HPLC-DAD analysis of Thymus serpyllum based natural pigments and investigation of their antimicrobial properties

Purpose – In this study, it was aimed to investigate the antibacterial properties of natural pigments prepared from Thymus serpyllum. Design/methodology/approach – Al (III), Fe (II), Sn (II) and Cu (II) complexed natural pigments were obtained by using a precipitation method and the main constituents in the pigments were identified with HPLC-DAD. Also FTIR analysis was performed for further structural characterization. Moreover, the thermal stability and thermal degradation properties of the pigments were analyzed by thermogravimetric analyses (TGA). The antimicrobial activity of the thyme plant-extracted pigments was evaluated by measuring the minimal inhibitory concentration. Findings – Apigenin and luteolin flavones were detected as the main components of the natural dyes. Thermal degradation behaviour of the pigments was determined by means of TGA. All pigments showed high char yields and it was attributed to the high complexation between the metal and the ligand species. The antimicrobial activity of the thyme plant-extracted pigments was measured and it was found that all pigments had high antimicrobial activity. Aluminum-thymus pigments showed the highest antimicrobial efficiency among other pigments used in this study. Originality/value – The obtained pigments have high antimicrobial activities, and therefore, they can be used for the production of antimicrobial textiles. Furthermore, Thymus-based natural pigments might have potential applications in coating, paint, plastic industries, etc.

It is well known that essential oils possess significant antimicrobial activity. This study was conducted to estimate the antimicrobial activity of various types of Biokill, a laboratory produced solution composed of several essential oils (Biokill dissolved in 96% ethanol; Biokill 96% further dissolved in DMSO; Biokill dissolved in 70% ethanol and Biokill 70% further dissolved in DMSO). The antimicrobial activity was evaluated against five selected fungal strains, Candida albicans ATCC 10231, Saccharomyces cerevisiae ATCC 9763, Aspergillus niger I.N. 1110, Aspergillus sojae CCF and Penicillium spp. FNS FCC 266. A variation of the microtiter plate-based antimicrobial assay was used in order to assess the antimicrobial activity of the solutions. By applying this assay minimal inhibitory concentrations (MIC) of the Biokill solutions were determined for each strain of the selected test microorganisms. The results demonstrated that all variations of Biokill showed antimicrobial activity at concentrations lower than 2.5?g/mL. Biokill 70% further dissolved in DMSO showed the best antimicrobial properties against all the selected strains with MICs less than 1.25?g/mL. These results indicated that Biokill could find application in the pharmaceutical industry, in food preservation and conservation, in the prevention and treat?ment of plants infected by certain phytopathogens, etc.

ABSTRACT Aim The present study was conducted to evaluate the efficacy of commercially available herbal toothpastes against the different periodontopathogens. Materials and methods Six herbal toothpastes that were commonly commercially available were included in the study. Colgate herbal, Babool, Meswak, Neem active, Dabur red toothpastes were tested for the study whereas sterile normal saline was used as control. Antimicrobial efficacies of dentifrices were evaluated against Streptococcus mutans and Actinobacillus actinomycetemcomitans. The antimicrobial properties of dentifrices were tested by measuring the maximum zone of inhibition at 24 hours on the Mueller Hinton Agar media inoculated with microbial strain using disk diffusion method. Each dentifrice was tested at 100% concentration (full strength). Results The study showed that all dentifrices selected for the study were effective against the entire test organism but to varying degree. Neem active tooth paste gave a reading of 25.4 mm as the zone of inhibition which was highest amongst all of the test dentifrices. Colgate Herbal and Meswak dentifrices recorded a larger maximum zone of inhibition, measuring 23 and 22.6 mm respectively, compared to other toothpastes. All other dentifrices showed the zone of inhibition to be between 17 and 19 mm respectively. Conclusion The antibacterial properties of six dentifrices were studied in vitro and concluded that almost all of the dentifrices available commercially had antibacterial properties to some extent to benefit dental health or antiplaque action. Purpose The purpose of this paper is to determine the antimicrobial compounds extracted from various types of soybean materials; raw soybean, soybean flour and roasted soybean. Bacteria growth inhibition was tested in E.coli and S. aureus by disk diffusion method. Efficiency of antimicrobial activities were compared among the extracted solution. Design/methodology/approach Soybean (Glycine max) isoflavones contain the ability to inhibit the growth of many strains of microorganisms. The solid-liquid extraction, ethanol-hexane extraction, was modified to isolate the inhibitory compounds from the three different types of soybean materials. All crude extracts at various concentrations performed under different extracted solutions (75, 50, 25, 12.5, 6.25 and 3.125 percent) were tested with E. coli. and S. aureus to determine the antimicrobial activities and the minimum inhibition concentration (MIC). Disk diffusion method was chosen to study the antimicrobial activity of isoflavones. Findings Soybean flour extract inhibited the growth of E. coli and S. aureus more effective than roasted soybean and raw soybean, respectively. The MIC of inhibitory compounds extracted from soybean flour was 6.25 percent (0.031 mg/ml), roasted soybean was 25 percent (0.125 mg/ml) and raw soybean was 50 percent (0.250 mg/ml). Crude extracts of soybean flour and roasted soybean showed better results than raw soybean. All the three of extracted soybeans inhibited the growth of E. coli better than S. aureus. Originality/value Ethanol-hexane extraction was successfully used to isolate the antimicrobial compounds from raw soybean, soybean flour and roasted soybean. Comparison of antimicrobial activity showed that soybean flour contained the highest inhibition activity.

Abstract Recently, natural products have been evaluated as a source of antimicrobial agent with efficacies against a variety of microorganisms. The antibacterial activities of the oak fruit,inner husk of oak fruit (Jaft), and Jaftex mouthwash have been studiedin several studies. Thisstudy aimedto review the studies ofthe effects of antibacterial properties of the oak fruit, oak fruit hull (Jaft), and Jaftex mouthwash. Materials and Methods In this review study, relevant articles related to the antimicrobial activity of the oak fruit, inner husk of oak fruit (Jaft), and 'Jaftex mouthwash' were searched from the current digital literature using electronic databases namely, SID, ScienceDirect, PubMed, Google Scholar, Magiran, Web of Science employing the same keywords from 1990 to 2019. Results Oakfruit, inner husk of oak fruit (Jaft), and Jaftex mouthwash have antimicrobial properties against many microorganisms. Discussion and Conclusion Antibacterial properties of oak fruit, inner husk ofoak fruit (Jaft) can be used to reverse the antibiotic sensitivity against pathogenic bacteria. Jaftex is recommended as an antibacterial and anti-plaque mouthwash.

Methicillin-resistant Staphylococcus aureus (MRSA) is a Gram-positive bacterium that causes community-acquired and health care-acquired infections. We previously demonstrated that clay phyllosilicates and customized aluminosilicates display antimicrobial activity against the MRSA strain SQL1. The SQL1 annotated genome reveals a USA300 lineage and contributes critical knowledge of the MRSA virulence factors associated with tissue infection.

A wide range of medicinal plant extracts has phytochemicals that possess antimicrobial properties and these plants are used to treat several infections. The study aimed to assess the antimicrobial activities of some spices extracts and to evaluate the phytochemicals present in them. The extracts of spices were prepared using Soxhlet apparatus refluxing with methanol and ethanol. The well diffusion technique was implemented for the evaluation of antimicrobial activities of the extracts and the zone of inhibitions was recorded in millimeters. The antimicrobial test was done against five bacterial isolates: Escherichia coli, Proteus mirabilis, Pseudomonas aeruginosa, Salmonella enterica serotype Typhi, and Staphylococcus aureus and a fungal isolate: Candida albicans. The extracts were concentrated by Rotary Vacuum Evaporator and a stock solution of 200 mg/mL was prepared by dissolving in 10 % DMSO. Concentrations of 40, 60, 80 and 100 mg/mL extracts were used for antimicrobial activity. The result of this study showed that clove extracts had the highest antimicrobial property against all the test microorganisms. Methanolic extract of clove had the highest inhibitory effect against Proteus mirabilis (24.21±0.15 mm), Pseudomonas aeruginosa (19.78±0.23 mm), and Candida albicans (20.07±0.08 mm) whereas ethanolic extract was effective against Escherichia coli (20.44±0.16 mm), Salmonella Typhi (21.66±0.31 mm) and Candida albicans (21.11±0.09 mm). Cinnamon and pepper extracts, leaving some exceptions, also had antimicrobial properties. The presence of phytochemicals: polyphenols, flavonoids, and tannins are the major components responsible for antimicrobial activity. Thereby, this study successfully demonstrated the possibilities of using spices extracts in the treatment of microbial infections.

Obtaining nanoscale materials has allowed for the miniaturization of components, which has led to the possibility of achieving more efficient devices with faster functions and much lower costs. While hydroxyapatite [HAp, Ca10(PO4)6(OH)2] is considered the most widely used material for medical applications in orthopedics, dentistry, and general surgery, the magnesium (Mg) is viewed as a promising biodegradable and biocompatible implant material. Furthermore, Mg is regarded as a strong candidate for developing medical implants due to its biocompatibility and antimicrobial properties against gram-positive and gram-negative bacteria. For this study, magnesium-doped hydroxyapatite (Ca10−xMgx (PO4)6 (OH)2, xMg = 0.1), 10MgHAp, suspensions were successfully obtained by an adapted and simple chemical co-precipitation method. The information regarding the stability of the nanosized 10MgHAp particles suspension obtained by ζ-potential analysis were confirmed for the first time by a non-destructive ultrasound-based technique. Structural and morphological studies of synthesized 10MgHAp were conducted by X-ray diffraction (XRD), Fourier-transform infrared (FTIR) spectroscopy in attenuated total reflectance (ATR) mode and scanning electron microscopy (SEM). The XRD analysis of the 10MgHAp samples confirmed that a single crystalline phase associated to HAp with an average grain size about 93.3 nm was obtained. The FTIR-ATR spectra revealed that the 10MgHAp sample presented broader IR bands with less visible peaks when compared to a well-crystallized pure HAp. The SEM results evidenced uniform MgHAp nanoparticles with spherical shape. The antimicrobial activity of the 10MgHAp suspension against gram-positive strains (Staphylococcus aureus ATCC 25923, Enterococcus faecalis ATCC 29212), gram-negative strains (Escherichia coli ATCC 25922, Pseudomonas aeruginosa ATCC 27853), as well as a fungal strain (Candida albicans ATCC 90029) were evaluated.

ABSTRACTCereulide and valinomycin are highly similar cyclic dodecadepsipeptides with potassium ionophoric properties. Cereulide, produced by members of theBacillus cereusgroup, is known mostly as emetic toxin, and no ecological function has been assigned. A comparative analysis of the antimicrobial activity of valinomycin produced byStreptomycesspp. and cereulide was performed at a pH range of pH 5.5 to pH 9.5, under anaerobic and aerobic conditions. Both compounds display pH-dependent activity against selected Gram-positive bacteria, includingStaphylococcus aureus,Listeria innocua,Listeria monocytogenes,Bacillus subtilis, andBacillus cereusATCC 10987. Notably,B. cereusstrain ATCC 14579 and the emeticB. cereusstrains F4810/72 and A529 showed reduced sensitivity to both compounds, with the latter two strains displaying full resistance to cereulide. Both compounds showed no activity against the selected Gram-negative bacteria. Antimicrobial activity against Gram-positive bacteria was highest at alkaline pH values, where the membrane potential (ΔΨ) is the main component of the proton motive force (PMF). Furthermore, inhibition of growth was observed in both aerobic and anaerobic conditions. Determination of the ΔΨ, using the membrane potential probe DiOC2(3) (in the presence of 50 mM KCl) in combination with flow cytometry, demonstrated for the first time the ability of cereulide to dissipate the ΔΨ in sensitive Gram-positive bacteria. The putative role of cereulide production in the ecology of emeticB. cereusis discussed.

Objective: The synthesis of novel benzimidazole-hydrazone derivatives has been carried out based on the previous findings that both these pharmacophores possess potent antimicrobial activities. The antibacterial properties of synthesized derivatives were screened against both Gram-positive and Gram-negative bacteria.Methods: O-phenylenediamine on condensation with substituted aromatic acids in polyphosphoric acid gave benzimidazole nucleus which on reaction with ethyl chloroacetate and hydrazine hydrate in two different steps resulted in the formation of substituted acetohydrazides. The targeted compounds 6a-l were synthesized by reaction of substituted acetohydrazides with aromatic aldehydes and screened for their antibacterial potential by cup-plate method.Results: The synthesized benzimidazole-hydrazones exhibited moderate to strong antibacterial activities against Staphylococcus aureus, Bacillus subtilis, Escherichia coli, and Pseudomonas aeruginosa. The compounds 6a-6f were found to be most effective against S. aureus, E. coli, and P. aeruginosa. Among all the synthesized compounds, the zone of inhibition of 6f in highest concentration, i.e., 100 μg/ml were found to be >31 mm against all the stains of bacteria.Conclusion: The antibacterial results revealed that the synthetized derivatives have significant antimicrobial properties and further structure activity relationship studies may develop more potent and less toxic molecules.

Multilayer films from hydrophobically modified poly(acrylic acid) (HMPA) and their complexes with cationic surfactants were successfully prepared using the layer-by-layer (LbL) method. Alkyl trimethylammonium bromide derivatives with various lengths of the hydrophobic chain (C10–C18) were used to interact with the HMPA polymer, generating highly hydrophobic domains in the films and contributing to the antimicrobial properties of the prepared coating. The antimicrobial efficiency against common pathogens such as Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, and Candida albicans was investigated in relationship with the morphology and composition of the thin films. The wettability and roughness of the multilayered systems were evaluated using atomic force microscopy (AFM) and contact angle measurements. The effects of the microbial exposure on the surface properties of the prepared films were investigated in order to assess the stability of the HMPA-deposited multilayers and the durability of the antimicrobial activity. The hydrophobically modified films exhibited antimicrobial activity against the studied pathogens. The best efficiency was registered in the case of S. aureus, which showed an inhibition of growth up to 100% after 2 h. C. albicans proved to be less sensitive to the effect of the multilayers deposited from HMPA–surfactant complexes. These results suggest that HMPA and HMPA–surfactant complex LbL multilayer films can be used as promising materials in antimicrobial surface coatings with increased resistance to pathogens during exposure.
