Kadam (Anthocephalus cadamba) is one types of three possible to develop in plantation forest or public forest to suply log necessity in Indonesia.  However, the silviculture information on kadam development is still limited especially in providing nurseries.  The objective of this research was to find out survival percentage, shooting ability, and rooting ability of kadam shoot cutting with some concentrations of Rootone-F addmited.  This research used completely randomized design with four treatments and five replications. Treatments were soaking the bud of kadam shoot cuttings with some of the following Rootone-F concentrations: 0 ppm, 100 ppm, 200 ppm, and 300 ppm.  Variables to observe were survival percentages, shoot lengths, shoot diameters, root lengths, and leaf numbers.  The results showed that Rootone-F addmited with 200 ppm concentration resulted in the best shoot height, root length, and leaf numbers of kadam shoot cuttings compared with concentrations of 1 ppm, 100 ppm, and 300 ppm.  The kadam shoot cutting with 200 ppm Rootone-F concentration had 96% survival, 20.47 cm shoot length, 19.60 root length, and 6.18 leafs.  The conclusion was that the concentration of 200 ppm Rootone-F gives the best results.

This study was aimed at determining the response of turmeric shoot explants after the provision of various types and concentrations of cytokinins in vitro. This experiment was conducted at the Tissue Culture Laboratory, Faculty of Agriculture, Padjadjaran University from January to April 2018. The data were analyzed using T-Test. The experimental method used in this research was Completely Randomized Design (CRD). Explant planting was carried out in Laminar Air Flow. The experiment consisted of 7 treatments consisting of 4 replications and each test consisted of 4 units. Observation of this experiment was carried out for 12 MST. The main observations were made on the data that were tested statistically namely the percentage of explant growing shoots, percentage of explant growing roots, shoot height, number of tuns, number of roots and root length. The treatments consisted of Control, 2.5 mg L-1 BAP, 5 mg L-1 BAP, 0.5 mg L-1 TDZ, 1 mg L-1 TDZ, 0.01 mg L-1 Zeatin and 0.1 mg L Zeatin -1. The results show that the treatment of 1 mg L-1 TDZ shows the best response to the growth of turmeric explants by increasing the number of turmeric shoot explants than the others.RESPONS EKSPLAN TUNAS KUNYIT SETELAH SITOKININ SECARA IN VITROTujuan dari penelitian ini yaitu untuk melihat respons eksplan tunas kunyit terhadap pemberian berbagai jenis dan konsentrasi sitokinin secara in vitro. Percobaan ini dilakukan di Laboratorium Kultur Jaringan, Fakultas Pertanian, Universitas Padjadjaran dari bulan Januari sampai April 2018. Hasil percobaan dianalisis dengan Sample T-Test. Metode percobaan yang digunakan dalam penelitian ini yaitu Rancangan Acak Lengkap (RAL). Penanaman eksplan dilakukan di dalam Laminar Air Flow. Percobaan terdiri dari 7 perlakuan sebanyak 4 ulangan dan setiap ulangan terdiri dari 4 unit. Pengamatan percobaan ini dilakukan selama 12 MST. Pengamatan utama dilakukan terhadap data-data yang diuji secara statistik yakni persentase eksplan tumbuh tunas, persentse eksplan tumbuh akar, tinggi tunas, jumlah tunas, jumlah akar dan panjang akar. Perlakuan terdiri dari Kontrol; 2,5 mg L-1 BAP; 5 mg L-1 BAP; 0,5 mg L-1 TDZ; 1 mg L-1 TDZ; 0,01 mg L-1 Zeatin; dan 0,1 mg L-1 Zeatin. Hasil penelitian menunjukkan bahwa perlakuan 1 mg L-1 TDZ menunjukkan respons yang lebih baik terhadap pertumbuhan eksplan kunyit dengan meningkatkan jumlah tunas eksplan tanaman kunyit daripada yang lainnya.

[STIMULATION EFFECTS OF BIOURINE CONCENTRATION AND THE STEM SEGMENT OF CUTTINGS ON  GROWTH OF TEA (Camellia sinensis) SEEDLINGS]. Tea seedlings can be prepared by the stem segment cutting and provide treatment to stimulate the growth of cuttings. This study aims to determine the optimal tea seedlings growth from different material of stem segment and biourine concentration. The experiment was conducted in February to June 2020 at PT. Sarana Mandiri Mukti Kelurahan Tangsi Baru, Kabawetan District, Kepahiang Regency, Bengkulu Province. This research uses factorial completely randomized design (CRD) with two factors. The first factor, biourine concentration consisting of 5 levels including 0%, 25%, 50%, 75%, 100%. The second factor, the stem segment of cuttings consisting of 3, 4, and 5 segments. The treatment was repeated 3 times, with 5 plants sample in each treatment. There was totally  225 plants. The results showed that biourine concentration of 100% has significantly different from the variable roots number and root length. The stem segment of the tea cuttings has no significantly difference  effect on all variables. The interaction between biourine concentration and stem segment of the tea cuttings has significantly different in variables of shoot length and seedlings fresh weight. The optimum growth of tea seedling was obtained in stem cutting of 3 segment and application of biourine concentration of 100%.

ABSTRACT: This study determined the effect of vermicompost humates as salinity mitigator in germination and morphometric characteristics of basil varieties seedlings, tolerant and sensitive to salinity, subjected to vermicompost humates diluted 1/60 (v/v) and 0, 50 and 100 mM NaCl in a completely randomized design with four replications. The rate and percentage of germination, root length, shoot height, root and shoot fresh and dry-weight were measured. The varieties showed differential response, highlighting Napoletano as the most tolerant. Vermicompost humates showed biostimulant effects on variables evaluated, allowing that tolerant variety to improve germination and growth, and increasing the salinity tolerance of the sensitive variety.

Abstract The demand for eucalypts has greatly increased since they are raw materials for timbers, plywoods, pulp and papers, and essential oil production. This study aimed to select the plus trees and determine the sprouting and rooting ability of E. pellita, E. brassiana, and their hybrids in Wanagama. Plus trees selection and girdling were conducted in December 2019. Cuttings were picked from the 3rd-4th nodes of sprouting at 8-10 cm in length. A completely randomized design was applied in a factorial experiment with three levels of Indole Butyric Acid (IBA) (100, 1000, and 4000 ppm) and two types of media (media 1: sand, rice husk charcoal and cocopeat (2:2:1), media 2: sand, topsoil and dung-fertilizer (2:2:1)), with ten replications. Results selected a total of 53 plus trees candidates (29 of E. pellita, 12 of E. brassiana, and 12 of hybrid). E. pellita is the best in sprouting ability (100% survival; 41-60 shoot/trees, 127-161cm shoot length, 0.53-0.57cm shoot diameter, and 194-252 leaves/shoot). Cuttings of E. pellita treated with 100 ppm IBA gained the highest survival and leaves production (96%-100%; 14-16 leaves), followed by E. brassiana (52%-66.67%; 7 leaves) and the hybrid (4% to 8.33%; 3-4 leaves). These results may contribute to arranging better strategies for mass production of selected eucalypts.

Abstrak: Tanaman sambung nyawa (Gynura procumbens) dikenal sebagai salah satu tanaman yang bermanfaat bagi kesehatan tubuh. Daun tanaman ini diketahui mempunyai efek anti peradangan yang dapat mengurangi bahkan mengobati berbagai macam penyakit. Penelitian ini berfokus pada pemilihan bagian batang tanaman sambung nyawa yang mempunyai kapasitas terbaik sebagai eksplan stek. Penelitian ini dilakukan dengan menanam 3 bagian batang tanaman sambung nyawa yang berbeda, yaitu bagian atas (pucuk sampai dengan 10 cm ke bawah), tengah (bagian batang yang berjarak 10cm dari pucuk sampai 20 cm), dan bawah (bagian batang yang berjarak 20 cm dari pucuk hingga 30 cm). Ketiga jenis eksplan ini ditanam di tanah yang mengandung humus tanpa diberi perlakuan penambahan ZPT. Penelitian ini menggunakan rancangan acak lengkap dengan sepuluh ulangan. Hasil penelitian menunjukkan bahwa tidak ada perbedaan yang nyata antara bagian batang dengan panjang daun, tetapi terdapat perbedaan yang nyata dengan lebar daun, panjang tunas, dan panjang akar. Hasil penelitian ini diharapkan dapat menjadi rujukan bagian batang “tengah” dan atau “bawah” sebagai eksplan terbaik dalam usaha perbanyakan tanaman sambung nyawa menggunakan stek. Kata kunci: organogenesis, perbanyakan, sambung nyawa, stek.Abstract: Sambung nyawa plant (Gynura procumbens) is known as one of the plants that are beneficial for human health. The leaves of this plant are known to have anti-inflammatory effects that can reduce and even treat various diseases. This research focused on the selection of the stem part of sambung nyawa plant which has the best capacity as explant cuttings. This research was carried out by planting three different parts of the stem of the plant, namely the upper part (shoots apical up to 10 cm down part), the middle (the stem part that is 10cm from the top to 20 cm down part), and the bottom (the part of the stem which is 20 cm apart from shoots up to 30 cm down part). These three types of explants were planted in soil containing humus without being treated with the addition of PGR(s). This study used a completely randomized design with ten replications. The results showed that there was no significant differences between the stem parts with leaf length parameter, but there were significant differences in leaf width, shoot length, and root length ones. The results of this study were expected to be a reference part of the stem "middle" and or "bottom" as the best explant in the effort to propagate the sambung nyawa plants using cuttings.Keywords: organogenesis, propagation, sambung nyawa, cuttings.

This study aims to determine the response of the immersion time of vanilla stem cuttings in the most root growth regulating agent and to know the length of soaking time of the vanilla stem cuttings in the most optimum root growth regulator. The design used in this study was a Completely Randomized Design (CRD) with six treatments soaking in minutes. Repeated four times, namely P1 (10 minutes), P2 (20 minutes), P3 (30 minutes), P4 (40 minutes), P5 (50 minutes), P6 (60 minutes). Variables observed including the number of days of first shoot growth, the number of roots, root length, shoot diameter, shoot length, wet weight of cuttings, and dry weight of cuttings.The results showed that the treatment of P6 gave a very significant effect. On the variable growth in the number of roots (55.59%), root length (58.86%), number of days of first shoot growth (28 HST), shoot diameter (70.15%) , shoot length (71.43%), wet weight of cuttings (37.14%) and dry weight of cuttings (65.04%) at each age of observation.

The purpose of this research is to determine the concentration of young coconut water that is appropriate for the growth of orchid plantlets (Dendrobium sp.) In vitro. This study used an experimental method, descriptive data analysis to compare several different concentrations of coconut water. The design of this study uses a completely randomized design (CRD). The treatments consist of 0% coconut water concentration (as a control), 15%, 30% and 60%. Each concentration was carried out 5 replications and each repetition consisted of 5 Dendrobium sp plantlets in each culture bottle conducted for 40 HST, for observing the root length carried out for 50 HST. The highest number of shoots and leaves were produced at the same concentration, namely 150 ml / L coconut water treatment (15% concentration) with an average of 2.8 shoots and the average number of leaves 10.8 leaves. The average number of roots and the longest root length was produced at a concentration of 600 ml / L coconut water (60% concentration) with an average of 6 roots, and the longest root length was 0.5 cm.Keywords: Young coconut water, (Cocos nucifera L.), Dendrobium sp., in vitro, growth.ABSTRAKTujuan penelitian ini ialah menentukan konsentrasi air kelapa muda yang tepat untuk pertumbuhan planlet anggrek (Dendrobium sp.) secara in vitro. Penelitian ini menggunakan metode eksperimen, analisis data secara deskriptif untuk membandingan beberapa konsentrasi air kelapa yang berbeda. Rancangan penelitian ini menggunakan Rancangan Acak Lengkap (RAL). Perlakukan terdiri dari konsentrasi air kelapa 0 % (sebagai kontrol), 15% , 30% dan 60%. Masing-masing konsentrasi dilakukan 5 kali ulangan dan setiap ulangan terdiri dari 5 planlet Dendrobium sp dalam setiap botol kultur yang dilakukan selama 40 HST, untuk pengamatan panjang akar dilakukan selama 50 HST. Jumlah tunas dan jumlah daun terbanyak dihasilkan pada konsentrasi yang sama, yaitu perlakuan air kelapa 150 ml/L (konsentrasi 15%)  dengan rata-rata jumlah tunas terbanyak 2,8 tunas dan rata-rata jumlah daun terbanyak 10,8 helai daun. Rata-rata jumlah akar terbanyak dan panjang akar terpanjang dihasilkan pada konsentrasi air kelapa 600 ml/L (Konsentrasi 60%) dengan rata-rata jumlah akar terbanyak sebanyak 6 akar, dan rata-rata panjang akar terpanjang 0,5 cm.Kata kunci : Air kelapa Muda (Cocos nucifera L.), Dendrobium sp., in vitro, pertumbuhan

This study aimed to determine the best dosage of goat manure as a natural fertilizer on Beha grass(Brachiaria humidicola). This study used 64 poles of Beha grass grown in polybags divided into 16plots, and it used a completely randomized design (CRD) to analyze the data. This study consist of 4treatments that is P0 (0 ton/ha of goat manure), P1 (10 ton/ha of goat manure), P2 (15 ton/ha of goatmanure), and P3 (20 ton/ha of goat manure). This study's variables were plant height, number of leaves,numbers of tillers, new production, and root length. Data analysis used ANOVA variance continued withreal honest difference test. The result of this study showed that averages of plant height (cm) wereP0=26, P1=50.5, P2=51.5, and P3=52.75. Averages of the number of leaves were P0=22, P1=110,P2=160, and P3=185. Averages of numbers of tillers were P0=7, P1=25, P2=37, and P3=38. Averages ofnew production (g) were P0=17.75, P1=123.5, P2=178, and P3=192.25. Averages of root length (cm)were P0=37.8, P1=39.5, P2=41.775, and P3=49.5. Fertilization with natural fertilizer from goat manurehas a genuine effect (P<0.01) on the growth of Beha grass. The best dosage of fertilization with goatmanure was 15 ton/ha.

An experiment was carried out at Navsari Agricultural University, Navsari during 2014 to evaluate the effect of pre-sowing treatments on survival percentage and growth of mango rootstocks. Mango stones were soaked in aqueous solutions of GA3 (100 and 200 ppm), Beejamruth (2 % and 3 %) and Thiourea (1 % and 2 %) for 12 and 24 hours. The trial was evaluated in Completely Randomized Design based on factorial concept and the treatments were replicated thrice. Imposition of treatments led to significant differences at 5 % level of significance for all parameters chosen in this study. Mango stones when treated with Thiourea at 1 % had the maximum shoot length (49.93 cm), root length (34.38 cm), shoot dry weight (21.08 g) and total dry weight (26.36 g). The highest number of lateral roots (10.90) and survival percentage (64.17) was observed in mango stones dipped in 100 ppm GA3. Between the two soaking duration, soaking mango stones for 24 hours recorded higher values for shoot length (45.03 cm), root length (32.79 cm), number of lateral roots (9.83), survival percentage (62.72), shoot root fresh weight ratio (4.30), shoot dry weight (21.33 g), total dry weight (26.28 g) and shoot root dry weight ratio (4.32). Thus, survival percentage and growth of mango rootstocks can be substantially improved by soaking mango stones in aqueous solutions of 100 ppm GA3 or Thiourea at 1 % for 24 hours before sowing.

<p>ABSTRACT</p><p>The objectives of this study were to study the effect of several concentrations of polyethylene glycol (PEG) on the in vitro growth of eggplant, to find the appropriate PEG concentration for in vitro selection to drought  tolerance  of eggplant  and the drought tolerant eggplant accessions. The experiment  was conducted  at  the  Laboratory  of  Tissue  Culture,  Department  of  Agronomy and Horticulture,  Bogor  Agricultural  University.  The  experiment  was arranged  in  a  completely randomized design with two factor. The first factor was concentration of PEG (0, 5, 10,  and  15%) while the second factor was eggplant accessions (Kania F1, 001, 007, 013, 016, 030, 034, 035, 055, 057, 069,  071,  072,  078,  085,  and  090).  The  results  showed  that  the addition  of PEG  to  in  vitro media significantly affected the survival percentage, the percentage of callus, developed the bud and the number of leaves of eggplant. Addition of PEG 10 and 15% in media can be used as the drought tolerance selective agent of eggplant in vitro. Penelitian ini dilaksanakan di laboratorium Kultur Jaringan,  Departemen Agronomi dan Hortikultura,  Institut  Pertanian  Bogor.  Penelitian  ini  disusun dalam  rancangan  acak  lengkap  dua faktor. Faktor pertama adalah konsentrasi PEG  terdiri atas  0, 5, 10, dan 15%.  Faktor kedua adalah nomor terung terdiri atas enam belas nomor (Kania F1, 001, 007, 013, 016, 030, 034, 035, 055, 057, 069,  071,  072,  078,  085,  dan  090).  Hasil  penelitian menunjukkan  bahwa  penambahan  PEG  pada media  in  vitro  memberikan pengaruh  nyata  dan  sangat  nyata  terhadap  persentase  hidup eksplan, persentase  eksplan  berkalus,  pertambahan  tinggi  tunas,  dan jumlah  daun  tanaman  terung.  Media PEG 10 dan 15% merupakan media yang dapat digunakan untuk seleksi kekeringan tanaman terung in vitro. Nomor terung Kania F1, 001, 007, 016, 034, 035, 055, 057, 069, 071, 072, 078, 085, dan 090 merupakan nomor-nomor terung yang toleran terhadap cekaman kekeringan.</p><p>Kata kunci: kultur jaringan, seleksi in vitro, solanaceae, toleran kekeringan</p>
