Introduction. According to the literature, nettle leaves contain polysaccharides and mucus as an integral part of the hydrophilic fraction. This group of biologically active substances (BAS) is involved in the manifestation of the physiological activity of the infusion. The traditional gravimetric method for the determination of polysaccharides in medicinal plant raw materials (PRM), pharmaceutical substances of plant origin and medicinal herbal preparations cannot provide a true picture of their content due to the presence of impurities. Therefore, it is advisable to determine the reducing sugars that are most fully extracted into the aqueous phase during the preparation of the infusion using modern physicochemical methods of analysis.Aim. The aim of the work was to determine the amount of polysaccharides and simple sugars in nettle leaves by various physico-chemical methods and their comparative characteristics.Materials and methods. A comparative determination of the sum of polysaccharides and simple sugars in the leaves of nettle by pharmacopoeial methods (picrine, anthrone and orcine) was carried out. Free and bound simple sugars were identified and quantified by thin layer chromatography (TLC). The composition and content of free simple sugars was studied by capillary electrophoresis (CE) in raw materials.Results and discussion. The content of free and bound sugars in raw materials has been established. The largest content of both the sum of polysaccharides and free simple sugars in the polysaccharide complex of nettle leaves, as well as the sum of free reducing simple sugars, according to experimental data, was obtained using the picrine method. The content of pentoses in the studied PRM was four times less, which is consistent with the literature data on the preferential construction of polysaccharides of plant objects from sugars of the hexose class. The resulting total content of the fraction of free sugars in the leaves, determined by the CE method, is an order of magnitude less compared with spectrophotometric methods. The obtained results of quantitative determination of the amount of free and bound simple sugars in the studied PRM are consistent with the determination data by the picrine pharmacopoeial method.Conclusion. In general, complete information, depending on the goals of the analysis, on the composition and quantitative content of simple and related sugars in the polysaccharide complex of plant objects can only be obtained by combining spectral methods with CE or TLC.

Introduction. Plant raw materials can be a source of biologically active substances and increase the nutritional value of food products. The present research objective was to determine the content of biologically active substances in powdered viburnum and barberry. Study objects and methods. The study featured viburnum (Viburnum opulus L.) and barberry (Berberis vulgaris L.), dried by convection and crushed into particles of 50 microns. Results and discussion. The total content of phenolic compounds in powdered viburnum was 3114.07 mg/100 g, in powdered barberry – 2272.7 mg/100 g. The content of flavonoids in powdered viburnum was 324.52 mg/100 g, in powdered barberry – 390.00 mg/100 g. The flavonoid profile of the powders included rutin, hyperoside, quercitrin, isoquercintrin, and astralagin. The total content of catechins was 446 mg/100 g for viburnum and 506 mg/100 g for barberry. The catechins included mainly epigallocatechin and catechin. In powdered viburnum, the catechin composition was as follows: epicatechin – 196, catechin – 118, and epigallocatechin – 89 mg/100 g; in powdered barberry: epigallocatechin – 173, catechin – 111, and epicatechin – 74 mg/100 g. The antiradical activity in relation to trolox equivalent was 7560 mg/100 g in powdered viburnum and 9460 mg/100 g in powdered barberry. Conclusion. The obtained viburnum and barberry powders can fortify food with biologically active substances and expand the range of functional products.

Introduction. Modern pharmacognostic research is aimed at searching for plant biologically active individual compounds (hereinafter referred to as RBAIS) isolated from plant extracts.Aim. Validation of HPLC-UV quantitative determination of sapogenin diosgenin in plant extracts from fenugreek seeds.Materials and methods. The object of study was raw materials-fenugreek seeds produced as medicinal plant raw materials by LLC «Sage» (Irkutsk). Validation of the method was carried out according to the parameters: specificity, linearity, correctness, precision in accordance with the requirements of SP XIV. One series of medicinal plant raw materials was used for the analysis, such as serial number – 010117, release date – 15 february 2017.Results and discussion. Validation characteristics were determined and their compliance with the necessary acceptance criteria was experimentally confirmed.Conclusion. It is established that the developed method of identification and quantitative determination of diosgenin in fenugreek seed extracts by HPLC-UV is correct, precise, specific and linear in the analytical field.

Introduction. As is known, the selection of optimal conditions for the analysis of extracts from medicinal plant raw materials (MPМ), as well as medicinal herbal preparations and pharmaceutical substances of plant origin, characterized by a complex variable composition of biologically active substances (BAS) by TLC, presents certain difficulties. For the design of mobile phases for the separation of mixtures of BAS of plant origin, in a thin layer of sorbent, the following approaches are used: literary sources; standard mobile phases; spot elution method; the scheme proposed by the firm Camag (Switzerland); model "PRISMA"; variocameras and others. In foreign literature, there are publications on the generalization of the available experimental data on the determination of various natural groups of BAS in objects of plant origin. However, such reviews did not reveal the regularities of the chromatographic behavior of individual BAS in a thin layer, as well as the influence of various factors on the reproducibility of R f values. The study of the possibility of a theoretical approach to the choice of optimal conditions for chromatography of groups of BAS of different polarity, allowing them to separate, identify and quantify by TLC is a relevant and poorly developed area of chromatography in general.Aim. The aim of this work was to develop a theoretical approach to the choice of optimal conditions for the chromatographic separation of various groups of BAS of plant origin in a thin layer of sorbent.Materials and methods. To study the regularities of chromatographic behavior in a thin layer of representatives of the main classes of BAS present in MPМ (amino acids, flavonoids, tannins, simple sugars, ascorbic acid, fat-soluble vitamins), the value of the main factor affecting the parameters of the efficiency of the chromatographic process, the polarity of the eluent, was studied. As objects of research, we used ready-made chopped raw material of nettle leaves, produced by a domestic manufacturer, that meets the requirements of regulatory documents, as well as sea buckthorn fruits collected on the territory of the Voronezh region, according to the rules for harvesting MPМ of various morphological groups in fresh and dried form.Results and discussion. The regularities of elution and mathematical models describing the chromatographic behavior of plant BAS in a thin layer of sorbent have been established. Based on the totality of the results obtained, from the standpoint of the efficiency of the chromatographic process, the optimal conditions for their TLC analysis were selected and theoretically substantiated. To study the qualitative composition of BAS and to achieve a clear separation of zones on chromatograms, TLC methods were developed and tested on the studied MPМ using simple, frontal or two-dimensional chromatography.Conclusion. It is shown that the determination and separation in a thin layer of the sorbent of hydrophilic and lipophilic BAS of MPМ in the presence of a joint requires different approaches and techniques. The paper proposes an algorithm for the selection of the mobile phase and methods of chromatography of BAS of medicinal products. The revealed mathematical models describing the chromatographic behavior of BAS will make it possible to select the conditions under which it is possible to determine the individual components of multicomponent mixtures without preliminary separation. The developed methods for the determination of BAS can also be used for standardization and quality assessment of other types of MPМ, phytopreparations and pharmaceutical substances of plant origin.

Carotenoids are of interest as precursors of vitamin A, which has a wide range of pharmacological effects — antioxidant, immunomodulatory, participating in photoreception, etc. The aim of the research was to develop a technique for the spectrophotometric determination of the A-vitamin activity of carotenoids-rich medicinal plant raw materials (MPRM). The objects of research were the traditional sources of carotenoids — sea buckthorn, carrot, pumpkin and wild rose. A method is proposed for determining the content of individual carotenoids in MPRM by optical density at wavelengths corresponding to the characteristic peaks of α-carotene, β-carotene and lycopene. It has been shown that it is preferable to use isopropyl alcohol as the most effective solvent. Taking into account the content of specific carotenoids makes it possible to determine the A-vitamin activity of the sum of carotenoids extracted from plant materials, and, accordingly, to increase the efficiency of the use of herbal preparations and biologically active additives for health improvement.

There was studied one- and three-fold solid-liquid extraction of natural phenolic compounds (NPC) (in terms of rutin and tannin) from various morphological parts of blue cornflower (Centaurea cyanus L.) on an ultrasonic device created in the research laboratory of the design bureau of the ISS (Voronezh). The optimal time of solid-liquid extraction at the ultrasonic unit has been determined, taking into account wetting and swelling of vegetable raw materials. The quantitative content of NPC (in terms of rutin and tannin) as a percentage of the total content in raw materials at the first stage is about 60%, at the second stage is 25%, and the third one is 18%. It was established that with threefold extraction, the yield of NPC from medicinal plant raw materials increases at the average of 8–10% compared with a one-time extraction. There is offered an improved technique of solid-liquid extraction on the ultrasound unit and UV spectrophotometric determination of NPC from different morphological parts of the blue cornflower. The technique is characterized by the absence of a systematic error, and the relative error at a confidence level of 0.95 does not exceed 5%. The duration of the analysis, taking into account the stage of wetting and osmotic swelling, triple extraction on the ultrasound unit and UV spectrophotometric determination of rutin and tannin in water-ethanol extracts is 40–45 minutes.

Flavonoids are an important component of plant materials; they have medicinal and biologically active properties. To use them as medicines, it is necessary to determine their qualitative and quantitative composition. The aim of this work is a comparative analysis of the identification and metrological characteristics of methods for the determination of flavonoids in plant raw materials and products of their processing. The paper presents a brief literature review of the main methods for the study of flavonoids. Certified and advanced techniques, such as NMR and mass spectrometry, have been discussed.

Increasing the yield of juice and biologically active components from plant raw materials is an urgent task in the production of functional beverages. For this, fruit, berry and vegetable pulp was treated with enzymatic preparations of pectolytic action and the yield of juice was determined. Samples of pulp of each raw material were treated with enzyme preparations: Fructocyme P6-L, Fructocyme P, Fructocyme MA, in an amount of 0.03% of the pulp mass. Enzymatic hydrolysis was carried out at 50 °C for two hours, after which the amount of obtained juice was compared with the control sample. As a result, when adding Fructocyme MA to the pulp of apples and carrots, the juice yield increases by 8% and 17%, respectively. When Fructocyme P6-L is added to the pulp of kiwi, blueberries, grapes, the juice yield increases by 6%, 12 and 10%, respectively. To determine the effect of enzyme preparations on the yield of biologically active components, the previously selected enzyme preparations were added to all samples in an amount of 0.01; 0.03; 0.05; 0.07% of the pulp mass, and the total content of polyphenols was determined by colorimetry and anthocyanins. As a result of the experiment, it was found that in terms of the amount of polyphenolic substances extracted from the pulp, fruit and berry raw materials can be divided into three groups: raw materials with loose pulp, with pulp of medium density and raw materials with dense pulp. As a result, individual enzyme preparations and optimal modes of application were selected for each type of raw material. The dependence of the content of polyphenolic compounds on the dosage of the addition of enzyme preparations has been established.

A large number of biologically active substances, organic acids, tannins, and pectin substances were detected during the study of fruits, leaves, and pulp of the fruits of mountain Ash. The content of these substances in leaves and pulp will significantly expand the arsenal of medicinal plant raw materials used. Given the wide distribution of mountain Ash in the territory of the Russian Federation, harvesting leaves from the same plants after harvesting the fruit and using pulp will make it possible to obtain new phytopreparations.

The article provides brief information about cardiotonic, sedative, cytostatic, diuretic, and antibacterial effects of biologically active compounds of Adonis L. (Ranunculaceae) species. Chemical studies allowed to identify the cardiac glycosides, or cardenolides: or cardenolides: adontoxin, adonitol, adonitoxigenin, acetyldigitoxin and others. In scientific medicine, it is currently allowed to use Adonis vernalis L. Other types of Adonis have a similar chemical composition and are offered as substitutes for this official species, for example, Adonis apennina L. Many Adonis species have limited natural resources, and in some regions are rare, requiring conservation of their natural populations. The search for alternative sources of medicinal plant raw materials, based on this, is relevant. The experimental part of our research was carried out using the morphological and geographical method with the involvement of information on ecology and phenology. For the first time summarizes the diagnostic features of Adonis flora of Russian flora. Previously unknown structural features (shape and size of anthers) were identified that characterize the subgenera Adonanthe and Adonis. Taxonomic study of the genus Adonis of the Russian flora allowed us to determine its species composition, clarify its systematic affiliation, and nomenclature synonyms. 9 species were identified. Of these, 6 are perennials belonging to the subgenus Adonanthe, section Consiligo, which includes 2 subsections: Amurenses (2 species) and Vernales, which is differentiated into 2 rows: Apenninae (2 species) and Vernales (2 species). Subgenus Adonis is represented by 2 sections: Adonis (1 species) and Lophocarpa with sections Aestivales (1 species) and Dentatae (1 species). For all the considered species and varieties, the main distribution areas are given. A key has been compiled to determine the wild Adonis species distributed in Russia.

Background. The deficiency of essential nutrients in the human body is the root cause of reduced efficiency, general resistance of the body to various diseases. That is an urgent problem in the context of the spread of coronavirus infection. In this regard, the search for opportunities to meet the needs of the population in food products that contain biologically active substances (BAS) through the use of high-value and at the same time available plant raw materials is extremely important. Marigolds attract special attention among such raw materials. The aim of the work is to study the chemical composition and evaluate the consumer properties of Tagetes L. varieties, zoned in the Kiev region, to prove the possibility of increasing the biological value of pasta by adding to their recipe powder from marigold inflorescences. Materials and methods. The object of research is pasta of B group (noodles). Inflorescences of marigolds of different varieties gathered during the flowering period in July-September 2019–2020 in the territory of Vasylkiv and Obukhiv districts of Kyiv region, Ukraine were used as enriching plant raw materials. The quality of finished pasta was assessed by organoleptic and physicochemical indexes (moisture content and titratable acidity). The content of mineral elements in the fine powder of marigold inflorescences was determined on the EXHERT-3L device, the quantitative composition and BAS identification – by spectrophotometric analysis on the device Specord-200 Analytic Jena UV-vis. The integrated quality index of pasta products was calculated taking into account the importance factors of individual indexes. Results. Comparing the obtained results, we can state that all the studied varieties – Gold Kopfen, Orange Flame, Hawaii, Equinox are characterized by high biological value, regardless of the area of collection. As a result of calculated and experimental studies it was determined that the addition of 7 % of the supplement does not provide the required supply of vitamins and minerals. Replacing the flour in the recipe by 20 % significantly improves the mineral composition, but deteriorates the taste and aromatic properties of pasta. The most optimal and rational amount of added additive is 15 %, as it improves the organoleptic properties of pasta and enriches their BAS. Conclusion. The usage of non-traditional vegetable raw materials, in particular powder from marigold inflorescences, which is rich in biologically active substances, allows not only to expand the range, improve the quality of finished products, but also increase the biological value of the product. The optimal amount of additive –powder from marigold inflorescences, was defined. It is 15 % by weight of flour in the production of noodles. This concentration allows not only to improve the organoleptic properties, but also to increase the content of biologically active substances – vitamins, micro- and macronutrients. The usage of non-traditional natural raw materials (powder from marigold inflorescences) allows to abandon artificial colorsand give the finished pasta a pleasant colorand aroma.
