Antibacterial and antioxidant potential of ethyl acetate extract from Streptomyces AIA12 and AIA17 isolated from gut of Chanos chanos. Biodiversitas 22: 3196-3206. Streptomyces has been recognized as a promising and productive source of antibacterial and antioxidant compounds. The phenotypic and genomic characterizations indicated AIA12 and AIA17 are closely related to the S. globisporus ARGB01 and S. misionensis S1-SC15, respectively. Incubation on yeast-malt extract broth for nine days (AIA12) and 11 days (AIA17) proved to be the best growth medium and optimum antibacterial production time for these two strains. Crude-extract of extracellular secondary metabolites, obtained by ethyl acetate extraction, demonstrated broad-spectrum inhibitory activity against Pseudomonas aeruginosa InaCC B52, Staphylococcus aureus ATCC 25923, Bacillus cereus ATCC 10876, Escherichia coli ATCC 25922, Salmonella Typhimurium ATCC 14028, and Listeria monocytogenes with minimum inhibitory and minimum bactericidal concentrations from 2.5 to 0.31 and 5.0 to 0.31 mg mL-1, respectively. Evaluation of antioxidant showed AIA17 crude-extract had moderate DPPH scavenging and antioxidant activities of 65.122% ± 0.56 and 28.178 ± 0.24 mg ascorbic acid equivalent antioxidant capacity (AEAC g-1), respectively. The identification of compounds through profiling with RP-HPLC showed optimum absorbance at 210 and 214 nm, which showed the presence of peptide groups in the constituent compounds' molecular structure. These findings indicate that Chanos chanos-derived Streptomyces produces valuable bioactive compounds with various promising biological activities.

Mallotus oppositifolius (Geiseler) Mull. Arg.  (Euphorbiaceae) is a predominant edible shrub in Nigeria. It is locally used for treatment of common infections and wounds. This study evaluated the toxicity and antioxidant potential of the crude extract and fractions of M. oppositifolius using invitro 1,1-diphenyl-2-picrylhydrazine (DPPH) radical scavenging assay. The result showed abundance of flavonoids (28.21), tannins (17.72), alkaloids (7.79) and Saponins (0.23) in the crude extract of M. oppositifolius leaf extract. Also, the result showed that the LD50 is above 5000 mg/kg, therefore the crude extract is safe for consumption. The result further showed that the ethyl-acetate and butanol had significant (p<0.05) antioxidant activity when compared with the control at 125, 62.5 and 31.125 µg/ml respectively while at 250 and 500 µg/ml there was no significant different in the level of reduction of oxidation between the butanol and ethyl acetate when compared with the control. The result further showed that the butanol and ethyl-acetate fraction of M. oppositifolius had the best antioxidant potential among the fractions studied. The result of GCMS studies of ethyl-acetate fraction showed the presence of 7,10,13-eicosatrienoic acid methyl ester, korormicin, agosterol, phyllanthin, ricinoic acid, 2,4,6-decatrienoic acid 1a,2,5,5a,6,9,10,10a-octahydro-5,5a-dihydroxy-4-(hydroxymethyl)-1,1,7,9, limonin, com-pounds that have shown to possess strong antioxidant activities. Also, the GCMS result of butanol fraction showed the presence of seven compounds which include: ferulylhistamine, hippeastrine, 10,13-eicosadienoic acid methyl ester, ethyl 5,8,11,14-eicosatetraenoate, 6-hydroxy-powelline-N-nitroso-7-demethoxy-aldehyde, 6,7-epoxypregn-4-ene-9,11,18-triol-3,20-dione 11,18-diacetate and 5Alpha-androstane-3,17-dione 17-monooxime,  compounds which may serve as a new lead for the treatment of diseases associated with the oxidative stress.

Muthupet mangrove forest soil sediment was the abundant resource of the actinomycetes with distinct nature of bioactive compounds. The soil sediment was collected at 1-3meter away from bank. The present study was focused on isolation, identification and antimicrobial activity of the actinomycetes from Muthupet mangrove soil samples. Totally 32 actinomycetes strains was isolated and screened for antimicrobial activity against bacterial and fungal pathogens. Among 32 isolates 16 have antibacterial activity and 10 have antifungal activity but MG-3 and MG-4 showed maximum activity against both all the test pathogens. These two strains are gram-positive, rod-shaped, MG-3 possessing an earthy characteristic odour and MG-4 produce purple color pigment. The isolates were confirmed as Streptomyces sp. based on morphological, cultural, biochemical and physiological observations, as well as identification using the 16S rRNA gene sequence, it showed 98% similarity with Streptomyces parvus for MG-3 and Streptomyces californicus for MG-4. Bioactive compounds were extracted from Streptomyces using different solvents such as ethyl acetate, methanol, chloroform, hexane and antibacterial activities were assayed against test pathogens, ethyl acetate extract showed maximum zone of inhibition when compared with other solvents. The Minimum inhibitory concentration of ethyl acetate extract was found ranged between 1.96-3.9 μg/ml. The invitro antioxidant capacity of the crude extract was estimated by DPPH, ferric reducing power assay, H202 radical scavenging assay, phosphomolybdenum assay and total antioxidant activities. The characterization of crude extracts was analyzed by FTIR and GC-MS. From the results, it is clear that the ethyl acetate crude extract of S.parvus MG-3 and S.californicus MG-4 possesses high antimicrobial and antioxidant activity and suggested that the isolated strains could be a potential for the nature resource of pharmaceutical.

  Objective: The aim of the present study was to investigate in vitro antibacterial, antifungal, and antioxidant activities of different solvent extracts of Garcinia mangostana leaves.Methods: The powdered leaf was subjected to sequential extraction using hexane, ethyl acetate (EA), and methanol. The extracts were subjected to quantitative and qualitative phytochemical analysis, antimicrobial, and antioxidant activities. The best solvent extract was subjected to gas chromatography–mass spectrometry (GC-MS) analysis.Results: The highest activity was recorded in EA extract which was subjected to GC-MS analysis revealing the presence of squalene (17.09%).Conclusion: From this present study, we conclude that EA is the best solvent for extracting antimicrobial and antioxidant compounds from the leaves of G. mangostana.

Poliomintha longiflora is a Mexican oregano, which has not been widely studied. This work aimed to describe the chemical composition, antimicrobial and antioxidant activities present in P. longiflora essential oil (EO), the hydrosol from EO extraction and extracts from waste solid residues (WSRs), identified as ethanol extract, ethyl acetate extract and the subfractions of ethanol and ethyl acetate extracts. The chemical characterization of the EO, hydrosol and WSR extracts was performed by GC–MS and HPLC. Their antioxidant activity was evaluated using two methods, and their antimicrobial activity was evaluated against Escherichia coli, Staphylococcus aureus, Listeria monocytogenes, Bacillus cereus, and Salmonella Typhimurium. Thirty-one chemical components were identified in the EO. The subfractions from the ethanol and ethyl acetate extracts contain methylmaleic anhydride, thymoquinone, thymol, carvacrol, thymol acetate, carvacrol acetate, and phenolic acids. The EO presented the highest biological activities for antioxidant (136.05 mg equivalent of ascorbic acid/g (AAE/g); IC50 83.70 μg/mL of 2, 2-diphenyl-1-picrylhydrazyl (DPPH)) and antimicrobial tests (minimal inhibitory concentration (MIC) value of 250–750 mg/L), while the hydrosol and the ethyl acetate extract from WSRs had the lowest antioxidant activity (14.16 and 12.29 mg AAE/g respectively), and the hydrosol had the lowest antimicrobial activity (MIC of 3000 mg/L). The data suggest that Mexican oregano P. longiflora hydrosol and extracts from waste solid residues can still have compounds with antimicrobial and antioxidant capacities.

Study to assess the larvicidal property of Lantana camara leaves against Aedes triseriatus larvae found that the ethyl acetate extract had profound larvicidal action with the crude extract having a LC50 value of 409.831ppm. GC-MS analysis of the ethyl acetate extract confirmed the presence of twenty-one compounds out of which beta-caryophyllene covered the highest percentage of the chromatogram area. Further tests with beta-caryophyllene against the mosquito larvae proved it to be the active ingredient of L. Camara with a LC50 value of 104.243ppm.

Background: Plant in Rhododendron genus that contains phenolic compounds has been used in traditional medicine and revealed considerable biological activities. Objective: Isolation and identification of antioxidant natural products from Rhododendron ungernii. Methods: Rhododendron ungernii Trautv. flowers were collected and dried in shade. The dried flowers were extracted with methanol for 3 days. The solvent was removed by reduced pressure to yield the extract which was subjected to column chromatography (Sephadex LH-20, C18 reversed phase column) to isolate catechin-7-O-glucoside (1), quercetin-3-O-β-galactoside (2), quercetin-3-O- β-xyloside (3), farrerol (4), myricetin (5), and quercetin (6). The structures of isolated compounds were elucidated by spectroscopic methods such as 1D-NMR, 2D-NMR, and LC-TOF/MS. DPPH scavenging effect, ABTS+ scavenging activity, and reducing power (FRAP) were performed for antioxidant assays of isolated natural compounds. Results: Isolated flavonoids displayed the outstanding antioxidant activities. Catechin-7-O-glucoside (1) and quercetin-3-O-β-galactoside (2) (IC50, 3.66 µg/mL) had the most DPPH• scavenging effect among the compounds. The highest ABTS•+ scavenging activity (IC50, 1.41 µg/mL) and reducing power effect (6.05 mmol TE/g comp) were observed for myricetin (5). Conclusion: R. ungernii extract and isolated compounds could be a promising antioxidant for food and pharmaceutical industries.

The present study proposes microwave-assisted extraction as a sustainable technique for the biosynthesis of bioactive compounds from rice fermented with Aspergillus flavus (koji). First, fermentation conditions (i.e., pH from 3–12, five temperatures from 20–40 °C, and four culture-fermentation media viz. wheat, wheat bran, malt and rice) were optimized for producing microbial bioactive compounds. Microwave extraction was performed at 2450 MHz and 500 W for 20, 30, and 40 s with seven solvents (distilled water, ethyl acetate, hexane, ethanol, chloroform, diethyl ether, and methanol). The obtained results revealed that ethyl acetate is the most appropriate solvent for extraction. Effects of this ethyl acetate extract were compared with a commercial synthetic antioxidant. Antioxidant properties were enhanced by preventing the oxidation of the linoleic acid (C18H32O2) with an inhibition rate (antioxidant efficacy) of 73.13%. Notably, the ferrous ion binding ability was marginally lower when compared to the disodium salt of ethylenediaminetetraacetic acid (EDTA). Additionally, the obtained total content of phenolic compounds in the ethyl acetate extract of fermented rice (koji) by Aspergillus flavus was 232.11 mg based on gallic acid/mL. Antioxidant compounds in the ethyl acetate extract of fermented rice showed stability under neutral conditions, as well as at high temperatures reaching 185 °C during 2 h, but were unstable under acidic and alkaline conditions. The results demonstrate the efficacy of novel microwave-assisted extraction technique for accelerating antioxidant production during rice fermentation.

The antioxidant activities of extracts (n-hexane, dichloromethane, ethyl acetate and methanol) from the leaves and stem barks of Artocarpus scortechinii were evaluated using various biochemical assays. The quantification of the Total Antioxidant Capacity was measured using ferric reducing antioxidant potential (FRAP) and 2,2'-azino-bis(3-ethyl-benzothiazoline-6-sulphonic acid) (ABTS) assays. While, the qualitative of The Total Phenolic Content (TPC) was determined via standard gallic acid calibration graph which was expressed as mg gallic acid equivalent (GAE)/g of dry weight (dw) using Folin Ciocalteau’s reagent. Among all the extracts tested, the methanolic extract of the stem barks showed the highest phenolic content with TPC value of 136.84 mg GAE/g dry weight (dw). FRAP results were expressed as mM equivalent to FeSO4.7H2O by calculating from the standard FeSO4.7H2O calibration graph. The ethyl acetate extract of the stem barks showed the most significant reducing potential in the range between 0.27-2.47 mM FRAP. ABTS+˙ radical scavenging capacity showed that the ethyl acetate extract of the stem barks had the highest scavenging capacity at concentration 1.0 mM with percentage of 90.9%.

ABSTRACT: In the present study, antibacterial and antioxidant [1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity] activities of a weed, slender amaranth (Amaranthus viridis L.) were investigated. Extracts of different plant parts were prepared in n-hexane, chloroform and ethyl acetate. Antibacterial activity was measured by using 100 mg mL-1 concentration extracts against 4 deadly phytopathogenic bacterial species viz. Pseudomonas syringae Van Hall, Ralstonia solanacearum Smith, Erwinia carotovora (Jones), Holland and Xanthomonas axonopodis Hasse. In antioxidants assays, 10, 20 and 30 mg mL-1 extracts were used keeping DPPH as control. In these bioassays, ethyl acetate fraction of A. viridis leaf exhibited the best antibacterial and antioxidants activity. Ethyl acetate leaf fraction showed the highest inhibition zone diameter (IZD) where it caused 21 mm IZD against P. syringae and 19 mm IZD against E. carotovora. This extract also showed 22, 52 and 84% antioxidant activity at 10, 20 and 30 mg mL-1 concentrations, respectively. Previously there is no report available that describes antibacterial activity of root extract of A. viridis against P. syringae. Moreover, antioxidant activity of stem and root extracts in n-hexane, chloroform and ethyl acetate was investigated first time in the world. It was concluded that the biological activities observed during the present investigation may be due to the presence of bioactive constituents that can be harnessed as natural antibacterials and antioxidants.

Background: Although the chemical constituents and biological activities of a large number of plants in the Croton genus have been studied, there are still recently discovered plants to be investigated. Objective: 1. To investigate the anti-bacterial, anti-HIV1-RT, and cytotoxicity activities of crude extracts from these plants. 2. To investigate the chemical constituents of Croton fluviatilis, Croton acutifolius, and Croton thorelii. Method: The anti-bacterial, anti-HIV1-RT, and cytotoxicity of the three plants were evaluated by standard techniques. Extraction, separation, and purification of extracts from the three plants were undertaken. Results: The ethyl acetate extract of C. fluviatilis showed low anti-bacterial activity against E. aerogenes, E. coli 0157: H7, and P. mirabilis, together with the ethyl acetate extract of C. acutifolius displayed low anti-bacterial activity against E. aerogenes, while all the crude extracts of C. thorelii were inactive. The ethyl acetate extracts of C. thorelii, and C. fluviatilis showed strong inhibited HIV1-RT, whereas the ethyl acetate extract of C. acutifolius, and the hexane extract of C. fluviatilis displayed moderate inhibited HIV1-RT. Cytotoxic properties of three Croton plants were specific to KKU-M213, MDA-MB-231, A-549, and MMNK-1. Especially, the ethyl acetate extract of C. acutifolius exhibited strong cytotoxic activities against MDA-MB-231, A-549, and MMNK-1. Furthermore, the ethyl acetate extract of C. thorelii showed high cytotoxic activities against KKU-M213, and MDA-MB-231. Compounds 1, and 4 were found in C. fluviatilis. Compounds 2 and 4 were also found in C. acutifolius. Moreover, compound 3 was only found in C. thorelii. Conclusion: The present study revealed that the three Croton species are good sources of flavonoid compounds and further investigation of the chemical constituents from these plants may prove to be fruitful to discover more active compounds to be tested as potential medicines.
