Pengaruh Tingkat Pengetahuan Petani  Pada Resistensi Beberapa Bahan Aktif Insektisida Kimia Untuk Mengendalikan Ulat Plutella Xylostella Skala Laboratorium The Effect of Level Farmers' Knowledge In Some  Active Chemical Insecticides Resistance to Control Plutella Xylostella Scale Laboratory Catur Hermanto, Rasiska Tarigan dan Fatiani Manik Balai Penelitian Tanaman Sayuran – Badan Penelitian dan Pengembangan Pertanian Jln Tangkuban Perahu No 517, Lembang, Bandung Barat 40391; e-mail: [email protected]   ABSTRAK  Petani dan pestisida merupakan dua sisi  yang saling berkaitan dalam mengendalikan serangan hama penyakit tanaman dilapangan Penggunaan pestisida oleh petani semakin hari kian meningkat khususnya didataran tinggi, namun  peningkatan ini tidak diimbangi dengan pemahaman dalam menggunakan pestisida. Penelitian ini bertujuan untuk mengetahui  pengaruh tingkat pengetahuan dan resistensi insektisida yang digunakan petani kubis di kabupaten Karo. Penelitian dilaksanakan di laboratorium Kebun Percobaan Berastagi dengan ketinggian tempat 1.340 mdpl yang dilaksanakan dari bulan september sampai november 2015. Pelaksanaan dibagi 2(dua) tahap. Tahap Pertama mengumpulkan makalah melalui teknik diskusi kelompok petani dan pengencer dan wawancara menggunakan kuisioner. Total jumlah responden 10 orang per 4 kecamatana. Dari hasil kuisioner dipilih jenis insektisida yang sering digunakan petani untuk diuji kepekaan ulat plutella xylostella terhadap insektisida yang diuji pada skala dilaboratoium.  Empat perlakuan dengan masing-masing terdiri atas 9 taraf konsentrasi menggunakan rancangan acak lengkap dan diulang 3 kali. Hasil penelitian menunjukkan  petani kubis dikabupaten karo melakukan pencampuran fungi dan insektisida > 2jenis dengan frekwensi penyemprotan 2 x 1 minggum serta dosis yang digunakan tidak sesuai dengan KF yang dianjurkan. Nilai LC 50 insektisida kimia dari bahan aktif klorantranilipro, prefenofos, sipemetrin dan kloropinofos berturut-turut 1,87 ml, 1,5 ml, 1,5 ml dan 2 ml. Dan waktu Waktu yang dibutuhkan untuk mematikan 50 % plutella xylostella (LT 50) dari keempat insektisida kimia berbahan aktif klorantranilipro, prefenofos, sipemetrin dan Kloropinfos berturut-turut adalah  39.20, 19.43, 23.57 dan 30.15 jam Kata kunci : Pengetahuan, Resistensi, Insektisida Kimia, Plutella xylostella , Laboratorium ABSTRACT. The Farmers and pesticides are important aspects that  interrelated tocontroll pests in the field of plant diseases as well as opportunities occur resistensi. The use of pesticides by farmers is constantly increasing, especially in the highlands, but this increase is not offset by an understanding in the use of pesticides. The aimed of the research to find the knowledge and the use of insecticide resistance cabbage farmers in Karo District.   The research was conducted at the laboratory of Berastagi experimental farm with altitude of 1,340 meters above sea level implemented from the month of September to November 2015. The implementation of divided by 2 (two) stages. The first is to collect  issues by discussion technique of farmers group and diluent and interview using a questionnaire. Total number of respondents  is ten peoples each four districts. From the questionnaire result so the selected types of insecticides often used by famers for the testing of  sensitivity plutella xylostella to insecticides test at laboratory scale with four treatment. each consisting of 9 degree of concentration using a completely randomized design and repeated 3 times. each consisting of 9 degree of concentration using a completely randomized design and repeated 3 times. The results showed cabbage farmers in the county karo mixing fungi and insecticides> 2 types with spraying frequency of 2 x 1 week as well as the doses used did not correspond to the recommended KF. LC 50 value of the chemical insecticide active ingredient klorantranilipro, prefenofos, sipemetrin and kloropinofos successively 1.87 ml, 1.5 ml, 1.5 ml and 2 ml. Time and time required to shut down 50% of Plutella xylostella (LT 50) of the four chemical insecticide active ingredient klorantranilipro, prefenofos, sipemetrin and Kloropinfos are respectively 39.20, 19:43, 23:57 and 30.15 hours

This study aims to determine the activity of plant-based pesticide formulations on martyred Parmarion semi-naked snails. The study conducted at the Laboratory of the Faculty of Agriculture, University of Flores, Sam Ratulangi Street, Ende Regency. The research was conducted from February to August 2019 using a Completely Randomized Design (CRD) with two stages of treatment. The first stage was testing the contact poison activity in 5 treatments namely P0 = Control, P1 = Dilution 1: 5, P2 = Dilution 1:10, P3 = Dilution 1:15, P4 = Pure extract. The second step is to test the stomach poison with five treatments namely R0 = Control, R1 = No immersion, R2 = Soak 10 minutes, R3 = Soak 20 minutes, R4 = Soak 30 minutes. All treatments are repeated five times so that they will get 25 units of experiments in every treatment. The results showed the concentration of vegetable pesticide extracts in contact poisons and stomach poisons the highest average mortality for contact poisons were found in P4 therapy with the highest mortality value of 68.00% and experienced the fastest death above 50.00% at 50.80 hours with single speed time is 0.55 hours. While the stomach poison in the R4 treatment is 66.66% with the time, the fastest mortality is above 50.00%, which is 55.40 hours with a speed of 0.57 hours per individual.

One of the entomopatogenous fungus types commonly observed and showed potency as myco-insectiside is Beauveria bassiana(Bals.) Vuill.In order to support effectiveness and patogenous activity of B. bassiana, it is necessary to add a carrying agent that protects its spores from ultra violet ray. This study aims to investigate the effect of storage temperature on viability of B. Bassianaspores on the carrier material. The observation was carried out in the Laboratory of Plant Diseases, Indonesian Coffee and Cocoa Research Institute. The research was arranged in completely randomized design by three factors. The first factor was carrier (C), that consists of C1 = rice flour, C2 = maize flour and C3 = tapioca flour. The second factor was dosage (D), that consists of D1 = 1 g B. bassiana+ 1 g carrier; D2 = 1 g B. bassiana+ 5 g carrier and D3 = 1 g B. bassiana+ 10 g carrier. The third factor was temperature of the storage (T),that consists of T1 = 5oC; T2 = 23oC and T3 = 29oC. Viability of B. Bassiana spores was examined by observing development of 100 blastopores randomly and determined under light microscope with 400 times magnification. Observation was conducted in two replicates after the spores of B. bassiana were kept in the storage for 2, 4, 8 and 16 weeks. The result showed that by adding 1 g tapioca flour and temperature of storage of 5oC was potentiall in keeping viability of B. bassianaspores at least for 2 months. It was due to that tapioca flour gave better effect than rice and maize flours in keeping the storage and appropriate low temperature. Viability of B. bassianaspores decreased with increasing carrier dosage, temperature and duration of the storage. Whereas, storage at 5oC was found to be a better condition in keeping viability of dry pure B. bassianaspores longer than conditions of 23o and 29oC. Kiwi fruit takes about 25 weeks from flower bloom until it reaches physiological maturity, so the time required to produce kiwi seeds from seeds in large quantities and uniform is very long. Tissue culture is one method that can be used to obtain a lot of kiwi seeds and uniforms with large quantities in a faster time. The purpose of this study was to examine various types of media compaction materials for the growth of kiwi shoots in vitro. This study was prepared based on Completely Randomized Design (RAL) with 5 treatments and repeated 4 times so that there were 20 experimental units, each experiment using 2 explants so that there are 40 eksplan. The treatments were: MA 1: Agar Swallow Globe 8 g / l, MA 2: Agar Swallow Globe 4 g / l + Agar Nutrijell 4 g / l, MA 3: Agar Swallow Globe 4 g / l + Agar Nutrijell 5 g / l, MA 4: Phytagel 2.2 g / l, MA 5: Agar Nutrijell 11 g / l. Observation variables are When shoots appear, Number of shoots, number of leaves, Number of Roots, number of root hair. The results showed Swallow Globe 4 g / l + Agar Nutrijell 4 g / l treatment gave the highest average number of shoots, the highest number of leaves and roots, this proved that the combination of Swallow Globe and Nutrijell agar gave a good condition for shoot growth kiwi plant.t.

ABSTRACT Chemical management is the most widely adopted technique to control weeds in sugarcane crops. Purple nutsedge (Cyperus rotundus L.) is among the most difficult species to control. Herbicides need to be absorbed and translocated in a sufficient amount for the tuber chain to be effective. Thus, this study aimed to evaluate the effectiveness of the herbicides sulfentrazone, diclosulam, imazapic, imazapyr, halosulfuron, ethoxysulfuron, monosodium methyl arsenate (MSMA), and 2,4-dichlorophenoxyacetic acid (2,4-D) in controlling C. rotundus and reducing the viability of its tubers. The experiment was carried out in a greenhouse in a completely randomized design, with nine treatments and four replications. The herbicides sulfentrazone (800 g active ingredient – ai·ha?1), diclosulam (193.17 g ai·ha?1), imazapic (133 g ai·ha?1), and imazapyr (500 g acid equivalent – ae·ha?1) were applied in preemergence at five days after planting the tubers, while halosulfuron (112.5 g ai·ha?1), ethoxysulfuron (135 g ai·ha?1), MSMA (1,975 g ai·ha?1), and 2,4-D (1,340 g ae·ha?1) were applied in postemergence (4 to 5 leaves). The parameters visual control, shoot dry matter, number of tubers and bulbs, weight of tubers + bulbs, number of epigeal manifestations, and tuber viability were analyzed. The herbicides diclosulam, halosulfuron, and ethoxysulfuron provided 100% control of the shoot at 90 days after application (DAA). All herbicides reduced the number of bulbs, weight of tubers + bulbs, and shoot dry matter. The herbicides sulfentrazone, imazapic, halosulfuron, ethoxysulfuron, and MSMA provided the highest reduction in tuber viability.

The increasing of pesticide use is in line with the increasing number of pest populations. However, the use of pesticides causes various negative impacts on the environment (soil, water, and air) such as pesticide-resistant pests, perishing of useful insects which are non-target pesticides, and the use of pesticide which can even lead to poisoning and death in humans. One of the environmentally-safe techniques to control pests is chemical, by the use soursop leaf extract. This study aims to determine the most effective concentration of soursop leaf extract (Annona muricata) in controlling leaf caterpillar (Plutella xylostella) in delima rose apples. The research was carried out in the plantation area of rose apples in tempuran village, Demak. The research method employed experimental research. The study was conducted using Completely Randomized Design. The data were obtained using probit analysis of LC50. The most effective concentration of soursop extract (Annona muricata) on leaf caterpillars’ mortality was 75 ppm. Soursop leaf extract (Annona muricata) has been proven to influence rose apples leaf caterpillars’ mortality index. The LC50 value of soursop leaf extract was 75,16 mg/L. The result showed that soursop leaf extract (Annona muricata) is effective as a biopesticide material to control leaf caterpillar pest for delima rose apples.

Abstract Berunok has been a commodity with low economic value, so it is necessary to use Berunok which has high nutritional value. This study aims to determine the proximate content of P. australis. Parameters measured were yield value and proximate analysis of berunok flour. This study consisted of two stages, namely sample preparation and manufacture of berunok flour, analysis of the chemical composition of berunok flour. The research design used a completely randomized design (CRD). The results showed that berunok flour had a dry texture, very smooth, and brownish in color. While the chemical composition contained in Berunok flour is water content of 8.19%, ash content of 37.21% (db), protein content of 48.78% (db ), fat content of 3.44% (db), and carbohydrate content by a difference of 10.57% (db). The average yield value of 83.34%.

Abstract— This research was conducted in two stages,  first stage, looking for the best concentration of natural sweetener with organoleptic. Second stage analyzed the physicochemical characteristics using the best concentration of first stage. This research was carried out using a Completely Randomized Design, with various natural sweeteners consisting 6 levels, namely: stevia sweetener (0.05%, 0.75%, 0.10%), sorghum sugar (10%, 15%, 20%), corn sugar (10%, 15%, 20%), coconut sugar (14%, 15%, 16%), palm sugar (7.5%, 10%, 12.5%), and honey (15%, 25%, 35%) and 3 replications so that 18 experimental units. The addition of various types of natural sweeteners significantly affected the hedonic test (taste and overall acceptance), water content, pH, total dissolved solids, total sugar, and color but did not significantly affect the hedonic quality test (texture), hedonic test (texture and color), crude fiber content, syneresis and gel hardness. Various problems have caused the decline in peanut production, among others the low resistance of peanut plants to various diseases. One of the obstacles in peanut production is the attack of plant diseases, such as the fungus Sclerotium rolfsii Sacc. The use of plant-based insecticides can be an alternative to disease control in peanuts. This study aimed to obtain the appropriate and effective dose of clove leaves and flower stalks in suppressing the in vitro growth of the fungus S. rolfsii Sacc causing damping off in peanuts, and to test the effectiveness of clove leaves and flower stalks on the formation of sklerotia of the fungus. This study used ‘tuni’ clove powder from Waai Village, Salahutu District. This experimental study employed a completely randomized design (CRD) with 8 treatments and 3 replications. The results showed that the growth of the fungus S. rolfsii Sacc. was inhibited by clove flower stalk powder at a dose of 1.5 g per100 mL of PDA, which was indicated by the number of sklerotia formed of 13.67% and propagule weight of 0.12 g, followed by a dose of clove leaf powder of 1.5 g per100 mL PDA, with amount of sklerotia formed of 27.33% and propagule weight of 0.17 g. These results were statistically shown on the parameters: colony diameter, Sklerotia formation time, number of Sklerotia, and weight of propagules. Keywords: Clove powder, dosage, in vitro, Sclerotium rolfsii Sacc.   ABSTRAK Berbagai masalah telah menyebabkan turunnya produksi kacang tanah, diantaranya masih rendahnya ketahanan tanaman kacang tanah terhadap berbagai penyakit. Salah satu kendala dalam produksi kacang tanah adalah serangan penyakit tanaman, seperti jamur Sclerotium rolfsii Sacc. Penggunaan insektisida nabati dapat menjadi alternatif pengendalian penyakit pada kacang tanah. Penelitian ini bertujuan untuk memperoleh dosis daun dan tangkai bunga cengkih yang tepat dan efektif dalam menekan pertumbuhan jamur S. rolfsii Sacc penyebab damping off pada kacang tanah secara in vitro, serta menguji efektifitas daun dan tangkai bunga cengkih terhadap pembentukan sklerotia jamur. Penelitian ini menggunakan bubuk cengkih tuni, dari Desa Waai, Kecamatan Salahutu. Penelitian eksperimental ini menggunakan Rancangan Acak Lengkap (RAL) dengan 8 perlakuan dan 3 ulangan. Hasil penelitian menunjukkan bahwa pertumbuhan jamur S. rolfsii Sacc. terhambat oleh bubuk tangkai bunga cengkih pada dosis 1,5 g per 100 mL PDA yang ditunjukan oleh jumlah sklerotia terbentuk 13,67% dan berat propagul 0,12 g dan diikuti oleh dosis bubuk daun cengkih 1,5 g per 100 mL PDA, dengan jumlah sklerotia 27,33% dan berat propagul 0,17 g. Hasil ini secara statistik ditunjukkan pada parameter: diameter koloni, waktu terbentuk sklerotia, jumlah sklerotia, dan berat propagul. Kata kunci: bubuk cengkih, dosis, in vitro, Sclerotium rolfsii Sacc.

DHF (Dengue Hemorrhagic Fever) is a serious problem in Indonesia. DHF disease control has been applied so far, one of which is the use of larvacide temephos (abate). However larvacide is a chemical insecticide that has a negative impact on human health and causes resistance. Therefore in this research, biological control is carried out by utilizing entomopathogenic fungi as a larvacide. The purpose of this study was to determine the effectiveness of entomopathogenic fungi isolated from Ae aegypti larvae. Against the death of Ae. Aegypti mosquito larvae with the moist chamber method. This study used a completely randomized design (CRD) with a factorial pattern and performed two repetitions. Factor A is a type of fungi with 3 levels, namely A1: Aspergillus sp1, A2: Aspergillus sp2, and A3: Syncephalastrum sp. Factor B is a dilution with 7 levels, namely B0: Control, B1: 100 (without dilution), B2: 10-1, B3: 10-2, B4: 10-3, B5: 10-4, B6: 10-5 with every treatment was applied in 2 repetitions. Observations were made 24 hours after treatment for 3 days. Data were analyzed using analysis of variances (anova) and continued with the Least Significant Difference Test (LSD) at 5%. The results indicate that fungi isolates are the most effective in killing Ae. Aegypti mosquito larvae is Aspergillus sp1 and Aspergillus sp2 on the treatment of spores without dilution. The use of hormones is important in improving the propagation process. The objective of the experiment was to identify the best dose of indole butyric acid (IBA) hormone for the rooting of coffee cuttings of the typical variety through clonal propagation in the Amazon region. The experiment was carried out in two stages. Both stages were carried out under a Completely Randomized Design (CRD). The percentage of rooting of cuttings and the number of roots were evaluated. The coffee tree clones were collected from rust tolerant elite plants. In the laboratory, the cuttings were immersed for 10 minutes in fungicide and then in five doses of IBA hormone (1000, 1500, 2000, 2500, and 3000 mg L−1) of IBA. They were then placed in a microtunnel. With the best dose obtained in stage 1 of IBA (T4: 2000 mg L−1), which achieved a rooting percentage of 52.50% and 5.8 roots on average, stage 2 was installed for the evaluation of acclimatization conditions (greenhouse and nursery). Thirty plants of the Típica variety were evaluated per treatment. Regarding the results after 90 days, the greenhouse obtained the highest values with 72.22% survival, a plant height of 9.48 cm, 4.32 mm stem diameter, 6.53 leaves per plant, 9.65 cm leaf length, and 3.83 cm leaf width. The IBA hormone in a controlled environment such as the greenhouse achieves good vegetative propagation for the Típica variety.
