Objectives: The study was designed to evaluate the hypoglycaemic effects ofCassia sophera’s powdered seeds and its different fractions. Study Design: An experimentalstudy. Setting: Faculty of Veterinary, University of Agriculture, Faisalabad, Pakistan. Period:November 2014 to February 2017. Methodology: Initial experiments were designed to find outthe hypoglycaemic effects of Cassia sophera powdered seeds in increasing doses i.e. 2, 3, 4g/kg body weight at 0, 2, 4, 8, 12 and 24 hours intervals. The effects of methanolic and aqueousextracts equivalent to 4g/kg body weight of Cassia sophera seeds were also studied in bothnormal and diabetic rabbits. Results: At 4 hours interval, 2grams/kg body weight of grindedseeds results no significant decrease in mean blood level of glucose. At 8 and 12 hours intervals,a significant decrease while at 4, 8 and 12 hours a highly significant reduction were observedwith both 3g/kg and 4g/kg body weight. With methanolic and aqueous extracts equivalent to4g/kg body weight, normal rabbits showed significant decrease i.e. with P value less than 0.05at 8 and 12 hours. In addition, the methanolic extract of Cassia sophera seeds produced betterhypoglycaemia as compared to aqueous extract. The effects of these extracts were comparableto Acetohexamide. The same study was also done in alloxan-induced diabetic rabbits and it wasnoticed that Cassia sophera powdered seeds caused significant decrease in blood glucoselevel at increasing doses 2, 3 and 4gram/kg of body weight and highly significant reduction(with P value less than 0.001) in blood level of glucose was noticed with methanolic extracts at4 hours interval which continued upto 8 hours while treatment with aqueous extract with Cassiasophera seeds showed highly significant decrease at 12 hours. Conclusion: It is concludedthat powdered seeds of Cassia sophera constitutes more than one type of hypoglycaemicelements, which have exerted a significant and consistent hypoglycaemic effects in normal andalloxan treated diabetic rabbits.

The current research was planned to measure the comparative phytochemical and anti-oxidative potential of aqueous and methanolic extracts of two indigenously grown millet varieties of Pakistan. The locally available millet varieties, i.e. MB-87 and AF-POP flours were chemically characterized through the proximate and mineral analysis. The antioxidant extract was prepared in water and methanol. The extracts were screened for saponins, terpenoids, flavonoids and tannins; methanolic extract of MB-87 and AF-POP showed flavonoids in an average quantity (++), whilst aqueous extract had only trace (+) values. The phytochemical screening showed the presence of saponins only in the aqueous extract of both varieties. However, terpenoids and tannins were present in both methanolic and aqueous extracts. During antioxidant evaluation, millet methanolic extracts showed higher level of TPC and DPPH of MB-87 and AF-POP as 0.30±0.001 & 0.23±0.0012 mg GAE/g and 48±0.96 &46±1.09%, respectively. However, the β-carotene activity of the aqueous extract of both varieties (MB-87 and AF-POP) was recorded higher. In a nutshell, the methanolic extract of MB-87 has an appreciable atioxidant profile. Furher research should be planned to screen the important constituents of Pakistani millet varieties.

Ayurvedic medicines are being used increasingly to fight or prevent common diseases. Udhara vayuhara churna is a polyherbal formulation containing Piper longum (Piperceae) Zingiber officinalis (Zingiberaceae), Elettaria cardamom (Zingiberaceae), Plumbago zeylanica (Plumbaginaceae), Carum carvi (Umbelliferae), Acorus calamus (Araceae) and Embilica officinalis (Euphorbiaceae). Methanolic and aqueous extracts of Udhara vayuhara churna were screened for their antimicrobial activity against Bacillus subtilis, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Aspergillus niger and Candida albicans. The results of antimicrobial activity of aqueous and methanolic extracts of the Udhara vayuhara churna indicated that methanolic extract inhibited the growth of one or more test pathogens than aqueous extract. Udhara vayuhara churna formulation extracts showed a broad spectrum of antimicrobial activity. Phytochemical investigation revealed the presence of tannins, saponins, alkaloids, glycosides, flavonoids and essential oils. This may be due to the multifunctional effect of all the seven plant ingredients of Udhara Vayuhara churn.

Introduction: Hepatic diseases remain the leading cause of death worldwide. Despite overall advancements in health care, mortality due to hepatic diseases is constantly growing. More than 2 million people globally are estimated to die each year from liver diseases, and current treatment offers little for its management. Thus, it is essential to find more effective and less toxic pharmaceutical alternatives for the treatment of liver diseases. Aims & Objectives: Tamarix dioica, a shrub broadly used in herbal medicine for the treatment and prevention of various diseases. The current study was designed to analyze the hepatoprotective effect of T. dioica in BALB?cmice against CCl4-induced acute liver damage. Place and duration of study: The study was conducted in NIH, Islamabad, Pakistan, for six months in 2016-2017. Material & Methods: For in vivo evaluation, the animals (n= 42) were randomly divided into seven groups (n=6), three control (i.e. Group, I or normal control, group II or induction control received 0.9% normal saline orally, and Group III or positive control received silymarin 100 mg/kg per oral), and four treatment groups (i.e. IV, V,VI and VII were treated with oral T.dioica 200 mg/kg/day, 300mg/kg/day methanol extract, 200mg/kg/day and 300mg/kg/day of aqueous extracts respectively for six days, followed by intraperitoneal administration of CCl4 on the seventh day. The blood samples were collected for analysis of LFTs, and hepatic tissue was taken for histological analysis. Data was analyzed using SPSS version 16, one-way ANOVA with Duncan’s Multiple Range Test (DMRT). Results: CCl4 induction in Group 2 resulted in severe hepatic derangement manifested as highly elevated mean LFTs (ALT 7245.56, AST 3292.11, ALP 340.09 U/L, bilirubin 4.64 mg/dl) as compared to healthy controls (ALT 38.97, AST 50.20, ALP 57.17 U/L, bilirubin 1.25 mg/dl: (Group 1) levels p<0.001. Pretreatment with different extracts of T.dioica for 6 days before CCl4 administration produced varying degrees of hepatoprotection. 300mg/kg aqueous extract T.dioica (Group7) prevented damage with maximal hepatoprotection, reduced LFTs (ALT: 339.95 , AST: 242.90 , ALP: 116.86 U/L, bilirubin: 1.38 mg/dl) and normalized liver histology as compared to Group 2 and standard drug silymarin 100mg/kg, (ALT: 6483.23, AST: 2567.69, ALP: 272.19 U/L, bilirubin: 2.84 mg/dl: Group 3) p<0.001. Lesser hepatoprotection was provided by T.dioica aqueous extract 200mg/kg (ALT: 439.93, AST: 367.87, ALP: 180.62 U/L bilirubin: 1.53 mg/dl: Group VI) and least by 300mg/kg & 200mg/kg methanolic extracts Groups V & IV (ALT: 6338.06, 6443.91, AST: 2800.81, 3012.34, ALP: 242, 248 U/L & bilirubin: 2.82 & 3.62 mg/dl) respectively. Further, no drug-induced toxicity symptoms were observed 24 hours after administration of the high dose oral T. dioica 2000 mg/kg/body weight aqueous and methanolic extracts were administered. Conclusion: Pretreatment with T. dioica extracts especially 300mg/kg aqueous extract reduced acute CCl4-mediated liver damage, ameliorated histopathological as well as biochemical parameters and was free of toxicity in 2000mg/kg /body weight dose in the mice experimental model. T. dioica has potential in hepatoprotective drug research.

OBJECTIVES: To evaluate the Immunomodulatory, apoptosis induction and antitumor effects of aqueous and methanolic extracts of Calvatia craniiformis regarding the size of tumour mass, caspase-8 expression and apoptotic index (AI%) in mice transfected with murine hepatocellular carcinoma cell line (H22) as an experimental therapeutic system for human hepatocellular carcinoma.MATERIAL AND METHODS: Forty-eight Balb/C albino mice were transfected in legs with H22 cells. Tumour size was measured twice a week. Caspase-8 protein expression and apoptotic index determination evaluated by Immunohistochemistry.RESULTS: Tumor size significantly differed between the two groups of mice transfected with H22 cells; the first was treated with C. craniiformis aqueous extract (0.3, 0.6, 1.2) mg/kg and the second group was treated with C. craniiformis methanolic extract (0.25, 0.5, 1.0) mg/kg compared with control group. The inhibitory activity of aqueous and methanolic extracts was dose and duration dependent. The size of the tumour mass was reduced up to 87.9% when treated with 1.2 mg/kg aqueous extract and 1 mg/kg for methanolic extract. Caspase-8 expression was increased in a dose-dependent manner among H22 bearing mice treated with C. craniiformis aqueous extract (0.3, 0.6, 1.2) mg/kg. At 0.3 mg/kg, the intensity of expression was strong in (33.33%) and very strong in (66.67%). While at 0.6 mg/kg and 1.2 mg/kg the intensity of expression was strong in (33.33%) and very strong in (100%) with a significant difference (P ≤ 0.001). H22 bearing mice treated with (0.25, 0.5, 1.0) mg/kg C. craniiformis methanolic extract shows increased caspase-8 expression in a dose-dependent manner. At 0.25 mg/kg, the intensity of expression was strong in (33.33%) and very strong in (66.67%). While at 0.5 mg/kg, the intensity of expression was strong in (33.33%) and very strong in (100%). At 1.0 mg/kg, the intensity of expression was strong in (16.67%) and very strong in (83.33%) with significant difference (P ≤ 0.001). AI% of H22 bearing mice treated with C. craniiformis aqueous and methanolic extracts were significantly increased (P ≤ 0.05) compared with the untreated control group. No significant difference was reported in AI% between aqueous and methanolic extracts treated groups.CONCLUSIONS: Extracts of C. craniiformis were highly efficient in tumour growth inhibition, causing a reduction in the tumour size clinically and increase the expression of caspase-8 gene product in tumour tissue, causing increase apoptotic index of H22 cells taken from the legs of inoculated mice leading to loss of legs due to bone necrosis. Antitumor activity of C. craniiformis aqueous, and the methanolic extract was dose and duration dependent.

The main aim of study is to evaluate the anti-arthritic effect of different extracts of Boerhaavia diffusa in arthritic rats. Different extracts were prepared by successive solvent extraction methods by using the various polar and non polar solvents and their % yields were calculated. Arthritis was induced by FCA induced arthritis model in rats and paw volume was measured on different days. Body weights of all animals were also measured simultaneously and at the end of experiment some haematological parameters were measured. On preliminary phytochemical studies extracts showed the presence of alkaloids, fatty acids, terpenoids, flavonoids and phenolic compounds. Among all extracts, methanolic extract significantly decreased the paw volume in all treated groups. Methanolic extracts also restored the body weight significantly. The results of our study revealed that all the extracts treated group’s causes significant alterations in the hematological parameters and maximal effects were observed at 400 mg/kg. Since methanolic extract showed best activity in arthritic model and its phytochemical study showed presence of flavonoids and phenolic compounds, so it may be possible that anti-arthritic activity of root extracts may be due to presence flavonoids.  Keywords: Arthritis, FCA induced arthritis, Boerhaavia diffusa, haematological parameters, and Body weight

Our studies were performed to investigate the effects of the aqueous extracts of Cecropia pachystachya and Larrea divaricata. These plants are used in folkloric medicine in infusion and were administered orally (0.76 g/kg) to male Albino Swiss mice for 16 days, on drink intake, organ weight/body weight (OW/BW × 100) ratio, histology, broqueoalveolar fluid (BALF) and elevated plus-maze (EPM). Feeding as well as body weight were unaffected by the consumption of these extracts. There were no signs of toxicity in BALF, morbidity or mortality during the study. C. pachystachya caused an increase in relative kidney OW/BW (p ≤ .05 vs control). The macroscopic and microscopic morphologic analyses of the organs were not altered by administration of these plants. A non-anxiolytic-like activity of an aqueous extract prepared from the leaves of C. pachystachya and L. divaricata in EPM was observed. We conclude that the crude aqueous extracts of leaves tested on mice orally did not produce signs of toxicity or behavioral changes in routine histological and clinical evaluation. However, knowledge of the biological activity of many herbal medicine used in Latin American is still deficient and more studies will be needed to elucidate the possible toxic effects.

Buchholzia coriacea (wonderful kola) is a medicinal plant that has been used worldwide as an alternative medication to promote human health. Comparative nephroprotective effects of crude seed powder, aqueous and methanolic extracts of B. coriacea in hepatotoxic rats was investigated for 56 days. The crude powder (BCCP), aqueous extract (BCAE) and methanolic extract (BCME) significantly reduced (p<0.05) the levels of creatinine and uric acid and decreased minimally throughout the treatment periods. Total protein increased significantly (p<0.05). Significant decrease (p<0.05) were observed with 200 mg/kg BCAE and BCCP respectively in urea and BUN levels. Noticeable nephroprotective effects may be attributed to the presence of phytochemicals like flavonoids and tannins which act as antioxidants. This study has demonstrated that B. coriacea crude seed powder, methanolic and aqueous extracts caused no adverse effect on the rat kidney and may be recommended for the management of nephrotoxicity Dhaka Univ. The anti-diabetic effect of ethanolic and aqueous extracts of Imperata cylindrical  rhizomes was investigated in alloxan-induced diabeties in rats. Diabetes was induced by a single 150 mg/kg intraperitoneal dose of alloxan. Rats were divided into five groups with six rats in each group i.e. the normal control group, diabetic control group, standard group (glibenclamide, 10mg/kg, p.o.), Test-I group (200 mg/kg ethanolic extract) and Test-II group (200 mg/kg aqueous extract). The above concerned groups were inoculated on 21st day. On the last day of the experiment, fasted rats were killed by cervical dislocation. The body weight was measured at the initial day and final day. The blood samples were collected for estimation of glucose. The loss of body weight in control group, but recovery was observed in drug treated group. The serum glucose level was significant increased in diabetic rats. However, significant improvement was observed in treated group. The biochemical parameters such as HDL and proteins level were decreased in the control group but maintained in drug treated group. LDL, cholesterol, triglyceride creatinine and urea were significant increase in control group however, reduced level in drug treated group. The present study concluded that ethanolic and aqueous extracts of I. cylindrical  rhizome showed an appreciable effect in reducing the hyperglycemia and the complications associated with diabetes. However, aqueous extract is found more significant in decreasing blood glucose level in comparison to the ethanolic extract. The study results justify the traditional use of the plant as anti-diabetic.

Background: Zingiber Officinale Roscoe (Zingiberaceae family) is knownas Ginger. It is famous for its antioxidant properties. Objectives: To evaluate the effects ofGinger aqueous extract on the serum creatinine and paired kidney weight in Alloxan induceddiabetic nephropathy of albino rats. Study Design: Experimental study. Period: 06 months01-01-2013 to 30 June 2013. Setting: Anatomy Department, Sheikh Zayed, PGMI Lahore.Materials and Methods: Diabetes mellitus was induced with Alloxan intraperitoneally (150 mg/kg body weight) in Experimental groups B & C. Then the rats of experimental group C received200mg/kg body weight of ginger aqueous extract by gavage daily for five weeks starting from8th day after Alloxan injection. Results: Serum creatinine levels increased more in experimentalgroup B as compared to experimental group C. Group wise comparison of creatinine levelrevealed that the difference among control (A group) and experimental (B & C Groups) wassignificant having p-value <0.001. We observed that Paired kidney weight in experimentalgroup B increased as compared to control group A. Less increase in the paired kidney weightwas observed in experimental group C as compared to experimental group B. The differenceof mean paired kidney weight among three groups was significant having p-value <0.001.Conclusion: The results of the present study indicated that the co-treatment of Ginger aqueousextract prevented alloxan induced diabetic nephropathy in albino rats. The aqueous extract ofGinger showed amazing results on paired kidney weight.

Meloidogyne javanica is considered as the most damaging nematode of vegetables in Morocco. Eco-friendly bionematicides are urgently required for its control. In vitro experiments were carried out to assess the direct effect of bioproducts of Peganum harmala against M. javanica. The bioassay showed extracts to be nematotoxic. Aqueous extracts of P. harmala exhibited reversible nematostatic activity. The estimated ID50 of the most active product in methanolic extracts was 368ppm. HPLC-MS of the methanolic extract revealed that total content of major alkaloids of P. harmala was approximately 12.162±0.637mg/g. Harmine (8.514±0.521mg/g) is the dominant alkaloid. In conclusion, Peganum harmala has a reversible nematostatic activity on second stage juveniles of M. javanica. The effect of P. harmala is due to its possession of a high content of β-carboline alkaloids, which warrant further experimentation. Bioproducts from P. harmala should be exploited through formulations for management of the root knot nematode.
