Comparison Among Five Eucalyptus Species Based on Their Leaf Contents of Some Primary and Secondary Metabolites

Background: Eucalyptus belongs to the Myrtaceae family. It is the most planted hardwood forest crop worldwide, representing a global renewable resource of fiber, pharmaceuticals and energy. Objective: To compare the five species, E. maidenii, E. robusta, E. citriodora, E. tereticornis and E. camaldulensis, seeking for the richest source of nutrients and pharmaceuticals. Methodology: Eucalyptus samples were subjected to some chemical determinations for both primary and secondary metabolites to verify their nutritional and pharmaceutical importance related to different extracts. GC-MS analysis was applied to detect the presence of some individual phenolic constituents in their leaves. Results: E. robusta recorded the maximum contents of carbohydrates (40.07%) and protein (31.91%). While E. camaldulensis contained the highest contents of total phenolic compounds (46.56 mg/g), tannins (40.01 mg/g) and antioxidant activities assayed by the phosphomolybednum method (57.60 mg/g), followed by E. citridora. However, E. tereticornis exhibited the highest reducing power ability (151.23 mg/g). The GC-MS highlighted 20 phenolic constituents and antioxidants which varied in their abundance in Eucalyptus leaves, 8 individual phenolics (hydroquinone, hesperitin, pyrogallol, resorcinol, protocatechuic acid, naringenin, chlorogenic acid and catechin) were maximally recorded with E. camaldulensis and secondly, with E. citridora in case of at least 5 components. Nevertheless, gallic and quinic acids were more abundant in the leaves of E. tereticornis, which may explain its high corresponding reducing powers. Conclusion: Acetone-water combination has enhanced phenolics extraction from Eucalyptus tissues. This is the first report aiming to compare between the aforementioned Eucalyptus species highlighting either their nutritional or medicinal importance.

AbstractThis study aimed to comprehensively analyze primary and secondary metabolites of three different-colored (white, pale green, and green) pak choi cultivars (Brassica rapa subsp. chinensis) using gas chromatography attached with time-of-flight mass spectrometry (GC-TOFMS) and high-performance liquid chromatography (HPLC). In total, 53 primary metabolites were identified and subjected to partial least-squares discriminant analysis. The result revealed a significant difference in the primary and secondary metabolites between the three pak choi cultivars. In addition, 49 hydrophilic metabolites were detected in different cultivars. Total phenolic and glucosinolate contents were highest in the pale green and green cultivars, respectively, whereas total carotenoid and chlorophyll contents were highest in the white cultivar. Superoxide dismutase activity, 2,2-diphenyl-1-picrylhydraz scavenging, and reducing power were slightly increased in the white, pale green, and green cultivars, respectively. In addition, a negative correlation between pigments and phenylpropanoids was discovered by metabolite correlation analysis. This approach will provide useful information for the development of strategies to enhance the biosynthesis of phenolics, glucosinolates, carotenoids, and chlorophyll, and to improve antioxidant activity in pak choi cultivars. In addition, this study supports the use of HPLC and GC-TOFMS-based metabolite profiling to explore differences in pak choi cultivars.

Triterpene acid and phenolic constituents from nine ancient varieties of apple (Malus domestica) fruits cultivated in Fanna, Friuli Venezia Giulia region, northeast Italy, were analyzed and compared with four commercial apples (‘Golden Delicious’, ‘Red Delicious’, ‘Granny Smith’ and ‘Royal Gala’). Total phenolic and flavonoid contents were measured by spectrophotometric assays. The quali-quantitative fingerprint of secondary metabolites including triterpene acid was obtained by LC-DAD-(ESI)-MS and LC-(APCI)-MS, respectively. Based on the two LC-MS datasets, multivariate analysis was used to compare the composition of ancient fruit varieties with those of four commercial apples. Significant differences related mainly to the pattern of triterpene acids were found. Pomolic, euscaphyc, maslinic and ursolic acids are the most abundant triterpene in ancient varieties pulps and peels, while ursolic and oleanolic acids were prevalent in the commercial fruits. Also, the content of the phenolic compounds phloretin-2-O-xyloglucoside and quercetin-3-O-arabinoside was greater in ancient apple varieties. The antioxidant (radical scavenging, reducing power, metal chelating and phosphomolybdenum assays) and enzyme inhibitory effects (against cholinesterase, tyrosinase, amylase and glucosidase) of the samples were investigated in vitro. Antioxidant assays showed that the peels were more active than pulps. However, all the samples exhibited similar enzyme inhibitory effects. Ancient Friuli Venezia Giulia apple cultivars can be a source of chlorogenic acid and various triterpene acids, which are known for their potential anti-inflammatory activity and beneficial effects on lipid and glucose metabolism. Our results make these ancient varieties suitable for the development of new nutraceutical ingredients.

Antioxidant activities of wampee peel extracts using five different solvents (ethanol, hexane, ethyl acetate, butanol and water) were determined by using in-vitro antioxidant models including total antioxidant capability, 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity, reducing power, and superoxide scavenging activity. Ethyl acetate fraction (EAF) exhibited the highest antioxidant activity compared to other fractions, even higher than synthetic antioxidant butylated hydroxyl toluene (BHT). In addition, the EAF exhibited strong anticancer activities against human gastric carcinoma (SGC-7901), human hepatocellular liver carcinoma (HepG-2) and human lung adenocarcinoma (A-549) cancer cell lines, higher than cisplatin, a conventional anticancer drug. The total phenolic content of wampee fraction was positively correlated with the antioxidant activity. This is the first report on the antioxidant and anticancer activities of the wampee peel extract. Thus, wampee peel can be used potentially as a readily accessible source of natural antioxidants and a possible pharmaceutical supplement.

The antioxidant activities of crude extract ofPhaulopsis fascisepalaleaf were evaluated and compared with α-tocopherol and BHT as synthetic antioxidants and ascorbic acid as natural-based antioxidant.In vitro, we studied its antioxidative activities, radical-scavenging effects, Fe2+-chelating ability and reducing power. The total phenolic content was determined and expressed in gallic acid equivalent. The extract showed variable activities in all of thesein vitrotests. The antioxidant effect ofP. fascisepalawas strongly dose dependent, increased with increasing leaf extract dose and then leveled off with further increase in extract dose. Compared to other antioxidants used in the study, α-Tocopherol, ascorbic acid and BHT,P. fascisepalaleaf extract showed less scavenging effect on α,α,-diphenyl-β-picrylhydrazyl (DPPH) radical and less reducing power on Fe3+/ferricyanide complex but better Fe2+-chelating ability. These results revealed thein vitroantioxidant activity ofP.fascisepala.Further investigations are necessary to verify these activitiesin vivo.

Acacia nilotica (L.) Delile is well known as “Desi Kikar”or Babul in India that possesses a wide range of pharmacological activities. In the present study, Acacia nilotica twig extract and its synthesized silver nanoparticles (AgNPs) were evaluated for total phenolic content (TPC), antioxidant activity and cytotoxic effects. Characterization of AgNPs was done by UV-Visible spectroscopy, Fourier-transform infrared spectroscopy (FTIR), Transmission electron microscopy (TEM) and Selected area electron diffraction (SAED) techniques. Antioxidant potential was determined using different assays including 2,2-diphenyl-1-picrylhydrazyl (DPPH), reducing power and β-carotene linoleic acid. Cytotoxicity was tested by 3-(4,5-dimethyl-2-yl)-2,5-diphynyl tetrazolium bromide (MTT) assay on Human embryonic kidney (HEK)-293 cell lines. The results indicated that AgNPs exhibited higher antioxidant activity (81.11 %) and TPC (57.35 mg of GAE/mL of extract) as compare to plant extract. A positive correlation was observed between the TPC and antioxidant activities. The inhibitory concentration (IC50) of A. nilotica extract and AgNPs was 52.08µg/mL and 56.82µg/mL respectively. Cytotoxicity against HEK-293 cell lines was dose dependent. Accordingly, it is summarized that A. nilotica based AgNPs could serve as a potential antioxidant for therapeutic purposes.

Zilla spinosa is commonly used in traditional medicine to treat gastrointestinal disorders and diabetes. In this study, aqueous ethanol (AE) and aqueous methanol (AM) extracts from aerial parts and roots of Z. spinosa were investigated. The total phenolic (TPC) and flavonoid (TFC) contents and antioxidant capacities in terms of 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging, hydrogen peroxide (H2O2), and ferric reducing antioxidant power (FRAP) assays were determined, and the correlations among the results were assessed using Pearson’s correlation. The antimicrobial activity was assessed through agar disc diffusion and broth microdilution assays. Phytochemical screening showed that Z. spinosa extracts had alkaloids, glycosides, saponins, triterpenoids, phenols, and flavonoids in different abundances. The aerial part-AE extract contained low TPC (30.17 ± 4.24 mg GAE/g) and TFC (7.40 ± 1.02 mg QE/g) and displayed significant antioxidant capacity in the DPPH (IC50 = 52.17 ± 7.30 μg/mL), H2O2 (91.22 ± 2.60 μg/mL), and FRAP (EC50 = 98.70 ± 2.21 μg/mL) assays. By contrast, the root-AM extract contained high amounts of TPC (87.72 ± 7.75 mg GAE/g) and TFC (25.60 ± 1.57 mg QE/g). It showed significantly high antioxidant activity with IC50 values of 12.33 ± 1.88 μg/mL in the DPPH and 39.37 ± 2.59 μg/mL in the H2O2 assays, as well as reducing power capacity with an EC50 value of 20.82 ± 1.14 μg/mL in the FRAP assay. Both TPC and TFC were exhibited negative correlations ( p < 0.01 ) with the IC50 and EC50 values obtained in the applied antioxidant assays. The aerial part-AM extract showed the highest inhibitory activity against Staphylococcus aureus (26.5 ± 0.20 mm), followed by Shigella flexneri (19.4 ± 0.40 mm) and Proteus mirabilis (17.7 ± 0.49 mm). S. aureus was the most affected microorganism with a minimum inhibitory concentration of 128 μg/mL against the aerial part-AM extract. Interestingly, all evaluated extracts showed potent antifungal activity against Candida albicans. However, aerial part-AM was the most effective, with an inhibition zone of 12.6 ± 0.17 mm. The results concluded that Z. spinosa possesses different phytochemicals displaying significant antioxidant and antimicrobial activities, thus lending credence to its use in traditional medicine.

In the present study, we aimed to develop a novel fermented tea (NFT) product and to evaluate their in vitro antioxidant potential and chemical composition. We found that NFT contained a high level of total phenolic compounds (102.98 mg gallic acid equivalents/g extract) and exhibited diverse antioxidant activities, such as scavenging of 1,1-diphenyl-2-picryl-hydrazyl (DPPH) and hydroxyl radicals, as well as reducing power. The total catechins in NFT were comparable to those of Lipton black tea (LBT), but lower than those of Boseong green tea (BGT) or Tieguanyin oolong tea (TOT). Among all catechins tested, epigallocatechin (EGC) and epigallocatechin-3-O-gallate (EGCG) were the predominant compounds in NFT. In particular, the contents of total theaflavins (TFs), theaflavin (TF), theaflavin-3-gallate (TF3G), and theaflavin-3’-gallate (TF3’G) in NFT were significantly higher than that of BGT, TOT, or LBT. NFT had the highest level of total essential amino acid and γ-aminobutyric acid (GABA) compared with BGT, TOT and LBT. Furthermore, the sensory evaluation results showed that NFT had satisfactory color, aroma, taste, and overall acceptability scores. Our results highlight the potential usefulness of this novel fermented tea as a nutraceutical food/ingredient with special functional activities.

A comparative study of antioxidant properties on two varieties of Phyllanthus (Phyllanthus niruiri L. & Phyllanthus urinaria L. leaves) growing in Bangladesh was done in the present study. Five complimentary test methods namely DPPH free radical scavenging activity, reducing power assay, total antioxidant capacity, total phenolic and flavonoid contents determination were used for the evaluation of antioxidant properties of two species of Phyllanthus. In the present investigation for the evaluation of antioxidant activities the concentration is expressed by the ratio of crude sample per solvent volume instead of extract weight per solvent volume. Based on these concentrations the investigated P. niruri L. and P. urinaria L. leaves extracts exhibited significant results. At 2 mg/ml concentration DPPH radical scavenging capacity of the methanol extract of both the plants was found to show significant (>90%) activity which is comparable to Ascorbic acid and BHA. In case of reducing power tests for both the extract, the activity of P. urinaria is comparable to that of BHA and AA and that of P. niruiri is comparable to BHA. P. niruiri showed better antioxidant potential than that of P. urinaria based on the observed results of their corresponding methanol extract. Hypochaeris laevigata var. hipponensis (Asteraceae) is an endemic plant from Algeria. In the current study, we analyzed for the first time its chemical composition, especially phenolic constituents of dichloromethane (DCM), ethyl acetate (EA), and n-butanol (BuOH) fractionsof the aerial parts of Hypochaeris laevigata var. hipponensis by liquid chromatography-mass spectrometry (LC-MS/MS). The number of phenolic compounds detected in DCM, EA, and BuOH fractions were found to be 9, 20, and 15, respectively. More specifically, 12 phenolic acids were detected. Among them, quinic acid, chlorogenic acid, and caffeic acid were the most abundant ones. Meanwhile, only seven flavonoids were detected. Among them, rutin, apigetrin, and isoquercitrin were the major ones. We also determined the total phenolic and flavonoid contents, and fraction EA showed the highest values, followed by BuOH, and DCM fractions. Furthermore, the antioxidant action was dictated by five methods and the tested plant fractions demonstrated a noteworthy antioxidant action.

This study addresses the favourable effects of drought stress imposed by polyethylene glycol (PEG) 6000 on the micropropagated shoots of Stevia rebaudiana. Various concentrations, i.e., 0, 0.5, 1, 2, and 4% of PEG 6000 were applied to the nodal shoot explants for four weeks, and the influence produced on shoots growth parameters, bioactive steviol glycosides (rebaudioside A and stevioside), and nonenzymatic antioxidant activities (total phenolic content (TPC), total flavonoid content (TFC), total antioxidant capacity (TAC), total reducing power (TRP) and 1,1-diphenyl-2-picrylhydrazyl(DPPH)-free radical scavenging activity (FRSA)) was elucidated. The significantly highest yield (92.4% direct shoot organogenesis) and secondary metabolites (2.94% Reb A, 2.52% ST, 95.3% DPPH-FRSA, 15.0% TPC, 13.0 µg/mg TFC, 22.3 µg/mg TAC, and 19.8 µg/mg TRP) production in response to abiotic stress elicitors was obtained in Murashige and Skoog (MS) medium treatment provided with 4% of PEG 6000. The overall trend was significant enhancement of growth dynamics and pharmaceutical compounds from control to 4% of PEG 6000 concentration as a defensive response against reactive oxygen species (ROS) produced in excess by water deficit. This is a very promising study to be employed in bioreactors to get markedly enhanced content of compounds of medicinal importance in the pharmaceutical market.
