<p>Kekeringan sangat berpengaruh terhadap pertumbuhan dan produksi tanaman kentang. Persilangan dengan tujuan toleran kekeringan telah dilakukan dan dilanjutkan dengan seleksi terbatas. Tujuan penelitian untuk mempelajari respon tanaman kentang terhadap kekeringan dan melakukan seleksi klon-klon hasil persilangan untuk sifat toleran kekeringan. Materi yang digunakan adalah 78 nomor hasil seleksi progeni kekeringan tahun 2015. Penelitian dilakukan dengan membandingkan antara tanaman pada kondisi kekeringan dan pengairan normal di dalam Rumah Kaca Balai Penelitian Tanaman Sayuran, Lembang, pada tahun 2016. Hasil penelitian menunjukkan bahwa cekaman kekeringan berpengaruh terhadap karakter pertumbuhan dan hasil. Pada kondisi kekeringan, vigor tanaman menurun dan menunjukkan gejala layu, menguning, serta daun menggulung ke atas. Kekeringan juga menyebabkan penurunan pada diameter batang (41,4%), jumlah batang (6,63%), tinggi tanaman (22,43%), diameter kanopi (18,76%), luas daun (53,7%), jumlah ubi pertanaman (17,54%), berat ubi pertanaman (70,35%), panjang ubi (44,45%) serta diameter ubi (42,85%). Respon tanaman terhadap kekeringan yang lain ditunjukkan oleh peningkatan kadar prolin daun dan klorofil. Seleksi berdasarkan perubahan karakter morfologi, pertumbuhan vegetatif serta produksi ubi, diperoleh 26 genotipe yang berpotensi memiliki sifat toleran terhadap kekeringan. Genotipe terseleksi tersebut memiliki kisaran jumlah ubi per tanaman 1,67 – 12,25, berat ubi per tanaman 26,45 – 80,775 g, panjang ubi 2,05 – 3,4 cm serta diameter ubi 1,43 – 3,06 cm. Hasil dari seleksi kekeringan ini akan dilanjutkan ke seleksi di lapangan untuk mendapatkan klon unggul kentang toleran terhadap kekeringan. Ketersediaan klon kentang toleran kekeringan dapat menjawab ancaman menurunnya produksi kentang akibat perubahan iklim.</p><p><strong>Keywords</strong></p><p>Solanum tuberosum L.; Produksi ubi; Prolin; Toleran kekeringan   </p><p><strong>Abstract</strong></p><p>Drought is very influential towards the growth and production of the potato crop. A crossing to drought-tolerant genotypes was conducted and continued with a progeny selection. The aims of this study were to study the response of the potato plant to drought stress and to select potato clones resulted from conventional crossing for drought tolerant. The genetic materials tested were 78 progenies resulted from drought selection in 2015. The study was conducted by comparing plants in drought and normal irrigation conditions in the Greenhouse of the Indonesian Vegetable Research Institute, Lembang, in 2016. Results showed that drought stress affected potato growth as well as tuber yield. On drought conditions, potato plants tend to be poor of plant vigor, showed wilting symptom, yellowing leaves and roll up of the leaves. Drought stress caused the decline of stem diameter of (41.4%), main stem number (6.63%), plant height (22.43%), canopy diameter  (18.76%), leaf area  (53.7%), per plant tuber number (17.54%), per plant tuber weight (70.35%), tuber length (44.45%) and tuber diameter (42.85%). Another response to drought was the increasing level of proline and chlorophyll in leaf. Based on morphological character changes, vegetative growth and tuber production, 26 genotypes demonstrated potential drought tolerance. The selected genotypes will be used to the next selection in the field to get stable drought-tolerant potato clones. The availability of drought-tolerant potato clones can respond to the threat of reduced potato production due to climate change. </p>

Potato (Solanum tuberosum L.) is an important vegetable crop in Jordan, occupying second position after olives. In 2012, potatoes were planted on about 6,000 ha with a production of about 141,000 t (2). Potato virus Y (PVY) is a serious problem for potato production worldwide. Recombinant strains of the virus were reported to cause tuber necrotic ringspot disease (PTNRD) in many potato-growing regions of the world. In the last few years, a new recombinant PVYNTN-NW that belongs to PVYZ (3) has been reported in the neighboring Syria. It included three recombination patterns, SYR-I, SYR-II, and SYR-III, and caused severe PTNRD (1). Since PVY is easily transmitted from one region to another by aphid vectors and infected potato seeds, this study was initiated to investigate the possible occurrence of PVY strains in Jordan. In October 2013, 33 leaf samples were collected from symptomatic potato plants cv. Spunta from Wadi Rum, Jordan (GPS coordinates 29°31′37.76″ N, 35°42′48.75″ E), the largest potato-producing area in Jordan. Sampled plants displayed leaf mottling and yellowing, symptoms similar to those caused by PVY. All samples were tested for PVY by DAS-ELISA using the ELISA kit (monoclonal cocktail) developed by BIOREBA (Reinach, Switzerland) to detect all PVY isolates. Twenty-nine samples were found positive for PVY by ELISA. To confirm virus infection, total RNA was extracted from all ELISA-positive samples and used as template in uniplex RT-PCR using strain-specific primers (1). The band pattern of PCR amplicons showed that 12 samples were infected with PVYNTN-NW genotype SYR-III and produced bands of 1,085, 441, and 278 bp. One sample was infected with PVYNTN (A) and produced bands of 1,307, 633, and 441 bp, and one other sample was infected with PVYNTN-NW genotype SYR-II and produced bands of 1,085 and 441 bp. Mixed infection with PVYNTN-NW genotype SYR-III and PVYNTN (B) was also detected in one sample producing bands of 278, 441, 1,085, and 1,307 bp. To confirm infection with the recombinant strains, PCR fragments of 278 bp amplified from three samples and 1,085 bp obtained from another three samples were directly sequenced and sequences were deposited in GenBank under accession numbers KJ159968, KJ159969, and KJ159970 for the 278-bp fragment and KJ159974, KJ159975, and KJ159976 for the 1,085-bp fragment. Sequence comparison with other PVY strains available in the NCBI database showed that the 278-bp fragment had the highest nucleotide sequence identity (100%) with PVY isolates SYR-III-A26 (AB461467) and SYR-III-2-4 (AB461457) from Syria. BLAST searches also showed that the 1,085-bp fragment shared 99% nucleotide identities with PVY isolates SYR-II-L3 (AB461482) and SYR-II-Be4 (AB461474) from Aleppo, Syria. To our knowledge, this is the first report of PVY recombinants in Jordan, and the first report of PVYNTN-NW recombinants infecting potato crop outside Syria. Since Europe is the main supplier of potato seeds for farmers in Jordan and Syria, the introduction of PVYNTN-NW to the region could have happened through infected potato seeds. Narrow profit margins, resource conservation issues and environmental concerns are the main driving forces to improve fertilizer uptake, especially for potatoes. Potatoes are a high value crop with a shallow, inefficient root system and high fertilizer rate requirements. Of all essential nutrients, nitrogen (N) is often limiting to potato production. A major concern in potato production is to minimize N leaching from the root zone. Therefore, the main objective of this study was to examine the potato crop characteristics under drip irrigation with low-discharge (0.6 L h−1) and to determine the optimal combination of irrigation (40, 60, 80, and 100%) and fertigation (0, 50, and 100%) doses. In this study, the 80% (438.6 mm) irrigation dose and a 50% (50 mg N L−1) fertigation dose (W80%F50%) showed that these doses are sufficient for optimal potato yield (about 40 ton ha−1) in conjunction with water and fertilizer savings. Moreover, this treatment did not exhibit any qualitative changes in the potato tuber compared to the 100% treatments. When considering water productivity and yield, one may select a harsher irrigation regime if the available agricultural soils are not a limiting factor. Thus, higher yields can be obtained with lower irrigation and fertigation doses and a larger area.

Background: In a Mediterranean agrosystem of low productivity, a study was carried out on the effects of municipal solid waste compost (MSWC) compared to other organic and inorganic amendments on the production, quality and yield of three potato varieties (Solanum tuberosum L.) and an advanced clone. Method: Simultaneously, the agronomic and nutritional parameters of the potato crop, the degree of bioavailability and the possible risks of heavy metal contamination were studied. Results: Two stages are observed in the yield and content of macro, micronutrients and heavy metals. The addition of all amendments and especially that of urban waste compost increased potato production and the content of macronutrients, micronutrients and heavy metals in the soils of all varieties, showing a progressive accumulation in tubers. Nevertheless, the performance is not maintained over time with a notable decrease during the second stage of its application. Conclusion: Highlighting the potato clone A7677 not only in its performance but also in the concentration of iron, zinc, copper, essential micronutrients for human consumption and especially for populations deficient in these trace elements.

The most serious insect pest problem in the potato crop in Colombia is the lepidopteran Tecia solanivora that causessignificant economic losses. In this research, we designedexpression cassettes based on the cry1Ba1 gene of Bacillusthuringiensis that could confer resistance to T. solanivora via the variety Pastusa Suprema. We selected the elements of thedesigned expression cassettes through an analysis of scientificliterature and patent databases; the considered factors were theproteolytic activation of the Cry1Ba1 protoxin, modification ofcodonic use, polyadenylation signals, and cryptic splicing sites. We used a tissue-specific patatine promoter to reduce potentialbiosafety risks, because it is expressed only in the tuber. Thefreedom to operate analysis suggests that the commercial use of the designed expression cassettes in transgenic potato plants does not affect the rights of third parties in Colombia.

ResumenAntecedentes: La papa (Solanum tuberosum L. grupo Andigena y Phureja) es un tubérculo de vital importancia en Colombia y en especial en el departamento de Boyacá. En la Universidad Pedagógica y Tecnológica de Colombia (UPTC) se cuenta con más de mil trabajos de grado, realizados en los 55 años de trayectoria del programa académico de Ingeniería Agronómica, entre los cuales 130 corresponden a investigaciones relacionadas con este cultivo. Objetivo: se recopiló información de dichos trabajos de grado desarrollados en este período y se analizaron 5 áreas de investigación: entomología, nutrición, fisiología vegetal, fitopatología y economía, en 3 lapsos de tiempo de 10 años cada uno y un último de 12 años. Resultados: En entomología se adelantaron 42 trabajos de grado (32,3%), siendo los de gusano blanco (Premnotrypes vorax Hustache) los más representativos. En el área de nutrición con 35 trabajos de grado (26,9%), se resaltaron los temas sobre correctivos de acidez y nutrición con elementos mayores N-P-K, haciendo énfasis en la evaluación del uso del fósforo. En fisiología vegetal se documentaron 20 trabajos de grado (15,4%), dentro de los cuales, se destacaron los temas sobre estadios fenológicos, crecimiento y desarrollo del cultivo de papa. En economía con 18 trabajos de grado (13,8%), fue más importante la evaluación económica y respuesta agronómica en fertilización, y finalmente en fitopatología con 15 trabajos de grado (11,5%), el tizón tardío (Phytophthora infestans) correspondió al patógeno más estudiado. Conclusión: Esta revisión brinda información a estudiantes, profesionales e instituciones involucradas con el sector papero colombiano y departamental, para la orientación y enfoque de futuros trabajos de investigación.Palabras clave: economía, entomología, nutrición, fisiología vegetal, fitopatología.AbstractBackground: Potato (Solanum tuberosum L. AbstractDrought is the most perilous abiotic stress that affects finger millet growth and productivity worldwide. For the successful production of finger millet, selection of drought tolerant varieties is necessary and critical stages under drought stress, germination and early seedling growth, ought to be fully understood. This study investigated the physiological and biochemical responses of six finger millet varieties (GBK043137, GBK043128, GBK043124, GBK043122, GBK043094 and GBK043050) under mannitol-induced drought stress. Seeds were germinated on sterile soil and irrigated with various concentrations of mannitol (200, 400 and 600 mM) for two weeks. Comparative analysis in terms of relative water content (RWC), chlorophyll, proline, and malondialdehyde (MDA) contents were measured the physiological and biochemical characteristics of drought stress. The results showed that increased level of drought stress seriously decreased germination and early seedling growth of finger millet varieties. However, root growth was increased. In addition, exposition to drought stress triggered a significant decrease in relative water content and chlorophyll content reduction the biochemical parameters assay showed less reduction of relative water content. Furthermore, oxidative damage indicating parameters such as proline concentration and MDA content increased. Varieties GBK043137 and GBK043094 were less affected by drought as shown by significant change in the physiological parameters. Our findings reveal the difference and linkage between the physiological responses of finger millet to drought and are vital for breeding and selection of drought tolerant varieties of finger millet. Further investigations on genomic and molecular to deeply insight the detail mechanisms of drought tolerance in finger millet need to explored.

Potato production is adversely affected by aphid infestation across the globe. Understanding the mechanism of host plant defense against aphids under drought stress is paramount for insect pest management. This study was conducted to examine the cross-talk of phytohormones in potato glycoalkaloids’ defense against green peach aphids under greenhouse conditions. A 3 × 2 × 2 factorial experiment comprising three potato cultivars (Qingshu 9, Longshu 3, and Atlantic) and two levels each of water availability and aphid infestation was conducted. The results show that under drought stress, green peach aphids thrive well on host plants, which contain a relatively high water content. The resistant cultivar DXY, which exhibited a higher level of phytohormones, also demonstrated higher α-chaconine and α-solanine contents in both leaf and root, under drought and aphid stress. Conversely, the susceptible cultivar QS9, which exhibited a lower level of phytohormones, also demonstrated low α-chaconine and α-solanine contents in both leaf and root, under drought and aphid stress. The DXY cultivar, which possessed high resistant traits such as α-chaconine and α-solanine, can be used in areas where green peach aphid infestation is a major setback.

ABSTRACT   This study aimed to examine the growth and production of potato crop varieties and varieties Granola Supejhon on two altitude, which is 750 m above sea level and 1200 m above sea level, studies using randomized block design with two factors. The first factor is the Granola variety and Supejhon. The second factor is the altitude, which is 750 m above sea level and 1200 m asl. The results of this study indicate that the rate Crop Growth Rate (LTT) and the Tuber Growth Rate (LTU) of Supejhon Granola varieties have a much higher altitude 1200 m asl (Modoinding) compared with altitude 750 m asl (Langowan). The altitude effect on the number of tubers / plant and production / plot, otherwise varieties had no effect on the number of tubers / plant, weight of tubers / plot and production / plot, and there is no interaction between variety and altitude. At altitude 750 m asl, both varieties yield an average production of tubers / plot of 1343.20 g (1.34 kg), whereas the altitude of 1200 m asl generate 7462.18 g (7.46 kg). Based on the average, the weight of tubers / plant and production / plots at very low reached by the two varieties of Granola and Supejhon well, so that the two varieties are not recommended to be cultivated at altitude 750 m above sea level. Further research is needed to examine other potato varieties that can be cultivated in areas with altitude of 750 m asl. Keywords: Potato, growth, production, level altitudes ABSTRAK Penelitian ini bertujuan untuk mengkaji pertumbuhan dan produksi tanaman kentang varietas Granola dan varietas Supejhon pada dua ketinggian tempat, yaitu 750 m dpl dan 1200 m dpl, penelitian  menggunakan Rancangan Acak Kelompok dengan dua faktor.  Faktor pertama adalah varietas  Granola dan Supejhon.  Faktor kedua adalah ketinggian tempat, yaitu 750 m dpl dan 1200 m dpl. Hasil penelitian ini menunjukkan bahwa Laju Tumbuh Tanaman (LTT) rata-rata dan Laju Tumbuh Umbi (LTU) rata-rata varietas Granola dan Supejhon memiliki nilai yang jauh lebih tinggi pada ketinggian tempat 1200 m dpl (Modoinding) dibandingkan dengan ketinggian tempat 750 m dpl (Langowan).  Ketinggian tempat berpengaruh terhadap jumlah umbi/tanaman dan produksi/petak, sebaliknya varietas tidak berpengaruh terhadap jumlah umbi/tanaman, bobot umbi/petak dan produksi/petak, serta tidak terdapat interaksi antara varietas dan ketinggian tempat. Pada ketinggian tempat 750 m dpl, kedua varietas menghasilkan rata-rata produksi umbi/petak sebesar 1343,20 g (1,34 kg), sedangkan pada ketinggian tempat 1200 m dpl menghasilkan 7462,18 g (7,46 kg).  Berdasarkan nilai rata-rata, bobot umbi/tanaman dan produksi/petak yang sangat rendah dicapai oleh kedua varietas baik Granola maupun Supejhon, sehingga kedua varietas tersebut tidak dianjurkan untuk dibudidayakan pada ketinggian 750 m dpl.  Perlu dilakukan penelitian lanjutan untuk menguji varietas kentang lainya yang dapat dibudidayakan pada daerah dengan ketinggian tempat 750 m dpl. Potato production plays an important role in Hungary and the other countries of Europe. Consumption of potato products has increased to a large extent during the past several years. We can satisfy market demands with high quality and virus-free varieties.Results of potato production depend on tolerance/resistance to abiotic stresses. In many cases, increased concentration of NaCl causes yield loss. Selection of salt tolerant varieties proved to be a difficult problem. Nowadays, the salt tolerance of potato varieties can be determined by cell/tissue/ protoplast techniques. Somaclonal variation provides a great potential for selection of lines resistant to salt stress. In vitro shoots and callus, derived plantlets selected for salt tolerance/resistance provide material for micropropagation.In vitro shoot development of potato (Solanum tuberosum L. cv. Kuroda) was investigated under salt stress (40 mM, 80 mM, 120 mM NaCl) conditions. Shoot heights of plantlets cultured under salt conditions were lower than the control through the investigation. However, the shoot development of plantlets originated from in vitro meristems was almost at the same level as the control under 40 mM NaCl concentration.There was no significant difference in the in vitro biomass production between control and treatment with 40 mM NaCl concentration. We measured a significant decrease in dry-matter mass under 120 mM NaCl concentration. There is a need for more investigation of different genotypes and for a conclusion as to whether in vitro tolerance could occur under in vivo circumstances in plants originated from somaclones as well.Under in vitro conditions, we investigated shoot and leaf callus initiation using different culture media with different 2,4-D concentrations. Under dark conditions, callus induction of shoot/leaf decreased as the 2,4-D concentrations increased.In light conditions, there was a little callus induction, while callus initiation from the shoot from 5 μM to 12 μM 2,4-D concentration showed a significant increase
