The aim of this study was to analyze the effects of ethanol extract of sambiloto (Andrographis paniculata) on fatty liver percentage, serum glutamic oxaloacetic transaminase (SGOT) level and lipid profile of wistar strain white rat exposed to high fat diet. The study used randomized post test only control group design. Total sample was 50 male wistar strain rats (Rattus norvegicus) divided randomly into 5 groups (randomization). The normality test used was Kolmogorov Smirnov test (a=0.05). The homogeneity test used was Levene test (a=0.05). The comparative test was done using Anova test (analysis of variance) (a=0.05) or Brown-Forsythe test (a=0.05). The correlation test was done using Pearson test (a=0.05). The administration of sambiloto ethanol extract with doses of 100, 200 and 400 milligrams (mg)/kilogram (kg) body weight (BW) decreased the percentage of fatty liver (r=-0.950), SGOT (r=-0.964)/SGPT (r=(R=-0.973)/LDL (low-density lipoprotein) (r=-0.960) and increased HDL (high-density lipoprotein) levels (r=-0.923)=0.956) in white rats exposed to a high-fat diet. In conclusion, increased dose of ethanol extract of sambiloto can decrease the percentage of fatty liver, SGOT/SGPT and total cholesterol/TG/LDL and increase HDL level of white rats exposed to high fat diet.

Background : The prevalence of hyperlipidemia in Indonesia is increasing from year to year, in 2008 it was recorded at 35.1%. Then in 2013 it increased to 35.9%(WHO, 2013). Increased hyperlipidemia triggers Non-Alcoholic Fatty Liver Disease (NAFLD)(Ghanaei et al., 2019). NAFLD conditions cause liver cell damage, causing increased levels of liver enzymes such as SGOT and SGPT (Prahastuti et al., 2020). According to Pradana et al. (2017) it is said that red spinach leaf extract has antioxidant activity that is able to inhibit the incision of the oxidation chain reaction because it contains flavonoid compounds, quercetin, tannins, and saponins which have the potential as a preventive therapy for fatty liver. Objective: To determine the effect of red spinach (Amaranthus tricolor L.) leaf extract on SGOT and SGPT levels in white rats (Rattus norvegicus) fed a high-fat diet. Research Methods: The research method uses a literature study. The database in the literature study was obtained from Google Scholar, PubMed and NCBI which was taken by Population, Intervention, Comparation, Output, Study, and Time (PICOST) from 5,166 journals and finally 8 journals were obtained. Results: Literature from 8 journals is known to have 8 that discuss red spinach leaf extract. Conclusion: From the literature obtained, there was an effect of giving red spinach leaf extract (Amaranthus tricolor L.) on the levels of SGOT and SGPT given a high-fat diet. The dose is 200mg/kgBB and 400mg/kgBB with a duration of administration of 7 to 29 days. Keyword : High fat diet, Red Spinach Extract, Hyperlipidemia, NAFLD, SGOT, SGPT, White Rat

Hyperlipidemia is a disease whichthe effect in increased fat levels in the blood. In hyperlipidemia, the condition of high oxidative stress is obtained due to the oxidation process of LDL (Low Density Lipoprotein) in the body, this condition showed by increased of MDA (Malondialdehyde) levels in serum. Bitter melon (Momordica Charantia L.) contains a lot of active ingredients, one of the ingredients of bitter melon that is thought to have antilipidemic effects is flavonoid compounds. This study was experimental study using a randomized posttest only control group design study with a sample size of 30 male white rats (Rattus norvegicus) wistar strain which was divided into 3 groups, namely K1, K2, and K3. In each group, different treatments are given. Negative control group (K1), given standard feed, water ad libitum; Positive control group (K2), given standard feed, high fat feed with a dose of 2.5 ml water ad libitum; The treatment group (K3) was given a standard diet, high-fat feed with a dose of 2.5 ml water ad libitum and extract of bitter melonat a dose of 250 mg / kg BB by orally every morning for 3 weeks. Data analysis using One Way Anova Test. From the results of the test shows the significance of p-value = 0.000 which is <α (0.05). The results showed that there was an effect of  administration ethanol extract of bitter melon (Momordica charantia L.) to decrease in serum MDA (malondialdehyde) levels in male white wistar rats (Rattus norvegicus) given a high-fat diet.

Eclipta Alba has been used in traditional and folklore medicine to treat Hyperlipidemia and hepatic disorders. The present study was aimed to investigate the Antihyperlipidemic and hepatoprotective potentials of Eclipta Alba in high-fat diet -induced Albino rats and to determine the underlying mechanism.  A total of 30 adult albino rats of Wistar strain weighing 165–215 g were utilized. Animals were treated with high-fat diet for 8 weeks followed by post-treatment of E. Alba for 1 week, 2 weeks, and 3 weeks, respectively. After 12 h of fasting on the last day of the experiment, serum blood samples were collected in EDTA vials and processed for biochemical analysis.  A significant decrease in levels of total cholesterol and triglycerides was noted on animals treated with E. alba compared to high-fat diet animals. Treatment of hypercholesterolemic rats with E. Alba showed a marked decrease of serum low-density lipoprotein (LDL) and very LDL cholesterol concentrations compared to the hypercholesterolemic rats. High-fat diet feeding worsened the levels of serum glutamic oxaloacetic transaminase, serum glutamic pyruvic transaminase, and alkaline phosphatase enzymes, whereas the same markers were significantly improved by supplementation with E. alba compared to the normal group.  E. alba acts as an antihyperlipidemic agent in hyperlipidemic conditions and helps for better health.

ABSTRACT This study aims to determine the antidiabetic activity of papaya flower ethanol extract (Carica papaya L.) against male white rats (Rattus norvegicus) induced by alloxan. This research is experimental. Fifteen rats were divided into 5 treatment groups, each group consisted of 3 rats. The first group was the negative control group given Aquades, the second group was positive control given Glibenklamid, the three groups were 200 mg dose variation groups, four groups were 400 mg dose variations, and the five groups were 800 mg dose variations. Previously, rats were examined fasting blood glucose levels, then mice were induced by an alloxan dose of 120 mg / kgBW intraperitoneally. On the 3rd day blood glucose levels were examined and then treated according to groups for 7 days, measurement of blood glucose levels after the treatment was carried out once every 3 days namely day 3, day 7, and day 10. Data obtained were then analyzed statistically using SPSS , including normality test (Shapiro-Wilk), homogeneity test (Levene), ANOVA test (One way). The results of this study indicate that Papaya Flower (Carica papaya L.) Ethanol Extract can provide the best blood glucose level reduction effect at a dose of 800 mg. Keywords : Antidiabetic, Papaya Flower Extract (Carica papaya L.), Diabettes Mellitus, Male White Rat (Rattus norvegicus), Aloxan.  ABSTRAK Penelitian ini bertujuan untuk mengetahui efektivitas Antidiabetik Ekstrak Etanol Bunga Pepaya (Carica papaya L.) Terhadap Tikus Putih Jantan (Rattus norvegicus) Yang Diinduksi Aloksan. Penelitian ini bersifat eksperimental. Sebanyak 15 ekor tikus dibagi ke dalam 5 kelompok perlakuan masing- masing kelompok terdiri dari 3 ekor tikus. Kelompok pertama kelompok Kontrol negatif yang diberikan Aquades, Kelompok kedua Kontrol Positif yang diberikan Glibenklamid, kelompok tiga kelompok variasi dosis 200 mg, kelompok empat kelompok variasi dosis 400 mg, dan kelompok lima kelompok variasi dosis 800 mg. Sebelumnya tikus dilakukan pemeriksaan kadar glukosa darah puasa, selanjutnya tikus diinduksi Aloksan dosis 120 mg/kgBB secara intraperitoneal. Pada hari ke 3 diperiksa kadar Glukosa darah kemudian diberikan perlakuan sesuai kelompok selama 7 hari, pengukuran kadar glukosa darah setelah perlakuan dilakukan 3 hari sekali yaitu hari ke 3, hari ke 7, dan hari ke 10. Data yang diperoleh kemudian dianalisis secara statistik menggunakan SPSS, meliputi uji normalitas (Shapiro-Wilk), uji homogenitas (Levene), uji ANOVA (One way). Hasil penelitian ini menunjukkan bahwa Ekstrak Etanol Bunga Pepaya (Carica papaya L.)  dapat memberikan efek penurunan kadar glukosa darah yang paling baik yaitu pada dosis 800 mg. Kata kunci : Antidiabetik, Ekstrak Bunga pepaya (Carica papaya L.), Diabettes Mellitus, TikusPutih Jantan (Rattus norvegicus), Aloksan.

The present study aimed to evaluate the potential synergistic and protective effects of ALM16, a mixture of Astragalus membranaceus (AM) and Lithospermum erythrorhizon (LE) extract in a ratio of 7 : 3, against hepatic steatosis in high fat diet (HFD)-induced nonalcoholic fatty liver disease (NAFLD) mice. Forty-eight mice were randomly divided into eight groups and orally administered daily for 6 weeks with a normal diet (ND) or high fat diet alone (HFD), HFD with AM (HFD + 100 mg/kg AM extract), HFD with LE (HFD + 100 mg/kg LE extract), HFD with ALM16 (HFD + 50, 100, and 200 mg/kg ALM16), or HFD with MT (HFD + 100 mg/kg Milk thistle extract) as a positive control. ALM16 significantly decreased the body and liver weight, serum and hepatic lipid profiles, including triglyceride (TG), total cholesterol (TC), high-density lipoprotein-cholesterol (HDL), and low-density lipoprotein-cholesterol (LDL), and serum glucose levels, compared to the HFD group. Moreover, ALM16 significantly ameliorated the HFD-induced increased hepatic injury markers, including aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), lactate dehydrogenase (LDH), and gamma-glutamyltransferase (GGT)-1. Furthermore, as compared to the mice fed HFD alone, ALM16 increased the levels of phosphorylated AMP-activated protein kinase (p-AMPK) and acetyl-CoA carboxylase (p-ACC), thereby upregulating the expression of carnitine palmitoyltransferase (CPT)-1 and downregulating the expression of sterol regulatory element-binding protein (SREBP)-1c and fatty acid synthase (FAS). These results demonstrated that ALM16 markedly inhibited HFD-induced hepatic steatosis in NAFLD mice by modulating AMPK and ACC signaling pathways, and may be more effective than the single extracts of AM or LE.

AbstractBackgroundNon-alcoholic fatty liver disease (NAFLD) is the main common cause of chronic liver disease. The aim of this study is to evaluate the effect of Shilajit, a medicine of Ayurveda, on the liver damage caused by NAFLD.Materials and methodsForty male Wistar rats, after being established as fatty liver models by feeding a high-fat diet (HFD, 12 weeks), were divided randomly into five groups as follows: control (standard diet), vehicle (HFD + distilled water), high-dose Shilajit (HFD + 250 mg/kg Shilajit), low-dose Shilajit (HFD + 150 mg/kg Shilajit) and pioglitazone (HFD + 10 mg/kg pioglitazone). The serum levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT), triglycerides (TG), total cholesterol (TC), low-density lipoprotein (LDL), glucose and liver glutathione peroxidase (GPx), superoxide dismutase (SOD) activity, malondialdehyde (MDA) levels, liver weight, and histopathological manifestation outcomes were measured after the 2-week intervention.ResultsShilajit treatment significantly reduced the values of AST and ALT, TG, TC, LDL, glucose, liver weight, and steatosis, and instead, increased high-density lipoprotein (HDL) compared with the vehicle group (p < 0.05). Further, Shilajit treatment improved the adverse effects of HFD-induced histopathological changes in the liver as compared with the vehicle group (p < 0.001). MDA level and GPx activity increased but SOD activity decreased in the vehicle group compared with the control group (p < 0.05), while treatment with Shilajit restored the antioxidant/oxidant balance toward a significant increase in the antioxidant system in the Shilajit group (p < 0.05).ConclusionsThese findings suggest that Shilajit improved the histopathological NAFLD changes in the liver and indicated the potential applicability of Shilajit as a potent agent for NAFLD treatment.

ABSTRACTSawilangit leaves (Vernonia Cinerea (L.) Less) is a plant that contains flavonoids as an antipyretic. The purpose of this study was to determine the effectiveness of flavonoid compounds from ethanol extract of sawilangit (Vernonia cinerea (L.) Less) leaves as an antipyretic in male white rats Wistar strain (Rattus norvegicus) induced by DPT vaccine. A total of 15 male rats were divided into 5 treatment groups, namely the negative control group (CMC 1%), the positive control group (paracetamol), and the ethanol extract group of sawilangit leaves with a dose of 200 mg, a dose of 400 mg, a dose of 800 mg. Each mouse was induced intramuscularly with a DPT vaccine at a dose of 0.3 ml. Temperature measurements were carried out before and after induction, then every 30 minutes after giving the test material from the 30th minute to the 180th minute. Data were analyzed statistically with One Way ANOVA and continued with LSD test to see significant differences between treatments. The results showed that the ethanol extract of sawilangit leaves had an antipyretic effect on male white rats Wistar strain..Key words: Antipyretics, Vernonia Cinerea, male white rats ABSTRAKDaun Sawilangit  (Vernonia Cinerea (L.) Less) sebagai antipiretik pada tikus putih jantan galur wistar (Rattus norvegicus) yang diinduksi vaksin DPT. Sebanyak 15 ekor tikus putih jantan dibagi 5 kelompok perlakuan yaitu kelompok kontrol negatif (CMC 1%), kelompok kontrol positif (parasetamol), dan kelompok ekstrak etanol daun sawilangit dosis 3,6 mg, dosis 7,2 mg, dosis 14,4 mg. setiap tikus diinduksi vaksin DPT dosis 0,3 ml secara intramuscular. Pengukuran suhu dilakuan sebelum dan sesudah induksi, kemudian setiap 30 menit  setelah pemberian bahan uji dari menit ke-30 sampai menit ke-180. Data dianalisis secara statistika dengan One Way ANOVA dan dilanjutkan uji LSD untuk melihat beda nyata antar perlakuan. Hasil penelitian menunjukan bahwa Ekstrak etanol daun sawilangit mempunyai efek antipiretik terhadap tikus putih jantan galur wistar. Kata kunci: Antipiretik, Vernonia Cinerea, tikus putih jantan

Enzyme treatment of the foods and herbs has been used to improve the absorption rate the efficiency of plant extracts by converting the glycosides of the plant into aglycones. In this study, we examined the obesity-inhibitory effect of Chrysanthemum indicum Linné (CI) treated with enzymes such as viscozyme and tannase, which are highly efficient in converting glycosides to aglycones and then compared with untreated CI extract. The enzyme-treated CI ethanol extract (CIVT) was administered orally at various doses for 7 weeks in the high fat diet (HFD)-fed male mice. CIVT administration reduced the body weights, the food efficiency and the serum levels of lipid metabolism-related biomarkers, such as triglycerides (TG), total cholesterol (TC), low-density lipoprotein cholesterol (LDL-c) and leptin in the dose-dependent manner but not those high-density lipoprotein cholesterol (HDL-c) and adiponectin. CIVT also reduced considerably the total lipid amount in the liver and the size of adipocytes in the epididymal white adipose tissue (eWAT). CIVT effectively downregulated the adipogenesis-related transcription factors such as peroxisome proliferation activated receptor (PPAR)-γ and CCAAT/enhancer binding protein-α (C/EBP-α) but up-regulated PPAR-α, in the liver and eWAT. In addition, when compared to the enzyme-untreated CI 50% ethanol extract (CIEE), CIVT enhanced the reduction of body weight and lipid accumulation. Moreover, the viscozyme and tannase treatment of CI increased the flavonoid contents of the aglycone form. Therefore, our results support that the enzymatic treatment induced the production of aglycones for potentially suppressing the adipogenesis and lipid accumulation in HFD-fed mice. It suggests that CIVT might be an effective candidate for attenuating the over-weight and its related diseases.
