 Paphiopedilum javanicum (Reinw. ex Lindl.) Pfitzer known as slipper orchid is an endemic terrestrial orchid from Indonesia. This orchid is threatened with extinction and listed under CITES Appendix I. In-vitro propagation of P. javanicum has been done in Bali Botanic Garden (BBG) for last six years but acclimatization stage has not given satisfied result. The purpose of this study was to know the ef-fect of liquid fertilizer on vegetative growth of P. javanicum planlet during acclimatization stage. Beyonic StarTmik liquid fertilizer ap-plied by five different doses considered as treatment of 0, 10, 20, 30 and 40 mL. They were 10 planlets for each treatment considered as replication. In this research we observe vegetative growth of P. ja-vanicum by quantitative and qualitative parameter. Quantitative data were analysis by ANOVA. Result showed, high percentage of planlet survival was observed of 98% and liquid fertilizer dose for 30 mL gave the optimal vegetative growth of P. javanicum planlet with aver-age height of 2.2 cm and leaves number of 13 pieces. There were no differences on qualitative parameter in all treatment that all leaves color was dark green and leaves tessellation appearance was obvious

SummaryThe APTT has been considered the most suitable candidate to monitor the anticoagulant activity of hirudin. However, its use is hampered by problems of standardization, which make the results heavily dependent on the responsiveness of the reagent used. Our aim was to investigate if this different responsiveness of different reagents when added in vitro is to be confirmed in an ex vivo study.Two different doses of r-hirudin (CGP 39393), 0.3 mg/kg and 1 mg/kg, were administered subcutaneously to 20 New Zealand male rabbits, and the differences in prolongation of APTT 2 and 12 h later were compared, using 8 widely used commercial reagents. All groups exhibited a significant prolongation of APTT 2 h after sc administration of hirudin, both at low and high doses. But this prolongation persisted 12 h later only when the PTTa reagent (Boehringer Mannheim) was used. In general, hirudin prolonged the APTT most with the silica- based reagents.In a further study, we compared the same APTT reagents in an in vitro study in which normal pooled plasma was mixed with increasing amount of hirudin. We failed to confirm a higher sensitivity for silica- containing reagents. Thus, we conclude that subcutaneous administration of hirudin prolongs the APTT most with the silica-based reagents, but this effect is exclusive for the ex vivo model.

A field experiment was conducted at the research field of Sher-e- Bangla Agricultural University, Dhaka during the period from December 1, 2016 to January 11, 2017 to investigate the effect of gibberellic acid (GA3) on vegetative growth, morphological attributes and foliage (leaf) yield of coriander (Coriandrum sativum L.). The experimental treatments consisted of eight different doses of GA3 viz., 0 (distilled water spray), 5, 10, 15, 20, 25, 30 and 35 ppm. The variety Rosina (Pahuja Co., India) was used as planting material and different doses of GA3 were applied on the plants at 25 and 30 days after sowing. The experiment was laid out in Randomized Complete Block Design with three replications. GA3 had significant effect on vegetative growth, morphology, foliage yield and yield contributing characters of coriander. Plant height, number of leaves plant-1, leaf length, plant spread and single plant weight were found maximum from the application of GA3 at 20 ppm, which was closely followed by 25 ppm GA3. Application of GA3 at 20 ppm produced the highest weight of foliage m-2. A strong positive correlation of foliage yield was observed with plant height, number of leaves plant-1, leaf length, plant spread, single plant weight and weight of foliage m-2. Application of GA3 at 20 ppm gave maximum foliage yield (9.34 t ha-1) which was followed by GA3 at 15 ppm (8.46 t ha-1) and 25 ppm (8.06 t ha-1), and the minimum foliage yield was recorded from control (distilled water spray) (4.90 t ha-1). Application of GA3 at 20 ppm increased foliage yield over control by 47.54 %. A quadratic relationship between applied GA3 concentration and foliage yield was found; the regression equation was y = 4.87 + 0.375x – 0.009x2 from which it came up to be optimum dose of GA3 as 20.83 ppm.Res. Female catkin primordia of black cottonwood (Populus trichocarpa T. & G. ex Hook.) were cultured for 70 days on a modified Murashige and Skoog's (1962) medium in vitro. Explants 2–3 mm long, and with bud scales removed, gave the best results, many of them developing floral structures characteristic of the female sex. There was a general tendency to callus formation with increasing age of the culture, occasionally followed by a reversal to vegetative growth. Catkin primordia raised on Wolter's medium without auxin or kinetin, but with 6-benzylaminopurine, and at 250 ft-c for a 16-h photoperiod, proliferated axillary shoots in loco of pistils.

Acrylamide (AA), a widely used industrial chemical, is shown to be neurotoxic, mutagenic and carcinogenic. This study was carried out to investigate the effects of different doses of AA on lipid peroxidation (LPO), haemolysis, methaemoglobin (MetHb) and antioxidant system in human erythrocytes in vitro. Erythrocyte solutions were incubated with 0.10, 0.25, 0.50 and 1.00 mM of AA at 37°C for 1 hour. At the end of the incubation, malondialdehyde (MDA), an end product of LPO, was determined by liquid chromatography (LC) while total glutathione, reduced glutathione (GSH) levels, activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px) enzymes and the rates of haemolysis and MetHb were determined by spectrophotometric methods. All of the studied concentrations of AA increased MetHb formation and SOD activity, and induced MDA formation and haemolysis due to the destruction of erythrocyte cell membrane. AA caused a decrease in the activities of GSH-Px, CAT and GSH levels. However, these effects of AA were seen only at higher concentrations than AA intake estimated for populations in many countries. We suggest that LPO process may not be involved in the toxic effects of AA in low concentrations, although the present results showed that the studied concentrations of AA exert deteriorating effects on antioxidant enzyme activities, LPO process and haemolysis.

Various regimens of treadmill exercise (0% slope) were used with rats: 60 min at 15 m/min (T-15), 180 min at 10 m/min (T-10), and 60 min/day at 15 m/min for 6 consecutive days (T-15-6). Exercise resulted in 1) decreases in the absolute number of mononuclear spleen cells in T-10 rats, 2) significant increases in in vitro splenic T-cell blastogenesis in response to phytohemagglutinin in T-10 rats, and 3) significant decreases in T-cell blastogenesis in T-15-6 rats. T-15-6 rats were given aminoglutethimide per os before exercise sessions to study the role of corticosteroids in the alteration of splenic T-cell blastogenesis. Aminoglutethimide significantly increased the T-cell blastogenesis in these T-15-6 rats compared with those not given aminoglutethimide, whereas it had no effect on immune parameters of sedentary rats. These results show that immunomodulations in the rat depend on the treadmill exercise regimen employed. If the mechanisms of the immunomodulation induced by isolated exercise of long duration are not elucidated, these data suggest that corticosteroids are involved in the alteration in T-cell blastogenesis induced by chronic muscular exercise.

Microbial control in integrated pest management (IPM) programs of coffee plantations is an important factor for the reduction of pest population densities. The use of selective pesticides can be associated with entomopathogens, increasing the efficiency of the control and reducing the use of required insecticides. The in vitro fungitoxic effect of insecticide formulations of Thiamethoxam, Cyfluthrin, Deltamethrin, Alpha-Cypermethrin, Triazophos, Chlorpyrifos, Fenpropathrin and Endosulfan and Beauveria bassiana (CG 425 strain) was evaluated at three concentrations (FR = average field recommendation; 0.5 ´ FR and 2 ´ FR). Effects of these products on conidia germination, vegetative growth and sporulation were compared. Only five insecticides, at the FR concentration, promoted conidia viability higher than 60%. Viability should be considered the most important factor to be evaluated since it is the first step of the infection process. The insecticide formulations of Alpha-Cypermethrin, Thiamethoxam and Cyfluthrin caused the lower inhibition level on conidia germination at the two lower concentrations, with no difference in relation to the control. With respect to vegetative growth analysis, Thiamethoxam at the two lower concentrations was not found to cause radial growth inhibition. Thiamethoxam caused the smallest inhibition level with regard to conidia production. The use of Alpha-Cypermethrin and Thiamethoxam formulations in coffee IPM programs for a B. bassiana inoculum conservation strategy are recommended, since these products were compatible with the entomopathogenic fungus Beauveria bassiana (CG 425), an important natural control agent of the coffee berry borer, Hypothenemus hampei.

The use of plant growth regulators is directly related to the success of in vitro propagation, which is an advantageous alternative to obtain seedlings on a commercial scale. This study aimed to evaluate the in vitro development of ‘IAC 95-5000’ sugarcane seedlings after the addition of different doses of ethephon (0, 25, 50, 100 and 200 mg L-1) or gibberellic acid (0, 2.5, 5.0, 7.5 and 10.0 mg L-1) to the culture medium. Ethephon increased the number of tillers (up to 231.70%), reduced height of the main tiller (44.66 to 60.47%), and did not affect the shoot´s fresh and dry mass. On the other hand, gibberellin decreased the number of tillers and negatively changed biomass partitioning. It is concluded that the use of ethephon is a potential strategy to enhance in vitro production of ‘IAC 95-5000’ sugarcane seedlings, since it increased the number of usable shoots in subsequent subcultures, and its effects on height reduction can be reversible. However, the use of the tested doses of gibberellic acid is not recommended, because it impaired seedling development of this sugarcane variety.

Background. Cilengitide is a selective αvβ3 and αvβ5 integrin inhibitor. We sought to investigate the effect of cilengitide on the neovascularization of abdominal aortic plaques in rabbits and explore its underlying antiangiogenic mechanism on human umbilical vein endothelial cells (HUVECs). Materials and Methods. For the in vivo experiment, the abdominal aortic plaque model of rabbits was established and injected with different doses of cilengitide or saline for 14 consecutive days. Conventional ultrasound (CUS) and contrast-enhanced ultrasound (CEUS) were applied to measure the vascular structure and blood flow parameters. CD31 immunofluorescence staining was performed for examining neovascularization. Relative expressions of vascular endothelial growth factor (VEGF) and integrin of the plaque were determined. For in vitro experiments, HUVECs were tested for proliferation, migration, apoptosis, and tube formation in the presence of different doses of cilengitide. Relative expressions of VEGF, integrin, and Ras/ERK/AKT signaling pathways were determined for the exploration of underlying mechanism. Results. CEUS showed modestly increased size and eccentricity index (EI) of plaques in the control group. Different degrees of reduced size and EI of plaques were observed in two cilengitide treatment groups. The expressions of VEGF and integrin in the plaque were inhibited after 14 days of cilengitide treatment. The neovascularization and apoptosis of the abdominal aorta were also significantly alleviated by cilengitide treatment. For in vitro experiments, cilengitide treatment was found to inhibit the proliferation, migration, and tube formation of HUVECs. However, cilengitide did not induce the apoptosis of HUVECs. A higher dose of cilengitide inhibited the mRNA expression of VEGF-A, β3, and β5, but not αV. Lastly, cilengitide treatment significantly inhibited the Ras/ERK/AKT pathway in the HUVECs. Conclusions. This study showed that cilengitide effectively inhibited the growth of plaque size by inhibiting the angiogenesis of the abdominal aortic plaques and blocking the VEGF-mediated angiogenic effect on HUVECs.

The purpose of the present work was to evaluate the vegetative growth of Mentha piperita L. cultivated under different water availability, as well its influence in content, chemical composition and in vitro antioxidant activity of its essential oil. Plants were propagated by mother plants microcutting and scions were transplanted to 5 L pots with soil and cattle manure. Afterward, were kept at field capacity for 30 days and under treatment for 40 days. It was treated with different levels of water deficit treatments: (T1): 100 of field capacity (FC); (T2): 80 of FC; (T3): 60 of FC; (T4) 40 of FC with 5 blocks. Vegetative growth was evaluated by dry matter contents of all part of plants and by root/aerial rate. The essential oil of the leaves was extracted by hydrodistillation, analyzed by GC-FID and GC-MS and in vitro antioxidant potential was evaluated. A significant decrease in the dry matter of leaves and stems accompanied with a decrease in the roots dry matter was observed with an increase in the water stress. Quantitative chemical differences were observed in the chemical composition of the essential oil, according water availability. Total antioxidant activity showed a gradual increase as water stress progressed.

Raspberry (Rubus idaeus L.) cultivar Willamette has long been the most commonly grown raspberry cultivar in Serbia, which is owing to high adaptability of the cultivar to respective agro-environmental conditions. Massive dieback of full bearing plantings is a major problem in raspberry growing hence quality planting material is a must when establishing new raspberry plantings. The study was conducted under protected conditions (in screenhouse) on plants obtained by micropropagation in vitro. In order to achieve optimal vegetative potential, plants were grown for two consecutive years (2004–2005) on two substrates (Steckmedium and Seedling) using three foliar fertilizers (Wuxal, Murtonik and Ferticare). The study revealed optimal vegetative growth in plants studied, excess manganese (150.60-214.52 mg/g), optimum iron content (94.00-123.50 mg/g), and zinc (28.60-31.00 mg/g) and copper (3.10-4.00 mg/g) deficiencies, based on the referent values of microelements content. The assessment of nutritional status of plants by the DOP index suggested significant differences in microelements imbalance when different foliar fertilizers and substrates are applied.
