The indices of the immune status and homocystein in persons working in the chemical industry are studied. As a result, the laboratory indications of activation of the immune system are revealed, as well as the increased activity of cellular and humoral immunity. Moderate homocysteinemia is found among men aging of 56—65 with an idiopathic hypertensia. Homocystein appears to be an independent risk factor of coronary, cerebral and peripheral vessels diseases.

The indicators of humoral immunity in clinically healthy women at the stage of pregnancy planning and in patients with pipe-peritoneal infertility in the pre-infantal period of vitro incense fertilization periods are studied. The initial immune status in women of the main group did not have pathological deviations. The identified shifts in the work of the immune system in women with infertility, depending on the outcomes of the auxiliary reproductive technologies, was carried out in a multidirectional nature and could serve as a factor in a preventive forecast, in order to ensure a more differentiated approach at the preparation stage and during extracorporeal fertilization.

Background: Elotuzumab, a humanized IgG1 monoclonal antibody targeting SLAMF7, is useful for the treatment of Relapsed or Refractory multiple myeloma (RRMM) in combination with Lenalidomide (LEN). However, cellular and molecular mechanisms underlying the immunomodulatory effects of elotuzumab still remain largely unclear. We have previously reported that LEN displays immunopotentiating activity that enhances Th2-mediated response at dendritic cell (DC) phase as upstream immune cascade associated with humoral immunity. DCs are pivotal cells in the sense of orchestrating both cell-mediated (linking with Th1) and humoral (linking with Th2) immunity as masters of the immune system. Series of analyses have clarified myeloid DCs (mDCs) play an important role in allergic immune response by the induction of Th2 response. Here, we focused on the effects of elotuzumab in combination with LEN on the function of human mDCs. Methods: Purified blood human CD11+ mDCs from healthy adult volunteers using cell sorting were cultured and analyzed by flow cytometry and ELISA. Serum were obtained from 16 MM patients with before and after elotuzumab therapy. This study was approved by the Institutional Review Board of Kansai Medical University. Results: We found that surface expression of SLAMF7 on mDCs was upregulated in response to Th2-inducing cytokine, thymic stromal lymphopoietin (TSLP) and the expression level was higher in response to TSLP than in response to toll-like receptor ligand R848. Elotuzumab at clinical in vivo plasma concentration of 30 to 300 µg/ml did not affect mDC survival and their CD86 and OX40-ligand expression when stimulated with 0.3 µM LEN and/or TSLP for 24 h. LEN enhanced TSLP-mediated Th2-recruiting chemokine CCL17/TARC from mDCs which functions as chemoattractant for memory Th2 cells and contribute to allergy and humoral immune responses, and elotuzumab significantly enhanced the LEN-mediated production of CCL17/TARC (TSLP+LEN as control vs. TSLP+LEN+100 µg/ml elotuzumab; 1.23 fold increase: p=0.003, and control vs. TSLP+LEN+300 µg/ml elotuzumab; 1.38 fold increase: p=0.038). This finding suggest elotuzumab enhances Th2-mediated immune profile at upstream phase of humoral immunity. In addition, serum CCL17 levels were analyzed in RRMM patients before and after 3 cycle elotuzumab administration (n=16). We found, serum CCL17 levels after elotuzumab administration were significantly higher compared with those before elotuzumab treatment (after; 1512 ± 459 pg/ml vs. before; 402.2 ± 87.4 pg/ml: p = 0.013). Conclusion: MM involves an element of humoral immune dysfunction. Immune status is important for the prognosis of MM, and clinical outcome can be improved by the recovery of immune status. In this context, our data showing the enhancement of Th2-mediated response by elotuzumab provide a plausible explanation for the observed clinical benefit of this antibody-drug in MM. This function of elotuzumab seems to be relevant to the treatment of MM patients under humoral immune dysfunction. Based on our data in focusing on DCs in the immune system, elotuzumab and IMiDs could function as immunostimulators of humoral immunity via mDCs, and this finding elucidated an additional cellular target of elotuzumab. The age of an individual is an important, independent risk factor for many of the most common diseases afflicting modern societies. Interleukin-7 (IL-7) plays a central, critical role in the homeostasis of the immune system. Recent studies support a critical role for IL-7 in the maintenance of a vigorous healthspan. We describe the role of IL-7 and its receptor in immunosenescence, the aging of the immune system. An understanding of the role that IL-7 plays in aging may permit parsimonious preventative or therapeutic solutions for diverse conditions. Perhaps IL-7 might be used to “tune” the immune system to optimize human healthspan and longevity.

The study found some features of the immune status of women with different levels of physical activity, living in climatic conditions of Khanty-Ugra, regarding the reference values. Marked changes in cellular and humoral immunity can be regarded as a manifestation of the adaptive immune response to ecological conditions of the region. Sport activities help to maintain and improve the adaptability of the female body to the climate-geographical conditions of Khanty-Ugra.

There are considered features of disorders of the immune response in chronic exposure to dust aerosols. The detected changes of indices of the immune status of employees of the dust dangerous occupations and patients with chronic dust pathology of the lungs were unidirectional in the character, which is probably caused by manifestations of nonspecific response of the immune system to the dust factor. The deterioration of cellular immunity, humoral immunity and cytokine profile predisposes to the occurrence of immunopathologic states, contributing to the development of caused by both worksite and occupation pathology.

BackgroundNon-metastatic renal cell carcinoma (RCC) with tumor thrombus showed a greater tendency for developing metastases after surgery. Early identification of patients with high risk of poor prognosis is especially important to explore adjuvant treatment of improving outcomes. Neutrophil-to-lymphocyte ratio (NLR) was a systemic inflammation marker and outcome predictor in RCC, reflecting the chaos in systemic immune status in cancer as myeloid cell expansion and lymphatic cell suppression. Neutrophil extracellular traps (NET) formation (NETosis) is the process of neutrophils generating an extracellular DNA net-like structure. NETosis in tumor was demonstrated to conduce to the subsequent metastases of tumor. However, the role of NLR for systemic immune status and tumor local immune infiltration, especially for neutrophil-associated NETs, in non-metastatic RCC with thrombus remains unclear.Patients and MethodsIn our clinical cohort, we enrolled the clinical, pathologic, and preoperative laboratory parameters of 214 RCC patients with tumor thrombus who were treated surgically. The clinical endpoint was defined as cancer-specific survival (CSS). In our basic research cohort, RNA-seq, TCR-seq, and scRNA-seq data were analyzed. Patients who reached the endpoint as recurrence-free survival (RFS) were defined as the “High-risk” group. Otherwise, they were separated into the “Low-risk” group.ResultsIn the clinical cohort, NLR≥4 was an independent risk factor for 203 localized RCC with tumor thrombus. In the basic research cohort, tumor thrombi were separated into NETosis-thrombi belonging to the “High-risk” group and non-NETosis-thrombi to the “Low-risk” group. NETs induced by tumor-derived G-CSF in tumor thrombus has a mechanistic role in unfavorable prognosis. Besides, NETs-score from single sample GSEA (ssGSEA) algorithm was an independent prognostic factor validated in the TCGA data. Apart from the neutrophils-associated NETosis, systemic immune perturbations of lymphocytes occurred in the “High-risk” group, represented with decreased TCR diversity and increasingly high proportion of CD4-positive effector memory T (Tem) cells, which indirectly represented the state of lymphopenia.ConclusionsOur findings firstly demonstrated that neutrophils-associated NETosis and systemic lymphocytes perturbations were considered as tumor progression in patients of localized RCC with tumor thrombus, which reflected NLR≥4 as an independent risk factor for patients.

SUMMARYThe immune status of a host infected withTrypanosomaspp. orLeishmaniaspp. can play an important role in successful chemotherapy. In animal models, treatment of African trypanosomiasis with difluoromethylornithine or melarsoprol requires an appropriate antibody-mediated immune response. An intact immune system is also necessary for rapid clearance of trypanosomes from the bloodstream following treatment with suramin or quinapyramine. Similarly, an efficient cell-mediated immune response is required for maximal efficacy of pentavalent antimonials in the treatment of leishmaniasis. However, the potential relationship between parasite-induced or acquired immunosuppression and effective chemotherapy has been poorly studied. Macrophages which have been activated by bacterial cell wall components or gamma-interferon are known to display increased activity againstLeishmania donovaniorTrypanosoma cruzi. In experimental and clinical visceral leishmaniasis, use of macrophage activators together with pentavalent antimonials has lowered the dose of antimony required to cure the infection.

The work is perfromed at the department of the hospital therapy GPMU, laboratory of llergiology and immunology of the research-practical Center of Dental faculty of SpbGMU named after I. Р. Pavlov. State of cellular and humoral immunity at the beginning and at the end of the training year of 15-17 years old young sportsmen of the first level and candidates masters of sports involved in an academic rowing and handball was studied. The athletes of both groups showed significant improvements as compared to the persons not involved in sports, lymphocyte apoptosis inducers (CD95+), the natural killer cells (CD16 + ) and B-lymphocytes (CD20 + ). Over time, in the athletes, regardless the sport, unidirectional changes are detected, encompassing the enhancing of the relative number of CD25+ with receptors for IL2, and immunoglobulin levels.

The aim. To evaluate the efficiency of physiotherapeutic methods and combine therapy on immune system and on oxidation process in patientes with chronic eczema. Materials and methods. There were 32 patients with severe chronic eczema, who were treated with standard systemic and topical therapy plus photophoresis with mometasone furoat (1 mg) and supracutaneus laser irradiation of blood (1 procedure). The efficiency of therapy was detected by the changes of EASI index, cell and humoral immunity parameters, antioxidation status. Results. After using of combine therapy in severe eczema patients positive changes in severity of cutaneous process and prolongation of remission periods were found out as well as normalization of immune status and levels of oxidation processes. Conclusion. The combine treatment of patients with severe chronic eczema is effective, available method of therapy of chronic eczema patients.

The article covers the topic Parvovirus infection in pregnant women caused the development of immunopathological reactions. 129 in fected pregnant women and 16 women with physiological pregnancy in the 1st, 2nd and 3rd trimesters were examined. In infected pregnant women, activation of the cellular level of immunity was observed, a significant increase in the number of natural killers in the peripheral blood and an increase in the immunoregulatory index were detected, with greater deviations from the control indicators found in the 1st trimester. In pregnant women, in which clinical complications were noted, the population composition of lymphocytes was characterized by an increased number of NK cells, low infection with the number of cytotoxic CD8+-lymphocytes and a shift in the ratio of CD4+/CD8+totheside, which indicates an increase in helper mechanisms. Increasing the number of T-lymphocytes, T-helpercells, NK/Т cellsandactivated CD25+-lymphocytes in pregnant women with out clinical complications at different periods of pregnancy can contribute to effective elimination of the virus and to prevent severe fetal lesions. Violation of the humoral link of the immune system in the infection of pregnant parvovirus infection was shown in an increase in the serum IgM content in different periods of pregnancy (by 79.1%, 54.2% and 38.5%), and a decrease in IgA levels in the 1st and 2nd trimesters (p>0.05), to a greater extent such changes were found in women with clinical complications of pregnancy and especially in the first trimester.
