MICROSCOPIC CHANGES IN THE INTERNAL ORGANS OF WHITE MICE DURING EXPERIMENTAL IRON (IV) CLATHROCHELATE TOXICOSIS

Iron (IV) clathrochelate based on a macrobicyclic ligand of the hexahydrazide type is a unique compound that contains iron in a rare high valence IV. Preclinical and clinical studies of this complex, which were started for the first time in Ukraine, have an important theoretical and practical consequence as this complex can be recommended as an active substance in iron-containing drugs with antianemic action. In conducting preclinical studies of new drugs, pathomorphological studies are important because they are a necessary step in studying the biological response of animals to the action of test substances. It was found that some pathological changes develop in the body of white mice under conditions of experimental acute and chronic iron (IV) clathrochelate intoxication. They correlated with the dose of the test compound. During chronic intoxication, the microscopic changes in the liver and kidney of white mice treated with iron (IV) clathrochelate at a dose of 1/10 DL50 were similar to the microscopic changes in the liver and kidney of mice treated with the experimental drug at a dose of 1/5 DL50. However, the severity of these changes was lower, reflecting a lower degree of organ damage. In the myocardium of mice treated with iron (IV) clathrochelate at a dose of 1/5 DL50 on the 10th day, as in acute iron (IV) clathrochelate poisoning, only edema was recorded. The prospects for further research are the study of microscopic changes in the organs of laboratory animals of other species during experimental iron (IV) clathrochelate toxicosis.

During the preclinical studies of new drugs, the study of the degree of manifestation of their irritant and allergenic effects affects subsequent clinical studies, for example the routes of administration, the need to add excipients to reduce irritation. The article presents the results of studies of the irritant effects and allergenic properties of the Iron in rare unconventional valence – IV. The irritant effect of the Iron(IV) clathrochelate complexes on the skin was studied comprehensively. 20 rabbits were divided into 4 groups (control and three experimental), 5 animals each. The  ointment on the vaseline and aqueous solution of the Iron(IV) clathrochelate complexes was applied to the skin of rabbits of the experimental groups. Also this solution was introduced subcutaneously. The investigated dosage forms were used at a dose of 1 ml/kg body weight (based on the active ingredient 500 mg/kg body weight of the laboratory animal). The results of the studies showed that of the Iron(IV) clathrochelate complexes has no irritant properties when used externally and is characterized by a lack of local reaction by subcutaneous injection. Determination of allergenic properties was performed by detecting itching and swelling in the guinea pigs in animals which were sensitized with this substance. In addition, in order to assess the severity of the inflammatory reaction, the skin temperature was determined before the experiment and on the 20th day of the experiment, and the skin fold thickness was measured using a caliper. The results of the studies showed that there is no allergic action of the Iron(IV) clathrochelate complexes. The results of the ophthalmic test on laboratory animals confirmed the data obtained.Therefore, comprehensive studies of the irritant and allergenic effects of the Iron(IV) clathrochelate complexes were performed for the first time. The Iron(IV) clathrochelate complexes in the form of ointment and solution does not irritate the skin and mucous membranes and has no allergenic properties to the body of the laboratory animals.

The prevalence of diabetes mellitus has reached epidemic proportion in Malaysia and worldwide. Scientific studies have shown that herbal plantPiper sarmentosumexhibits an antidiabetic property. Despite the extensive usage and studies of this herb as alternative medicine, there is paucity of the literature on the safety information of this plant. Thus, the present study aimed to observe the subacute toxic effects ofPiper sarmentosumaqueous extract (PSAE) on the haematological profile, liver, and kidney in rats. The extract was administered by oral gavage to 6 male and femaleSprague Dawleyrats in daily dose of 50 mg/kg, 300 mg/kg, and 2000 mg/kg for 28 consecutive days. The control group received normal saline. General behavior of the rats, adverse effects, and mortality were observed for 28 days. The haematological and biochemical parameters were determined at baseline and after the treatment. PSAE did not show abnormality on the body weight and gross observation of internal organs. The haematological, biochemical and histopathological profiles showed minimal changes and variation within normal clinical range except for significant increase in serum potassium level that suggests the need of regular monitoring. Nevertheless, these findings suggested that PSAE up to 2000 mg/kg/day did not show subacute toxicity inSprague Dawleyrats.

Goods of plastic, due to their durability, universality and economical properties are broadly used in all spheres of life. On the whole, polymers are inert and nontoxic, but in the process of their production, various additives are used, which on contact or introduction into an organism has a negative effect on it. In our study, we determined the impact of some types of plastic (polyvinyl chloride, polysterene and polyethylene) on the organism of laboratory animals according to changes in their body weight, indices of mass of the internal organs, and blood parameters. For the experiment, we formed four groups of white male mice at the age of 3 weeks and average body weight of 50 g. For each group, we used different litter. For group I, the litter was sawdust; and for the other groups we added plastic products in different volumes to the sawdust; for group II finely cut polyvinyl chloride, for group III cut polyethylene, and for group IV granules of polystyrene. Every 3 days, we determined the body weight of the animals, and 32 days later we determined mass of the organs, clinical and biochemical parameters of the blood. Addition of polyvinyl chloride, polyethylene, and polystyrene into the substrate for mice did not have a significant effect on tempi of growth of body weight, and also relative mass of heart and lungs. Polyvinyl chloride and polystyrene have an immune-suppressive effect, and polyvinyl chloride affects both central and peripheral organs, and polystyrene mostly harms the peripheral organs. All used types of plastic cause leukocytopenia, following which neutrophilia of band neutrophils and monocytosis takes place as a result of damage to the biological barriers. We determined the systemic toxic effect of the studied types of plastic on the internal organs, which manifested in increase in their mass (liver, kidneys), steep increase in the activity of liver enzymes (AST, ALT), simultaneous decrease in activity of alkaline phosphatase and content of cholysterol and glucose in the blood serum of the mice. Also polyvinyl chlorine, polyethylene and polystyrene cause degeneration of the epithelium of the uriniferous tubule, which is manifested in reduction of globulins and creatinine in the blood of animals from the experimental groups following increase in relative mass of the kidneys. The results of our research allow us to state that different types of plastic can cause toxic effect on animals, as well as people who are in frequent contact with them.

Repeated introduction into the body of an animal of a medicinal substance can lead to its accumulation in the tissues of the body. Therefore, knowledge of the cumulative properties of substances or conditions that may lead to cumulations, are especially important for understanding the pathogenesis of intoxication, because this phenomenon often lies at the basis of acute and chronic poisoning. The purpose of the work was to study the degree of cumulation of the “Bendamin” preparation in the body of white rats. The properties of the cumulative drug were studied in 12 white rats weighing 150–160 g. Rats were divided into 2 groups: control and experimental. To determine the cumulative properties of Bendamin, it was administered to laboratory animals starting at a dose of 0.1 DL50, with a sequential increase in the dose of 1.5 times every 4 days. It was found that the cumulative coefficient of Bendamin was 8.31 units, indicating that the drug does not exhibit cumulative effects. In determining the mass coefficients of the internal organs in the animals of the experimental group, the tendency to increase the weight and liver mass coefficients was determined, respectively, by 5.2 and 5.1%. The weight ratio of the heart of the animals of the experimental group increased by 5.7% relative to the control group. After studying the morphological parameters of blood of rats at 24 days of the experiment for studying the cumulative properties of the “Bendamin” drug, an increase in the number of erythrocytes to 6.13 ± 0.27 T/L, white blood cells to 8.42 ± 0.54 G/L and hemoglobin up to 132.4 ± 2.44 g/l. In the analysis of leukocyte profile in rats, there was a decrease in the number of eosinophils and monocytes, respectively, at 1.1 and 1.3% relative to the control group. The number of lymphocytes in the blood of the experimental group of rats increased by 1.5%, while neutrophils by 0.9% compared to the control. When studying the level of total protein, its small increase was determined by 5.2%. The activity of aminotransferases in the blood serum of experimental rats on the 24th day of the experiment for the study of cumulative properties of the drug also increased by 13 and 8% relative to the control group. Also, creatinine and total bilirubin levels in their rat blood were decreased by 3.1 and 8.8%, respectively.

Mycotoxins have been formed in feed are secondary metabolites of fungi and are quite stable substances that have teratogenic, mutagenic and carcinogenic effects. An effective way to combat mycotoxins in feed is the use of feed additives that adsorb toxins, prevent their absorption in the gastrointestinal tract of the animal and are excreted from the body. The infl uence of coconut enterosorbent Shelltic Es on fattening and meat qualities of young pigs has been studied. It has been found in the process of fattening that the highest feed digestibility was in pigs of the experimental group, in the diet of which enterosorbent has been added. The results of studies of the chemical composition and technological properties of pork have been provided. The positive eff ect of enterosorbent on precocity, absolute average daily gain of live weight and reduction of feed expenditures per 1 kg of gain has been revealed. In pigs from the experimental group the average thickness of the fat was higher and the area of the “muscle eye” was lower compared with animals of the control group, which indicates that higher rates of carcass yield have been obtained due to faster accumulation of fat tissue. The weight of internal organs of pigs indicates the intensity of metabolic processes in the body. In experimental animals the weight of the lungs was 0,1 kg lower, and the liver and heart were higher by 0,13 and 0,01 kg, respectively. Enterosorbent had no effect on the content of vitamins in the liver of pigs of the compared groups. The content of impurities of organochlorine toxicants and toxic elements in the meat and liver of animals of the experimental group was lower than that of control analogues. Therefore, the use of enterosorbent Shelltic Es promotes more active excretion of them from the body.

The article presents in an experiment obtained principal results based on repeated low-level inhalation exposures of laboratory animals (white rats, outbred) to nickel oxide nanoparticles with a diameter of (23 ± 5) nm, 4 hours a day, 5 times a week for up to 10 months in a «nose only» installation. It was shown that non-specific body reactions to the action of NiO NPs include: diverse manifestations of systemic toxicity with a particularly pronounced influence on liver and kidney function, redox balance, damage to some areas of brain tissue, associated with proven movement of the nanoparticles themselves from the nasal mucosa along the olfactory tract; some cytological signs of probable development for allergic syndrome; paradoxically low severity of pulmonary pathology by pneumoconiotic type explained by a small chronic delay of nanoparticles in the lungs; the genotoxic effect of the organismal level, even at those low levels of chronic exposure, at which systemic toxicity is rather poorly. Along with that, NiO NPs also induce phase-stimulation of erythropoiesis, which is relatively specific for the toxic nickel effects.

Background: The liver disease problem prompts investigators to search for new methods of liver treatment. Introduction: Silymarin (Sil) protects the liver by reducing the concentration of free radicals and the extent of damage to the cell membranes. A particularly interesting method to increase the bioavailability of Sil is to use synthesized gold nanoparticles (AuNPs) as reagents. The study considered whether it was possible to use the silymarin-AuNP conjugate as a potential liver-protecting drug. Method: AuNPs were conjugated to Sil and examine the liver-protecting activity of the conjugate. Experimental hepatitis and hepatocyte cytolysis after carbon tetrachloride actionwere used as a model system, and the experiments were conducted on laboratory animals. Result: For the first time, silymarin was conjugated to colloidal gold nanoparticles (AuNPs). Electron microscopy showed that the resultant preparations were monodisperse and that the mean conjugate diameter was 18–30 nm ± 0.5 nm (mean diameter of the native nanoparticles, 15 ± 0.5 nm). In experimental hepatitis in mice, conjugate administration interfered with glutathione depletion in hepatocytes in response to carbon tetrachloride was conducive to an increase in energy metabolism, and stimulated the monocyte–macrophage function of the liver. The results were confirmed by the high respiratory activity of the hepatocytes in cell culture. Conclusion: We conclude that the silymarin-AuNP conjugate holds promise as a liver-protecting agent in acute liver disease caused by carbon tetrachloride poisoning.

Background: Helicobacter pylori infection and its treatment still remains a challenge to human health worldwide. A variety of antibiotics and combination therapies are currently used to treat H. pylori induced ulcers and carcinoma; however, no effective treatment is available to eliminate the pathogen from the body. Additionally, antibiotic resistance is also one of the main reasons for prolonged and persistent infection. Aim of the study: Until new drugs are available for this infection, vaccinology seems the only alternative opportunity to exploit against H. pylori induced diseases. Methods: Multiple epitopes prioritized in our previous study have been tested for their possible antigenic combinations, and results in 169-mer and 183-mer peptide vaccines containing the amino acid sequences of 3 and 4 epitopes respectively, along with adjuvant (Cholera Toxin Subunit B adjuvant at 5’ end) and linkers (GPGPG and EAAAK). Results: Poly-epitope proteins proposed as potential vaccine candidates against H. pylori include SabAHP0289-Omp16-VacA (SHOV), VacA-Omp16-HP0289-FecA (VOHF), VacA-Omp16-HP0289-SabA (VOHS), VacA-Omp16-HP0289-BabA (VOHB), VacA-Omp16-HP0289-SabA-FecA (VOHSF), VacAOmp16-HP0289-SabA-BabA (VOHSB) and VacA-Omp16-HP0289-BabA-SabA (VOHBS). Structures of these poly-epitope peptide vaccines have been modelled and checked for their affinity with HLA alleles and receptors. These proposed poly-epitope vaccine candidates bind efficiently with A2, A3, B7 and DR1 superfamilies of HLA alleles. They can also form stable and significant interactions with Toll-like receptor 2 and Toll-like receptor 4. Conclusion: Results suggest that these multi-epitopic vaccines can elicit a significant immune response against H. pylori and can be tested further for efficient vaccine development.

AbstractIn vivo bioprinting has recently emerged as a direct fabrication technique to create artificial tissues and medical devices on target sites within the body, enabling advanced clinical strategies. However, existing in vivo bioprinting methods are often limited to applications near the skin or require open surgery for printing on internal organs. Here, we report a ferromagnetic soft catheter robot (FSCR) system capable of in situ computer-controlled bioprinting in a minimally invasive manner based on magnetic actuation. The FSCR is designed by dispersing ferromagnetic particles in a fiber-reinforced polymer matrix. This design results in stable ink extrusion and allows for printing various materials with different rheological properties and functionalities. A superimposed magnetic field drives the FSCR to achieve digitally controlled printing with high accuracy. We demonstrate printing multiple patterns on planar surfaces, and considering the non-planar surface of natural organs, we then develop an in situ printing strategy for curved surfaces and demonstrate minimally invasive in vivo bioprinting of hydrogels in a rat model. Our catheter robot will permit intelligent and minimally invasive bio-fabrication.

AbstractContinuous multi-channel monitoring of biopotential signals is vital in understanding the body as a whole, facilitating accurate models and predictions in neural research. The current state of the art in wireless technologies for untethered biopotential recordings rely on radiative electromagnetic (EM) fields. In such transmissions, only a small fraction of this energy is received since the EM fields are widely radiated resulting in lossy inefficient systems. Using the body as a communication medium (similar to a ’wire’) allows for the containment of the energy within the body, yielding order(s) of magnitude lower energy than radiative EM communication. In this work, we introduce Animal Body Communication (ABC), which utilizes the concept of using the body as a medium into the domain of untethered animal biopotential recording. This work, for the first time, develops the theory and models for animal body communication circuitry and channel loss. Using this theoretical model, a sub-inch$$^3$$ 3 [1″ × 1″ × 0.4″], custom-designed sensor node is built using off the shelf components which is capable of sensing and transmitting biopotential signals, through the body of the rat at significantly lower powers compared to traditional wireless transmissions. In-vivo experimental analysis proves that ABC successfully transmits acquired electrocardiogram (EKG) signals through the body with correlation $$>99\%$$ > 99 % when compared to traditional wireless communication modalities, with a 50$$\times$$ × reduction in power consumption.
