The stone of Cornus mas L. remains the least known morphological part of this plant, whereas the fruit is appreciated for both consumption purposes and biological activity. The stone is considered to be a byproduct of fruit processing and very little is known about its phytochemical composition and biological properties. In this study, the complete qualitative determination of hydrolyzable tannins, their quantitative analysis, total polyphenolic content, and antioxidant properties of the stone of C. mas are presented for the first time. The 37 identified compounds included the following: various gallotannins (11), monomeric ellagitannins (7), dimeric ellagitannins (10), and trimeric ellagitannins (7). The presence of free gallic acid and ellagic acid was also reported. Our results demonstrate that C. mas stone is a source of various bioactive hydrolyzable tannins and shows high antioxidant activity which could allow potential utilization of this raw material for recovery of valuable pharmaceutical or nutraceutical substances. The principal novelty of our findings is that hydrolyzable tannins, unlike other polyphenols, have been earlier omitted in the evaluation of the biological activities of C. mas. Additionally, the potential recovery of these bioactive chemicals from the byproduct is in line with the ideas of green chemistry and sustainable production.

AbstractThe genus Santolina is a taxonomically complex group of plant species widely distributed in the Mediterranean flora and used in traditional medicine since ancient times for their biological properties, including antimicrobial, anti-inflammatory, antispasmodic, digestive, and analgesic activities. Phytochemical investigations of Santolina species have revealed the presence of terpenoids as the main bioactive constituents of the genus. Coumarins and flavonoids were also identified. This review deals, for the first time, with information on the traditional uses, chemical profile, and biological properties of plants of the genus Santolina in order to provide input for future research prospects.

Objective: Study the influence of the mechanical preparation methods (grinding, fluidization) of solid pharmaceutical substances (PS) and herbal raw material on their physicochemical properties and biological activities. Methods: Test substances and solvents-Lactose monohydrate (DFE Pharma, Germany). Sodium chloride, bendazol hydrochloride (all Sigma-Aldrich, USA) and herbal raw material (Callisia fragrans). The dispersity and native structure of pharmaceutical substances were analyzed by several methods: optical microscopy–Altami BIO 2 microscope (Russia); low angle laser light scattering (LALLS) method (Malvern Instruments, UK); Spirotox method–Quasichemical kinetic of cell transition of cellular biosensor Spirostomum ambiguum; Fourier-transform infrared spectroscopy–the analysis in the middle IR region was carried out using an IR Cary 630 Fourier spectrometer (Agilent Technologies, USA). The analysis of dried leaves of C. fragrans before and after mechanical activation was performed using Shimadzu EDX-7000 X-ray fluorescence spectrophotometer without mineralization (Shimadzu, Japan). Results: It was established that the mechanical change, such as dispersion and drying, alters the biological activity of PS and herbal raw materials. The observed increase in the influence of the dispersed substance on the biosensor S. ambiguum is quantitatively estimated from the values of the activation energy (obsEa), which turns to be valued 1,5 (P≤0,05) times more than for the native form substance. In the study of the dependence of the availability of chemical elements K, Ca, Zn on the degree of dispersion of herbal raw materials was established a quantitative 4-fold (P≤0,05) increase in the concentration of elements in mechano-activated raw materials. Conclusion: By the example of the biological model of Spirotox (single-celled biosensor S. ambiguum) and herbal raw materials obtained from C. fragrans, the increase of biological activity of PS at the dispersion of initial preparations was proved.

Isatis tinctoria L. (Brassicaceae), which is commonly known as woad, is a species with an ancient and well-documented history as an indigo dye and medicinal plant. Currently, I. tinctoria is utilized more often as medicinal remedy and also as a cosmetic ingredient. In 2011, I. tinctoria root was accepted in the official European phytotherapy by introducing its monograph in the European Pharmacopoeia. The biological properties of raw material have been known from Traditional Chinese Medicine (TCM). Over recent decades, I. tinctoria has been investigated both from a phytochemical and a biological point of view. The modern in vitro and in vivo scientific studies proved anti-inflammatory, anti-tumour, antimicrobial, antiviral, analgesic, and antioxidant activities. The phytochemical composition of I. tinctoria has been thoroughly investigated and the plant was proven to contain many valuable biologically active compounds, including several alkaloids, among which tryptanthrin, indirubin, indolinone, phenolic compounds, and polysaccharides as well as glucosinolates, carotenoids, volatile constituents, and fatty acids. This article provides a general botanical and ethnobotanical overview that summarizes the up-to-date knowledge on the phytochemistry and biological properties of this valuable plant in order to support its therapeutic potential. Moreover, the biotechnological studies on I. tinctoria, which mainly focused on hairy root cultures for the enhanced production of flavonoids and alkaloids as well as on the establishment of shoot cultures and micropropagation protocols, were reviewed. They provide input for future research prospects.

This research was aimed at investigating polyphenolic content, antioxidant and anti-neurodegenerative activities of post-distillation waste extracts of Macedonian Salvia amplexicaulis, S. jurisicii and S. ringens, for the first time. Total phenolic and flavonoid contents varied in a broad range (3.92-146.49 mg GAE/g and 7.11-67.51 mg QE/g, respectively), with the highest values obtained for S. amplexicaulis and S. ringens extracts. Certain S. amplexicaulis and S. ringens extracts neutralized more than 80% of DPPH radicals at the highest concentration, while S. amplexicaulis extracts showed up to ≈50% inhibition of β-carotene oxidation in β-carotene/linoleic acid assay. Post-distillation waste extracts inhibited acetylcholinesterase (25.94-38.15%) and tyrosinase (18.84-59.52%), with the strongest inhibition of S. amplexicaulis extracts. The obtained results suggest that post-distillation waste of tested species, especially of S. amplexicaulis, show antioxidant and anti-neurodegenerative activities and could be considered as potential raw material rich in polyphenols.

1,3-Oxazole chemicals are a unique class of five-membered monocyclic heteroarenes, containing a nitrogen atom and an oxygen. These alkaloids have attracted extensive attention from medicinal chemists and pharmacologists owing to their diverse arrays of chemical structures and biological activities, and a series of 1,3-oxazole derivatives has been developed into therapeutic agents (e.g., almoxatone, befloxatone, cabotegravir, delpazolid, fenpipalone, haloxazolam, inavolisib). A growing amount of evidence indicates that marine organisms are one of important sources of 1,3-oxazole-containing alkaloids. To improve our knowledge regarding these marine-derived substances, as many as 285 compounds are summarized in this review, which, for the first time, highlights their sources, structural features and biological properties, as well as their biosynthesis and chemical synthesis. Perspective for the future discovery of new 1,3-oxazole compounds from marine organisms is also provided.

In this work, the phytochemical profile and the biological properties of Colchicum triphyllum (an unexplored Turkish cultivar belonging to Colchicaceae) have been comprehensively investigated for the first time. Herein, we focused on the evaluation of the in vitro antioxidant and enzyme inhibitory effects of flower, tuber, and leaf extracts, obtained using different extraction methods, namely maceration (both aqueous and methanolic), infusion, and Soxhlet. Besides, the complete phenolic and alkaloid untargeted metabolomic profiling of the different extracts was investigated. In this regard, ultra-high-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UHPLC-QTOF-MS) allowed us to putatively annotate 285 compounds when considering the different matrix extracts, including mainly alkaloids, flavonoids, lignans, phenolic acids, and tyrosol equivalents. The most abundant polyphenols were flavonoids (119 compounds), while colchicine, demecolcine, and lumicolchicine isomers were some of the most widespread alkaloids in each extract analyzed. In addition, our findings showed that C. triphyllum tuber extracts were a superior source of both total alkaloids and total polyphenols, being on average 2.89 and 10.41 mg/g, respectively. Multivariate statistics following metabolomics allowed for the detection of those compounds most affected by the different extraction methods. Overall, C. triphyllum leaf extracts showed a strong in vitro antioxidant capacity, in terms of cupric reducing antioxidant power (CUPRAC; on average 96.45 mg Trolox Equivalents (TE)/g) and ferric reducing antioxidant power (FRAP) reducing power (on average 66.86 mg TE/g). Interestingly, each C. triphyllum methanolic extract analyzed (i.e., from tuber, leaf, and flower) was active against the tyrosinase in terms of inhibition, recording the higher values for methanolic macerated leaves (i.e., 125.78 mg kojic acid equivalent (KAE)/g). On the other hand, moderate inhibitory activities were observed against AChE and α-amylase. Strong correlations (p < 0.01) were also observed between the phytochemical profiles and the biological activities determined. Therefore, our findings highlighted, for the first time, the potential of C. triphhyllum extracts in food and pharmaceutical applications.

Twenty-nine mushrooms collected in the mountainous areas of Nepal were analyzed for antioxidant activity by different methods, including Folin-Ciocalteu, ORAC, ABTS, and DPPH assays. Intracellular H2O2-scavenging activity was also performed on HaCaT cells. The results showed that phenolic compounds are the main antioxidant of the mushrooms. Among studied samples,Inonotus andersonii, andPhellinus gilvusexhibited very high antioxidant activity with the phenolic contents up to 310.8 and 258.7 mg GAE/g extracts, respectively. The H2O2-scavenging assay on cells also revealed the potential of these mushrooms in the prevention of oxidative stress. In term of ACE-inhibition, results showed thatPhlebia tremellosawould be a novel and promising candidate for antihypertensive studies. This mushroom exhibited even higherin vitroACE-inhibition activity thanGanoderma lingzhi, with the IC50values of the two mushrooms being 32 μg/mL and 2 μg/mL, respectively. This is the first time biological activities of mushrooms collected in Nepal were reported. Information from this study should be a valuable reference for future studies on antioxidant and ACE-inhibitory activities of mushrooms.

Nitrogen containing heterocyclic compounds such as oxindoles especially isatins and their derivatives have excellent biological properties such as anti-inflammatory, antimicrobial, anticancer activity. In addition, this is first time ever we would like to report the antioxidant properties of the said class of compounds. To determine such an important bioactivity a number of halogenated oxindoles 1-3, their diaza-biphenyl derivatives (4-6), ring extended biphenyl (7) and bis-amide (8) were synthesized by classical heating method. The antioxidant activity of all the synthesized compounds was screened by DPPH method with respect to ascorbic acid. Aims: Boscia agustifolia and Gardenia erubescens are two medicinal plants widely used in the Mouhoun region. The strong use of these two plants in traditional medicine would be linked to their therapeutic virtues. Study Design: The purpose of this work was to carry out a preliminary biological study on two plants widely used by the population of the Mouhoun region (Burkina Faso) against certain diseases. Place and Duration of Study: The harvest of plant material was made in the Mouhoun region in March 2019. The phytochemistry and the antioxidant tests were carried out at LABIOCA in June 2019. The microorganism tests were carried out at CRSBAN. Methodology: The extracts were obtained by ethanolic maceration. The FRAP method and DPPH were used to evaluate the antioxidant properties of the extracts. The antibiomicrobial activity of the extracts was determined using five microbial strains. Results: The ethanolic extract of Gardenia erubescens bark had antioxidant activity through the iron ion reduction capacity (6.71 ± 1.16 mMol EAA/100 g extract. Boscia angustifolia showed inhibitory activity on the five microbial strains. Conclusion: The biological activities obtained with these extracts could be justified by the presence of active phytochemicals such as flavonoids. These biological properties to constitute a reason based on the strong use of these two plants in traditional medicine.

The seaweed collected after stranding on beaches of Latvia is underexploited natural resource, which has a potential as raw material for biologically active compound extraction for cosmetic and pharmacy and fertilizer in sustainable agriculture. The aim of the present study was development of the approach for the processing of brown alga Fucus vesiculosus biomass, collected from the Gulf of Riga. The thorough characterization of the chemical composition of Fucus vesiculosus has shown that it is a potential source for obtaining of nitrogen-containing fertilizers, and biologically active compounds. One of the proposed approaches for the processing of the Fucus vesiculosus biomass under study includes algae extraction with organic solvents and CaCl2 solution and obtaining soil organic amendment on the basis of the extract-free residue. The ethyl acetate extract was rich in phenolic compounds (430 ± 30 GAE mg/g) with high antioxidant activity in DPPH• and ABTS•+ tests. The ethanol extract contained significant amounts of phlorotannins that was confirmed by the data of LC-MS/MS analysis. The CaCl2 extract was used for the obtaining of sulphated polysaccharide fucoidane (yield ≈7% on the basis of oven dry matter, o.d.m.), which has numerous biological activities. The extract-free residue didn’t show phytotoxicity. The extract free algal biomass exhibited positive effect in root elongation tests with oat (Avena sativa). As alternative approach for processing of the algal biomass, the mechano-chemical treatment of algal biomass with lignin was proposed. EPR analyses confirmed interaction between algal biomass and lignin.
