Evaluation the Prevalence of Toxoplasma Gondii between Local Cows, and its Effect to Renal and Liver Function in Salah Adeen

The study was conducted to evaluate the epidemiological of toxoplasma gondii in local cows in Salah Adeen, and its effect to renal and liver function. One hundred twenty six sample of serum were collected from cows n different cows farm in Salah Adeen, include Samarra (60 sample), Al-Mutasim (28 sample), Baled (23 sample) and Aldejil (15 sample), from the period between July 2019 until February 2020. The study include detection of serum anti T. gondii IgG antibody by using ELISA Toxoplasma IgG kit, and also determination of serum liver enzymes activity which include (Transaminases enzymes: aspartate transaminase-AST and alanine transaminase-ALT) and also the concentration of serum total protein-TP, albumin, globulin, urea and creatinine. The results indicate that the total percentage for the prevalence of T.gondii between cows in Saleh Eldean was 51.58%, distributed according to the region (68.33% in Samarra, 50% in Al-Mutasim, 26.08% in Baled and 26.66 in Aldejil). While the total percentage for infection with the parasite according to the gender of animals were 58.33% (28 from 48) in male animal and 47.43% (37from 78) in female animal. The results also showed that the level of TP, Albumin, globulin were significantly higher P≤0.05 in sera of infected animals, and also the activity of liver enzymes (AST and ALT) were significantly higher P≤0.05 in sera of infected animals with T.gondii with no significant difference in the level of urea and creatinine in sera of infected group as compared with non-infected group as control group. From all the above results we can conclude that the prevalence of T. gondii was high among animal under investigation, and this infection may be effect to the physiological function especially liver function.

Aims: The aim of this study was to investigate the toxic effect of Echium amoenum plants on the liver and kidney of animal model. Background: Echium amoenum is one of the medicinal plants containing pyrrolizidine alkaloids with several properties which has widely consumed among different communities. Objective: The toxic effects of Echium amoenum on the liver and kidney were investigated in this study. Methods: Sixty mice were kept for 28 days under the appropriate laboratory conditions. Echium amoenum extract (25, 12.5, 50 mg / kg, ip.) was administered for 28 days. At the end of experiment, blood samples were drawn and liver and kidneys were removed for evaluating hepatotoxicity and nephrotoxicity of extract. Additionally, experiments were conducted to assay the enzymatic and oxidative activities. Results: There was no significant difference in the levels of copper ion in the liver and kidneys among all groups. There was a significant difference in the levels of lipid peroxidation in the liver of treated groups versus control group. The significant difference was not observed in the levels of glutathione of the liver of all groups. However, the levels of glutathione of the kidney significantly decreased in the treated groups versus control group. There was no significant difference in the liver enzymes including ALP, SGOT, and SGPT between all groups. This indicates that damage increase with enhancing the time and concentrations of extract. Biochemical analysis showed the creatinine and urea levels did not change in the treated groups versus control group. Conclusion: According to the present findings, it is suggested that Echium amoenum causes hepatotoxicity and nephrotoxicity effects in dose and time dependent manner.

ABSTRACTObjective: As an antiobesity therapy, combination extracts of Detam 1 soybean and Jati Belanda will be consumed for a long time; therefore, theirtoxicities to the liver need to be investigated. To determine the effect of subchronic toxicity test of combination of ethanol extract of Detam 1 soybean(EEDS) and ethanol extract of Jati Belanda (EEJB) on liver function with parameters: Alanine transaminase (ALT), macroscopic, and histopathologicalof liver.Methods: This study was conducted on 120 Wistar rats (60 males and 60 females), 90 days (treatment group) and 120 days (satellite group). Ratswere divided into six treatment groups (3 test materials, 1 control, and 2 satellites); each group included 10 males and 10 females.Results: ALT levels of treatment groups (low dose, medium, and high), both males and females were lower than the control group (p<0.05). Thetreatment groups demonstrated a good effects effect on liver function. Liver weight of all groups showed no significant difference compared with thecontrol group (p>0.05). Results of histopathological score interpretation of male and female liver rats of low dose groups were not disturbed; middledose groups were slightly disturbed and high dose groups were damaged. Satellite high doses of male groups were disrupted, while female groupswere not.Conclusion: The combination of EEDS and EEJB has a good effect on liver function, did not lead to change organ weight and at low doses did not causerenal histopathology damage in rats after 90 days administration.Keywords: Combination of soybean Jati Belanda, Toxicity subchronic test, Function, Weight, Histopathology, Liver.

Introduction: Toxoplasmosis is a zoonosis that represents a serious public health problem, caused by Toxoplasma gondii, which affects 20-90% of the world human population [1,2]. It is a serious problem especially when considering the congenital transmission due to congenital sequels. Treatment with highly diluted substances is one of the alternative/complementary medicines most employed in the world [3,4]. The current ethical rules regarding the number of animals used in animal experimental protocols with the use of more conservative statistical methods [5] can not enhance the biological effects of highly diluted substances observed by the experience of the researcher. Aim: To evaluate the minimum number of animals per group to achieve a significant difference among the groups of animals treated with biotherapic T. gondii and infected with the protozoan regarding the number of cysts observed in the brain. Material and methods: A blind randomized controlled trial was performed using eleven Swiss male mice, aged 57 days, divided into two groups: BIOT-200DH - treated with biotherapic (n=6) and CONTROL - treated with hydroalcoholic solution 7% (n=7).The animals of the group BIOT-200DH were treated for 3 consecutive days in a single dose 0.1ml/dose/day. The animals of BIOT – 200DH group were orally infected with 20 cysts of ME49-T. gondii. The animals of the control group were treated with cereal alcohol 7% (n=7) for 3 consecutive days and then were infected with 20 cysts of ME49 -T. gondii orally. The biotherapic 200DH T. gondii was prepared with homogenized mouse brain, with 20 cysts of T. gondii / 100μL according to the Brazilian Homeopathic Pharmacopoeia [6] in laminar flow. After 60 days post-infection the animals were killed in a chamber saturated with halothane, the brains were homogenized and resuspended in 1 ml of saline solution. Cysts were counted in 25 ml of this suspension, covered with a 24x24 mm coverglass, examined in its full length. This study was approved by the Ethics Committee for animal experimentation of the UEM - Protocol 036/2009. The data were compared using the tests Mann Whitney and Bootstrap [7] with the statistical software BioStat 5.0. Results and discussion: There was no significant difference when analyzed with the Mann-Whitney, even multiplying the "n" ten times (p=0.0618). The number of cysts observed in BIOT 200DH group was 4.5 ± 3.3 and 12.8 ± 9.7 in the CONTROL group. Table 1 shows the results obtained using the bootstrap analysis for each data changed from 2n until 2n+5, and their respective p-values. With the inclusion of more elements in the different groups, tested one by one, randomly, increasing gradually the samples, we observed the sample size needed to statistically confirm the results seen experimentally. Using 17 mice in group BIOT 200DH and 19 in the CONTROL group we have already observed statistical significance. This result suggests that experiments involving highly diluted substances and infection of mice with T. gondii should work with experimental groups with 17 animals at least. Despite the current and relevant ethical discussions about the number of animals used for experimental procedures the number of animals involved in each experiment must meet the characteristics of each item to be studied. In the case of experiments involving highly diluted substances, experimental animal models are still rudimentary and the biological effects observed appear to be also individualized, as described in literature for homeopathy [8]. The fact that the statistical significance was achieved by increasing the sample observed in this trial, tell us about a rare event, with a strong individual behavior, difficult to demonstrate in a result set, treated simply with a comparison of means or medians. Conclusion: Bootstrap seems to be an interesting methodology for the analysis of data obtained from experiments with highly diluted substances. Experiments involving highly diluted substances and infection of mice with T. gondii should be better work with experimental groups using 17 animals at least.

Aims: Damage to liver tissue and its dysfunction is very important and if left untreated, it can cause serious problems and even death. In this study, we aimed to investigate the effects of the hydroalcoholic extracts of Linum usitatissimum and Rosa damascena on liver enzymes, total protein, bilirubin, albumin, and serum glucose levels. Materials and Methods: This is a non-randomized clinical trial conducted on 42 male rats divided into 6 groups; control group (group 1) received only sufficient water and food, groups 1 and 2 received 300 and 500 mg/ kgB.W Linum usitatissimum extract, groups 3 and 4 received 500 and 1000 mg/ kgB.W Rosa damascena, and group 6 received 100 mg/ kgB.W Linum usitatissimum plus 250 mg/ kgB.W Rosa damascena extracts intraperitoneally for 28 days. After the last injection, the rats were weighed and their blood samples were collected. The study parameters were measured using a colorimetric method by a spectrophotometer, and then were analyzed using ANOVA and Tukey’s test in SPSS V. 25 at a significance level of P<0.05. Findings: There was no significant difference between alanine aminotransferase, alkaline phosphatase, total and direct bilirubin levels in the control group in comparison with other groups (P>0.05). In the groups received Rosa damascena extract, there was a significant difference between total protein and albumin levels compared to the control group (P<0.05). Moreover, there was a significant difference between serum glucose and aspartate aminotransferase in the control group compared to other groups (P<0.05). Conclusion: Linum usitatissimum and Rosa damascena have no negative effect on the liver function. The probability of diarrhea occurrence and the possible effects on the total protein and serum albumin after using Rosa damascena, and the effects of different doses of Linum usitatissimum on the glucose levels should be taken into account.

AbstractThe objective of this randomized controlled trial (RCT) was to assess the impact of rifaximin on the course of liver function, liver regeneration and volumetric recovery in patients undergoing major hepatectomy. The ARROW trial was an investigator initiated, single-center, open-label, phase 3 RCT with two parallel treatment groups, conducted at our hepatobiliary center from 03/2016 to 07/2020. Patients undergoing major hepatectomy were eligible and randomly assigned 1:1 to receive oral rifaximin (550 mg twice daily for 7–10 or 14–21 days in case of portal vein embolization preoperatively and 7 days postoperatively) versus no intervention. Primary endpoint was the relative increase in postoperative liver function measured by LiMAx from postoperative day (POD) 4 to 7. Secondary endpoint were the course of liver function and liver volume during the study period as well as postoperative morbidity and mortality. Between 2016 and 2020, 45 patients were randomized and 35 patients (16 individuals in the rifaximin and 19 individuals in the control group) were eligible for per-protocol analysis. The study was prematurely terminated following interim analysis, due to the unlikelihood of reaching a significant primary endpoint. The median relative increase in liver function from POD 4 to POD 7 was 27% in the rifaximin group and 41% in the control group (p = 0.399). Further, no significant difference was found in terms of any other endpoints of functional liver- and volume regeneration or perioperative surgical complications following the application of rifaximin versus no intervention. Perioperative application of rifaximin has no effect on functional or volumetric regeneration after major hepatectomy (NCT02555293; EudraCT 2013-004644-28).

Abstract Objectives This study was designed to evaluate the role of anti-CD74 antibodies in diagnosis of axial spondyloarthritis (axSpA) and their relationship to disease duration and disease activity. Methods Fifty patients with axSpA, 15 patients with RA and 15 healthy subjects were included in the study. Clinical examination and laboratory tests were done. The ESR, CRP level and ASDAS were measured as markers of the disease activity. Quantitative determination of human CD74 IgG antibodies was done. Results The mean age of the patients was 38.22 (S.D.12.20) years. The level of CD74 autoantibodies was significantly higher in axSpA in comparison to control groups. Most patients with positive articular and extra-articular manifestations were positive for CD74 autoantibodies. In patients with inactive disease, 33.3% were positive for CD74 autoantibodies, as were 83% with active disease. High percentages of patients with early and late axSPA were CD74 autoantibody positive. The majority of patients with positive disease activity in early and late axSpA were CD74 autoantibody positive. CD74 autoantibodies had 80% sensitivity vs both control groups with 87% specificity vs the healthy control group and 80% vs the RA control group in the diagnosis of axSpA. Conclusions The frequency of positive anti-CD74 IgG antibodies was as high in patients with early axSpA as in those with late axSpA, with no significant differences. There was a significant difference in the frequency of positive anti-CD74 IgG antibodies between patients with positive and negative disease activity. Based on the sensitivity and specificity of anti-CD74 IgG, this is a promising diagnostic tool to support the clinical diagnosis of axSpA.

Background: Toxoplasmosis is a common parasitic disease, which leads to serious disease symptoms in immunocompromised hosts and infants. Recent epidemiologic surveys showed that microbial agents may be associated with some cases of multiple sclerosis (MS). Objectives: This case-control study aimed to examine the role of Toxoplasma gondii in MS by evaluating sero-frequency of anti-T. gondii IgG (ATXAb) antibody between patients with MS and ordinary people in Shiraz Province, south of Iran during 2016 - 2018. Methods: Serum samples obtained from MS patients (n = 130), and a group of age and gender-matched controls (n = 130) with the same socioeconomic status with the patients’ group were collected to evaluate the prevalence of T. gondii IgG antibodies (ATXAb). Moreover, the presence of the ATXAb antibody of the patients and controls was determined by the ELISA test. SPSS 20 software was used to perform the statistical analysis (SPSS, inc., Chicago, USA). Results: Out of 130 (35.4%) cases with MS and 130 (13.8%) controls, 46 and 18 subjects were seropositive for ATXAb, respectively. Based on the chi-square test, a significant difference was observed in terms of the positivity rates of ATXAb between the MS patient group and the control group (P = 0.001). The mean ± standard deviation ATXAb levels in controls and MS patients were found to be 61 ± 34 and 114 ± 47 IU/mL, respectively. Also, there were statistically significant differences between the levels of these two groups (P = 0.001). Conclusions: Given the relationship between toxoplasmosis and MS, it is possible that the prevalence of MS decreases by increasing hygiene and preventing toxoplasmosis.

Abstract: Central obesity is considered as a risk factor that strongly associated with several chronic diseases. Men with waist circumference ≥ 90 cm and women with waist circumference ≥80 cm are stated as central obesity. This study aimed to determine the effect of brisk walking on liver function in central obesity. This was an experimental field study with a non-randomized pre-post one group test and control group. This study used consecutive sampling method. There were 30 samples divided into two groups; 15 samples were given intervention and 15 samples as the control group. Data were analyzed with Wilcoxon signed ranks test.   The results showed that brisk walking for 1 month decreased the SGPT and SGOT levels, however, statistically there was no significant difference before and after intervention with a p value >0.05. Conclusion: There was no significant influence of brisk walking on liver function in central obesity. Penelitian ini bertujuan untuk mengetahui pengaruh brisk walking (jalan cepat) terhadap fungsi hati pada obesitas sentral.  Metode penelitian ini yaitu eksperimental lapangan dengan rancangan non – randomized pre-post test with control. Sampel ditentukan dengan teknik pengambilan sampel konsekutif sampling. Terdapat 30 sampel yang dibagi menjadi 2 kelompok yakni 15 sampel yang diberi perlakuan dan 15 sampel sebagai kontrol. Data dianalisis dengan SPSS 20 dan uji Wilcoxon Signed Ranks Test. Hasil penelitian memperlihatkan terjadi penurunan kadar enzim hati SGPT dan SGOT pada sampel yang melakukan brisk walking selama 1 bulan tetapi secara statistik tidak ada perbedaan bermakna sebelum dan sesudah mendapatkan perlakuan dengan nilai p > 0,05.  Simpulan: Tidak terdapat pengaruh bermakna dari brisk walking (jalan cepat) terhadap fungsi hati pada obesitas sentral. Aim: To evaluate the anti-oxidant enzymes, lipid peroxidation, lipid profile and liver function in albino rats orally administered tartrazine. Study Design: A total number of 63 female albino rats weighing approximately 0.2 kg were used for this study. The study was divided into two phases, phase 1 which lasted for the first 30 days, comprised of 35 rats, 20 rats were used as test group while 15 rats served as the control group. Phase 2 of the study was for 60 days and 28 rats were used with 16 as test group and 12 as the control. The test groups were orally administered with 7.5 mg/kg of tartrazine (ADI) daily over the specified periods while the control groups were not treated with tartrazine but given only food and water. Place and Duration of Study: The study was carried out in the Department of Medical Laboratory Science, Rivers State University, Port Harcourt, Nigeria within a period of 12 months (Feb., 2019 – Jan., 2020). Methodology: At the end of the study, 5 mls of whole blood specimens was collected by means of cardiac puncture into plain bottles. To obtain the serum, the whole blood samples were allowed to clot and later dislodged and spun at 3500 rpm for 10 minutes. The collected serum specimens were used to analyze SOD, MDA, GPX, ALT, GGT, ALP, TG, TCHOL, and HDL-C, while LDL-C was calculated using Friedwald equation. Results: The chronic treatment of rats with tartrazine azo food dye at the ADI dose caused an increase in MDA levels after 30 and 60 days test rats compared to the control, while TCHOL and HDL-C showed significant decrease after 30 and 60 days of treatment in the test group compared to the control group. In addition, ALT indicated significant increase in test group after 60 days of treatment compared to control group. ALP, GGT, TG, LDL-C, SOD and GPX showed no significant difference after 30, and 60 days of treatment at ADI doses. Histologic examination of the liver indicated hydropic dilation, degenerating hepatocyes and infiltration of central vein with parenchymal materials alongside kupffer cells. Conclusion: The results from this study revealed that orally administered tartrazine at the recommended ADI dose increased lipid peroxidation as seen in the elevated MDA levels. Hepatic derangements were also seen as revealed by increased ALT and histologic distortions as well fall in TCHOL and HDL-C lipid fractions.

This experiment was premeditated to probe the toxic impact of Silver Nanoparticles (AgNPs) upon liver function parameters in male rats, as well as to demonstrate the protective effect of Tinospora cordifolia (T.C) against liver function disorders caused by AgNPs. Twenty-four rats were classified into four packs, six ones for every pack. Rats were included by control animals without treatment, while the AgNPs group included rats treated with 50μl/kg / day AgNPs. In the AgNPs + T.C group, rats were co-administered of AgNPs at a dose of 50μl/kg with T.C at a dose of 400 mg/kg. Whereas, animals of the T.C pack were treated with 400 mg/kg of T.C. The duration of the experiment was 28 days. After blood samples were taken at the conclusion of the experiment. The levels of biochemical parameters (liver enzymes) such as alanine aminotransferase, aspartate aminotransferase, and alkaline phosphatase were evaluated to detect liver function abnormalities. Thwe Pw A t e dewoe t ste user e dnoAiwoe se dsnd disnte dnirease in the activity of serum liver enzymes in comparison to the control group (p < 0.05).While T. cordifolia co-treatment significantly reduced the levels of activities of those enzymes. Therefore, it can be considered that T. cordifolia effectively contributes to the protection against liver dysfunction caused by AgNPs in rats.
