Abstract Background: Currently, there are relatively few studies on the effects of changes in estrogen and androgen levels on prostatic MVD.This article aimed to study the changes of prostatic MVD in SD rats after castration under the effect of estrogen/androgen at different concentrations.Methods: Male Sprague-Dawley(SD) rats aged 3-4 months were randomly divided into the control group, castration group, and different concentrations of estrogen/ androgen treatment after castration. Dihydrotestosterone(DHT) and estradiol(E) were administered daily by subcutaneous injection for one month. All the rats were sacrificed by cervical dislocation after one month, and the serum DHT and E concentrations of the rats in each group were measured by ELISA assay. Prostate tissues specimens were immunohistochemically stained with monoclonal antibodies against CD-34 and factor VIII for the MVD.Results: Compared with the control group, the MVD decreased significantly in the castration group (P<0.05). When the exogenous E concentration was constant, in general, the MVD of rats in all the groups increased with the increase of exogenous DHT concentration; Among them, compared with the castration group, the MVD increased significantly in the E0.05+DHT0.015 mg/kg group, E0.05+DHT0.05 mg/kg group, E0.05+DHT0.15 mg/kg group, E0.05+DHT0.5 mg/kg group, and E0.05+DHT1.5 mg/kg group (P<0.05). In addition, when the exogenous DHT concentration was constant, the MVD increased with the increase of exogenous E concentration in all the groups; Among them, compared with the control and castration group, the MVD increased significantly in the DHT0.15+E0.015 mg/kg group, DHT0.15+E0.15 mg/kg group, and DHT0.15+E0.5 mg/kg group (P<0.05).Conclusions: Androgens carried an important role in the regulation of prostatic MVD in SD rats, and the decrease of DHT concentration can induce a decrease in prostatic MVD. In contrast, prostatic MVD can be increased with the increase of DHT concentration. In addition, prostatic MVD can be increased gradually with the increase of estrogen concentration.

PURPOSE: To investigate synergistic suppression of donor liver pre-perfusion with recipient serum (RS) and cobra venom factor (CVF) treatment on hyperacute rejection (HAR) following liver xenotransplantation. METHODS: Guinea-pigs (GP, n=24) and Sprague-Dawley rats (SD, n=24) were recruited. Before transplantation, serum was collected from SD rats and used for preparation of inactivated complements. GP and SD rats were randomly assigned into four groups (n=6), respectively: RS group, CVF group, RS+CVF group and control group. Orthotopic liver xenotransplantation was performed with modified two-cuff technique. The survival time and liver function of recipients, morphological and pathological changes in rat livers were investigated. RESULTS: There was no piebald like change in the recipient livers in all experiment groups. The survival time of recipients in all experiment groups was longer than that in control group (p<0.05). Moreover, the survival time in the RS+CVF group was markedly longer than that in the RS group (p<0.01) and CVF group (p<0.05). The serum ALT level in all experiment groups were lower than that in the control group (p<0.05). Furthermore, the ALT level in the RS+CVF group was significantly lower than that in the CVF group (p<0.05) and RS group (p<0.01). The histological damages were significantly improved when compared with the control group, and the histological damages in the RS+CVF group were milder than those in the remaining groups (p<0.05) CONCLUSION: Pre-perfusion of donor liver with recipient serum and cobra venom factor treatment can exert synergistic suppressive effects on the hyperacute rejection following liver xenotransplantation.

Dexmedetomidine (DMED), an alpha-2 adrenoreceptor agonist, has been widely used in regional anesthesia procedures. However, the effect of DMED on local anesthetic cardiotoxicity has not been well delineated. This study consisted of two experiments. In experiment A, 42 Sprague–Dawley (SD) rats were randomly divided into 6 groups ( n = 7), each group was pretreated with DMED 0 μg kg−1 (D0 group), 1 μg kg−1 (D1 group), 3 μg kg−1 (D3 group), 6 μg kg−1 (D6 group), 12 μg kg−1 (D12 group), and 24 μg kg−1 (D24 group), administered through the right femoral vein. In experiment B, 20 SD rats were randomly divided into 4 groups ( n = 5), such as control group, DMED group, yohimbine (YOH) group, and DMED + YOH group. Each subgroup in experiment B was also pretreated similarly as in experiment A. After pretreatment of rats as described above (in experiments A and B), bupivacaine 2.5 mg kg−1 min−1 was infused to induce cardiac arrest. In experiment A, the lethal dose threshold of bupivacaine and plasma bupivacaine concentration in D3 and D6 group were higher than the other groups. In experiment B, there was no interaction between DMED and YOH in lethal dose threshold, arrhythmia time, plasma concentration of bupivacaine, and myocardial content of bupivacaine. DMED doses of 3–6 μg kg−1 elevated the lethal dose threshold of bupivacaine without involvement of the alpha-2 adrenoceptors.

Observe the therapeutic effect of Danshensu on lung injury for rats, as well as explore the mechanism of Danshensu in TGF-β1/Smads signaling. Thirty Sprague-Dawley (SD) rats were intratracheally instilled with bleomycin to induce lung injury and interstitial fibrosis. Divided Thirty rats into three groups. DA group (η = 10): Inject 15 mg/kg Danshensu into the abdominal cavity; DXM group (η = 10): Inject 1 mg/kg dexamethasone into the abdominal cavity; BLM group (η = 10): Inject 2 mL physiological saline into the abdominal cavity. Then ten SD rats were intratracheally instilled with physiological saline as normal control group, NC group: Inject 2 mL physiological saline into the abdominal cavity. After a period of 28 days, the degree of pulmonary alveolitis was evaluated using hematoxylin-eosin (HE) staining, and the degree of lung fibrosis was evaluated using Masson?s trichrome (MT) staining. The immunohistochemistry was used to determine the expression of α-SMA. Magnetic nanoparticles+rtQ-PCR was used to determine the mRNA expressions for TGF-β1, Smad3, and Smad7. The alveolitis and pulmonary fibrosis in DA rats were obviously less than those in BLM rats and DXM rats. The expression of α-SMA in DA rats was obviously less than that of in BLM rats and DXM rats; the mRNA expression of TGF-β1 and Smad3 in DA rats were obviously reduced; the Smad7 mRNA expression was obviously up-regulated. DA can alleviate rat lung injury caused by bleomycin. Inhibiting the TGF-β1 and Smad3 mRNA expression, as well as boosting the Smad7 mRNA expression is one of the mechanisms by which Danshensu reduces lung injury.

AbstractIntroduction:Tiletamine-xylazine-tramadol (XFM) has few side effects and can provide good sedation and analgesia. Adenosine 5’-monophosphate-activated protein kinase (AMPK) can attenuate trigeminal neuralgia. The study aimed to investigate the effects of XFM and its specific antagonist on AMPK in different regions of the brain.Material and Methods:A model of XFM in the rat was established. A total of 72 Sprague Dawley (SD) rats were randomly divided into three equally sized groups: XFM anaesthesia (M group), antagonist (W group), and XFM with antagonist interactive groups (MW group). Eighteen SD rats were in the control group and were injected intraperitoneally with saline (C group). The rats were sacrificed and the cerebral cortex, cerebellum, hippocampus, thalamus, and brain stem were immediately separated, in order to detect AMPKα mRNA expression by quantitative PCR.Results:XFM was able to increase the mRNA expression of AMPKα1 and AMPKα2 in all brain regions, and the antagonist caused the opposite effect, although the effects of XFM could not be completely reversed in some areas.Conclusion:XFM can influence the expression of AMPK in the central nervous system of the rat, which can provide a reference for the future development of anaesthetics for animals.

ABSTRACT Talinum paniculatum (som Java) reported to have many pharmacological activities such as increasing spermatozoa quality, cell regenerations, fertility, and antibacterial effect both empirically and scientifically. This study aimed to prove the combination infusion effects of som Java, gotu kola, and temulawak on introduction, climbing, and coitus levels as well as testosterone levels in male Sprague Dawley (SD) rats. SD rats were divided into 2 groups, control group received aquadest, treatment group received formula a mixture of som java Talinum paniculatum) root, gotu kola (Centella asiatica) herb, and Javanese turmeric (Curcuma xanthorrhiza) rhizome with the dose of 491,2 mg/200 g BW for 7 consecutive days. Artificial estrus of female SD rats were induced by administering estradiol valerat 2 mg/200 g BW 48 hours before observation. The determined parameters were levels of testosterone as well as introduction, climbing and coitus frequencies. Administration of estradiol valerat led to artificial estrus induction of female SD rats. The administration of the combination infusion dose 491.2 mg/200 g BW significantly increased testosterone level as well as introduction and climbing frequencies of male SD rats as of 27.77%; 86.39% and 69.17% respectively. ABSTRAK Som jawa (Talinum paniculatum) secara empiris dan ilmiah terbukti telah banyak dimanfaatkan oleh masyarakat untuk mengatasi berbagai masalah kesehatan diantaranya untuk meningkatkan kualitas dan kuantitas spermatozoa, regenerasi sel, fertilitas, dan sebagai antibakteri. Penelitian ini dilakukan untuk membuktikan efek peningkatan kadar hormon testosteron serta frekuensi introduction, climbing, dan coitus tikus jantan SD dari pengaruh infusa kombinasi som jawa, pegagan, dan temulawak. Hewan uji tikus galur SD dibagi menjadi 2 kelompok, yaitu kelompok kontrol diberikan akuades, sedangkan kelompok perlakuan diberikan infusa ramuan akar som jawa, herba pegagan, dan rimpang temulawak dosis 491,2 mg/200 g bb selama 7 hari berturut-turut. Induksi estrus artifisial tikus betina dengan pemberian estradiol valerat 2 mg/200 g bb dilakukan 48 jam sebelum pengamatan. Parameter yang diukur meliputi kadar testosterone, frekuensi introduction, climbing, dan coitus. Dibandingkan dengan kelompok kontrol, pemberian infusa ramuan tidak berefek pada frekunsi coitus, namun mampu meningkatkan kadar testosteron serta frekuensi introduction dan climbing tikus jantan sebesar 27,77; 86,39 dan 69,17%.

Exposure to paraquat leads to acute lung injury and oxidative stress is widely accepted as a contributor to paraquat-induced acute lung injury. Recent studies have reported that consumption of water with dissolved molecular hydrogen to a saturated level (hydrogen water) prevents oxidative stress-induced diseases. Here, we investigated whether consumption of saturated hydrogen saline protects rats against paraquat-induced acute lung injury. Adult male Sprague-Dawley (SD) rats were randomly divided into four groups: Control group; hydrogen water-only group (HW group); paraquat-only group (PQ group); paraquat and hydrogen water group (PQ  +  HW group). The rats in control group and HW group drank pure water or hydrogen water; the rats in PQ group and PQ  +  HW group were intraperitonealy injected with paraquat (35 mg/kg) and then provided pure water or hydrogen water. Both biochemical and histological lung alterations were measured. The results showed that hydrogen water ameliorated these alterations, demonstrating that hydrogen water alleviated paraquat-induced acute lung injury possibly by inhibition of oxidative damage.

Abstract BackgroundThere is considerable evidence that many patients concurrently take dietary supplements with conventional drugs, with a risk of potential drug-supplement interaction. The aim of this study was to determine the effect of Cellgevity® supplement on selected rat liver cytochrome P450 (CYP) enzymes and on the pharmacokinetics of carbamazepine.MethodsSprague-Dawley (SD) rats were put into 5 groups and modulation of CYP enzyme activity by Cellgevity® was determined by comparing the enzyme activity of Cellgevity-treated groups with the negative control group after 30 days of treatment. For the effect of Cellgevity® on the pharmacokinetics of carbamazepine, 12 SD rats were put into 2 groups; one group received an oral administration of carbamazepine plus Cellgevity®, and the other carbamazepine plus normal saline. Blood samples were collected at specific time points and analyzed for levels of carbamazepine. ResultsActivities of CYP1A1/2, CYP2C9 and CYP2D6 were significantly increased by Cellgevity®. The pharmacokinetic parameters for rats administered carbamazepine with Cellgevity® vis-a-vis carbamazepine with normal saline were changed as follows: Cmax; 20 μmol/L vs 11 μmol/L, AUC0→24; 347 μmol.h/L vs 170 μmol.h/L, Ke; 0.28 h-1 vs 0.41 h-1, and t1/2; 2.3 h vs 1.7 h, respectively.ConclusionsCellgevity® increased the activity of rat CYP1A1/2, CYP2C9 and CYP2D6, and also altered the pharmacokinetics of carbamazepine in rats.

The current study aims to investigate the influence of five rare earth elements (REEs) (i.e., lanthanum (La), cerium (Ce), praseodymium (Pr), neodymium (Nd), and gadolinium (Gd)) on the growth of Sprague–Dawley (SD) rats, and to explore the accumulation characteristics of REEs in tissues and organs with different doses as well as the detoxification and elimination of high-dose REEs. Fifty healthy male SD rats (140~160 g) were randomly divided into five groups and four of them were given gavage of sodium citrate solution with REEs in different doses, one of which was the control group. Hair, blood, and bone samples along with specific viscera tissue samples from the spleen and the liver were collected for detection of REEs by Inductively Coupled Plasma-Mass Spectrometry (ICP-MS). Treated rats expressed higher concentrations of REEs in the bones, the liver, and spleen samples than the control group (P < 0.05). Few differences were found in relative abundance of La, Ce, Pr, Nd, and Gd in the hair and the liver samples, although different administration doses were given. The relative abundance of Ce in bone samples was significantly lower in the low-dose group and control group, whereas the relative abundance of La and Pr in the bone samples were highest among all groups. Although in the REEs solution, which was given to rats in high-dose group, the La element had a higher relative abundance than Ce element, it ended up with higher Ce element relative abundance than La element in the spleen samples. REEs had a hormetic effect on body weight gain of SD rats. The accumulation of the measured REEs were reversible to low concentrations in the blood and hair, but non-reversible in the bones, the spleen, and the liver. Different tissues and organs can selectively absorb and accumulate REEs. Further inter-disciplinary studies about REEs are urgently needed to identify their toxic effects on both ecosystems and organisms.

Objective. To study the effects of Huanglian Jiedu Decoction (HJD) on gut microflora of SD rats. Method. After 3 days of adaptive feeding, 36 rats were randomly divided into 4 groups, namely, the normal control group (NC, 10 mL/kg, distilled water), high_HJD dose group (H_HJD, 6.25 g/kg, weight ratio between crude drug and rat), medium_HJD dose group (M_HJD, 3.125 g/kg), and low_HJD dose group (L_HJD, 1.56 g/kg), and each group consisted of 9 mice. The HJD groups were then treated with orally administered HJD for 21 days, while the NC group was treated with distilled water. After 7 days, 14 days, and 21 days of the experiment and after 12 hours of fasting and water deprivation, 3 SD rats in each group were randomly sacrificed by cervical dislocation and sterile operation to collect stool faces. Sample DNA was extracted by Fecal total DNA extraction kit, sequenced using Illumina MiSeq platform, and analyzed. Results. The abundance of Romboutsia, Turicibacter, Lactobacillus, and Gemella decreased, while that of Escherichia_Shigella, Coprobacillus, Blautia, Akkermansia, Klebsiella, Rhodococcus, Parabacteroides, Citrobacter, Enterococcus, Bacteroides, and Erysipelotrichaceae_incertae_sedis increased after using H_HJD. The abundance of Gemella, Turicibacter, Romboutsia, and Lactobacillus decreased, while that of Blautia, Akkermansia, Escherichia_Shigella, Thiobacillus, Rothia, Enterococcus, Lactobacillus, and Erysipelotrichaceae-incertae-sedis increased after using M_HJD. The abundance of Romboutsia, Turicibacter, Lactobacillus, and Gemella decreased, while that of Escherichia_Shigella, Bacteroides, Akkermansia, Enterococcus, Rhodococcus, Parabacteroides, Desulfovibrio, Blautia, Fusobacterium, Rothia, and Streptococcus increased after using L_HJD. Conclusion. HJD can regulate gut microbiota, and its effect varies with different dosage and duration of medication.

ABSTRACTObjective: To investigate the anti-inflammatory effect of aqueous extract of Zingiber officinale on carrageenan-induced inflammation on SpragueDawley (SD) rats.Methods: SD rats were divided into six of five groups and allowed to acclimatize for 1 week. Inflammation was induced on the animal by injecting theright hand paw with carrageenan (0.1 ml of 1%). Group 1 was given normal saline and served as a control. Group 2 was fed with food and water andserved as the carrageenan control. Group 3 was given 200 mg/kg aqueous extract of ginger, Group 4 with 400 mg/kg aqueous extract of ginger, andGroup 5 with 150 mg/kg diclofenac sodium (reference drug for inflammation).Results: The paw edema in carrageenan-induced SD rats was considerably reduced by treating with 400 mg/kg aqueous ginger extracts whencompared to the untreated SD rats (p<0.001).Conclusion: This study indicates that aqueous extract of Z. officinale possesses anti-inflammatory properties.Keywords: Anti-inflammatory, Sprague Dawley rats, Zingiber officinale, Carrageenan, Edema
