino rats. The samples collection was taken at day 15 and data were collected and analyzed by Analysis of Variance (ANOVA) test. The result of collagen and PMN cells score in the control group (P0) were 1.80 ± 0.16 and 1.6 ± 0.08 with mild angiogenesis and no fibrosis. The results in the treatment (P2) group, where the collagen and PMN scores were 1.60 ± 0.08 and 1.5 ± 0.8, accompanied by mild angiogenesis and fibrosis. Meanwhile, in the treatment (P1) group, the results of scoring of collagen and PMN were lower than the other groups, 0, 80 ± 0.08 and 0.6 ± 0.16 accompanied by moderate angiogenesis and fibrosis. There was a significant difference of collagen scoring, PMN cells number, fibrosis and angiogenesis among the treatment groups (p<0.05). P1 treatment group has significant different with treatment groups P0 and P2 (p<0.05), but there is no significant difference between treatment groups P0 and P2 (p>0.05).  From the results, it can be concluded that the topical application of ellagic acid ointment was proved to provide healing process on incision wound. Topical administration of ellagic acid ointment for 14 days was able to improve the skin condition and promoted the wound healing.Keywords: Ellagic acid ; wound healing ; collagen ; angiogenesis ; fibrosis ; anti-inflammatory ; antioxidant ; antibacterial.

AbstractObjectivesThe main objective of the present study is to evaluate the mitigative effect of hydroalcoholic extract of Momordica cymbalaria fruits against sodium fluoride (NaF) induced hepatotoxicity.MethodsIn this study, Wistar male albino rats were randomly divided into five groups of six rats each. Group I and II served as normal and toxic controls. Group III as plant control received extract at a dose of 400 mg/kg b. wt, p.o and Groups IV and V as treatment groups received extract at a dose 200 and 400 mg/kg b. wt, p.o for 30 days. All groups except Groups I and III received 100 ppm of NaF through drinking water. After completion of the study, blood collected for the estimation of liver blood serum biomarkers such as aspartate aminotransferases (AST), alanine aminotransferases (ALT), alkaline Phosphatase (ALP), direct and total bilirubin, total protein and albumin. The liver tissue homogenate was for estimation of lipid peroxidation, catalase, and reduced glutathione levels.ResultsThe results showed that NaF intoxication caused elevation of liver blood serum levels and lipid peroxidation; decreased levels of serum total protein, albumin and liver reduced glutathione, and catalase observed. The treatment groups showed decreased elevated serum biomarkers (ALT, AST, and ALP), liver lipid peroxidation and increased serum total protein and albumin, liver reduced glutathione and catalase levels in a dose-dependent manner. Histopathological studies also further strongly supported for mitigative effects of the plant.ConclusionsIn conclusion, our findings of the study indicated that M. cymbalaria fruits were a potential drug candidate in the treatment of NaF induced hepatotoxicity.

The aim of this study was to compare the efficacy of trabeculectomy (TE), single XEN microstent implantation (solo XEN) or combined XEN implantation and cataract surgery (combined XEN) in primary open-angle glaucoma cases, naïve to prior surgical treatment, using a monocentric retrospective comparative cohort study. Intraocular pressure (IOP) and the number of IOP-lowering drugs (Meds) were monitored during the first 24 months after surgery. Further disease progression was monitored using peripapillary retinal nerve fiber layer (RNFL) thickness examinations using spectral domain optical coherence tomography (OCT) as well as visual acuity (VA) and visual field (VF) tests. In the TE group (52 eyes), the mean IOP decreased from 24.9 ± 5.9 to 13.9 ± 4.2 mmHg (p < 0.001) and Meds decreased from 3.2 ± 1.2 to 0.5 ± 1.1 (p < 0.001). In the solo XEN (38 eyes) and the combined XEN groups, the mean IOP decreased from 24.1 ± 4.7 to 15.7 ± 3.0 mmHg (p < 0.001) and 25.4 ± 5.6 to 14.7 ± 3.2 mmHg (p < 0.001), while Meds decreased from 3.3 ± 0.8 to 0.8 ± 1.2 (p < 0.001) and 2.7 ± 1.2 to 0.4 ± 1.0 (p < 0.001), respectively. The VF and VA indices showed no sign of further deterioration, the RNFL thickness further decreased in all treatment groups after surgery. TE and XEN led to comparable reductions in IOP and Meds. Although the VA and VF indices remained unaltered, the RNFL thickness continuously decreased in all treatment groups during the 24-month follow-up.

Pregnant female albino rats were randomly assigned to one of three groups which were administered either 15 mg/kg diphenylhydantoin, 5 mg/kg phenobarbital or an equal volume of distilled water. All administrations occurred daily in one of four time frames, Days 1–7, Days 8–14, Days 15-birth, Days 1-birth. At 75 days of age one male and one female offspring from 48 different litters were tested on avoidance conditioning. Few birth defects were noted in the offspring, and abnormalities were not noted with regard to physical development. The results of avoidance-conditioning testing indicate that the administration of diphenylhydantoin during the third period of pregnancy and the administration of phenobarbital during the first period are most influential in affecting learning in a detrimental direction. Caution seems advisable in the usage of these drugs in pregnancy.

Abstract Introduction Burn is one of the most severe traumas that causes coagulative destruction of the skin. The use of various products that accelerate wound healing in patients with burn may affect the patient’s survival and reduce the complications that may be seen. In the present study we aimed effects of subcutaneous ozone injection on second degree burn wound. Methods A total of 72 Sprague-Dawley male rats included in the study were divided randomly into three groups (control group (CG), silver sulfadiazine group (SG), ozone group (OG)) and each group was divided randomly two subgroups (as sacrificed on d7 and on d14).A deep second degree scald burns were created on the lower back. In CG subcutaneous 0.9% serum saline was injected daily into the burn area. In SG, burns were dressed with silver sulfadiazine daily and in OG subcutaneous ozone was injected daily into the burn area. Tissue hydroxyproline level measurement and histopathological evaluation were done. Results When the groups were compared in terms of weight change, no significant difference was found on the 7th and 14th days. In the evaluation made in terms of tissue hydroxyproline, tissue hydroxyproline level in OG was found to be significantly higher on both the 7th and 14th days (p &lt; 0.001). In histopathological evaluations, it was determined that wound healing in OG was significantly higher than in the other groups. Conclusions According to the results, subcutaneous ozone therapy is more effective than silver sulphadiazine in the healing process of second-degree burn wounds and it can be safely used in the treatment of burn wounds.

The exogenous lipoic acid (LA) is successfully used as a drug in the treatment of many diseases. It is assumed that after administration, LA is transported to the intracellular compartments and reduced to dihydrolipoic acid (DHLA) which is catalyzed by NAD(P)H-dependent enzymes. The purpose of this study was to investigate whether LA can attenuate cardiovascular disturbances induced by ethanol (EtOH) and disulfiram (DSF) administration separately or jointly in rats. For this purpose, we measured systolic and diastolic blood pressure, recorded electrocardiogram (ECG), and estimated mortality of rats. We also studied the activity of aldehyde dehydrogenase (ALDH) in the rat liver. It was shown for the first time that LA partially attenuated the cardiac arrhythmia (extrasystoles and atrioventricular blocks) induced by EtOH and reduced the EtOH-induced mortality of animals, which suggests that LA may have a potential for use in cardiac disturbance in conditions of acute EtOH intoxication. The administration of EtOH, LA, and DSF separately or jointly affected the ALDH activity in the rat liver since a significant decrease in the activity of the enzyme was observed in all treatment groups. The results indicating that LA is an inhibitor of ALDH activity are very surprising.

Background: Among venomous elapid snakes, cobras have the highest public awareness, as their venom represents a combination of proteins, peptides, and enzymes that have a range of biochemical and pharmacological roles and are also the main constitutes of biological activity and lethal toxicity. Objectives: The study aimed to evaluate the effect of the venom of Egyptian Spitting Cobra, Naja nubiae, on the vascular permeability based on the extravasation of the azo dye Evans blue (EB) into the tissues of the liver and kidneys of animals envenomed with low (¼ LD50; 0.32 mg/kg) and high (½ LD50; 0.65 mg/ kg) doses at three sampling times (30, 120, 360 min) post-injection of the venom. Methods: Fifty-four adult male Albino rats (8 weeks old and 180±2 0 g body weight) were divided into three main groups (n=6). In the control group, rats were subcutaneously (SC) injected with saline solution. Envenomed groups were SC injected, one group with 0.32 mg/kg and the other group with 0.65 mg/kg body weight of crude venom, respectively. Rats were I.V injected with EB dye 20 minutes before SC injection with saline solution as control animals and with Naja nubiae venom as treatment groups. Results: The results illustrated a high significant rate of EB extravasation to hepatic and renal tissues by the colorimetric determination of EB dye concentration. Conclusion: The venom of Naja nubiae can cause increased hepatic and renal vascular permeability which may explain the inflammatory effect induced by this venom.

Three-dimensional clumps of mesenchymal stem cells (MSCs)/extracellular matrix (ECM) complexes (C-MSCs) can be transplanted into tissue defect site with no artificial scaffold. Importantly, most bone formation in the developing process or fracture healing proceeds via endochondral ossification. Accordingly, this present study investigated whether C-MSCs generated with chondro-inductive medium (CIM) can induce successful bone regeneration and assessed its healing process. Human bone marrow-derived MSCs were cultured with xeno-free/serum-free (XF) growth medium. To obtain C-MSCs, confluent cells that had formed on the cellular sheet were scratched using a micropipette tip and then torn off. The sheet was rolled to make a round clump of cells. The cell clumps, i.e., C-MSCs, were maintained in XF-CIM. C-MSCs generated with XF-CIM showed enlarged round cells, cartilage matrix, and hypertrophic chondrocytes genes elevation in vitro. Transplantation of C-MSCs generated with XF-CIM induced successful bone regeneration in the SCID mouse calvaria defect model. Immunofluorescence staining for human-specific vimentin demonstrated that donor human and host mouse cells cooperatively contributed the bone formation. Besides, the replacement of the cartilage matrix into bone was observed in the early period. These findings suggested that cartilaginous C-MSCs generated with XF-CIM can induce bone regeneration via endochondral ossification.
