Vitamin E ameliorates disturbances in testosterone pathway and sperm quality of male rats induced by the glycosides vicine and convicine of Vicia faba

Purpose This paper aims to design to evaluate the protective effect of vitamin E to ameliorate the disturbances in testosterone pathway and sperm quality of male rats induced by the glycosides vicine (V) and convicine (C) of Vicia faba. Design/methodology/approach Forty male albino rats were divided into five equal groups; control, paraffin oil, V (400 mg/kg) C (150 mg/kg)-treated group, vitamin E (100 mg/kg) + VC-treated group, and vitamin E (200 mg/kg) + VC-treated groups which injected intraperioneally (IP) with 0.5-ml saline, 0.5-ml paraffin oil,V (400 mg/kg) and C (150 mg/kg) of Vicia faba, vitamin E (100 mg/kg) + VC-treated groups, and Vitamin E(200 mg/kg) + VC-treated groups, respectively. Blood and testicular tissue were obtained after one month of the study. The male genital organs were calculated. Testosterone (Ts), luteinizing hormone (LH), follicle stimulating hormone (FSH), dehydroepiandrosterone sulfate (DHEA-SO4), sex hormone binding globulin (SHBG),?-glutamyl transpeptidase (?-GT), glucose-6-phosphate dehydrogenase (G6PD), 3ß-hydroxysteroid dehydrogenase (3ßHSD), lactate dehydrogenase (LDH), spermatozoa concentration, percent of mortality and abnormal sperms were evaluated. Findings The VC-treated group showed significant decrease (p < 0.01) in Ts, DHEA-SO4, G6PD, spermatozoa number and mortality percent, as well as, the male genital organs (testes, epidydemis, seminal vesicle, prostate and vasa deferentia) while significant increase (p < 0.01) was found in LH, FSH, SHBG, LDH, ?-GT, sperms monoclonal Ki-67, and abnormal spermatocytes levels compared with control group. Vitamin E co-injection with VC-treated group returned all these parameters to the normal values. The higher dose of vitamin E (200 mg/kg) was more effect than the lower dose (100 mg/kg). Originality/value Vicia faba contains V and C glycosides. The V and C glycosides in Vicia faba are hydrolyzed by intestinal microflora to aglycones divicine and isouramil, respectively. Divicine and isouramil are highly reactive compounds generating free radicals where divicine and isouramil are the main factors of favism. The V and C glycosides induced disturbances in testosterone pathway and sperm quality of male rats and vitamin E ameliorates these disturbances.

Abstract Background: Artificial insemination technology is the production technology used in more than 99% of the world's pig farms. As the frozen sperm damage is very serious, the preservation of pig sperm is mainly carried out at 17°C. However, the accumulation of a large amount of reactive oxygen species (ROS) during the preservation process is the main reason for the deterioration of sperm quality. In our research, by adding different concentrations of hydroxytyrosol to the diluent during the storage of pig sperm at 17°C, we compared them with the traditional diluent to study the cumulative amount of ROS, the extent of membrane damage, the sperm acrosome integrity, the activity of antioxidant enzymes, and other indicators to evaluate the effect of hydroxytyrosol on the changes of active oxygen species and sperm quality during storage. After we determined the optimal amount of hydroxytyrosol in the diluent, proteomics was used to monitor difference in sperm protein expression between the control group and the addition of 120 μmol/L hydroxytyrosol group (optimum concentration). Ultimately, we selected the control group and the 120 μmol/L hydroxytyrosol group to test the effect of artificial insemination.Results: The results of our research showed that during storage of pig sperm at 17 ℃, the sperm quality and antioxidant capacity of the hydroxytyrosol-treated group was significantly improved (P < 0.05). Proteomics sequencing analysis proved that the addition of hydroxytyrosol treatment group has potential value in improving sperm quality. Besides, the significant increase in sow pregnancy rate and piglet birth weight also confirmed that sperm quality during storage can be of vital importance in actual production (P < 0.05).Conclusions: Based on our results, we demonstrate that the addition of hydroxytyrosol to the diluent could improve the quality of pig sperm and the efficiency of artificial insemination.

Diabetes mellitus is a metabolic disorder that often affected fertility health, specifically in men. Gynura procumbens (G. procumbens) has been used as traditional medicine due to its ability in treating various types of illness. Hence, this study was carried out to determine the potential beneficial effect of G. procumbens as anti-hyperglycaemia, pro-fertility and libido agent towards diabetes-induced male rats. A total of 42 male Sprague Dawley rats were randomly assigned into six groups; normal, negative, and positive control, and three treated groups of different dosages of G. procumbens aqueous extract (GPAE); 150 mg/kg, 300 mg/kg and 450 mg/kg. Each group was given treatment via oral gavage for seven consecutive days. The rats were sacrificed on day eighth for further analysis. The fasting blood glucose (FBG) level in all GPAE treated group showed significant decrease, and 450 mg/kg treated group showed significant reduction to a normal blood glucose level compared to all groups. There were significant increases in the sperm quality of GPAE treated groups after seven days of treatment compared to the negative and positive control group, specifically in 450 mg/kg treated group. Testis histology showed that GPAE treated groups produced a significant result whereby the seminiferous tubules were seen packed with sperm and successive stage of spermatogenesis were shown compared to control groups. Hormone analysis suggested that the luteneizing hormone (LH), follicle stimulating hormone (FSH) and testosterone hormone of the treated groups were elevated after seven days of treatment compared to the negative and positive control groups. Libido analysis of seven days treatment showed that GPAE treated groups, significantly improved in 450 mg/kg dose compared to other groups, with the highest number of mounting frequency and shortest mounting latency. Fertility test revealed a significant increase in number of implantation sites produced in GPAE treated groups compared to positive and negative control groups. In conclusion, GPAE exhibited a potential beneficial effect as an anti-diabetic and also as fertility agent in diabetic-induced male rats.

Abstrak Tingginya angka infertilitas pada pria disebabkan antara lain oleh kualitas produksi spermatozoa dan gangguan hormonal. Tujuan penelitian ini adalah untuk mengetahui pengaruh gangguan tidur terhadap kadar hormon testosteron dan jumlah spermatozoa. Jenis penelitian adalah eksperimen laboratorium dengan desain post test only control groupdesign. Populasi tikus jantan Wistar berumur 2-3 bulan, dan berat badan 300 – 350 gr. Sampel sebanyak 24 ekor dibagi atas 4 kelompok yaitu: 1 kelompok kontrol dan 3 kelompok perlakuan. Variabel independen adalah gangguan tidur dan variabel dependen adalah kadar hormon testosteron dan jumlah spermatozoa. Analisa data mengunakan metode ANOVA dan dilanjutkan dengan uji statistik Multiple Comparisons jenis Bonferroni. Hasil penelitian menunjukkan ada perbedaan bermakna rerata kadar hormon testosteron kelompok kontrol dan perlakuan dengan nilaip=0,000. Gangguan tidur memberikan perbedaan bermakna terhadap kadar hormon testosteron. Terdapat perbedaan bermakna rerata jumlah spermatozoa kelompok kontrol dengan perlakuan dengan nilai p=0.000. Gangguan tidur juga memberikan perbedaan bermakna terhadap jumlah spermatozoa. Kesimpulan penelitian ini ialah terdapat pengaruh yang bermakna antara gangguan tidur terhadap kadar hormon testosteron dan jumlah spermatozoa pada tikus jantan wistar. The objective of this study was to find out the effect of sleeping disturbance to the quality of testosterone hormone and the number of sperm. This was a laboratory experimental research with post-test only control group design. The populations were 2-3 months, 300-350 grams of weight, male rats. The sample was 24 rats which consisted of 4 groups: one control group and three experimental groups. Sleeping disturbance was an independent variable, whilethe quality of sperm testosterone hormone and the number of sperm were dependent variables. The data of the research were analyzed statistically by  ANOVA method and Bonferroni multiple comparison. There was a significant different of the mean of testosterone hormone quality between control group and experimental group with p=0,000. Sleeping disturbance gave significant different to the quality of testosterone hormone. There was a significant different on the number of sperm quality between control group and experimental group with p=0,000. Sleeping disturbancegave significant different to the number of sperm quality. The conclusion from this study is that there is effect of sleeping disturbance to testosterone hormone quality and the number of sperm on the wistar male rats. Background and objectives: The hepatoprotective activity of vitamin E and C is evident due to their ability of modulating the antioxidant pathway. In this study, we have evaluated the effects of &alpha;-tocopherol and ascorbic acid on paracetamol induced liver damage with offsetting various levels of drug treatment following an in vivo experimental protocol on Wistar albino male rats. Materials and Methods: The level of lipid peroxidation as well as histological examination of liver tissues were observed among 50 Wistar albino male rats to evaluate hepatoprotective effect of &alpha;-tocopherol and ascorbic acid on hepatocytes. The experiment was divided into 5 groups (10 rats in each group)- Basal control group (Group-I, with propylene glycol), Paracetamol treated control group (Group &ndash;II), &alpha;-tocopherol pretreated &amp; paracetamol treated group (Group &ndash;III), Ascorbic acid pretreated &amp; paracetamol treated group (Group &ndash;IV) and Ascorbic acid pretreated &amp; paracetamol treated group (Group &ndash;IV). Results: The mean (&plusmn; SD) Malondialdehyde (MDA) concentration were significantly reduced in &alpha;-tocopherol pretreated and paracetamol treated group (P&lt;0.001), Ascorbic acid pretreated and paracetamol treated group (P&le;0.05) and combined &alpha;-tocopherol with ascorbic acid pretreated &amp; paracetamol treated group (P&lt;0.001). Statistically significant differences in histological findings of rat liver were observed in paracetamol treated control group (P&lt;0.001), ascorbic acid pretreated and paracetamol treated group (P&lt;0.001). The serum alanine aminotransferase (ALT) level was also significantly higher in paracetamol treated group (P&lt;0.001), &alpha;-tocopherol pretreated plus paracetamol treated group (P&le;0.05) and in ascorbic acid pretreated plus paracetamol treated group (P&lt;0.001). Conclusion: The combined pretreatment of &alpha;-tocopherol &amp; ascorbic acid have better hepatoprotective effects than &alpha;-tocopherol or ascorbic acid alone against paracetamol induced liver damage. The decrement of free radicals produced by vitamin E could be a better hepatoprotective antioxidant than vitamin C in paracetamol induced toxicity.

Lipopolysaccharide (LPS) as an endotoxin forms part of the cell wall of gram-negative bacteria and is responsible for initiating an acute inflammation after entering the living tissue. In this study, male rats were divided into eight groups: control group, vitamin E (VE) treatment group (200 mg/kg body weight (b.w.)), sodium selenite (SS) treatment (0.35 mg/kg b.w.) group, VE + SS treatment group (200 + 0.35 mg/kg b.w.), LPS treatment group (10 mg/kg b.w.), LPS + VE (10+200 mg/kg b.w.), LPS + SS treatment (10 + 0.35 mg/kg b.w.), and LPS + SS + VE treatment (10 + 0.35 + 200 mg/kg b.w.) group. Oxidative stress parameters, pathological changes, immunohistochemical analyses, terminal deoxynucleotidyl transferase deoxyuridine triphosphate nick end-labeling (TUNEL) assay, and changes in DNA structure with comet assay of the kidney were investigated at the end 6 h comparatively with the control group. When LPS-treated group was compared with the control group, antioxidant enzyme activities were decreased and malondialdehyde (MDA) levels, changes in histological and DNA structure and apoptosis were increased significantly at the end of 6 h. However, when LPS + SS and/or VE-treated group were compared with the LPS-treated group, superoxide dismutase, catalase, glutathione peroxidase, and glutathione- S-transferase activities were increased and MDA levels were decreased significantly at the end of the treatment period. Light investigations figured out pathological changes in kidneys of LPS- and LPS + SS and/or VE-treated groups. There was a decrease in the number of proliferating cell nuclear antigen-positive cells and an increase in the number of TUNEL-positive apoptotic cells in the wall of the distal and proximal tubules. As a result, it was observed that the combined use of antioxidants was more protective than their use alone against LPS.

After we determined the optimal amount of hydroxytyrosol in the diluent, Proteomics was used to monitor difference in sperm protein expression between the control group and the addition of 120 μmol/L hydroxytyrosol (optimum concentration) group after three days’ storage. Ultimately, we selected the control group and the addition of 120 μmol/L hydroxytyrosol group to test the effect of artificial insemination.Results: During storage of pig semen at room temperature, the sperm quality of the hydroxytyrosol-treated group was significantly improved (P < 0.05). Besides, its antioxidant capacity was also significantly improved (P < 0.05). Proteomics sequencing analysis proved that the addition of hydroxytyrosol treatment group has potential value in improving sperm cell quality. The significant increase in sow pregnancy rate and the increase in piglet birth weight also confirmed that sperm quality during storage can be of vital importance in actual production (P < 0.05).Conclusions: Based on our results, we demonstrated that the addition of hydroxytyrosol to the diluent can not only improve the quality of pig sperm, but also has an irreplaceable role in improving the efficiency of artificial insemination.

Abstract. It has been suggested that treatment of intact male rats with oestradiol benzoate (OeB) causes an interference with testosterone (T) production by the testes by a direct inhibitory effect on steroidogenesis. To test this hypothesis, different doses (5, 10 or 25 IU) of hCG were administered concomitantly with 50 μg of OeB to adult intact or hypophysectomized male rats. The testicular and plasma testosterone, and serum hCG levels were determined. The sex accessory weights were recorded. In the intact OeB-treated group of animals, hCG stimulated both the secondary sex organs and plasma testosterone levels above the intact control group. However, in hypophysectomized animals, although plasma testosterone levels increased above that of intact controls, their secondary sex organ weights did not. Moreover, inspite of high circulating hCG levels, the testicular testosterone content and concentration remained suppressed in OeB-treated animals. The reason for such dichotomy of hCG action on OeB-treated animals is not clear at present.

✓ The adverse effect of a minimal cerebral blood flow (CBF) in models of global ischemia has been noted by many investigators. One factor believed important in this situation is the level of blood glucose, since a continued supply of this metabolite results in increased tissue lactate, decreased brain pH, and increased cell damage. The authors have extended these observations to a model of focal incomplete ischemia. Brain pH was measured in fasted squirrel monkeys in regions of focal incomplete ischemia after transorbital occlusion of the middle cerebral artery (MCA). In both control and hyperglycemic animals, CBF was reduced to less than 30% of baseline. At 3 hours after MCA occlusion, brain pH in the control group was 6.66 ± 0.68 as compared to 6.27 ± 0.26 in the glucose-treated group. This difference was statistically significant by Student's unpaired t-test (p < 0.05). Thus, hyperglycemia results in decreased tissue pH in regions of focal incomplete cerebral ischemia in monkeys.

Background: Infertility has been more common problems among couple of reproductive age. One of the factors causing this disorder is unhealthy environmental factors including exposure to cigarette smoke. Polynuclear aromatic hydrocarbons (PAHs) in cigarette smoke can cause testicular atrophy, while the free radicals can inhibit the stages of spermatogenesis, and nicotine in cigarettes affects the brain dopamine levels affecting the levels of GnRH, and subsequently affect the levels of FSH and LH needed in spermatogenesis. The use of Mucuna pruriens seed extract containing antioxidants and L-dopa is expected to improve the quality of sperm after exposure to cigarette smoke. Objective: This study aimed to determine the effect of Mucuna pruriens seed extract on the sperm quality in mice exposed to cigarette smoke. Methods: This study was an experimental study with a post test only control group design. A total of 20 mice were divided into 4 groups of five mice each. All groups were exposed to cigarette smoke. Group 1 was the negative control exposed to cigarette smoke. Groups 2, 3, 4 were exposed to cigarettes smoke and given Mucuna pruriens seed extracts at the dose of 250; 300; and 350 mg/Kg BW/day. Parameters of sperm quality included concentration, morphology, motility and viability. Results: Post hoc tests showed there were significant differences among treatment groups. Conclusion: the administration of Mucuna pruriens seed extract affects the sperm quality of BALB/c mice exposed to cigarettes smoke. Bangladesh Journal of Medical Science Vol.20(4) 2021 p.768-773
