The phenolic compound and other active compounds were extracted from cinnamon, in anattempt to produce a drug from Medical and herbal Plants for the diabetes on rats. Theextraction process included two methods the first carrid out with 2% v/v acetic acid andpropanol, and in the second process 70% v/v ethanol and chloroform were used. In the detectionof extracts it was noticed that both conteined phenols, flavonoids, tannins and Glycosides andCoumarins, while the saponin showed up only in the alcoholic extract of cinnamon. The phenoliccompounds in the extracts of cinnamon were determined by using Ultraviolet Spectroscopy (UV)and other function groups such as phenols, aldehyedes, estars, hydrocarbons, alcohols andcarboxylic compounds were diagnosed by using Infrared spectroscopy (IR). The study of the halflethal dose (midl lethal) (LD50) was also examined by testing the toxicity of the two cinnamon 2015 مجلة مركز بحوث التقنيات االحيائية المجلد التاسع- العدد االول42extracts. Rats were used as laboratory animals in conducting the experiments. The rats were fedby the dose (100, 200, 300,400, 500 mg / kg of body weight). The results have shown that there isno toxic effect in the cinnamon extracts. Similar concentrations were used to determine theeffective dose of the extracts in reducing the level of glucose in the blood of healthy animals. Thedose (300) mg / kg of body weight of the animal body has proved to be the the most efficient as itdecreased. The fasting glucose (Glu) serum consider by following of the cinnamon extracts withthe adopted groups during the duration of (30) days in comparison with the control group (theinfected) and outperformed the treated given Glibenelinide (daonil) group. The biochemical testsshowed that the serum concentrations of each of cholesterol (TC),Triglyceride (TG), low densitylipoprotein (LDL-C) was lowered in each of the groups treated with both cinnamon extracts incomparison with the positive groups (infected) throughout the duration (30) days and overcomethe daonil treated group. However the serum high density lipoprotein (HDL-C) level wasincreased in cinnamon extract treated groups and decreased in the control group (infected).The serum concentrations of enzymes (GOT) and (GPT) were measured to test the liver functionafter where their levels were lowered after (30) days in the cinnamon extracts administeredgroups compared with the two other groups, the control (positive) and the Daonil. The Urea andCreatinine levels were measured to determine or test the kidney functions, where it was observedas from the early days of the infection in that animals infected by induced diabetes developedsymptoms of the illness, apparent in the high levels of urine which affected the functions ofkidney, but the long duration (30 days) of treatment with these extracts of cinnamon proved theirefficiency over the control group (positive).

Excess body weight is a major risk factor for type 2 diabetes (T2D) and associated metabolic complications, and weight loss has been shown to improve glycemic control and decrease morbidity and mortality in T2D patients. Weight-loss strategies using dietary interventions produce a significant decrease in diabetes-related metabolic disturbance. We have previously reported that the supplementation of low molecular chitosan oligosaccharide (GO2KA1) significantly inhibited blood glucose levels in both animals and humans. However, the effect of GO2KA1 on obesity still remains unclear. The aim of the study was to evaluate the anti-obesity effect of GO2KA1 on lipid accumulation and adipogenic gene expression using 3T3-L1 adipocytes in vitro and plasma lipid profiles using a Sprague-Dawley (SD) rat model. Murine 3T3-L1 preadipocytes were stimulated to differentiate under the adipogenic stimulation in the presence and absence of varying concentrations of GO2KA1. Adipocyte differentiation was confirmed by Oil Red O staining of lipids and the expression of adipogenic gene expression. Compared to control group, the cells treated with GO2KA1 significantly decreased in intracellular lipid accumulation with concomitant decreases in the expression of key transcription factors, peroxisome proliferator-activated receptor gamma (PPARγ) and CCAAT/enhancer-binding protein alpha (CEBP/α). Consistently, the mRNA expression of downstream adipogenic target genes such as fatty acid binding protein 4 (FABP4), fatty acid synthase (FAS), were significantly lower in the GO2KA1-treated group than in the control group. In vivo, male SD rats were fed a high fat diet (HFD) for 6 weeks to induced obesity, followed by oral administration of GO2KA1 at 0.1 g/kg/body weight or vehicle control in HFD. We assessed body weight, food intake, plasma lipids, levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) for liver function, and serum level of adiponectin, a marker for obesity-mediated metabolic syndrome. Compared to control group GO2KA1 significantly suppressed body weight gain (185.8 ± 8.8 g vs. 211.6 ± 20.1 g, p < 0.05) with no significant difference in food intake. The serum total cholesterol, triglyceride, and low-density lipoprotein (LDL) levels were significantly lower in the GO2KA1-treated group than in the control group, whereas the high-density lipoprotein (HDL) level was higher in the GO2KA1 group. The GO2KA1-treated group also showed a significant reduction in ALT and AST levels compared to the control. Moreover, serum adiponectin levels were significantly 1.5-folder higher than the control group. These in vivo and in vitro findings suggest that dietary supplementation of GO2KA1 may prevent diet-induced weight gain and the anti-obesity effect is mediated in part by inhibiting adipogenesis and increasing adiponectin level.

Aim of the Study: This study was undertaken to ascertain if Hibiscus sabdariffa extract can affect the lipid profile (Total cholesterol (TC), triglycerides (TG), high density lipoprotein (HDL), very low density lipoprotein (VLDL), and low density lipoprotein (LDL)) levels in a paracetamol- induced hepatotoxicity using albino Wistar rat as a model. Materials and Methods: Thirty (30) rats used for this study were divided into three groups. Group A (n=10) served as control. Group B (n=10) was administered paracetamol only at a dose of 750 mg/kg body weight. Group C (n=10) was administered paracetamol (dose 750 mg/kg body weight) and aqueous extract of H. sabdariffa (dose 10 ml/kg body weight) of the animal for 3 weeks. All animals were allowed free access to clean drinking water and normal rat chow. Results: Results of the study revealed that TC was significantly lower (p<0.05) in the paracetamol + H. sabdariffa-treated group as compared to paracetamol-treated group and control respectively. Similar trend was observed with TG, VLDL-c, LDL-c and HDL-c. However, the decrease in HDL-c was not statistically significant when compared to control. Conclusion: The presence of bioactive constituents vis; anthocyanins, flavonoids, polyvenols and free radical scavenging properties in H. sabdariffa enabled a hypolipidemic effect on the animals by lowering the levels of serum TG, VLDL-c, LDL-c despite challenge on the liver. However, it was unable to produce significant effect on HDL concentration -very important cholesterol required in high level to maintain homeostasis inside the body. This may be due to the challenge on the liver as a result of the paracetamol abuse.

Objective: To evaluate anti-obesity effect of aqueous P. nigrum extract in a murine model of high-fat diet (HFD) induced obesity. Methods: Male Wistar rats were fed with a high-fat diet (HFD) (20g/day/rat) for a period of 50 d to induce obesity. Aqueous P. nigrum extract (20 mg/kg) administered orally to high-fat diet (HFD) fed rats from 8th day to 50th day (total 42 d). The parameters like gain in body weight, serum lipids, insulin and leptin were measured. Results: The rats treated with extract showed a significant reduction in body weight gain, serum insulin, leptin, lipids as compared to rats fed with only a high-fat diet (HFD). In addition, the extract-treated group showed a considerable rise in high-density lipoprotein (HDL-C) level (29.61±7.68 mg/dl) as compared to the control group (23.23±9.69 mg/dl). Conclusion: The results indicate that aqueous P. nigrum extract possess the potential to reduce obesity markers in a high-fat diet (HFD) fed rats.

Background: Cardiotoxicity can be developed due to prolonged use of higher doses of some drugs, exposure to some chemicals, toxins or infectious agents and also by some disease conditions. Natural plant food such as peanut (Arachis hypogaea L.) may have free radical scavenging and lipid lowering activity, thereby, can be used for the prevention and management of heart disease. Objective: To observe the protective effect of peanut (Arachis hypogaea L) and its combined action with propranolol on dyslipidemia in Isoproterenol induced cardiotoxic rats. Methods: This experimental study was carried out in the Department of Physiology, Sir Salimullah Medical College (SSMC), Dhaka from January to December, 2012. Twenty Wistar albino rats, age 85-100 days, weighing 120 to 150g (initial body weight) were included in the experimental group (Group B, with peanut). They were further sub-divided into group B1 (isoproterenol treated group after peanut treatment), and group B2 (isoproterenol treated group after combined treatment of peanut and propranolol). Age and weight matched 30 Wistar albino rats without any peanut supplementation was taken as control (group A) and divided into three sub-groups, group A1 (baseline control), group A2 (isoproterenol treated control) and group A3 (isoproterenol treated control after propranolol treatment). Each subgroup consisted of 10 rats. After taking final body weight all the rats were sacrificed on 22nd day. Blood was collected from heart and supernatant serum was preserved in deep freeze until analysis. For assessment of lipid profile status, serum total cholesterol (TC), low density lipoprotein cholesterol (LDL- C) and high density lipoprotein cholesterol (HDL-C) were estimated by standard method. The statistical analysis was done by one way ANOVA and Bonferroni test as applicable. Results: In this study, percent change from initial body weight to final body weight was significantly (p<0.01) lower both in isoproterenol treated group after peanut treatment and in isoproterenol treated group after combined treatment of peanut and propranolol as compared to that of baseline control. Again, this value was almost similar and the difference was not statistically significant between isoproterenol treated group after peanut treatment and isoproterenol treated group after combined treatment of peanut and propranolol. Again, the mean serum TC (p<0.01) and LDL-C (p<0.05) were significantly lower in isoproterenol treated group after peanut treatment and isoproterenol treated group after combined treatment of peanut and propranolol in comparison to those of isoproterenol treated control group. Moreover, the mean serum HDL-C was significantly (p<0.01) higher in isoproterenol treated group after combined treatment of peanut and propranolol, in comparison to that of isoproterenol treated control group. Furthermore, the mean serum TC and LDL-C were non significantly higher and serum HDL-C was significantly (p<0.01) higher in isoproterenol treated group after combined treatment of peanut and propranolol when compared to those of isoproterenol treated group after peanut treatment. Conclusion: The present study revealed that peanut alone can maintain blood lipid level by decreasing TC and LDL-C levels and by increasing HDL-C level in isoproterenol induced cardiotoxic rats. However, the combined therapy of peanut with propranolol showed synergistic effect on preventing dyslipidemia. We investigated the change of tryptophan-niacin metabolism in rats with puromycin aminonucleoside PAN-induced nephrosis, the mechanisms responsible for their change of urinary excretion of nicotinamide and its metabolites, and the role of the kidney in tryptophan-niacin conversion. PAN-treated rats were intraperitoneally injected once with a 1.0% (w/v) solution of PAN at a dose of 100 mg/kg body weight. The collection of 24-hour urine was conducted 8 days after PAN injection. Daily urinary excretion of nicotinamide and its metabolites, liver and blood NAD, and key enzyme activities of tryptophan-niacin metabolism were determined. In PAN-treated rats, the sum of urinary excretion of nicotinamide and its metabolites was significantly lower compared with controls. The kidneyα-amino-β-carboxymuconate-ε-semialdehyde decarboxylase (ACMSD) activity in the PAN-treated group was significantly decreased by 50%, compared with the control group. Although kidney ACMSD activity was reduced, the conversion of tryptophan to niacin tended to be lower in the PAN-treated rats. A decrease in urinary excretion of niacin and the conversion of tryptophan to niacin in nephrotic rats may contribute to a low level of blood tryptophan. The role of kidney ACMSD activity may be minimal concerning tryptophan-niacin conversion under this experimental condition.

Background: Ficus glomerata (F. glomerata) Linn. Family Moraceace is a large tree found all over India including outer Himalayan ranges, Punjab, Chota Nagpur, Bihar, Orissa, West Bengal, Rajasthan, Deccan and also as a common plant in South India. It is planted around the home and temples. It is cultivated throughout the year, distributed in evergreen forests and moist localities. Objective: The Ethanolic Extract of roots of F. Glomerata (EEFG) belonging to the family Moraceace, was investigated for its antidiabetic activity using alloxan induced diabetic rats. Methods: Thirty rats were divided into 5 groups having 6 rats in each group. The alloxan was administered to the rats of all groups except normal control group through intraperitoneal route at a concentration of 140mg/kg body weight. A dose of 100mg/kg and 200 mg/kg body weight of EEFG was administered to alloxan induced diabetic rats. The administration of the extract was lasted for 11 days. Effectiveness of the extract on glucose, cholesterol, triglycerides, and high density lipoprotein and protein concentrations was analyzed. Results: Significant (p<0.05) reduction in the levels of glucose, cholesterol, triglyceride of the diabetic rats was observed after treatment with ethanolic extract. After subjecting to oral glucose tolerance test EEFG also showed significant improvement in glucose tolerance. Conclusion: F. glomerata root ethanolic extract showed that it possesses antidiabetic effect and can be found useful for the management of diabetes mellitus.

Objective. To observe the effect of preventive acupuncture and moxibustion on blood lipid of menopause rats.Methods. Seventy 10-month-old SD rats with estrous cycle disorders were divided into three control groups and four treatment groups (n=10/group) and another ten 3.5-month-old female SD rats were chosen as young control group. Preventive acupuncture and moxibustion were applied at Guanyuan (CV 4). Body weight growth rate has been recorded. Plasma total cholesterol (TC), triglyceride (TG), low density lipoprotein (LDL), and high density lipoprotein (HDL) levels and uterusE2level were measured.Results. Compared to young control group, plasma TC and LDL increased and uterusE2reduced significantly in 12-month-old control group. Compared to 12-month-old control group, plasma TC and LDL level and body weight growth rate decreased while HDL level increased remarkably in preventive acupuncture 12-month-old group. Compared to 14-month-old control group, plasma TC level and body weight growth rate decreased remarkably in preventive moxibustion 14-month-old group.Conclusions. Preventive acupuncture and moxibustion can significantly decrease the plasma TG and LDL, increase the plasma HDL, and prevent fat accumulation. Our finding suggests that preventive acupuncture and moxibustion have beneficial effects on blood lipid. Different treatment effects were found between preventive acupuncture and preventive moxibustion.

Abstract Objectives Several studies have established the ethnobotanical benefits of Pupalia lappacea (PL) in laboratory animals without extensive toxicological evaluation of its safety profiles. Thus, an extensive toxicological investigation of sub-chronic oral administration of the hydroethanol leaf extract of P. lappacea in rodents was carried out in this study. Methods Different groups of rats were treated orally with the extract (10, 50 and 250 mg/kg) daily for 90 consecutive days. The control group received distilled water (10 mL/kg). After 90 days, some rats were left for additional 30 days without treatment for reversibility study. Blood and organs samples were collected for different evaluations at the end of study periods. Results The extract decreased the bodyweights, feeding and water intakes in female rats. PL increased the weights of the liver and kidney in male rats. PL increased the red blood cell (RBC), packed cell volume (PCV), hemoglobin (Hb), triglycerides (TRIG), cholesterol and high density lipoprotein (HDL) contents in rats. PL (250 mg/kg) significantly reduced the sperm motility and serum testosterone level. Cyto-architectural distortions of the testes, liver and spleen were visible. Conclusions The findings showed that P. lappacea is relatively safe at lower doses but cautions should be taken at higher dose.

Objective: Evaluation of the anti-obesity effect of aqueous extract of Mucuna pruriens seeds on rats.Methods: Male Sprague-Dawley (SD) rats were subjected to high-fat diet (HFD) for 12 wk. L-DOPA (12.5 mg/kg, p. o.) as standard drug and aqueous extract of Mucuna pruriens (AEMP) seeds (200 mg/kg, p. o. and 400 mg/kg, p. o.) as test drugs were administered in last 4 wk along with HFD. Body weight, food intake, body mass index (BMI), serum total cholesterol (TC), triglyceride (TG) and high-density lipoprotein (HDL) levels were measured at the end of fourth, eighth and twelfth wk, while white adipose tissue (WAT) mass and brain dopamine levels were measured at the end of the twelfth wk.Results: AEMP (200 mg/kg, p. o.) and (400 mg/kg, p. o.) treated groups showed a significant decrease in food intake and weight gain without altering BMI. Moreover, TG levels were lower in treated groups as compared to the HFD group, but no significant changes were observed in TC and HDL levels. L-DOPA-treated group showed a significant decrease in body weight, food intake, BMI and WAT. Both AEMP and L-DOPA-treated groups showed an increase in brain dopamine levels as compared to disease control group (p<0.05).Conclusion: L-DOPA and AEMP showed anti-obesity activity by reducing body weight gains, food intake and WAT weights; modulating TG with increased brain dopamine level which correlates to the inhibitory action of dopamine on reward mechanism.

<p class="Abstract">The aim of this study was to evaluate the diuretic activity of <em>Eysenhardtia polystachya</em> bark aqueous extract at different doses in a rat model. Different doses of <em>E. polystachya</em> (125, 250, 500 and 750 mg/kg body weight), furosemide (4 mg/kg) and vehicle were administered per os to female rats (n=6 animals per group). After 6 hours in metabolic cages, the effect on urinary flow, glomerular filtration rate and electrolyte balance of sodium and potassium were assessed in all animals. <em>E. polystachya</em> at the doses of 500 and 750 mg/kg induced diuretic activity, since markedly increased (p&lt;0.05) the urinary flow rate, similar to that of furosemide treated group. Only the dose of 750 mg/kg produced an increment in urinary excretion of sodium but not of potassium compared with control group. These findings indicate that<em> E. polystachya</em> bark-induced diuretic activity, providing evidence for its folkloric use.</p><p> </p>
