Introduction: Expansion appliance that can imply tooth movement and opening maxilla suture might affect the blood vessels, bone cells, and formation of the new bone. HBOT accelerated the rate of osteoblast differentiation leading to an increase in bone formation during osteonecrosis. Research about HBOT during maxillary suture expansion has never been investigated. The purpose of this research was to correlate the blood vessels number, osteoblast by administered 2.4 ATA HBOT from day 8-14 to imply new bone formation during maxillary suture expansion. Methods: Eighteen male guinea pigs with randomised post-test only control group design divided into 3 groups. Negative control group K(-), positive control with expansion appliance K(+), expansion appliance and HBOT (P). After 14 days guinea pigs were decapitated. Histological slide of the blood vessels number, osteoblast, and new bone formation were examined under a light microscope then were analysed by regression test. Results: Descriptive test showed increasing blood vessels number, osteoblast, and new bone formation on the administration of HBOT during maxillary suture expansion. Regression test showed that there was a strong correlation between the blood vessels number and osteoblast to the new bone formation with R=6.95 during maxillary suture expansion. Conclusion: 2.4 ATA HBOT influences the intercausal relationship between increasing blood vessels number, increasing osteoblast, and new bone formation during maxillary suture expansion.

Introduction. Knowledge about the effects of medications, vitamins, and various supplements on orthodontic tooth movement (OTM) is imperative for orthodontists. This study aimed to assess the effect of methotrexate (MTX) injection on OTM in rats. Materials and Methods. Twenty-eight male Wistar rats were randomized into four groups (n = 7). The first molar and central incisor were connected using a nickel-titanium (NiTi) coil spring with a 50 g load in each rat. The two experimental groups received 0.75 mg/kg and 1.5 mg/kg MTX, respectively, intraperitoneally for 21 days. The negative control group did not receive any injection and did not undergo orthodontic treatment. The positive control group underwent orthodontic treatment and received 0.9% saline (NaCl) injections for 21 days. All rats were sacrificed with chloroform inhalation after 21 days; their maxilla was resected, and the mean number of Howship’s lacunae, blood vessels, osteoclasts, and resorption lacunae was counted. The reduction in bone volume (bone volume to total volume ratio (BV/TV)) at the site of the maxillary molar was quantified by microcomputed tomography (micro-CT). Results. OTM, the number of osteoclasts, and the number of blood vessels significantly increased in rats treated with MTX ( P < 0.05 ). However, the increase in the number of Howship’s lacunae and resorption lacunae was not significant ( P > 0.05 ). Lower BV/TV in the MTX groups was in agreement with the increased number of osteoclasts. Conclusion. Injection of MTX can significantly increase OTM and decrease root resorption in rats.

Objective: to compare the amount of new bone produced by Bio-Oss® Collagen to that produced by collagen matrix in vivo. Method: eighteen bone defects, 5mm by 10mm were created in the parietal bone of 9 New Zealand White rabbits. 6 defects were grafted with Bio-Oss® Collagen. 6 defects were grafted with collagen matrix alone (positive control) and 6 were left empty (negative control). Animals were killed on day 14 and the defects were dissected and prepared for histological assessment. Quantitative analysis of new bone formation was made on 100 sections (50 sections for each group) using image analysis. Results: A total of 339% more new bone was present in defects grafted with Bio-Oss® Collagen than those grafted with collagen matrix (positive control). No bone was formed in the negative control group. Conclusion: Bio-Oss® Collagen has the effect of stimulating new bone formation locally compared with collagen matrix in vivo. Bio-Oss® Collagen may be utilized as a bone graft material.

<p><strong><em>Background:</em></strong><em> Orthodontic tooth movement is a </em><em>interaction prosess</em><em> of resorption and deposition of bone remodeling. Orthodontic tooth movement by mechanical strength causes changes in alveolar bone. Osteocyte is an essential cell to respond bone remodelling. Hyperbaric Oxygen Therapy affects production of osteocyte because it can release Reactive Oxygen Species (ROS) and Nitrid Oxide (NO).  <strong>Purpose: </strong>To determine the difference number  of osteocyte in pressure and tension area during tooth movement by adjuvant of Hyperbaric Oxygen 2,4 ATA during 7 days starting on day 8 to day 14. <strong>Materials and Methods</strong>: This research used Completery Randomized Control Group Post Test Only Design. 36 cavia cobaya (male)  were divided into 3 groups randomly : the negative control groups, positive control group, and treatment group. Preparat staining used Hematoxylin Eosin (HE) and calculated on microscop 1000x with 20 field of view. Data analyses used one way ANOVA and LSD test then compared each area by using paired T test. <strong>Result:</strong> The data showed that the treatment group (P=10,67) tension area has the highest number of osteocyte than  negative control group (K-=3,67), positive control (K+=7,42). ABSTRACTPeanut shell (PS) infusion has been shown to be antithrombocytopenia, but there has been no research on safety standards. This study aims to identify the symptoms of toxic effects, the potency of toxicity and histopathology of liver male Balb/C mice after a single dose of PS infusion. This research uses randomized matched posttest only control group design. Twenty five mice were divided into 5 orally dosage groups, ie, PS infusion with a dose of 0,026; 0.052; 0.104; 0.208 g/20gBW; and negative control of CMC Na 0.5%. The observation period is for 14 days. The results showed that single dose of PS infusion had a pseudo LD50 value ie > 0.208g/20gBW which was practically non toxic. Symptoms to watch out for the BW infusion were passive behavior, bradycnea, hair color change, hair loss, and weight loss at doses of 3 and 4. It is unclear whether liver damage ie inflammation, necrosis, and albuminous degeneration caused by PS infusion or other causes.keywords: acute toxicity, infusion, peanut shell

One major limitation for the vascularization of bone substitutes used for filling is the presence of mineral blocks. The newly-formed blood vessels are stopped or have to circumvent the mineral blocks, resulting in inefficient delivery of oxygen and nutrients to the implant. This leads to necrosis within the implant and to poor engraftment of the bone substitute. The aim of the present study is to provide a bone substitute currently used in the clinic with suitably guided vascularization properties. This therapeutic hybrid bone filling, containing a mineral and a polymeric component, is fortified with pro-angiogenic smart nano-therapeutics that allow the release of angiogenic molecules. Our data showed that the improved vasculature within the implant promoted new bone formation and that the newly-formed bone swapped the mineral blocks of the bone substitutes much more efficiently than in non-functionalized bone substitutes. Therefore, we demonstrated that our therapeutic bone substitute is an advanced therapeutical medicinal product, with great potential to recuperate and guide vascularization that is stopped by mineral blocks, and can improve the regeneration of critical-sized bone defects. We have also elucidated the mechanism to understand how the newly-formed vessels can no longer encounter mineral blocks and pursue their course of vasculature, giving our advanced therapeutical bone filling great potential to be used in many applications, by combining filling and nano-regenerative medicine that currently fall short because of problems related to the lack of oxygen and nutrients.

Abstract Sequestration of Plasmodium berghei ANKA-infected erythrocytes occurs in BALB/c mice as characteristic of  Plasmodium falciparum infection in humans. Animals’ bile has been widely used for centuries in Traditional Chinese Medicine. Goat bile has been used in healing infectious and non-infectious diseases; however, no report on the use of goat bile against malaria infection and sequestration. The purpose of this study was to analyze the correlation between parasitemia and sequestration in the liver of P.berghei ANKA-infected BALB/c mice treated with goat bile. This research was an in vivo experimental study using the post-test control group design. The male BALB/c mice aged ± 6 weeks, body weight 20-25 g were used. The mice were divided into five groups where Group 1-3 were mice treated with goat bile 25%, 50%, and 100%, respectively. Group 4-5 were negative (sterile water) and positive controls (DHP). Parasitemia was observed daily from each mouse and the number of sequestered infected erythrocytes on the endothelium of sinusoids. The data were analyzed using t independent test. Antimalarial activity of goat bile was shown by the lower parasitemia in goat bile-treated mice compared with the negative control. The average number of sequestration was goat bile concentration-dependent manner. The higher the concentration, the lower the number of sequestration. Sequestration was correlated with parasitemia (p=0,0001). Sequestration of P.berghei ANKA-infected erythrocytes correlated with parasitemia, and was goat bile concentration-dependent manner. Keywords: Malaria, parasitemia, sequestration, goat bileCorrespondence: [email protected]

ABSTRACT Background : Gastrointestinal infections, especially caused by Shigella dysenteriae bacteria is still a health issue in Indonesia. The number of ampicillin resistance to S. dysenteriae has been widely reported. Garlic (Allium sativum) is a traditional medicine that efficacy as an antibacterial. Garlic contains an organosulfur compound  Allicin. Research Purpose: This study aimed to determine the antibacterial activity of garlic rule extract on the growth of S. dysenteriae bacteria that are resistant to ampicillin. Research Methods: This study used the experimental with post test only control group design. The antibacterial activity test was determined based on the diameter of clear zone formed. Test  were performed  on garlic filtrate, ciprofloxacin as a positive control and aquades as a negative control. Each treatment was performed three times. Research Result: This research showed that rule extract of garlic (A. sativum)  had a clear zone against S. dysenteriae that was resistant to ampicillin. Conclusion: The conclusion of this research was that rule extract of garlic (A. sativum) had an antibacterial activity against S. dysenteriae which resistant to ampicillin.  Keywords : Antibacterial activity, Garlic (A. sativum), S. dysenteriae

The using of plant-based medicine is a popular approach to health care. A study presented by American Hearth Association showing that brown rice can lower cholesterol level in bood serum. To know the benefi ts of Angkak’s steeping water to decrease serum cholesterol levels in mice and determine the effects of Angkak’s steeping water to decrease serum cholesterol levels in rats compared with simvastatin. This study used an experimental research design, and using pre and post test control group design, performed in the laboratory of biomedical III FK UMS on November 1, 2012 to date of January 1, 2013. The purposive sampling technique, sampel determinaton using formula Federer by the number of sample of 30 male white rats wistar strain. After the data is collected, analyzed using the Shapiro-Wilk test, ANOVA, LSD, and the unpaired t test. The result of this study demnstrate hypothesis testing using ANOVA test with signifi cant level 95% of the negative control group, the treatment group a dose of 1, 2, and 3, it was found p<0,05 were signifi cantly different meaning. In the test dose of LSD between treatment groups 2 and 3 the value of p=0,839 (p>0,05) wich means no signifi cant difference. In the unpaired T test between the treatment group a dose of 1, 2, and 3 with positive control group were signifi cant difference. The unpaired T test between the treatment group a dose of 1, 2, and 3 with white rice also found signifi cant difference. Conclusion: Angkak’s steeping water can lower serum cholesterol levels of white rat as well as the water steepimg Angkak has the same effectiveness with simvastatin to decrease serum cholesterol levels of mice.Keyword: Cholesterol, Angkak

Moringa Oleifera is a plant that contains chemical compounds that are useful, such as flavonoids. The ability of this flavonoid compound that can capture free radicals cause damage and hepatoprotektan hepar. Purpose of study was to determined levels of Moringa leaf extract which can overcome the effects of liver damage caused by toxic doses of paracetamol through MDA, SGOT and SGPT Method used in this laboratory experimental study is a Randomized Post Test Only Control Group Design with the following stages: 1. Moringa Leaf Extraction with Ethanol 96%; Try 2.Preparasi animals, 3. Treatment of Animals Try the extract of leaves of Moringa 3 dose is: 250mg / 200BB rat (dose of A), 500mg / 200BB mice (dose B), 1000mg / 200BB mice (dose C) for 14 days in combination with paracetamol 2 g / 200BB mice, compared to the negative control group (group given just paracetamol 2 g / 200BB rat) and the positive control group (the group who were given regular feed) for 14 days.Results : turned out to be no difference in the reduction in SGOT levels are statistically significant between the negative control group with high-dose treatment group ie the dose C with =0,016 smaller than 0.05, whereas a decrease in ALT levels were significantly decreased in the treatment group high dose is the dose C with =0,009 smaller than 0.05. While MDA group treated with the negative control group experienced an overall decline for the dose A with =0,05, dose B with =0,0011 and dose C with =0,001. Conclusion of this study showed that the extract of Moringa leaves can be potentially as an antioxidant in all doses at once can be as hepatoprotektor at high doses is 1000mg / 200BB Rattus Novergicus.

Abstract: One of the treatment options of periodontal abscess caused by Fusobacterium nucleatum is administration of antibiotics. However, long-term antibiotics consumption can cause negative side effects. Therefore, alternative treatments that have low side effects and easy to be obtained are needed. Nike fish (Awaous melanocephalus) is one of the endemic fish of North Sulawesi province which has antibacterial properties. This study was aimed to evaluate the inhibition effect of nike fish extract on the growth of Fusobacterium nucleatum. This was a true experimental study with a posttest only control group design. We used modified Kirby-Bauer method with filter papers. Ciprofloxacin was used as the positive control and aquadest as the negative control. Extract of nike fish and stock of pure bacteria Fusobacterium nucleatum were prepared. The results showed that the average diameters of the inhibition zones formed in the nike fish extract after three repetitions, were as follows: for extract concentration of 12.5% was 2.91 mm; 25% was 4.16 mm; 50% was 8.41 mm; and 100% was 9.58 mm. In conclusion, nike fish extract (Awaous melanocephalus) at concentrations of 50% and 100% had a weak inhibitory effect (Himedia category) on the growth of Fusobacterium nucleatum meanwhile at concentrations of 12.5% and 25% there was no activity of zone of inhibition.Keywords: extract of nike fish (Awaous melanocephalus); Fusobacterium nucleatum; inhibitory effect Abstrak: Salah satu opsi pengobatan abses periodontal yang disebabkan oleh bakteri Fusobacterium nucleatum yaitu dengan penggunaan antibiotik namun mengonsumsi antibiotik jangka panjang dapat menimbulkan efek samping negatif. Oleh karena itu, diperlukan pengobatan alternatif yang memiliki efek samping rendah serta mudah didapat. Ikan nike merupakan salah satu ikan endemik Provinsi Sulawesi Utara yang berkhasiat sebagai antibakteri. Penelitian ini bertujuan untuk mengetahui daya hambat ekstrak ikan nike (Awaous melanocephalus) terhadap pertumbuhan bakteri Fusobacterium nucleatum. Jenis penelitian ialah eksperimental murni dengan post test only control group design. Metode yang digunakan yaitu metode modifikasi Kirby-Bauer dengan menggunakan paper disk. Kontrol positif menggunakan antibakteri ciprofloxacin dan kontrol negatif menggunakan akuades. Pada penelitian ini digunakan ekstrak ikan nike dan stok bakteri murni Fusobacterium nucleatum. Hasil penelitian menunjukkan bahwa rerata diameter zona hambat yang terbentuk pada ekstrak ikan nike setelah tiga kali pengulangan yaitu untuk konsentrasi 12,5% sebesar 2,91 mm; 25% sebesar 4,16 mm; 50% sebesar 8,41 mm; dan 100% sebesar 9,58 mm. Simpulan penelitian ini ialah ekstrak ikan nike (Awaous melanocephalus) pada konsentrasi 50% dan 100% memiliki daya hambat kategori lemah (Himedia) terhadap pertumbuhan bakteri Fusobacterium nucleatum sedangkan pada konsentrasi 12,5% dan 25% dikategorikan tidak terdapat aktivitas zona hambat. Kata kunci: ekstrak ikan nike (Awaous melanocephalus); Fusobacterium nucleatum; daya hambat
