The possibility of using various methods for determining the viability of cultures of microalgae Dunaliella salina and Chlorococcum dissectum before and after freezing-warming was investigated and analyzed. It has been established that the selection of an effective method should be carried out individually for each culture. For an integral assessment of the proliferative and metabolic activity of cells of both species of the studied microalgae, Alamar Blue-test and the ability to grow on liquid nutrient media can be used. The use of the Koch plate method, MTT-test and TTC staining is possible only for the microalga C. dissectum. Vital staining with trypan blue was found to be incorrect.

AbstractWe report the development of a lab-on-a-chip system, that facilitates coupled dielectrophoretic detection (DEP-D) and impedimetric counting (IM-C), for investigating drug resistance in K562 and CCRF-CEM leukemia cells without (immuno) labeling. Two IM-C units were placed upstream and downstream of the DEP-D unit for enumeration, respectively, before and after the cells were treated in DEP-D unit, where the difference in cell count gave the total number of trapped cells based on their DEP characteristics. Conductivity of the running buffer was matched the conductivity of cytoplasm of wild type K562 and CCRF-CEM cells. Results showed that DEP responses of drug resistant and wild type K562 cells were statistically discriminative (at p = 0.05 level) at 200 mS/m buffer conductivity and at 8.6 MHz working frequency of DEP-D unit. For CCRF-CEM cells, conductivity and frequency values were 160 mS/m and 6.2 MHz, respectively. Our approach enabled discrimination of resistant cells in a group by setting up a threshold provided by the conductivity of running buffer. Subsequent selection of drug resistant cells can be applied to investigate variations in gene expressions and occurrence of mutations related to drug resistance.

Abstract Twelve hyper-β carotene-producing strains of algae assigned to the genus Dunaliella salina have been isolated from various hypersaline environments in Israel, South Africa, Namibia and Spain. Intron-sizing of the SSU rDNA and phylogenetic analysis of these isolates were undertaken using four commonly employed markers for genotyping, LSU rDNA, ITS, rbcL and tufA and their application to the study of Dunaliella evaluated. Novel isolates have been identified and phylogenetic analyses have shown the need for clarification on the taxonomy of Dunaliella salina. We propose the division of D. salina into four sub-clades as defined by a robust phylogeny based on the concatenation of four genes. This study further demonstrates the considerable genetic diversity within D. salina and the potential of genetic analyses for aiding in the selection of prospective economically important strains.

Abstract The exploration and transportation of fluids from corrosive fields or reserves demand corrosion-resistant pipelines. The pipelines for sour-service application demand higher resistance to H2S corrosion because of higher sulphur content observed in the geographical locations. The pipelines laid in the corrosive and marshy lands demand high wall thickness to allow for wall thinning due to corrosion. The linepipes manufactured using TMCP plates are widely accepted for sour-service applications. The use of cut-to-length sheets from hot rolled coils for the manufacturing of LSAW pipes is economical but having limited acceptance by the pipeline operators. Welspun took an initiative to develop the linepipes for an onshore sour-service application using API 5L X60MS grade cut-to-length sheets from TMCP hot rolled coils procured from an approved steel mill. The LSAW pipes of 24” OD × 14.27 mm WT were manufactured by the JCO-E press at Welspun Pipes Mill in Anjar-Gujarat-India. The experience of 24”OD × 14.30 mm WT, API 5L X60MO grade pipes manufacturing using TMCP plates procured from another approved steel mill, was used to decide factors such as alloy design, cleanliness, strength, DWTT, hardness, HIC, SSCC, CVN for the selection of cut-to-length sheets. This paper deals with sheet-to-pipe behavioural changes in properties before and after cold mechanical expansion. Wherever possible, the comparison was also made with plate-pipe behavioural changes in properties.

Effect of  role playing macro methods on children's speaking ability in group B in Nurul Rohmah Bekasi. This research is motivated by the lack of ability to speak children. This is because the method used in the learning process is more likely to use conventional methods. The selection of methods to role playing macro so that children can be motivated and interested in learning and can stimulate the ability to speak children. The population in this study were all children of group B in kindergarten Nurul Rohmah while the study sample was B3 group 11 children for the experimental group and B1 group 11 children for the control group. Data collection techniques through test techniques, and observation. Data analysis techniques used were descriptive statistical analysis and nonparametric statistical analysis. Based on the results of the Wilcoxon test calculation, the calculated T value is 66 and T table 11 then the results obtained T count (66)> T table (11) H1 is accepted and Ho is rejected means that there is an effect of role playing macro  methods on children's speaking ability. Whereas the calculated Z value obtained is 2.93 and 1.645 table Z then the result of Z arithmetic (2.93)> Z table (1.645) H1 is accepted and Ho is rejected which means that there is an effect of role playing macro method on children's speaking ability. These results indicate that there is a change in the value of the child's speaking ability before and after getting learning based on the role playing macro method.

This paper presents the results of an intervention study exploring the effectiveness of an implementation of a selection of digital learning resources (DLR) in lower secondary science classrooms. Eight teachers participated in a quasi-experimental intervention, teaching three different subjects with and without DLRs. The data presented are from pupil tests before and after the teaching combined with observation of lessons and interviews with both pupils and teachers. In two of the three subjects the groups using DLRs did slightly better than the comparison group. The results indicate that successful ICT based learning is just as dependent on teacher competence as the quality and features of the resources.

Skin color is an important economic trait in meat-type chickens. A uniform bright skin color can increase the sales value of chicken. Chickens with bright yellow skin are more popular in China, especially in the broiler market of South China. However, the skin color of chickens can vary because of differences in breeds, diet, health, and individual genetics. To obtain greater insight into the genetic factors associated with the process of skin pigmentation in chickens, we used a colorimeter and high-resolution skin photographs to measure and analyze the skin color of chickens. By analyzing 534 chickens of the same breed, age, and feed condition, we found that the yellowness values of the chickens varied within this population. A significant positive correlation was found between the cloacal skin yellowness values before and after slaughter, and the cloacal skin yellowness value of live chickens was positively correlated with the overall body skin yellowness value. Additionally, chicken skin yellowness exhibited low heritability, ranging from 0.07 to 0.27. Through RNA sequencing, 882 genes were found to be differentially expressed between the skin with the highest and lowest yellowness values. Some of these differentially expressed genes may play an important role in yellow pigment deposition in chicken skin, which included TLR2B, IYD, SMOC1, ALDH1A3, CYP11A1, FHL2, TECRL, ACACB, TYR, PMEL, and GPR143. In addition, we found that the expression and variations of the BCO2 gene, which is referred to as the yellow skin gene, cannot be used to estimate the skin yellowness value of chickens in this population. These data will help to further our understanding of chicken skin yellowness and might contribute to the selection of specific chicken strains with consistent skin coloration.

This paper estimates the impact of the capture of leaders of criminal organizations on the labor market in municipalities where these organizations operated between 2004 and 2006. The difference-in-difference analysis compares different employment outcomes in cartel locations and the rest, before and after the capture of cartel leaders. The results show that captures caused a decrease in nominal wages and paid employment in cartel municipalities. Using Economic Census Data, I find that captures also caused a fall in the number of establishments and had a negative impact on other establishment outcomes. This document focuses exclusively on the impact of the capture of leaders of criminal organizations on the labor market until 2011 without studying other possible consequences, and thus does not make an integral assessment of this policy

This research aims to describe the level of competence of farmers in composting, and to draft extension and try out the implementation of counseling about composting in group of livestock of Mekar. The research was conducted in sub-district Dagangan of Madiun Regency. Selection of livestock group is done by purposive that is the only group of livestock that have made compost that is group of livestock of Mekar which is located in Kepet village. The research method used is case study with quantitative descriptive approach with scoring. The population is 44 people, the sample is based on the census. The results showed that: 1) the level of competence of farmers in composting is classified with the details of the competence of the preparation stage is low, the competence of the manufacturing process is low, and the competence of the storage stage is classified; and 2) drafting the extension plan based on the target and the purpose of extension so that the breeder know, understand and able to increase the C-Organic content and C / N Ratio on the compost according to the standard. Success shows the improvement of compost quality before and after counseling in accordance with the purpose of counseling.   Keywords: competence of breeder, livestock group of Mekar, draft extension, composting, extension

This article is devoted to issues of interaction of stakeholders of construction enterprises. First of all, the peculiarities of the construction industry are defined, which form additional requirements for the development of an integrated system for assessing interaction with stakeholders. Second, approaches to the formation of a register of potential stakeholders have been identified. A template for analysis has been developed, and individual examples are grouped by interaction directions. The methodology of identifying potential stakeholders by means of a three-level process of applying different versions of expert assessment methods in order to minimize their negative factor – subjectivity, is proposed. Methods of direct selection of experts and algorithms of their assessment are defined. Proposals to rank stakeholders according to their priority and potential result from their interaction with the construction enterprise have been developed. According to the integral assessment, ways of controlling the processes of interaction with stakeholders of different ranking levels are proposed, recommendations for building a further strategy for managing these processes are given. A system of modeling interaction between construction enterprises and stakeholders to ensure planning, organization, monitoring, and control processes in optimistic, realistic, and pessimistic scenarios has been created. Recommendations for further use of the methodology of integral assessment of the interaction of stakeholders of construction enterprises in practice are given.
