The Organophosphate Paraoxon Has No Demonstrable Effect on the Murine Immune System following Subchronic Low Dose Exposure

Paraoxon is the bioactive metabolite of the organophosphate pesticide parathion. Desulphuration of parathion by liver enzymes or sunlight results in the formation of paraoxon which inhibits acetylcholine esterase (AChE) activity. In the present study, we analyzed the effect of a 6-week, subchronic treatment with two different daily intraperitoneal doses (30 or 40 nmol) of paraoxon on the immune system of BALB/c mice. At a dose of 30 nmol/day, body weight of treated animals was unchanged compared to the controls. In contrast, the higher dose (40 nmol/day) induced a reduction in body growth, particularly in the first 3 weeks of treatment, peaking at week 2 when the saline group showed a 14.2-fold increase in body weight gain compared to paraoxon-treated animals. Moreover, mice treated with either dose of paraoxon had a >50% reduction in AChE activity during the first 3 weeks of treatment, but by the end of the treatment (week 6), AChE activity returned to normal. With regard to immunological parameters, there was no significant difference in either total spleen weight or in the ratios of various spleen cell populations between control and paraoxon-treated animals. Furthermore, no changes were observed in mitogen-induced cytokine secretion from splenocytes of paraoxon-treated mice. Finally, subchronic exposure to paraoxon did not alter mortality of mice exposed to a bacterial infection with Salmonella typhimurium. These data suggest that although subchronic exposure to paraoxon induced a transient inhibition in AChE activity, it had no demonstrable effect on the host immune system.

Poultry farming holds a special place in ensuring the products that the consumers demand, it provides the population with essential food products,such as eggs and meat,that contain vital micro and macronutrients, proteins, lipids, and vitamins. Therefore, the issues of rational, economically feasible feeding of meat poultry, namely broiler chickens, are an urgent task. It is also essential to find effective methods of their application in order to correct the natural resistance and immune and biological reactivity of birds. The purpose of this research is to study the effect of the biological stimulant-Nucleostim on the growth and development of chickens, hematological, and immunological parameters of the blood of birds.This Biostimulant is a purified bovine spleen extract containing at least 1 mg / ml of low molecular weight peptides (nucleotides and nucleosides) formed as a result of autolysis, using dry whey and diatomite as fillers. Onthe application ofNucleostim, the gain in live weight of chickens was increased by 9.7%. At the end of the experiment, the livability of the chicks of the experimental group treated with Nucleostimcame up to 88%, compared with the 72% of the control group. The use of biostimulant had a stimulating effect on the liver of chickens confirmed by the research results presented in the article, as well as contributed to the development of the thymus in the setting of general dystrophy. Thus, it improved chicklivability and increased body weight gain. The biological stimulant-Nucleostim as an adaptogenic, anabolic, and immunostimulatory agent is promising for finding new drugs that improve the health and productivity of poultry.

ABSTRACT. Tempe of soybean is fermented soybean with Rhizopus ologosporus fungus. From a result of researchers, it is the fact that tempe can increase valuable of nutrion and digest capacity and also produce antibiotic . Utilization of abtibiotic in chiken food as stimulus growth in not recommended because it can cause resistance a certain germ and it is presume there are antibiotic residu in egg and chiken meat. It is hope that the use of fermented soybean oil meal can increase nutrient value and digest capacity as well as produce antibiotic, so that it can get a better growth and over come a problem of using antibiotic in chiken food. The study was conducted to evaluate the effect of fermented soybean oil meal utilization in the ration on body weight gain, feed consumption, feed conversion and mortality. The study use 80 unsex day old chick broiler CP-707 and conducted at Poulty Laboratory Product Agriculture Faculty, Syiah Kuala University. Chiken were putting in 27 houses unit, each unit contain of four chiken. The design of experiment was Completely Randomized Design, four treatments and five replication. The treatments were ration contained 0%, 5%, 10% and 15% fermented soybean oil meal as replace the use of soybean oil meal. The protein and energy content of all dietary treatments were equally composed from six weeks investigation, were body weight gain, feed consumption, feed conversion and mortality. The result of the study indicated that up to 10% graded of fermented soybean oil meal in ration were significant difference influenced increase the body weight gain and decrease the feed conversion. The fermented soybeanoil meal) utilization more than 10% were significant difference decrease the body weight gain and increase the feed conversion. If the inclusion was increase the feed consumption would tendency decline. In this study there were no mortality in all treatment. In can be advised from this study that fermented soybean oil meal could be used in broiler ration or in all poulty ration.

As of today, 20 disease-modifying drugs (DMDs) have been approved for the treatment of relapsing multiple sclerosis (MS) and, based on their efficacy, they can be grouped into moderate-efficacy DMDs and high-efficacy DMDs. The choice of the drug mostly relies on the judgment and experience of neurologists and the evaluation of the therapeutic response can only be obtained by monitoring the clinical and magnetic resonance imaging (MRI) status during follow up. In an era where therapies are focused on personalization, this study aims to develop a modeling infrastructure to predict the evolution of relapsing MS and the response to treatments. We built a computational modeling infrastructure named Universal Immune System Simulator (UISS), which can simulate the main features and dynamics of the immune system activities. We extended UISS to simulate all the underlying MS pathogenesis and its interaction with the host immune system. This simulator is a multi-scale, multi-organ, agent-based simulator with an attached module capable of simulating the dynamics of specific biological pathways at the molecular level. We simulated six MS patients with different relapsing–remitting courses. These patients were characterized based on their age, sex, presence of oligoclonal bands, therapy, and MRI lesion load at the onset. The simulator framework is made freely available and can be used following the links provided in the availability section. Even though the model can be further personalized employing immunological parameters and genetic information, we generated a few simulation scenarios for each patient based on the available data. Among these simulations, it was possible to find the scenarios that realistically matched the real clinical and MRI history. Moreover, for two patients, the simulator anticipated the timing of subsequent relapses, which occurred, suggesting that UISS may have the potential to assist MS specialists in predicting the course of the disease and the response to treatment.

The effect of different levels of supplemented dietary protein on body weight, certain haematological values and meat yield characteristics in 20 "Shaver Star Bro" broilers was studied during the period from 02 March to 12 April 2002. The broilers were randomly assigned to four equal groups (A to D) each consisting of 5 birds. Group A was considered as control, fed only with commercial ration and other three groups were treated with extra dietary protein supplement @ 5%, 10% and 15% as group B, C and D respectively. Increased body weight was recorded in group B and the body weight decreases as the level of protein increased from 5% to 10% and 15% and the decrease was linear. No differences in haematological values were observed among the groups but the TEC varied significantly (p < 0.05). The mean weight of breast meat, drum stick meat varied significantly (p < 0.01). The shank weight varied significantly (p < 0.05) but there was no significant difference among the mean weight of thigh meat and wing meat corresponding to the different levels of supplemented dietary protein. It can be concluded that 5% supplemented dietary protein with commercial ration gives better result in respect to body weight gain, haematological values and meat yield characteristics.Key words: dietary protein; haematological values; meat yield; broiler birdsdoi: 10.3329/bjvm.v2i2.2543Bangl. Abstract The fundamental basis for immunotherapy of leukemia is that leukemia cells express specific antigens that are not expressed by normal hematopoietic cells. However, the host immune system appears to be tolerant of leukemia cells. To overcome this immune tolerance, we transduced WEHI-3B mouse monocyte leukemia cells (1) with a transmembrane form of GM-CSF (tmGM-CSF). The tmGM-CSF was constructed using the pDisplay vector for cell-surface targeting (Invitrogen) into the pLOX lentivirus gene transfer vector (2). After infection of WEHI-3B cells with a recombinant lentivirus encoding tmGM-CSF, nearly all the transduced cells expressed tmGM-CSF on the cell-surface, as determined by flow cytometry analysis using anti-GM-CSF. To determine whether vaccination with tmGM-CSF expressing WEHI-3B cells would prevent leukemia formation, immunocompetent BALB/c mice were immunized with lethally-irradiated WEHI-3B cells (106, 3 times 7 day intervals), which express tmGM-CSF, prior to challenging vaccinated mice with WEHI-3B cells (5x104) that express GFP as a marker. 100% of vaccinated mice were protected from leukemia. Non-vaccinated mice succumbed to leukemia within 50–55 days. Vaccination of mice with lethally-irradiated WEHI-3B cells expressing CD40L protected 80% of the mice from leukemia. In contrast, mice immunized with lethally-irradiated WEHI-3B/GFP cells lacking tmGM-CSF were not protected. Mice vaccinated three times at 5,12, 19 days after challenge with WEHI-3B/GFP cells had a significant increase in survival in that 60% of mice were alive and healthy at 16 days (to this date) after all control non-vaccinated mice had died. Similar vaccine studies were performed with BCR-ABL (b3a2)+ 32D cells (106) in immunocompetent C3H/HeJ mice (3). These mice die of leukemia within 35 days. After infection of BCR-ABL+ 32D cells with the lentivirus encoding tmGM-CSF/GFP, tmGM-CSF was expressed on the cell-surface. The C3H/HeJ mice challenged with BCR-ABL+32D/GFP cells (106) showed a significant level of protection by vaccination with lethally-irradiated tmGM-CSF+ 32D BCR-ABL cells (106, 2 times at 7 day intervals); 40% of the vaccinated mice remained healthy; all non-vaccinated mice died of leukemia. There was a significant difference in survival (P=0.03) between the vaccinated and non-vaccinated groups. Interestingly, the spleens of vaccinated C3H/HeJ mice that died of leukemia at the same time as non-vaccinated mice approached normal size whereas non-vaccinated mice had enlarged spleens. Our findings suggest that over-expression of cell-surface tmGM-CSF in leukemia cells can overcome immune tolerance, allowing the immune system to efficiently recognize and destroy the leukemia cells, providing extended survival of vaccinated mice. Because significant protection from death was achieved by vaccination after challenge with leukemia cells, tmGM-CSF expression in leukemia cells has potential as a therapeutic strategy for treatment of leukemia.

Two groups of one-day-old Peking ducklings (Groups I and II, 12 birds/group) were inoculated orally withBrachyspira pilosicoliand two groups withB. alvinipulli(Groups III and IV, 12 birds/group). T-2 toxin was added to the feed of Groups II and IV in a dose of 1 mg/kg of feed. Groups V and VI served as uninfected control groups (ducks of Group VI received T-2 toxin). The body weight gain of the ducks was measured and clinical signs were monitored continuously. The birds were sacrificed and necropsied on days 7, 14, 21, and 28 post infection (PI). The liver, spleen, kidney, thymus, bursa of Fabricius, ileum, caecum and colon were examined histologically. Culturing ofBrachyspiraspp. and immunohistochemistry were performed from the sampled parts of the intestines as well. No gross pathological or histological lesions that could be associated withB. pilosicoliorB. alvinipulliwere detectable in the intestinal mucous membrane including the colonised intestinal glands. Mortality did not occur during the experimental period. Decrease in body weight gain was significant in the T-2-toxin-treated groups, and it was slight (not significant) in theBrachyspira-infected groups. Crust on the beaks, necrosis, crusting and ulceration in the mucous membrane of the oral cavity and on the skin of the feet, atrophy of the thymus and bursa of Fabricius due to the effect of T-2 toxin, accompanied by lymphocyte depletion, were observed. These lesions were most prominent on days 14 and 21 PI but were seen on day 28 PI as well. Immunohistochemical detection and reisolation ofB. pilosicoliandB. alvinipulliwere successful on days 7, 14, 21 and 28 days from different segments of the intestine of certain birds, but no significant difference was observed in the colonisation rate between the T-2-toxin-treated and the untreated groups.

Objective: The study developed mushroom stalk residues as feed additives in the broiler diet for improving the growth performance and immunity of broilers as well as to increase the value of mushroom stalk residues.Methods: In total, 300 ROSS 308 broilers were randomly allocated into fifteen pens with five dietary treatments: i) control, basal diet; ii) CMWM, supplemented with 1% <i>Cordyceps militaris</i> waster medium (CM); iii) CMPE, supplemented with 0.5% CM+0.5% <i>Pleurotus eryngii</i> stalk residue (PE); iv) CMPS, supplemented with 0.5% CM+0.5% <i>Pleurotus sajorcaju</i> stalk residue (PS); v) CMFV, supplemented with 0.5% CM+0.5% <i>Fammulina velutipes</i> stalk residue (FV).Results: The chemical analysis results showed that CM extracts, PE extracts, PS extracts, and FV extracts contain functional components such as polysaccharides and phenols and have both 2, 2‐diphenyl‐1‐picryl‐hydrazyl‐hydrate scavenging and Ferrous scavenging capacities. The group CMWM saw increased body weight gain and feed conversion rate and the promotion of jejunum villus growth, but there is no significant difference in the intestinal bacteria phase. Antioxidant genes in the nuclear factor (erythroid-derived 2)-like 2 (Nrf2)- antioxidant responsive element pathway among the groups are significantly higher than that of the control group, especially in group CMWM.Conclusion: The mushroom stalk residues have antioxidant functional components, can improve the intestinal health and body weight gain of chickens, and can activate the antioxidant pathway of Nrf2 to increase the heme oxygenase-1 expression. The treatment with 1% CM was the most promising as a feed additive.

ABSTRAKPenelitian ini bertujuan untuk memanfaatkan Indigofera zollingeriana dalam ransum terhadap kinerja ayam sentul. Seratus empat puluh empat ekor ayam Sentul dialokasikan ke dalam 6 perlakuan dan 4 ulangan. Unggas tersebut dipelihara dari mulai day old chick (DOC) sampai umur 3 minggu dengan diberi ransum BR 501 CP, selanjutnya selama 7 minggu diberi ransum perlakuan. Ransum perlakuan tersebut adalah 1) R1 = ransum komersial (Sinta SP 22), 2) R2 = ransum yang biasa digunakan peternak, 3) R3 = ransum mengandung Indigofera zollingeriana 20% tanpa jagung, 4) R4 = ransum mengandung Indigofera zollingeriana 10%, 5) R5 = ransum mengandung Indigofera zollingeriana 15%, 6) R6 = ransum mengandung Indigofera zollingeriana 20% dan mengandung jagung. Penelitian menggunakan rancangan acak lengkap dan data yang terkumpul dianalisis dengan uji Duncan. Hasil menunjukkan bahwa konsumsi dan konversi ransum tidak menunjukan perbedaan yang nyata, tapi pertambahan bobot badan tertinggi (P<0,05) dicapai pada R1 dengan bobot badan akhir sebesar 833,25 g pada umur 10 minggu. Ransum R4 yang mengandung Indigofera zollingeriana 10% menghasilkan pertambahan bobot badan yang sama dengan R1 dengan bobot akhir penelitian sebesar 722 g. Kesimpulan Indigofera zollingeriana sebanyak 10% dapat diberikan dalam ransum ayam sentul.Kata kunci:  ayam sentul, performa, Indigofera zollingerianaABSTRACTThis study was aimed to utilize Indigofera zollingeriana in diets on the performance of sentul chicken. One hundred and fourty four sentul chickens were allocated into 6 treatments and 4 repetitions. This poultry was kept from DOC until the age of 3 weeks with BR 501 CP diets, next for 7 weeks fed treatment diets. The treatment diet was 1) R1 = commercial diet (Sinta SP 22), 2) R2 = diet commonly used by breeder, 3) R3 = diet containing Indigofera zollingeriana 20% without maize, 4) R4 = diet containing Indigofera zollingeriana 10%,  5) R5 = diet containing Indigofera zollingeriana 15%, 6) R6 = diet containing Indigofera zollingeriana 20% and containing maize. The study used a complete randomized design and the collected data were analyzed by Duncan test. The results showed that the consumption and conversion of diets did not show any significant difference, but the highest body weight gain (P <0.05) was achieved in R1 with final bodyweight of 833.25 g at 10 weeks. The R4 diet containing 10% Indigofera zollingeriana resulted in the same weight gain as R1 with the final weights of 722 g. Conclusion Indigofera zollingeriana as much as 10% can be given in sentul chicken diets.Keywords: performance, Indigofera zollingeriana, sentul chicken

A trial was conducted with 32 indigenous goat kids (Capra prisca) 8,5 weeks old to examine the effectiveness of dietary avoparcin as growth promoting factor. The kids were randomly allocated into two groups (control- and avoparcin group with initial mean body weight of 11,61 ±1,16 kg and 11,57±1,47 kg, respectively) comprising of 16 kids (8 male and 8 female) each. For a period of 12 weeks, kids in the control group were given lucerne (alfalfa) hay (mean daily consumption 170 g/head) and an appropriate compound diet (given ad libitum), while kids in the avoparcin group were fed the same diet except that the compound diet was supplemented with avoparcin (20 mg/kg). Avoparcin supplementation significantly (P<0.05) improved final mean body weight by 10.57% (19.24 ± 2.33 kg vs. 17.40 ± 2.30 kg), body weight gain (BWG) by 32.30% (7.66 ± 1.42 kg vs. 5.79 ± 1.74 kg), mean daily feed consumption in dry matter basis (DM) by 14.24% (551.80 ± 31.60 g DM vs. 483.02 ± 28.94 g DM), feed conversion ratio by 13.62% (6.09 ± 0.47 kg DM consumption/kg BWG vs. 7.05 ± 0.56 kg DM consumption/kg BWG) and carcass weight by 14.86% (11.44 ± 1.48 kg vs. 9.96 ± 1.76 kg). Moreover, no significant differences (P>0.05) were noticed neither in carcass yield and carcass chemical composition nor in percentages of small intestine-, major omentum-, lung-,heart-, liver- and spleen weight in the body weight.

Background and aims The aim of this study is to exploratively evaluate the effect of Tsumura Daikenchuto Extract Granules (DKT, TJ-100) on abdominal symptoms, body weight, and nutritional function following colorectal cancer surgery. Methods The subjects included 20 patients for curative resection of colorectal cancer. A TJ-100 administration group (n = 10) and non-administration group (n = 10) were randomized and compared. In the administration group, TJ-100 was administered from 2 days prior to surgery up to 12 weeks following surgery. The endpoints included body weight gain, Gastrointestinal Symptom Rating Scale (GSRS), and blood biochemical factors. For the purpose of observing safety, drug adverse events were evaluated including liver function tests. Results Excluding one patient, we compared 9 cases in the administration group and 10 cases in the non-administration group. No obvious adverse events were observed in any of the cases. In the comparison of body weight gain, the TJ-100 administration group showed significantly higher values at 2, 4, and 12 weeks following the surgery. There was a tendency for lower stable GSRS scores in the administration group overall, with no statistically significant difference. Conclusion It is suggested that TJ-100 can be safely administered in the perioperative period for cases undergoing colorectal cancer surgery, potentially preventing weight loss during the early postoperative period.
