ABSTRACTUtilization of marine life is not just consumptive but can be used as raw material for medicines, one of which is sea cucumber. Sea cucumbers (H.atra) are marine invertebrates belonging to the Echinoderms phylum which have been studied containing antioxidants. Therefore, a study was conducted to determine the antioxidant activity of sea cucumber (H.atra) extract taken from the waters of Manado Bay using the DPPH (1,1-diphenyl-2-picrylhydrazyl) method. Sea cucumber samples were extracted using the maceration method. The ethanol extract of sea cucumber showed high antioxidant activity at a concentration of 8 μg / ml with 77.83% inhibition percentage. Keywords: H.atra, Antioxidant, DPPH (1,1-diphenyl-2-picrylhydrazyl).  ABSTRAKPemanfaatan biota laut bukan hanya sekedar konsumtif saja tapi dapat berpotensi sebagai bahan baku obat-obatan salah satunya teripang. Teripang (H.atra) adalah hewan invertebrata laut yang termasuk ke dalam filum Echinodermata  yang pernah diteliti mengandung antioksidan. Oleh karena itu, dilakukan penelitian untuk mengatehui aktivitas antioksidan dari ekstrak teripang (H.atra) yang diambil dari perairan Teluk Manado dengan menggunakan metode DPPH (1,1-diphenyl-2-picrylhydrazyl). Sampel teripang diekstraksi menggunakan metode maserasi. <strong>Extraction and Oral Suspension Formulation of Sea Cucumber Holothuria scabra as Source of Antioxidants.</strong> Sea cucumbers have long been utilized in the food and medicine for Asia and Middle East communities. Indonesia is one of the main suppliers in the trade of sea cucumbers in Asia. Sea cucumber utilization in Indonesia is generally done only in the form of raw materials. Holothuria scabra is one of the sea cucumber that can be explored as a potential source of antioxidants. This study aims to determine the antioxidant activity of sea cucumber H.scabra in extract and suspension. Sea cucumber was extracted with methanol 96% and ethanol 96%, then both antioxidant activity was tested. Extract with higher antioxidant activity were made into oral suspension preparation for further evaluation of the stability (organoleptic, pH, microorganisms) and antioxidant testing. The results showed that ethanol extracts had higher antioxidant activity of methanol extract, but still lower than ascorbic acid. Based on the calculation, IC<sub>50 </sub>of methanol extract was 316.85 ppm and ethanol extract was 130.54 ppm, while ascorbic acid showed IC<sub>50</sub> 34.06 ppm. The most stable formula (F1a) shows that there is still an antioxidant activity that is equal to 42.11 %.

Diseases mediated by the immune system are difficult problems to treat such as human immunodeficiency virus (HIV) and other lethal viruses. Infections that occur in normal people are generally brief and rarely leave permanent damage. Treatment of this disease requires an aggressive and innovative approach to the development of new treatments so that it requires the role of immunomodulators to improve the immune system. A substance that acts as an enhancer or immune enhancer can be obtained by using herbs that are efficacious as immunostimulants. One of the herbs used is herbal binara (Artemisia vulgaris L) which has been studied as a potential immunomodulator with high antioxidant activity. Previous research also stated that red shoots (Syzygium oleana) were studied as potential immunomodulators with high antioxidant activity. Several other species such as Syzygium samarangense have 16 flavonoida compounds which show pharmacological immunological activity. The purpose of this study was to determine the content of secondary metabolites of ethanol extract of herbal binara (Artemisia vulgaris L.) with red shoots (Syzygium oleana) and to determine the best dose of extract from the ethanol extract of herbal binara (Artemisia vulgaris L.) with red shoots (Syzygium oleana) can reduce the volume of swelling of mouse feet. Examination of the chemical content of secondary metabolites from the ethanol extract of herbal binara (Artemisia vulgaris L.) with red shoots (Syzygium oleana) is carried out by chemical screening and characterization of simplicia and extract. The method used is the slow type hypersensitivity method. In this test the independent variable is the secondary metabolite of ethanol extract of herb binara (Artemisia vulgaris L.) with red shoots (Syzygium oleana) with four concentrations (50, 100, 200 and 400 mg / kgBB). The positive control used by Stimuno dose is 32.5 mg / kgBB

ABSTRACTHabitat of sponge Stylissa sp., were under the sea and these sponges contain active compounds, which are more active than the compounds produced by terrestrial plants. Antioxidants are inhibitors of oxidation reactions due to the free radicals, which can cause weak damage to unsaturated cells, cell wall membranes, blood vessels, DNA bases, and lipid tissue, that causing disease. The study was conducted to determine the antioxidant activity of ethanol extracts  of sponge Stylissa sp., which is located on the Bangka Island, Likupang District, North Minahasa Regency. Sponge Stylissa sp., was extracted by maceration with using ethanol. Testing of antioxidant activity was carried out by the DPPH method measured by a UV-Vis spectrophotometer. The results of the study showed that ethanol extracts of sponge Stylissa sp., has antioxidant activity in each concentration and the highest at a concentration of 100 mg / L. The conclusion is a ethanol extract of sponge Stylissa sp. have high antioxidant activity with a concentration of 25 mg/L (77,40%), concentration of 50 mg/L (85,63%), concentration of 75 mg/L (88,66%), and concentration 100 mg/L (88,96%). Key words: Stylissa sp. Antioksidan adalah zat penghambat reaksi oksidasi akibat radikal bebas yang dapat menyebabkan kerusakan lemah tak jenuh, membran dinding sel, pembuluh darah, basa DNA, dan jaringan lipid sehingga menimbulkan penyakit. Penelitian dilakukan untuk mengetahui aktivitas antioksidan ekstrak etanol biota laut spons Stylissa sp. yang terdapat di pulau Bangka, Kecamatan Likupang, Kabupaten Minahasa Utara. Spons Stylissa sp. ini diekstrak dengan metode maserasi menggunakan etanol. Pengujian aktivitas antioksidan ini dilakukan dengan metode DPPH yang diukur dengan alat spektrofotometer UV-Vis. Hasil dari penelitian menunjukan bahwa ekstrak etanol spons Stylissa sp. memiliki aktivitas antioksidan disetiap konsentrasi dan yang tertinggi pada konsentrasi 100 mg/L. Kesimpulan yang didapat bahwa ekstrak etanol spons Stylissa sp. memiliki aktivitas antioksidan yang tinggi dengan konsentrasi 25 mg/L (77,40%), konsentrasi 50 mg/L (85,63%), konsentrasi 75 mg/L (88,66%), dan konsentrasi 100 mg/L (88,96%). Kata Kunci : Spons Stylissa sp., Antioksidan, DPPH (1,1-difenil-2-pikrilhidrazil)

Abstract Marine invertebrates that move too slowly to evade unfavorable environmental change may instead exhibit phenotypic plasticity, allowing them to adjust to varying conditions. The orange-footed sea cucumber Cucumaria frondosa is a slow-moving suspension feeder that is preyed on by the purple sunstar Solaster endeca. The sea cucumber’s antipredator behavior involves changing shape and detaching from the substratum, which might increase its probability of being displaced by water motion into an unsuitable environment. We hypothesized that sea cucumbers’ antipredator responses would be diminished under stronger hydrodynamic forces, and that behavioral strategies would be flexible so that individuals could adjust to frequent changes in water flows. In a natural orange-footed sea cucumber habitat, individuals lived along a pronounced hydrodynamic gradient, allowing us to measure antipredator behavior under different water flow strengths. We placed purple sunstars in physical contact with sea cucumbers living at various points along the gradient to elicit antipredator responses. We then repeated this procedure in a laboratory mesocosm that generated weak and strong hydrodynamic forces similar to those observed at the field site. Subjects in the mesocosm experiment were tested in both wave conditions to determine if their antipredator behavior would change in response to sudden environmental change, as would be experienced under deteriorating sea conditions. Antipredator responses did not covary with hydrodynamic forces in the field. However, antipredator responses in the mesocosm experiment increased when individuals were transplanted from strong to weak forces and decreased when transplanted from weak to strong forces. Overall, our results indicate environmentally induced plasticity in the antipredator behavior of the orange-footed sea cucumber.

The aim of the present study was to determine the influence of the winemaking process on the antioxidant potential and content of phenolic compounds and L-ascorbic acid in wines from the fruits of Rosa rugosa. The results obtained in this study clearly indicate that the fruits of the Rosa rugosa are a desirable raw material for the production of fruit wine. The parameters of the technological process of producing wines from rose fruits had a diversified influence on the tested quality characteristics. Aged wines contained phenolics levels of 473–958 mg/100 mL GAE. The final concentrations of ascorbic acid ranged from 61 to 155 mg/100 mL for the different variants of the wine. Wines revealed high antioxidant activity in assay with DPPH. On the basis of the obtained results, it can be assumed that all the applied variants of the winemaking process are suitable for rose fruit wine. Each variant ensured at least the stability of the antioxidant capacity.

Henslowia frutescens is a semi-parasitic plant, although considered harmful, it has medicinal and cosmetic potential. This plant has secondary metabolite compounds, namely phenolic compounds, flavonoids, saponins, tannins, and steroids which have high antioxidant activity. The purpose of this study was to determine the sunscreen activity of the ethanol extract of Henslowia frutescens based on the in vitro SPF (Sun Protecting Factor) value and to determine its antioxidant activity. The research stages included sampling, sampling, maceration extraction with 95% ethanol, yield calculation, phytochemical screening, sunscreen activity test based on SPF value, and antioxidant activity test. Data analysis was carried out descriptively based on the measurement results processed according to the calculation formula. The results showed that the sunscreen activity of the ethanol extract of Henslowia frutescens based on the in vitro SPF value was 14.57 - 25.20 with moderate protection category. The antioxidant activity of the ethanol extract of Henslowia frutescens showed a very strong category with an IC value of 22.82 ± 1.33.

Teripang merupakan hewan jenis Echinodermata. Beberapa species teripang, misalnya H. scabra, H. fuscogilva dan T. ananas memiliki nilai jual yang tinggi. Meskipun tidak semua spesies memiliki nilai jual yang tinggi, teripang berpotensi untuk dikembangkan dalam bidang kesehatan, terutama dalam kemampuannya untuk meredam radikal bebas dan mencegah berbagai penyakit degeneratif. Tujuan dari penelitian ini yaitu untuk mengidentifikasi, menentukan aktivitas antioksidan, kandungan total fenol, serta kadar karotenoid pada ekstrak beberapa teripang yang berasal dari Perairan Karimunjawa Jepara. Hasil identifikasi dari ke empat spesies adalah Stichopus cf. quadrifasciatus, Pearsonothuria graeffei, Bohadschia vitiensis, dan Holothuria atra. Ekstraksi dilakukan dengan teknik maserasi dinding tubuh teripang menggunakan pelarut metanol. Uji aktivitas antioksidan dilakukan dengan menggunakan metode DPPH (1,1-difenil-pikrilhidrazil) sebagai agen radikal bebas. Penentuan total fenolik dilakukan menggunakan metode folin-ciocalteu, dan penentuan kadar karotenoid dilakukan dengan menggunakan spektrofotometer. Hasil menunjukkan bahwa aktivitas antioksidan Bohadschia vitiensis, Stichopus cf. quadrifasciatus, Pearsonothuria graeffei, dan Holothuria atra tergolong sangat lemah dengan nilai IC50 secara berturut-turut adalah sebesar 454,28 ± 14,4; 713,51 ± 9,1; 801,57 ± 8,4 dan 1535,47 ppm. Kandungan total fenol sebesar 21,08 ± 0,49; 14,325 ± 0,21; 14,033 ± 0,33 dan 10,67 ± 0,12 mg GAE/g sampel, dan kadar karotenoidnya sebesar 23,28 ± 0,07; 25,78 ± 0,56; 11,85 ± 0,04; dan 41,44 ± 0,008 µmol/g sampel.Kata kunci: Teripang, Antioksidan, DPPH, Fenolik, Karotenoid Sea cucumbers are sea benthos from phylum Echinoderms phylum. Some sea cucumbers such as H. scabra, H. fuscogilva, T. Ananas have a high economical price. Eventhough, some sea cucumbers have the potency of being a healthy food. It it scientifically proven that some sea cucumbers have the ability to reduce free radicals and prevent various degenerative diseases caused by the excessive free radicals. The aims of this study were to identified, determine antioxidant activity, total phenol content, and carotenoid levels in sea cucumber extracts originally from Karimunjawa waters. Some of the sea cucumbers obtained are Stichopus cf. quadrifasciatus, Pearsonothuria graeffei, Bohadschia vitiensis, and Holothuria atra. The extraction was done by macerating the body walls with methanol. The antioxidant activity test were tested using the DPPH method as free radicals, the total phenolic content tested using the folin-ciocalteu method, and determined the carotenoid levels spectrophotometrically. Based on the results, antioxidant activity of  B. vitiensis, S. cf. quadrifasciatus, P. graeffei, and H. atra were  classified as very weak with IC50 454.28 ± 14.4; 713.51 ± 9.1; 801.57 ± 8.4 dan 1535.47 ppm. Total phenolic content were 21.08 ± 0.49; 14.325 ± 0.21; 14.033 ± 0.33 dan 10.67 ± 0.12 mg GAE / g samples, while carotenoid levels were 23.28 ± 0.07; 25.78 ± 0.56; 11.85 ± 0.04; dan 41.44 ± 0.008 µmol / g sample.Keyword : Sea cucumber, Antioxidant, DPPH, Phenolic, Carotenoid

The seaweed collected after stranding on beaches of Latvia is underexploited natural resource, which has a potential as raw material for biologically active compound extraction for cosmetic and pharmacy and fertilizer in sustainable agriculture. The aim of the present study was development of the approach for the processing of brown alga Fucus vesiculosus biomass, collected from the Gulf of Riga. The thorough characterization of the chemical composition of Fucus vesiculosus has shown that it is a potential source for obtaining of nitrogen-containing fertilizers, and biologically active compounds. One of the proposed approaches for the processing of the Fucus vesiculosus biomass under study includes algae extraction with organic solvents and CaCl2 solution and obtaining soil organic amendment on the basis of the extract-free residue. The ethyl acetate extract was rich in phenolic compounds (430 ± 30 GAE mg/g) with high antioxidant activity in DPPH• and ABTS•+ tests. The ethanol extract contained significant amounts of phlorotannins that was confirmed by the data of LC-MS/MS analysis. The CaCl2 extract was used for the obtaining of sulphated polysaccharide fucoidane (yield ≈7% on the basis of oven dry matter, o.d.m.), which has numerous biological activities. The extract-free residue didn’t show phytotoxicity. The extract free algal biomass exhibited positive effect in root elongation tests with oat (Avena sativa). As alternative approach for processing of the algal biomass, the mechano-chemical treatment of algal biomass with lignin was proposed. EPR analyses confirmed interaction between algal biomass and lignin.

Drying is the most important step to keep the compound stability in simplicia and is very influential to produce good quality igredients in the antioxidant activity contained in the simplicia. Antioxidants are oxidation-free inhibitors of free radical reactions. Parijoto is one of the plants that have antioxidant compounds.This study aims to determine the effect of drying, concentration of antioxidant activity of parijoto (Medinilla speciosa Blume) fruit. The drying method used is direct sunlight drying, indirect sunlight and oven. The antioxidant activity of ethanol extract on drying method was determined by 2,2 diphenyl-1-picrylhidrazyl (DPPH) method which is free radical. The qualitative test DPPH showed that the higher levels of ethanol extract from the three drying methods further diminished the color of DPPH solution in ethanol.The result showed thet the drying method gave different effect to the antioxidant activity of parijoto fruit which is significant(p0,05). Drying methods wich have high antioxidant activity are given by drying using oven method with an IC50 value 33,75µg/ml.

Sea cucumbers belonging to the phylum Echinodermata are important marine invertebrates, generally occur in the sandy substrates and rocky shores near the bottom of sea. This review work has been attempted to make an interlink between biological and socio-economical perspectives of sea cucumber because of the high demand in the present world. The outer body part of sea cucumber (bêche-de-mer) has been recognized for producing secondary metabolites including vitamins and triterpenoid glycoside saponins that have profound ecological and bio-medicinal significance. Sea cucumbers are also used to remove arthritis pain, exert wound healing, and nowadays, are utilized in human health therapeutics and Asian folk medicine. Due to higher pharmaceutical, cosmeceutical and nutraceutical values, sea cucumber has become one of the most essential products, and fetched higher market prices and therefore, provide a significant earning source and employment opportunity to the coastal communities in many countries. The sea cucumber and its product have been considered to be the excellent bioresource in the invertebrate fishery, whose harvestings, transporting, handlings, processing and maintenance are easy, convenient and cost-effective. Historically, bêche-de-mer were harvested from the multi-species capture fishery and stored for long times for selling to the businessmen and traders directly on the site. As like other lucrative and expensive food items viz., fish-maws, shark-fins and abalones, sea cucumbers are taken into the account as the high-valued products in the world seafood markets. Nevertheless, bêche-de-mer product supply in the South-East Asian countries has been decreasing day by day because of unstrained harvesting and overexploitation. On the other hand, the marketing structure in these countries is typically ineffective and the trading networks are multifaceted. Due to the lack of proper information, fishermen are not usually getting proper values and actual prices from the sea cucumber products, whereas other distributors and value-chain members have made complex systems, which have led to high cost of the products. The sea cucumber products will still have higher possibility of trading throughout the world markets, that might eventually contribute to the global economy in a very worthwhile, significant and sustainable manner.
