Aqueous Ethanolic extract of vegetative (leaf) part of herbal plant Erythrina subumbrans (Hassk.) was assessed for Anthelmintic and Antioxidant activity. The obtained crude extract was prepared in different concentration, i.e., 50 and 100 mg/mL, against the standard Piperazine citrate, i.e., 10mg/mL for anthelmintic activity. Pheretima posthuma test worms were used during the study and the anthelmintic activity was evaluated based on Paralysis and Death time. Three groups of the test specimen and freshly prepared solutions were used during the study. The average paralysis and death time of standard is 1.4 & 39.59 min (10mg/mL) compared to sample 5.3 & 65.09 min (50mg/mL); 2.5 & 54.08 min (100mg/mL). From the data, aqueous ethanolic extract possesses promising anthelmintic activity compared to piperazine citrate standard. Similarly, antioxidant activity was evaluated for sample at different concentration ranging from 5, 10, 20, 40, 80, 160, and 320 µg/ml premixed with 2.5mL of 0.0135mM DPPH solution. Ascorbic acid was used as a standard and treated in a similar way as that of a sample. Control samples were also prepared without standard and sample solutions. Prepared solutions were vortexed and kept at benchtop condition for 30 minutes and absorbance was measured at 517 nm in a UV-Vis spectrophotometer. The absorbance pattern of the sample is not comparable with that of the standard solution at the same concentration. Aqueous ethanolic extract with IC50 (half maximal inhibitory concentration) value is >320 µg/mL against standard Ascorbic acid with an IC50 value of 15.5 µg/ml. Which witness the crude extract possess less antioxidant activity.

Sintoc (Cinnamomum sintoc Bl.) is a plant which is used as medicine. This plant has been known to have an analgesic antiinflamatory activity, therefore it is predicted to have an antioxidant activity. An investigation on antioxidant activity of sintoc essential oils and ethanolic extract of its cortex using ascorbic acid as standard has been carried out. Essential oils and ethanol extract of sintoc cortex was tested using DPPH (1,1-diphenyl-2-pikril-hidrazil) by measuring absorbance using visible spectrophotometer at 518 nm. The methods of this research were distillation of essential oils and extraction of sintoc cortex, determination of the essential oil and extract concentrations required for 50% inhibition of DPPH radical scavenging effect (IC50) with ascorbic acid as the possitive control. The variation concentration  of essential oils are 15, 5, 1, 0.1, 0.5 ppm and 25, 20, 17, 15, 10 ppm for ethanolic extracts. The results showed that the essential oil showed antioxidant activity with IC50 value was 16.29 ppm (5 times lower than ascorbic acid) and then ethanolic extract showed IC50 value 38.89 ppm (11 times lower than ascorbic acid, IC50 of ascorbic acid was 3.35 ppm).

The present study was undertaken to investigate the anthelmintic activity of extract of Embelia tsjeriam-Cottam using earthworm. Different concentrations of standard drug (Albendazole) and extract of Embeliatsjeriam-Cottam fruits were employed and the average time required for paralysis and death was noted. It was found that the Paralysis time & Death time was the lowest for 5% concentration of Ethanolic extract and Death time was slightly better than the Albendazole Standard solution. Though Ethanolic Extract can be compared to the Standard hence establishing the pharmacological antihelminthic activity of Embelia tsjeriam-Cottam.

α-amylase inhibitors present in pancreatic region has an operative strategy by controlling the breakdown of starch and helps to minimize the post-prandial hyperglycemia levels. In this study, vegetative (leaf) part of herbal plant Erythrina subumbrans (Hassk.) was assessed for anti-diabetic activity. Aqueous ethanol (80 %) extract was prepared in the different concentration (10, 20, 40, 80, 160, and 320 µg/ml). Acarbose was used as a standard and treated in similar way as that of sample. Control samples were also prepared without standard and sample solutions. A known volume of α-amylase solution was added (0.1mg/mL) was added to standard, sample, control solutions which were preincubated at 37 °C for 15 minutes. Further, known volume of starch solution was added and incubated for 60 min to initiate the reaction. Hydrochloric acid (HCl) and iodine reagent was added to the test tubes and absorbance was measured at 580 nm in UV-Vis spectrophotometer. A strong pancreatic amylase inhibitory activity (>50 %) was obtained from aqueous ethanolic extract with IC50 (half maximal inhibitory concentration) value of 23 µg/ml against standard acarbose with IC50 value of 27 µg/ml. The values endorse Erythrina subumbrans (Hassk.) for further experiments on their potential for managing Diabetes.

Free radicals and reactive species are widely believed to contribute to the development of several diseases by causing oxidative stress and eventually oxidative. Vernonia amygdalina (Astereacea) is a small shrub or tree between 1 and 5m high growing throughout tropical Africa. Plants are generally known as bitter leaves is well cultivated and is a general market for merchandise in several countries. The purpose of this study was to determine the antioxidant activity of hexane fraction from ethanol extract od Frican leaves (Vernonia amygdalina Del.). The method used in this study was the DPPH (1,1-Diphenil-2-Picrylhydrazyl) method. The result of phytochemical screening showed that ethanolic extract of African leaves contained a composition of secondary metabolites of alkaloids, flavonoids, tannins, steroids/triterpenoids and saponins. The antioxidant activity of the extract of n-hexane fraction was classified as very weak with an IC50 value of 317.98 ppm.

The antioxidant activities of Japanese knotweed rhizome bark extracts, prepared with eight different solvents or solvent mixtures (water, methanol, 80% methanol(aq), acetone, 70% acetone(aq), ethanol, 70% ethanol(aq), and 90% ethyl acetate(aq)), were determined using a 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical-scavenging assay. Low half maximal inhibitory concentration (IC50) values (2.632–3.720 µg mL−1) for all the extracts were in the range of the IC50 value of the known antioxidant ascorbic acid at t0 (3.115 µg mL−1). Due to the highest extraction yield (~44%), 70% ethanol(aq) was selected for the preparation of the extract for further investigations. The IC50 value calculated for its antioxidant activity remained stable for at least 14 days, while the IC50 of ascorbic acid increased over time. The stability study showed that the container material was of great importance for the light-protected storage of the ascorbic acid(aq) solution in a refrigerator. Size exclusion–high-performance liquid chromatography (SEC-HPLC)–UV and reversed phase (RP)-HPLC-UV coupled with multistage mass spectrometry (MSn) were developed for fractionation of the 70% ethanol(aq) extract and for further compound identification, respectively. In the most potent antioxidant SEC fraction, determined using an on-line post-column SEC-HPLC-DPPH assay, epicatechin, resveratrol malonyl hexoside, and its in-source fragments (resveratrol and resveratrol acetyl hexoside) were tentatively identified by RP-HPLC-MSn. Moreover, epicatechin was additionally confirmed by two orthogonal methods, SEC-HPLC-UV and high-performance thin-layer chromatography (HPTLC) coupled with densitometry. Finally, the latter technique enabled the identification of (−)-epicatechin. (−)-Epicatechin demonstrated potent and stable time-dependent antioxidant activity (IC50 value ~1.5 µg mL−1) for at least 14 days.

Paku atai plant (Angiopteris ferox Copel), specifically the tuber has been used empirically as an anticancer and antidote materials by Dayaknese in West Kutai, with limited scientific study. Thus, this research had been conducted to determine and evaluate the total phenolic, flavonoid content of this plant along with the antioxidant and toxicity profile. The research used several extracts’ solvents including extraction with ethanol (crude extract, CE) and subsequently partitioned with n-hexane (HF); ethyl acetate (EF) and ethanol aqueous (EAF). The resulted crude extract and fractions were then analyzed through colorimetric method to determine the phenolic and flavonoid total; with DPPH and FRAP to observe the antioxidant activity; and using BSLT method to evaluate the toxicity activity. The results showed that the EF fraction provided the strongest antioxidant activity with IC50 value of 13.79 μg/mL and iron reduction with FRAP value of 387.5±6.41 µM/g. However, a high correlation was observed between the antioxidant and the total phenolic content (r2 = 0.970 - 0.974) but little correlation in total flavonoid (r2 = 0.345 - 0.373). Furthermore, the EF (19.56±7.35) showed the highest toxicity activity followed by CE (22.42±2.10), HF (39.52±7.38), and EAF (41.75±5.10). Therefore, the paku atai tuber can be potentially developed as a natural antioxidant and anticancer material.

The presence of natural antioxidant in plants is well known. Plant phenolics constitute one of the major groups of components that act as antioxidant and free radical terminator. Hence, this study focused on investigating the antioxidant activity of Celery plant (Apium graveolens L). The fresh leaves were collected, crushed and extracted with ethanol and acetone by maceration. The radical scavenging properties of the extracts were determined by measuring changes in absorbance of DPPH radical at a wave lenght of 517 nm by UV and ascorbic acid is used as the standard. It showed that the crude ethanolic extract has higher antioxidant activity compared to ascorbic acid and acetone extract with less scavenging activity. The values were (IC50 114.6 µg/mL) for ascorbic acid, (IC50 112 µg/mL) for the crude ethanolic extract and (IC50172 µg/mL) for crude acetone extract. The result shows that Celery plant grown in Jos possess good antioxidant properties which may be linked to the presence of phenolics and flavonoids in the plant, which justifies its use as a medicinal plant. This can be further investigated for the isolation and identification of active compounds of medicinal utilities.

PurposeMoringa oleifera (MO) is a herbal remedy that is rich in essential amino acids and phytochemicals in the leaves, seeds and pods. It is becoming a famous food in various ethnic groups, and the consumption is increasing worldwide. However, there is no safety profile of the hydroalcoholic MO leaf extracts, or documented teratogenicity aspect. This study was sought to compare the antioxidant activity and the teratogenicity of hydroalcoholic MO leaf extracts in the zebrafish embryo.Design/methodology/approachMO leaves were extracted with aqueous alcohols (ethanol or methanol: water, 80:20 v/v) using the Soxhlet method. The antioxidant capacity was determined using DPPH assay. The concentrations of extracts ranging from 3 to 1,000 µg/mL were used to examine the toxicity and teratogenicity on the zebrafish embryo.FindingsBoth hydroalcoholic MO leaf extracts were positive for alkaloids, terpenoids, steroids, tannins, flavonoids, phenols, coumarins, quinones and glycosides, except saponins only absent in aqueous methanol extract. The antioxidant capacity based on lC50, was 15.92 ± 3.62 mg/mL for aqueous methanolic and 25.28 ± 2.89 mg/mL for aqueous ethanolic extract. For acute toxicity, the aqueous methanolic extract has lower LC50 (163.87 ± 12.88 µg/mL) compared to aqueous ethanolic extract (337.48 ± 30.04 µg/mL). The embryo treated with aqueous methanolic extract showed phenotypic defect but not on the aqueous ethanolic extract.Originality/valueThis study suggests that aqueous methanol of MO leaves extract has better antioxidant capacity compared to ethanol, and ethanolic leaf extract is safer (higher LC50 and no teratogenicity) than methanolic extract.

<p><strong>Objective</strong>:<strong> </strong>The present study aims to investigate <em>in vitro</em> anthelmintic and anti-amylase properties of the ethanolic fruit extract of <em>Garcinia pedunculata</em>.</p><p><strong>Methods</strong>:<strong> </strong>For the study, mature fruits of <em>G. pedunculata</em> were collected from local markets during April-May. Fleshy pericarps of fruits were chopped into small pieces, dried and extracted by using a Soxhlet apparatus. Ethanol extract of <em>G. pedunculata</em> was used for evaluation of <em>in vitro</em> anthelmintic and anti-amylase activities. <em>In vitro</em> anthelmintic activity was evaluated in animal models, <em>Pheretima posthuma</em>, an earthworm species. <em>In vitro</em> anti-amylase activity was evaluated by using zymographic, achromic point analysis (Starch-Iodine method) and spectrophotometric method [Di Nitro Salicylic acid (DNS)–Maltose method].</p><p><strong>Results</strong>:<strong> </strong>Ethanolic extract of <em>G. pedunculata</em> showed anthelmintic activity at a concentration of 75 mg/ml, paralysis and death timing was reported at 0.62±0.26 min and 1.42±0.07 min, respectively. The reference standard (Albendazole) showed paralysis time: 2.13±0.28 min and death time: 5.12±0.29 min. In the anti-amylase study, a zymographic density analysis of <em>G. pedunculata</em> showed significant variation in band intensity as compared to Starch–Iodine achromic point analysis and DNS–Maltose method. A concentration of 1.5 mg/ml of extract showed inhibition of amylase: 67.65±1.53 % as compared to other concentrations and control sets.</p><strong>Conclusion</strong>:<strong> </strong>It could be concluded that ethanolic extract of <em>G. pedunculata</em>has biological properties which could be utilised in medicine by characterising its bioactive components.<p> </p>

Objectives: The objectives of this study were to determine the antioxidant activity and cardioprotective activity of bangun-bangun leaves ethanolic extract.Methods: Bangun-bangun leaves ethanolic extract was obtained by maceration process. The antioxidant activity test was performed by 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) free radical scavenging method with various concentrations of extract. The absorbance was measured by visible spectrophotometric method and calculated the inhibitory concentration (IC50) value for antioxidant activity analysis. Cardioprotective activity test was performed by measuring the cardiac troponin T (cTnT) level, creatine kinase-muscle/brain (CK-MB) level, and histology of the heart tissue. Animals induced with doxorubicin at the 8th day and the 9th day, bangun-bangun leaves ethanolic extract was administered from the 1st day to the 9th day with various doses of extract.Results: Bangun-bangun leaves ethanolic extract had IC50 value of 57.79 μg/mL. Difference dose of bangun-bangun leaves ethanolic extract shows difference cardioprotective activity. Bangun-bangun leaves ethanolic extract at dose 300 mg/kg bw did not differ significantly to the positive control group and normal group. The higher the dose of an extract the greater the decrease in cTnT and CK-MB levels and increase protection against heart damage.Conclusion: Bangun-bangun leaves ethanolic extract had strong antioxidant and had cardioprotective activity.
