The poultry industry is one of the main providers of protein for the world's population, but it faces great challenges including coccidiosis, one of the diseases with the most impact on productive performance. Coccidiosis is caused by protozoan parasites of the genus Eimeria, which are a group of monoxenous obligate intracellular parasites. Seven species of this genus can affect chickens (Gallus gallus), each with different pathogenic characteristics and targeting a specific intestinal location. Eimeria alters the function of the intestinal tract, generating deficiencies in the absorption of nutrients and lowering productive performance, leading to economic losses. The objective of this manuscript is to review basic concepts of coccidiosis, the different Eimeria species that infect chickens, their life cycle, and the most sustainable and holistic methods available to control the disease.

Avian coccidiosis is a disease causing considerable economic losses in the poultry industry. It is caused by Eimeria spp., protozoan parasites characterized by an exogenous–endogenous lifecycle. In vitro research on these pathogens is very complicated and lacks standardization. This review provides a description of the main in vitro protocols so far assessed focusing on the exogenous phase, with oocyst viability and sporulation assays, and on the endogenous phase, with invasion and developmental assays in cell cultures and in ovo. An overview of these in vitro applications to screen both old and new remedies and to understand the relative mode of action is also discussed.

SUMMARY The Apicomplexa are a phylum of diverse obligate intracellular parasites including Plasmodium spp., the cause of malaria; Toxoplasma gondii and Cryptosporidium parvum, opportunistic pathogens of immunocompromised individuals; and Eimeria spp. and Theileria spp., parasites of considerable agricultural importance. These protozoan parasites share distinctive morphological features, cytoskeletal organization, and modes of replication, motility, and invasion. This review summarizes our current understanding of the cytoskeletal elements, the properties of cytoskeletal proteins, and the role of the cytoskeleton in polarity, motility, invasion, and replication. We discuss the unusual properties of actin and myosin in the Apicomplexa, the highly stereotyped microtubule populations in apicomplexans, and a network of recently discovered novel intermediate filament-like elements in these parasites.

Apicomplexan parasites are responsible for devastating diseases, including malaria, toxoplasmosis, and cryptosporidiosis. Current treatments are limited by emerging resistance to, as well as the high cost and toxicity of existing drugs. As obligate intracellular parasites, apicomplexans rely on the uptake of many essential metabolites from their host. Toxoplasma gondii, the causative agent of toxoplasmosis, is auxotrophic for several metabolites, including sugars (e.g., myo-inositol), amino acids (e.g., tyrosine), lipidic compounds and lipid precursors (cholesterol, choline), vitamins, cofactors (thiamine) and others. To date, only few apicomplexan metabolite transporters have been characterized and assigned a substrate. Here, we set out to investigate whether untargeted metabolomics can be used to identify the substrate of an uncharacterized transporter. Based on existing genome- and proteome-wide datasets, we have identified an essential plasma membrane transporter of the major facilitator superfamily in T. gondii—previously termed TgApiAT6-1. Using an inducible system based on RNA degradation, TgApiAT6-1 was depleted, and the mutant parasite’s metabolome was compared to that of non-depleted parasites. The most significantly reduced metabolite in parasites depleted in TgApiAT6-1 was identified as the amino acid lysine, for which T. gondii is predicted to be auxotrophic. Using stable isotope-labeled amino acids, we confirmed that TgApiAT6-1 is required for efficient lysine uptake. Our findings highlight untargeted metabolomics as a powerful tool to identify the substrate of orphan transporters.

AbstractA life cycle framework for a new integrated classification system for buildings and the identification of renovation strategies that lead to an optimal balance between reduction of seismic vulnerability and increase of energy efficiency, considering both economic losses and environmental impacts, is discussed through a parametric application to an exemplificative case-study building. Such framework accounts for the economic and environmental contributions of initial construction, operational energy consumption, earthquake-induced damage repair activities, retrofitting interventions, and demolition. One-off and annual monetary expenses and environmental impacts through the building life cycle are suggested as meaningful performance metrics to develop an integrated classification system for buildings and to identify the optimal renovation strategy leading to a combined reduction of economic and environmental impacts, depending on the climatic conditions and the seismic hazard at the site of interest. The illustrative application of the framework to an existing school building is then carried out, investigating alternative retrofitting solutions, including either sole structural retrofitting options or sole energy refurbishments, as well as integrated strategies that target both objectives, with a view to demonstrate its practicality and to explore its ensuing results. The influence of seismic hazard and climatic conditions is quantitatively investigated, by assuming the building to be located into different geographic locations.

Zoospores of the obligately parasitic chytrid Rozella allomycis which settle upon hyphae of the water mold host, Allomyces arbuscula, encyst and germinate before their protoplasts penetrate into the host cytoplasm. This process has been examined by light and electron microscopy. Three stages which follow the attachment to the host and the retraction of the zoospore's flagellum are described: (1) the early cyst lacks a wall; it is discoid, and its shape is maintained by the coil of the retracted axoneme which forms its rim; (2) a cyst wall is formed while multivesicular bodies occur at the cell periphery and eventually disappear; a germ tube starts to grow at the point of attachment; and (3) the firm-walled cyst is spheroidal; it has a fully developed germ tube with a specialized class of vesicles; it also forms a distal, flattened vacuole whose swelling eventually injects the Rozella protoplast into the host; at this stage the retracted axoneme has disappeared and the cell's organelles have undergone extensive changes. Electron-dense, "gamma-like" granules enclosed in vacuoles may play a major role in the formation of both the cyst wall and the distal vacuole. These granules appear to give rise to small vesicles, and thus to multivesicular bodies; the distal vacuole appears to form by coalescense of gamma-like vacuoles.The general sequence of encystment and germination resembles that found in other Chytridiomycetes, both saprophytic and parasitic. However, the distal vacuole and the vesicles in the germ tube appear to be parasitic adaptations and are shared by obligate intracellular parasites from several unrelated groups of zoosporic fungi.

Abstract Background Immunological stress decreases feed intake, suppresses growth and induces economic losses. However, the underlying molecular mechanism remains unclear. Label-free liquid chromatography and mass spectrometry (LC-MS) proteomics techniques were employed to investigate effects of immune stress on the hepatic proteome changes of Arbor Acres broilers (Gallus Gallus domesticus) challenged with Escherichia coli lipopolysaccharide (LPS). Results Proteomic analysis indicated that 111 proteins were differentially expressed in the liver of broiler chickens from the immune stress group. Of these, 28 proteins were down-regulated, and 83 proteins were up-regulated in the immune stress group. Enrichment analysis showed that immune stress upregulated the expression of hepatic proteins involved in defense function, amino acid catabolism, ion transport, wound healing, and hormone secretion. Furthermore, immune stress increased valine, leucine and isoleucine degradation pathways. Conclusion The data suggests that growth depression of broiler chickens induced by immune stress is triggered by hepatic proteome alterations, and provides a new insight into the mechanism by which immune challenge impairs poultry production.

Chairman Blattner: During recent years, there has been increasing interest shown in diseases caused by filterable viruses, and significant work has been accomplished in this comparatively new and absorbing field of endeavor. With the advent of chemotherapeutic agents and antibiotics, the presence and action of these infectious agents has become more apparent. Viral diseases, therefore, have assumed increasing importance in medical literature in general and in pediatric literature in particular. By way of review, it is well to bear in mind that viruses are filter-passing agents, obligate intracellular parasites, capable of reproducing themselves and of producing disease in plants and animals, including man. While these agents cannot be seen except by the most elaborate methods, their presence can be detected by their injurious effects. The pathologic picture produced by viral agents is rather characteristic and can be recognized readily by experienced observers acquainted with tissue response. In some instances, inclusion bodies are produced which may be intranuclear or intracytoplasmic, and represent cytologic changes which are considered typical of the pathologic response to viral invasion. When inclusion bodies are present they may serve as sign posts for the recognition of the type of infectious agent. The nature of a filterable virus is as yet unknown. Viruses may be a form of life similar to bacteria, but infinitely smaller in size. It is conceivable that viruses are enzymes capable of reproducing themselves and capable of producing cellular response. They may be non-living, crystallizable substances, such as the Stanley tobacco-mosaic virus; or a form of life, the definite nature of which is as yet unrecognized. Dr. Thomas M. Rivers has stated : "Viruses are a heterogeneous collection of diverse agents which happen to induce a state of broad similarity." He points out that the reaction of the tissues in general, and of the cells in particular, determines the nature of the pathologic process about as much as the infectious agent itself.

ABSTRACT: The aim of this research was to evaluate the changes in intestinal tract microbiota, hematological parameters and zootechnical indexes of yellowtail lambaris (Astyanax bimaculatus) fed with different probiotic supply frequencies (Lactobacillus spp.). Lambaris (1,200) were distributed in twelve boxes, divided in three treatments, 100% supply; 25% and 0% (control) of feeds supplemented with probiotic. After 87 days, fish fed with supplemented diet, regardless of frequency (100% and 25%), showed an increase in the total lactic acid bacteria count of 7.99±0.15 log CFU mL-1 and 7.04±1, 29 log UFC mL-¹ respectively, in the intestinal tract and decrease of Pseudomonas spp. in both 100% and 25% (4.09±0.84log UFC mL-1 and 4.00±1.12log UFC mL-1, respectively) in relation to the control treatment (6.74±1.04log UFC mL-1). Fish fed with 100% of diets supplemented with probiotics had a higher leukocyte count (47.7±6.7x103μL-1), lymphocytes (36.0±2.6x103μL1), monocytes (12.0±4.2x103μl-1), as well as higher mean final weight (10.60±0.89g) and weekly growth rate (0.80±0.08g.week-1) than the other treatments (25% of probiotic offered and control). Probiotic (Lactobacillus spp.) offered in 25% of feeds (or more) alters the yellowtail lambari microbiota (A. bimaculatus); however, must be offer in 100% of feeds to improve the hematology profile and productive performance.

The poultry industry is characterized for its constant search for productivity and profitability, which are based on flock health status. Brazilian Commercial laying hens (Gallus gallus domesticus) have been impacted significantly by mite infestations. This review aims to compile the literature on the occurrence, economic losses, biology, epidemiology and control of mite species considered important for the Brazilian laying poultry industry. The national experience was compared with practices of other countries and a scarcity of studies on this subject in Brazil was evident. The poultry industry has prioritized the use of pesticides to control infestations with little regard for the adverse effects. In this context, the integrated control programs using several strategies simultaneously constitute the best alternative to mite control. Integrated control programs involve measures of chemical, physical and biological nature, as well as attention to cultural aspects. However, studies should be performed aiming at the development of new control methods, evaluating the adequacy of practices developed in other countries to the national reality.

ObjectivesThe woody breast (WB) myopathy has caused economic losses in excess of $200 million annually to the poultry industry due to undesirable textural attributes and decreased functionality. This hardened muscle is also associated with other undesirable traits, such as white striping. This research was conducted to evaluate the impact of WB severity and genetic strain on consumer acceptability and sensory attributes of baked and fried broiler breast meat and elucidate the consumer acceptability of tumble-marinated, fajita meat made from broilers with normal (NOR), moderate (MOD) and severe (SEV) WB meat.Materials and MethodsFor descriptive analysis (n = 7 panelists, 10 panels) on baked and fried chicken, 3 × 5 factorial arrangements within randomized complete block designs with four replications were utilized to evaluate three severities of woody breast and the five different genetic strains that are most commonly used in the poultry industry. When significant differences (P < 0.05) occurred among treatments, Duncan’s multiple range test was utilized to separate treatment means. For consumer acceptability of baked chicken (n = 123 panelists), fried chicken (n = 125 panelists), and fajita meat (n = 127 panelists), randomized complete block designs with two replications were used to determine the impact of strain and severity on acceptability.ResultsFor baked chicken, SEV breasts were chewier, juicier, crunchier, and more cohesive (P < 0.05) than NOR and MOD breast samples. For fried chicken, SEV breasts were less tender and chewier (P < 0.05) than NOR breasts. In addition, SEV breasts were more cohesive and juicier, but less mushy (P < 0.05) than NOR and MOD breasts. For fried chicken samples, SEV breasts were crunchier (P < 0.05) than MOD breasts, which were crunchier (P < 0.05) than NOR breasts. The texture and overall acceptability of NOR baked breasts and fajita meat were preferred by consumers (P < 0.05) over SEV breasts. In contrast, the SEV breasts were preferred (P < 0.05) over the NOR breast meat for the fried chicken formulation. No differences existed (P > 0.05) in acceptability among genetic strains in baked or fried chicken breasts. The baked chicken consumer panelists were divided into 7 distinct clusters based on their sensory evaluation ratings. Cluster analysis indicated that 49% of panelists preferred NOR breast fillets, 21% preferred SEV, and 30% had no preference between NOR and WB (MOD, SEV) samples. The fried chicken consumer panelists were divided into 5 clusters, of which 65% preferred WB (MOD, SEV) over NOR, 29% preferred strain B over strain A, and 11% preferred strain A over strain B. The fajita chicken meat consumer panelists were divided into 5 clusters, of which 75% of panelists liked NOR breast samples, 72% liked MOD samples, and 45% liked SEV samples.ConclusionResults indicated that WB severity had a greater impact on sensory attributes and consumer acceptability than genetic strain. Higher WB severity created an undesirable texture that negatively impacted the acceptability of baked meat. However, the increased crunchiness and cohesiveness due to woodiness had a positive impact on the fried chicken acceptability. Results indicated that a large percentage of consumers rated baked, fried, and fajita samples as acceptable regardless of whether NOR or WB (MOD, SEV) meat was used, but some consumers did not like baked or fajita meat that was made from SEV WB meat.
