The current research was planned to measure the comparative phytochemical and anti-oxidative potential of aqueous and methanolic extracts of two indigenously grown millet varieties of Pakistan. The locally available millet varieties, i.e. MB-87 and AF-POP flours were chemically characterized through the proximate and mineral analysis. The antioxidant extract was prepared in water and methanol. The extracts were screened for saponins, terpenoids, flavonoids and tannins; methanolic extract of MB-87 and AF-POP showed flavonoids in an average quantity (++), whilst aqueous extract had only trace (+) values. The phytochemical screening showed the presence of saponins only in the aqueous extract of both varieties. However, terpenoids and tannins were present in both methanolic and aqueous extracts. During antioxidant evaluation, millet methanolic extracts showed higher level of TPC and DPPH of MB-87 and AF-POP as 0.30±0.001 & 0.23±0.0012 mg GAE/g and 48±0.96 &46±1.09%, respectively. However, the β-carotene activity of the aqueous extract of both varieties (MB-87 and AF-POP) was recorded higher. In a nutshell, the methanolic extract of MB-87 has an appreciable atioxidant profile. Furher research should be planned to screen the important constituents of Pakistani millet varieties.

<p class="Abstract">The present study is aimed to investigate the phytochemical screening and biological activities of methanolic extract of <em>Cyperus scariosus</em> roots. Dried plant was grounded and extracted with methanol to prepare methanol crud extract. In vitro biological tests were conducted using this methanolic extracts according to the standard procedure. 100% death rate of brine shrimp was perceived at 3 mg/mL of plant extract after 72 hours. The extract showed action against <em>Aspergillus flavius</em> i.e. 90% followed by A. niger (91%) while the highest activity was shown against <em>A. fumegatrus</em> (94%). Important scavenging results were detected during scavenging of free radicals viz; 92.2% against DPPH, 82.2% to ABTS, 75.8% to hydrogen peroxide, 88.1% to β-carotene, 86.1% to hydroxyl radical and 89.4% against phosphomolybdate at 3 mg/mL were obtained. The results obtained in this study point out that extract showed significant biological activities which might be due to the presence of bioactive constituents.</p><p> </p>

Objectives: The study was designed to evaluate the hypoglycaemic effects ofCassia sophera’s powdered seeds and its different fractions. Study Design: An experimentalstudy. Setting: Faculty of Veterinary, University of Agriculture, Faisalabad, Pakistan. Period:November 2014 to February 2017. Methodology: Initial experiments were designed to find outthe hypoglycaemic effects of Cassia sophera powdered seeds in increasing doses i.e. 2, 3, 4g/kg body weight at 0, 2, 4, 8, 12 and 24 hours intervals. The effects of methanolic and aqueousextracts equivalent to 4g/kg body weight of Cassia sophera seeds were also studied in bothnormal and diabetic rabbits. Results: At 4 hours interval, 2grams/kg body weight of grindedseeds results no significant decrease in mean blood level of glucose. At 8 and 12 hours intervals,a significant decrease while at 4, 8 and 12 hours a highly significant reduction were observedwith both 3g/kg and 4g/kg body weight. With methanolic and aqueous extracts equivalent to4g/kg body weight, normal rabbits showed significant decrease i.e. with P value less than 0.05at 8 and 12 hours. In addition, the methanolic extract of Cassia sophera seeds produced betterhypoglycaemia as compared to aqueous extract. The effects of these extracts were comparableto Acetohexamide. The same study was also done in alloxan-induced diabetic rabbits and it wasnoticed that Cassia sophera powdered seeds caused significant decrease in blood glucoselevel at increasing doses 2, 3 and 4gram/kg of body weight and highly significant reduction(with P value less than 0.001) in blood level of glucose was noticed with methanolic extracts at4 hours interval which continued upto 8 hours while treatment with aqueous extract with Cassiasophera seeds showed highly significant decrease at 12 hours. Conclusion: It is concludedthat powdered seeds of Cassia sophera constitutes more than one type of hypoglycaemicelements, which have exerted a significant and consistent hypoglycaemic effects in normal andalloxan treated diabetic rabbits.

Ayurvedic medicines are being used increasingly to fight or prevent common diseases. Udhara vayuhara churna is a polyherbal formulation containing Piper longum (Piperceae) Zingiber officinalis (Zingiberaceae), Elettaria cardamom (Zingiberaceae), Plumbago zeylanica (Plumbaginaceae), Carum carvi (Umbelliferae), Acorus calamus (Araceae) and Embilica officinalis (Euphorbiaceae). Methanolic and aqueous extracts of Udhara vayuhara churna were screened for their antimicrobial activity against Bacillus subtilis, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Aspergillus niger and Candida albicans. The results of antimicrobial activity of aqueous and methanolic extracts of the Udhara vayuhara churna indicated that methanolic extract inhibited the growth of one or more test pathogens than aqueous extract. Udhara vayuhara churna formulation extracts showed a broad spectrum of antimicrobial activity. Phytochemical investigation revealed the presence of tannins, saponins, alkaloids, glycosides, flavonoids and essential oils. This may be due to the multifunctional effect of all the seven plant ingredients of Udhara Vayuhara churn.

OBJECTIVES: To evaluate the Immunomodulatory, apoptosis induction and antitumor effects of aqueous and methanolic extracts of Calvatia craniiformis regarding the size of tumour mass, caspase-8 expression and apoptotic index (AI%) in mice transfected with murine hepatocellular carcinoma cell line (H22) as an experimental therapeutic system for human hepatocellular carcinoma.MATERIAL AND METHODS: Forty-eight Balb/C albino mice were transfected in legs with H22 cells. Tumour size was measured twice a week. Caspase-8 protein expression and apoptotic index determination evaluated by Immunohistochemistry.RESULTS: Tumor size significantly differed between the two groups of mice transfected with H22 cells; the first was treated with C. craniiformis aqueous extract (0.3, 0.6, 1.2) mg/kg and the second group was treated with C. craniiformis methanolic extract (0.25, 0.5, 1.0) mg/kg compared with control group. The inhibitory activity of aqueous and methanolic extracts was dose and duration dependent. The size of the tumour mass was reduced up to 87.9% when treated with 1.2 mg/kg aqueous extract and 1 mg/kg for methanolic extract. Caspase-8 expression was increased in a dose-dependent manner among H22 bearing mice treated with C. craniiformis aqueous extract (0.3, 0.6, 1.2) mg/kg. At 0.3 mg/kg, the intensity of expression was strong in (33.33%) and very strong in (66.67%). While at 0.6 mg/kg and 1.2 mg/kg the intensity of expression was strong in (33.33%) and very strong in (100%) with a significant difference (P ≤ 0.001). H22 bearing mice treated with (0.25, 0.5, 1.0) mg/kg C. craniiformis methanolic extract shows increased caspase-8 expression in a dose-dependent manner. At 0.25 mg/kg, the intensity of expression was strong in (33.33%) and very strong in (66.67%). While at 0.5 mg/kg, the intensity of expression was strong in (33.33%) and very strong in (100%). At 1.0 mg/kg, the intensity of expression was strong in (16.67%) and very strong in (83.33%) with significant difference (P ≤ 0.001). AI% of H22 bearing mice treated with C. craniiformis aqueous and methanolic extracts were significantly increased (P ≤ 0.05) compared with the untreated control group. No significant difference was reported in AI% between aqueous and methanolic extracts treated groups.CONCLUSIONS: Extracts of C. craniiformis were highly efficient in tumour growth inhibition, causing a reduction in the tumour size clinically and increase the expression of caspase-8 gene product in tumour tissue, causing increase apoptotic index of H22 cells taken from the legs of inoculated mice leading to loss of legs due to bone necrosis. Antitumor activity of C. craniiformis aqueous, and the methanolic extract was dose and duration dependent.

Buchholzia coriacea (wonderful kola) is a medicinal plant that has been used worldwide as an alternative medication to promote human health. Comparative nephroprotective effects of crude seed powder, aqueous and methanolic extracts of B. coriacea in hepatotoxic rats was investigated for 56 days. The crude powder (BCCP), aqueous extract (BCAE) and methanolic extract (BCME) significantly reduced (p<0.05) the levels of creatinine and uric acid and decreased minimally throughout the treatment periods. Total protein increased significantly (p<0.05). Significant decrease (p<0.05) were observed with 200 mg/kg BCAE and BCCP respectively in urea and BUN levels. Noticeable nephroprotective effects may be attributed to the presence of phytochemicals like flavonoids and tannins which act as antioxidants. This study has demonstrated that B. coriacea crude seed powder, methanolic and aqueous extracts caused no adverse effect on the rat kidney and may be recommended for the management of nephrotoxicity Dhaka Univ. The study was designed to evaluate the phytoconstituents present in the methanolic extract of aerial parts of Abelmoschus moschatus. The qualitative phytochemical screening of different extracts of aerial parts revealed the presence of some bioactive compounds. GC – MS analysis was performed using Shimadzu Gas chromatography-mass spectroscopy (Model Number: QP2010S) instrument. GC-MS detection of phytoconstituents was done by computer evaluation of mass spectra of samples through National Institute Standard and Technology (NIST II) and WILEY 8 library. GC – MS analysis detected the presence of 14 compounds. GC – MS profile of the methanolic extract revealed the presence of megastigmatrienone, phytol, loliolide, farnesyl acetate, methyl linoleates, gamma-sitosterol, cis, cis, cis-7,10,13-Hexadecatrienal, thymine, pyranone, coumarin, 2 – methoxy 4 – vinyl phenol, guanosine, chinasaure and 3- cyclopentyl propionic acid 2 dimethyl aminoethyl ester. The current study suggests that methanolic extracts of aerial parts of Abelmoschus moschatus contain phytoconstituents with antioxidant and cytoprotective activity. The study results will pave a way for the production of therapeutic agents which can be used for the treatment of various diseases.

Staphylococcus aureus is a ubiquitous bacteria that causes a serious health problem because of its multi-resistance to antibiotics. The aimed of this study was to evaluate the antibacterial activity of P. kotschyi root against S. aureus ATCC 29213 and S. aureus, a clinical strain. The phytochemical compound was sought in the extracts by standard staining tests and extractions were carried out by fractionation using solvents depletion method with increasing polarity. The method of dilution in liquid medium was used for the antibacterial tests. The results showed that the aqueous extract of the bark was effective against both S. aureus with MICs of 0.39 mg.mL-1. The MICs of ethanolic extract were 0.39 and 0.78 mg.mL-1, respectively on S. aureus ATCC 29213 and S. aureus. The steles of P. kotschyi roots were less effective. The ethanolic and aqueous extracts at concentrations = 2MICs, have been respectively bacteriostatic and bactericidal effects on tested germs. The kinetics of inhibition showed that the aqueous extract of the bark at 0.78 mg.mL-1, completely destroyed the two germs respectively in 4 h and 5 h. The action of ethanolic extract at 0.78 and 1.56 mg.mL-1 occurs late against both S. aureus at 6 h and 7 h. Phytochemical screening revealed the presence of alkaloids, tannins, flavonoids, anthracenes, glycosides, saponosides and sterols, some of that may be responsible for the observed antimicrobial activity. This study proved the antibacterial activity of P. kotschyi roots that can be exploited as an antibiotic in the treatment of S. aureus infections.

Meloidogyne javanica is considered as the most damaging nematode of vegetables in Morocco. Eco-friendly bionematicides are urgently required for its control. In vitro experiments were carried out to assess the direct effect of bioproducts of Peganum harmala against M. javanica. The bioassay showed extracts to be nematotoxic. Aqueous extracts of P. harmala exhibited reversible nematostatic activity. The estimated ID50 of the most active product in methanolic extracts was 368ppm. HPLC-MS of the methanolic extract revealed that total content of major alkaloids of P. harmala was approximately 12.162±0.637mg/g. Harmine (8.514±0.521mg/g) is the dominant alkaloid. In conclusion, Peganum harmala has a reversible nematostatic activity on second stage juveniles of M. javanica. The effect of P. harmala is due to its possession of a high content of β-carboline alkaloids, which warrant further experimentation. Bioproducts from P. harmala should be exploited through formulations for management of the root knot nematode.

The phytochemical screening of Dipsacus mitis D. Don and Woodfordia fruticosa (L.) Kurz revealed the presence of bioactive secondary metabolites in both of the species but their concentrations were variable. Flavanoids and saponins were found in remarkable amount in both of the samples. The volatile compounds of both plants were extracted by Simultaneous Steam Distillation and Extraction (SDE) apparatus and analyzed by Gas Chromatography/Mass Spectrometery (GC-MS). Total 53 volatile compounds were tentatively identified from roots of Dipsacus mitis D. Don and 78 were identified from flowers of Woodfordia fruticosa (L.) Kurz. Alcohols were dominant in both the plants accounting 45% and 32% respectively. Numerous bioactive volatile compounds were detected in both herbs. The effects of increasing concentrations of the methanolic extracts on the amplitude and frequency of spontaneously contracting uterine tissues were tested. The effect of extracts on the smooth muscle strips from rat uterus showes the dramatic muscular relaxation on spontaneous contractility and was determided most effective at a concentration 6500 µg/ml of methanolic extract of Dipsacus mitis D.DOI: http://dx.doi.org/10.3126/jnpa.v26i1.6627JNPA. Objective: The present study undertakes a comparative analysis of the level of secondary metabolites present in the leaf, flower and stem of the two ornamental plants, Allamanda blanchetii and Allamanda cathartica.Methods: The two plant species, Allamanda blanchetii and Allamanda cathartica were collected, washed, shade dried in room temperature and powered in mechanical grinder. Phytochemicals were extracted from the power with methanol and double distilled water. The estimation of flavonoids, polyphenols, polysaccharide were done by standard methods and the anti-oxidant activity was measured by 1,1-diphenyl-2-picrylhydrazyl (DPPH) discoloration assay.Results: Our study reveals that the flower of both species contain highest amount of secondary metabolites in crude methanolic and aqueous extracts. In case of leaf, the methanolic extracts contain higher amount of polyphenol, flavonoid and anti-oxidant property in comparison to aqueous extracts, where as the aqueous extract contain higher amount of polysaccharide content than its counterpart. In stem, crude organic extract has higher amount of polyphenol and flavonoid and the aqueous extract has higher amount of polysaccharide and anti-oxidant property.Conclusion: The flower of Allamanda cathartica and Allamanda blanchetii has higher amount of flavonoids, polyphenols, polysaccharide and the floral extracts display comparatively higher anti-oxidant property.
