Background: Rotenone treatment causes oxidative stress in neurons and forms the basis of animal models of Parkinson's disease. The reduced form of glutathione is predicted to detoxify rotenone from neurons in the brainstem. This study aims to measure the concentration of total glutathione and analyze the formation of protofibril in the brainstem of Wistar rats treated with rotenone. Methods: Seventy-two male Wistar rats aged 8–9 weeks weighing 200–250 g were divided into two investigations: total glutathione determination and protofibril analysis. The independent variables were treatment group, observation time, and location in the brainstem. The dependent variables were the concentration of total glutathione and protofibril density. Results: The concentration of total glutathione was not significantly different among treatment groups (p: 0.084), observation time (p: 0.608), or the location in the brainstem (p: 0.372). Protofibril density was different in the treatment groups (p: 0.001), observation time (p: 0.001), and between the upper and lower brainstem (p: 0.001). Rotenone treatment subcortically induced the concentration of total glutathione in the brainstem to decrease, but protofibril density tended to increase. Conclusions: The total glutathione concentration is inversely proportional to protofibril density. Total glutathione might be an early marker of neuronal damage.

Objective: To evaluate transcallosal changes after a local ischemic injury in rats by using the monoclonal marker anti-NeuN (Mouse anti-neuronal nuclei). Methods: Twenty eight adult, male, Wistar rats were subjected to focal injury in the right hemisphere. The technique used was the experimental model of focal ischemic injury through intraluminal suture of the middle cerebral artery. Analyses were made for the five groups: and after the lesion (control), at 24 hours, 96 hours, 10 days and 20 days. Exofocal neuronal damage was inferred from neuronal immunoreactivity changes to NeuN. Results: In the cortex contralateral to the lesion, immunoreactivity was diminished. This was most notable in the supragranular layers 24 hours post ischemia. After 96 hours, there was a generalized diminishment of the inmmunoreactivity in supra and infragranular layers. At 10 and 20 days, the tissue recovered some NeuN immunoreactivity, but there were set changes in the VI layer. Conclusion: The immunoreactive changes to NeuN support the process of interhemispheric diaschisis. Changes in immunoreactivity could indicate metabolic stress secondary to the disruption in connectivity to the site of lesion.

BACKGROUND<br />Non-alcoholic steatohepatitis (NASH) is a chronic liver disease associated with increased free fatty acids, trans fatty acids and free radical compounds due to the consumption of deep frying oil (DFO). Ginger has hypolipidemic and antioxidant properties, and acts as an insulin sensitizer. The present study was conducted to evaluate the hepatoprotective effect of an ethanol extract of Zingiber officinale Roxb var rubrum (EEZO) in DFO-fed male Wistar rats.<br /><br />METHODS<br />A study of experimental design was conducted involving 24 non-alcoholic steatohepatitis (NASH) male Wistar rats that were randomly divided into 4 groups. Group K1 (control) was given DFO-3 (3 times heated DFO) at a dose of 10 ìl/g BW/day. The treatment groups (P1, P2, P3) were given DFO-3 and red ginger rhizome extract at doses of 100, 200, and 400 mg/kg BW orally for 30 days. The histopathology examination assessed fatty degeneration, and degree of fibrosis. The data were analyzed with the Kruskal-Wallis and Mann-whitney test.<br /><br />RESULTS    <br />The Kruskal-Wallis test showed that fatty degeneration (steatosis, lobular inflammation, hepatocyte ballooning and NASH) were significantly different among the four groups (p&lt;0.05). Mann-Whitney test proved that fatty degeneration was not significantly different between control and treatment groups (p&gt;0.05).<br /><br />CONCLUSION<br />Deep frying oil increased fatty degeneration and NASH, and none of red ginger rhizome extract doses was able to inhibit fatty degeneration significantly. For the future research it is intended to prolong the induction time and the administration of red ginger rhizome extract.

Background: Although ethanol exerts its neurotoxic effect on the brain through inflammatory and oxidative processes, the effect of Riboceine on the brain following ethanol neurotoxicity is yet to be elucidated. Therefore, this study was designed to evaluate the effects of riboceine on the cellular, behavioral, and molecular impairments induced by ethanol toxicity in rats. Methods: A total of 24 male Wistar rats weighing between 160-170 grams were used for the study, and were divided into four groups of six rats each. After completion of the administration of ethanol and riboceine, and testing for motor impairment, the rats were sacrificed. The cerebellum was excised and processed for oxidative stress analyses, based on oxidative stress markers and histological examinations. The immunohistochemical expression of astrocytes in the cerebellum was examined, using Glial Fibrillary Acidic Protein (GFAP) stain. Results: This study demonstrated that ethanol-induced neurotoxicity in the cerebellum, characterized by increased oxidative stress profile, astrocyte activation, and neuronal death in the cerebellum, especially the Purkinje layer. Necrosis, significant decrease in Superoxide Dismutase (SOD), Catalase (CAT) and Gluathione (GSH) activities (P<0.05) as well as astrogliosis was associated with ethanol treatment. However, riboceine was observed to significantly increase the cerebellar SOD, CAT and GSH activities with significantly reduced Malondialdehyde (MDA) levels (P<0.05). It also attenuated the histomorphological alteration of the cerebellum and reduced the cerebellar astrocytes activation following ethanol-induced neurotoxicity, thus leading to the attenuation of motor impairment. Conclusion: Riboceine attenuated motor impairment caused by chronic ethanol-induced neurotoxicity, suggestive of its anti-oxidative and anti-inflammatory properties.

Critical Hepatitisis is the inflammation of the liver that is caused by viruses, alchol and drugs which are characterized by the increasing values of SGOT SGPT. The purpose of this study is to determine the effectiveness of fig leaves (Ficuss carica L) steeping water towards the decrease of SGOT SGPT serum on male wistar rats in critical hepatitis model. The object of this study is 30 male wistar rats with the age of 2-3 months and weight 180-200 grams and randomly divide into 3 groups, which are the negative group that is only by giving an usual food and drink, the positive group and the treatment groups are given paracetamol 120mg/ oral for 7 days. The treatment group is given 0.65 gram/ day of fig leaves steeping water for 7 days. One way ANOVA of SPSS version 24 is used to analyze this research. The result of the study shows that there is a significant difference in SGOT serum level between the treatment group, positive group and negative group (p<0,05). And there is a significant difference  in SGPT level between the treatment group and the positive group (p<0,05). The conclusion of this study is tin leaves (Ficus carica L) steeping water has an effect towards the decrease of SGOT SGPT in male Wistar rats in critical hepatitis model.

The rising threat of Plasmodium falciparum resistance to Monotherapies has prompted the world health organization (WHO) 2006 guidelines to recommend the use of different anti-malarias. In this study, the anti-malaria and hypoglycaemic activities of Diosgenin, a potent, yet poorly reported saponin was investigated on P. falciparum inoculated and Alloxan-Induced, Diabetic Wistar Rats. Fort two (42) adult male wistar rats of between 100g and 150g were procured, acclimatized (for two weeks), and grouped into seven of six (6) rats per group. While Group 1 (Normal control) received normal rat chow and water ad libitum, groups 2 – 4 received no treatment (untreated), 10 mg/kg body weight of anti-diabetic Metformin and 25 mg/kg body weight of diosgenin respectively after inducing diabetes mellitus (DM) with alloxan monohydrate; whereas, groups 5-7 (all malaria infected) were untreated (negative malaria control), 25 mg/kg body weight of diosgenin and 56 mg/kg body weight of anti-malaria coartem respectively. Following treatment period, blood samples were obtained and assayed for fasting blood sugar, packed cell volume (PCV) and total white blood cell count (TWBCC). From the result, P. falciparum exposed rats showed lowered PCV values than control with observed improvements in coartem (significant at p < 0.05) and diosgenin (insignificant) treatment groups. Also, diabetic, diosgenin treated rats showed an insignificant reduction in blood sugar levels compared to control, even though this change was apparently improved compared to diabetic, untreated group. Again, TWBCC caused notable decrease in diosgenin treated, though this decrease signified a huge recovery compared to untreated rats. Corroborative studies on diosgenin with other systems is recommended.

ABSTRACT Excessive energi consumption of high-fat diet increase the body’s adipose tissue which also affect the increased secretion of leptin, this is because leptin secretion is proportional to the mass of adipose tissue. Leptin activates the lateral hypothalamus as the center of hunger and feeding activity. The aim of this study was to obtain the relationship between high trans fat feeding on male Wistar rats and feeding activity. This method uses male Wistar rats (Rattus norvegicus) as an animal model. Sixteen rats were grouped into 4 treatment groups. The first group was given additional feed 20% trans fat, the second group was given additional feed 40% trans fat, a third group was given additional feed 60% trans fat, and the last as a control group. This feed was given once a day for 70 days. Observation of activities perform using Opto-varimex® auto-track system ver. 4.31. The results showed an corellation between activity based doses of trans fat. Keywords: Opto-varimex®, Trans fat, Activity, Wistar rat.

Background: There is a folk belief that drinking Ramsar spring mineral water (RSW) is a remedy for renal stones. Objectives: This study aimed to evaluate the effect of RSW on prevention or treatment of nephrolithiasis in male Wistar rats and identify the constituents of the water. Methods: Nephrolithiasis was induced by providing drinking water containing 1% ethylene glycol (EG) for two weeks in male Wistar rats. The animals were divided into prevention and treatment groups, each contained five sub-groups (10 rats for each). Prevention groups were treated for two weeks: control group (without treatment), EG group (1% ethylene glycol in drinking water), RSW 5, 10, and 15% groups (1% ethylene glycol + RSW 5, 10, and 15% in drinking water). The treatment groups received 1% EG for two weeks, then it was discontinued, and the RSW (10, 15, and 20%) was added to the drinking water for the next two weeks. The calcium oxalate (CaOx) depositions in the kidneys' tubules were evaluated by Hematoxylin and Eosin staining. The spring water was analyzed for its constituents. Results: Calcium oxalate crystals were significantly increased in EG group as compared to the control group (P < 0.05). Moreover, RSW did not significantly prevent CaOx crystals but alleviated CaOx crystals at 15 and 20% concentrations in the treatment protocol (P < 0.001). Conclusions: The hard water of Ramsar spring reduced CaOx crystals in nephrolithiatic rats, which may be due to its high calcium and magnesium content.

Objective: Smoking are known to have detrimental effect on sperm. The effects are suspiciously come from nicotine as its main pharmacologic component. This study is carried out to analyze the effect of nicotine exposure per inhalation to the difference of motile sperm percentage and normal morphology sperm percentage of male wistar rats. Material & Methods: Thirty male rats were used for this research. They were randomly divided into 5 groups and treated with nicotine inhalation 0.5 mg/kgBW,  1 mg/kgBW, 2 mg/kgBW and 4 mg/kgBW for 30 days while the control rats were not given any treatment. At the end of experiment, sperm analysis was carried out to measure the percentage of motile sperm and percentage of sperm with normal morphology. Results: Percentage of motile sperm significantly decreased (p<0.05) in all treatment groups compared to control group. Comparison between treatment groups revealed that the percentage of motile sperm significantly decreased in 0.5 mg/kgBW vs 2 mg/kgBW, 0.5 mg/kgBW vs 4 mg/kgBW, and 1 mg/kgBW vs 4 mg/kgBW. Percentage of normal morphology sperm decreased significantly (p<0.05) in 1 mg/kgBW, 2 mg/kgBW, and 4 mg/kgBW compared to control groups. Comparison between treatment groups revealed that there were no significant decreased between each treatment groups. Conclusion: This research conclude that nicotine exposure per inhalation can deteriorate the percentage of motile sperm and the percentage of normal morphology sperm of male wistar rats.

Background: Physical exercise aims to improve or maintain physical fitness. However, excessive physical exercise may cause increase of oxidative stress which leads to cellular injury, including in the proximal tubules of kidney. This research aims to find out the effect of single- and repeated- excessive swimming exercise to the kidney histopathology of male wistar rats (Rattus norvegicus). Methods: This research was an experimental study with complete random and post test only control group design. Twenty seven male wistar rats were divided into three treatment groups: control group, single- excessive swimming exercise group (45 minutes for one day), and repeated- excessive swimming exercise group (45 minutes/day for seven days). In the end of the treatment, the kidney of the rats were taken to be analyzed by using hematoxylin-eosin stain. The mean of proximal tubules injury of kidneys were counted by two observers using blinded method. Data were analyzed by using one way anova test. Result: The analysis showed there was a significant difference in the mean percentage of proximal tubules injury between control group and single swimming group as well as the repeated swimming group (Anova p=0,000), also there was significant difference between the treatment groups (p=0,020). The highest average of proximal tubules injury was in the single swimming group. Conclusion: Single- and repeated- excessive swimming exercise causes the proximal tubules injury of kidney.

The purpose of this study was to determine the effects of DLBS1033 on blood pressure reduction in hypertension--induced wistar strain rats. Design used in this study was true experimental design. The study was conducted at Biomedical Laboratory, Physiology, Faculty of Dentistry, Jember University, and Clinical and Community Pharmacy Laboratory, Pharmacy Faculty, Jember University. Subjects were taken from male Wistar rats aged 2-3 months, weighing 150-200 grams, as many as 22 rats. Intervention samples were induced into hypertension by giving prednisone in a dose of 1.5 mg/kg/day orally and 2% NaCl followed DLBS1033 of 0.5 grams/kgbw/day orally in the treatment group. T test results on systolic blood pressure (p = 0.00), which was <0.05, showing significant differences in systolic blood pressure in treatment groups. It can be concluded that DLBS1033 can lower systolic blood pressure in wistar rats with induced hypertension using prednisone 1.5 mg/kgbw/day and NaCl 2%.
