Lichens represent a significant source of antioxidants due to numerous metabolites that can reduce free radicals. Usnea barbata (L.) F.H. Wigg. has been recognized and used since ancient times for its therapeutic effects, some of which are based on its antioxidant properties. The present study aims to analyze the phytochemical profile and to evaluate the antioxidant and cytotoxic potential of this lichen species. Five dry extracts of U. barbata (UBDE) in different solvents (acetone, ethyl acetate, ethanol, methanol, water) were prepared by refluxing at Soxhlet to achieve these proposed objectives and to identify which solvent is the most effective for the extraction. The usnic acid content (UAC) was quantified by ultra-high performance liquid chromatography (UHPLC). The total polyphenols content (TPC) and tannins content (TC) were evaluated by spectrophotometry, and the total polysaccharides (PSC) were extracted by a gravimetric method. The 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) free radical method was used to assess the antioxidant activity (AA) and the Brine Shrimp Lethality (BSL) assay was the biotest for cytotoxic activity evaluation. The ethyl acetate extract had the highest usnic acid content, and acetone extract had the highest content of total polyphenols and tannins. The most significant antioxidant effect was reported to methanol extract, and all the extracts proved high cytotoxicity. The water extract has the lowest cytotoxicity because usnic acid is slightly soluble in this solvent, and it was not found at UHPLC analysis. All extracts recorded a moderate correlation between the content of usnic acid, polyphenols, tannins, and AA; furthermore, it has been observed that the cytotoxicity varies inversely with the antioxidant effect.

Nowadays, knowledge of ancient botanical medicinal practices and application of modern phytochemical techniques have provided the excellent tools for the purification and structural elucidation of various phyto-compounds, which, in turn, has given insights into their mode of action on the human body. This study has been designed to investigate for the first time the antioxidant effects of the ethyl acetate, methanolic, and water extracts of Kitaibelia balansae. Different chemical methods were performed and the observed abilities depend on the solvent used. The best antioxidant ability was noted in water extract, followed by methanol and ethyl acetate extracts. The highest level of phenolic was also detected in water extract. The present findings suggest that K. balansae can be considered as a potential source of bioactive compounds for novel phytopharmaceuticals development

Bersama abyssinica, belonging to the Melianthaceae family, is distributed across Sub Saharan Africa. Decoctions of the bark, leaves, and roots of B. abyssinica have been extensively used in traditional medicine to manage many stomach complications such as colic, diarrhea, dysentery, and intestinal worms. In this study, we examined three extracts (ethyl acetate, methanol and water) obtained from B. abyssinica stem barks in terms of antioxidant properties. The antioxidant abilities were investigated by different chemical methods, including free radical scavenging (DPPH and ABTS), reducing power (CUPRAC and FRAP), metal chelating and phosphomolybdenum assay. In addition, total phenolic and flavonoid contents in the extracts were calculated. The highest level of phenolics was determined in water extract (230.83 mg GAE/g extract), followed by methanol (216.79 mg GAE/g extract) and ethyl acetate (100.57 mg GAE/g extract). In same line with total phenolic content, the best antioxidant properties were noted for water and methanol extracts. Our findings suggested that B. abyssinica stem bark extracts could be considered as promising sources of natural antioxidants.

In this study, ethyl acetate, methanol, and water extracts of Bersama abyssinica (Melianthaceae) stem bark were screened for enzyme inhibitory and antioxidant properties. The water extract possessed the highest concentration of phenols (230.83 mg gallic acid equivalent/g extract), while the methanol extract was rich in flavonoids (75.82 mg rutin equivalent/g extract), and the ethyl acetate extract possessed the highest amount of saponins (97.37 mg quillaja equivalent/g). The aim of this study was to investigate the antiproliferative effects against the human colon cancer HCT116 cell line challenged with serotonin (5-HT) as a stimulating-proliferation factor. The level of HCT116 cell-deriving pool of kynurenic acid (KA) was also assessed. The UHPLC results confirmed the presence of 58, 68, and 63 compounds in the ethyl acetate, methanol, and water extracts, respectively. Mangiferin, vitexin and its isomer isovitexin were tentatively identified in all extracts and KA (m/z 190.05042 [M−H]+) was also tentatively identified in the methanol and water extracts. The methanol extract (1464.08 mg Trolox equivalent [TE]/g extract) showed the highest activity in the CUPRAC assay, whereas the water extract (1063.70 mg TE/g extract) showed the highest activity with the FRAP technique. The ethyl acetate extract was the most active acetylcholinesterase (4.43 mg galantamine equivalent/g extract) and α-glucosidase (mmol acarbose equivalent /g extract) inhibitor. The water extract was able to inhibit 5-HT-stimulated viability of HCT116 cells, and blunt 5-HT-induced reduction of cell-deriving KA. The scientific data generated in this study provide baseline data regarding the biological properties of B. abyssinica stem bark, highlighting its potential use for the development of new pharmaceutic and cosmetic agents.

<p>In this work, we are interested in both the study of the phenolic compounds and the evaluation of the antioxidant properties of the extracts of <em>Ammi visnaga L. </em>collected from the Middle Atlas in Morocco. Phytochemical screening was carried out to highlight the qualitative composition of the secondary metabolites contained in this plant. Also, the extraction of total polyphenols was carried out by maceration using hydromethanolic and hydroacetonic mixtures. Then, the fractionation of the crude extracts was conducted by liquid-liquid extraction using successively two organic solvents with different polarities: ethyl acetate and n-butanol. The polyphenols contents were spectrophotometrically estimated using the Folin-Ciocalteau method. It showed that the ethyl acetate fraction is more abundant in phenolic compounds than the other fractions. The qualitative analysis was performed by high-pressure liquid chromatography with U.V. and electrospray ionization coupled to mass spectrometry (HPLC/UV-ESI-MS), which confirmed the richness of this species in polyphenols and particularly in flavonoids, especially by the identification of kaempferol, rhamnetin, biochanin A, ammiol. These molecules belong to flavones, flavonols, and furochromones, which are also identified both in the form of genin and of heteroside.<strong> </strong>The antioxidant activity of the different fractions was evaluated by diphenyl-picryl-hydrazyl (DPPH<sup>●</sup>). The IC<sub>50</sub> is equal to 12.87 μg / mL for the ethyl acetate fraction, against 8.8 μg / mL for ascorbic acid and 10.8 μg / mL for BHA which are used as reference compounds.</p>

Objective: Phytochemical constituents of various solvent extracts of leaf and stem bark of Polyalthia fragrans (Dalz.) Bedd were screened. Antimicrobial and antioxidant activities of the extracts were evaluated.Methods: Crude extracts of methanol, ethyl acetate, chloroform and aqueous of leaf and stem bark were evaluated for antimicrobial activity by disc diffusion method and antioxidant activity by DPPH (diphenyl-2-picrylhydrazyl) radical scavenging and reducing power assay. Quantitative analysis of total phenolics was done by the Folin-Ciocalteau method and total flavonoids by aluminum chloride method.Results: The study revealed the presence of several physiologically active phytochemicals such as alkaloids, phenols, flavonoids and tannins. The highest antibacterial activity of methanol extract of the leaf was observed against Bacillus subtilis with 14.33±0.57 mm inhibition zone and water extract of stem bark against Proteus vulgaris with 17.67±1.52 mm inhibition zone among other extracts. The lowest activity of leaf methanol and leaf water extract was observed against Bacillus subtilis. The IC50 values of 58.18±2.04 µg/ml, 116.58±2.43 µg/ml, 134.78±3.64 µg/ml and 387.28±2.9 µg/ml for DPPH activity were observed in methanol stem bark, ethyl acetate stem bark, methanol leaf and ethyl acetate leaf extracts respectively. The reducing power of extracts was very potent which increased with increasing concentrations of the sample.Conclusion: The results indicate that phytochemicals like alkaloids, phenols, flavonoids and tannins may be responsible for the antimicrobial and antioxidant properties. The results suggest that P. fragrans can be further researched to be used as a natural source of a potent drug by mankind.

The antioxidant properties of Glechoma hederacea L. (Lamiaceae), of Serbian origin, were studied in respect to its potential use in foodstuffs. Ethanol-water (8:2, v/v) and purified ethyl acetate extracts of the plant were found to possess significant antioxidant activity. Tests were performed on two different substrates, prime steam pork lard and active-carbon-treated edible sunflower oil, using Schaal oven test storage conditions at 60°C. The ethanol-water and purified ethyl acetate extracts of G. hederacea showed strong concentration-dependent antioxidant activity. On the contrary, under the Rancimat method conditions at 120°C, the ethanol-water extract showed significantly stronger antioxidant activity, in comparison with the other tested extracts. All activities were compared with commercial antioxidants, such as BHA and a tocopherol mixture, respectively. For the first time, the activity of the flavonol quercetagetin was determined.

Aim: This study was designed to investigate the phytochemical profile and the cytotoxic activities of the eco-friendly extracts of olive leaves from Chemlali cultivar. Materials and Methods: The Phenolic composition of olive leaves extracts, the antioxidant activity and the cytotoxic effects against MCF-7 and HepG2 cells were determined. Results: Olive leaves extracts showed relevant total polyphenols contents. Oleuropein was the major detected phenolic compound reaching a concentration of 16.9 mg/ml. The antioxidant potential of the studied extracts varied from 23.7 to 46.5mM Trolox equivalents as revealed by DPPH and ABTS assays. Cytotoxicity experiments showed similar trends for both HepG2 and MCF-7 cells with the infusion extract being the most active. Conclusion: This study denotes that olive leaves may have great potential as endless bioresource of valuable bioactive compounds which may have a wide application.

Background: Snakebite envenomation is a global priority ranked top among other neglected tropical diseases. There is a folkloric claim that Uvaria chamae is beneficial for the management of snakebite and wounds in African ethnobotanical surveys. Besides, there are many registered patents asserting the health benefits of U. chamae. Objective: This study aimed to investigate U. chamae’s potentials and identify candidates for the development of tools for the treatment and management of N. nigricollis envenomation. Methods: Freshly collected U. chamae leaves were air-dried, powdered, and extracted in methanol. The median lethal dose of the extract was determined and further fractionated with n-hexane, n-butanol and ethyl acetate. Each fraction was tested for neutralizing effect against venom-induced haemolytic, fibrinolytic, hemorrhagic, and cytotoxic activities. Results: U. chamae fractions significantly (p<0.05) neutralized the haemolytic activity of N. nigricollis venom in n-butanol; 31.40%, n-hexane; 33%, aqueous residue; 39.60% and ethyl acetate; 40.70% at the concentration of 100mg/ml of each fraction against 10mg/ml of the snake venom when compared to the positive control. The fibrinolytic activity of N. nigricollis venom was significantly (p<0.05) neutralized in n-hexane at 73.88%, n-butanol; 72.22% and aqueous residue; 72.22% by the fractions of U. chamae. In addition, haemorrhagic activity of N. nigricollis venom was significantly (p<0.05) neutralized by U. chamae fractions at the concentrations of 100mg/ml, 200mg/ml and 400mg/ml except for n-butanol and aqueous residues at 400 mg/ml. Conclusion: U. chamae leaves fractions possess a high level of protection against N. nigricollis venoms-induced lethality and thus validate the pharmacological rationale for its usage in the management of N. nigricollis envenomation.

Sarcopoterium spinosum (L.) is a medicinal plant traditionally used for the treatment of various diseases including cancer in the Near- and Middle East. The fractions and constituents of the ethanol extract of S. spinosum were screened for in vitro cytotoxic activities on Jurkat (acute T-lymphoblastic leukemia), HeLa (cervical adenocarcinoma), MCF-7 (mammary gland adenocarcinoma), Caco-2 (human colorectal adenocarcinoma), and MDA-MB-231 (mammary gland adenocarcinoma) cell lines using the MTT (3-(dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay. The ethanol extract was subsequently re-extracted with ethyl acetate and in its sub-fraction obtained by column chromatography three compounds (stachydrine, benzalkonium chloride and rutine) were the first time identified by nuclear magnetic resonance (NMR) analyses. The most active subfraction showed cytotoxic activity against HeLa, MCF-7, and Caco-2 cell lines. The three compounds mentioned, as standards of high-performance liquid chromatography (HPLC) quality, were studied individually and in combination. Cytotoxic activity observed might be due to the presence of benzalkonium chloride and rutin. Benzalkonium chloride showed the strongest growth suppression effect against HeLa cells (IC50 8.10−7 M) and MCF-7 cells (IC50 5.10−6 M). The mixture of stachydrine and benzalkonium chloride allowed a synergistic cytotoxic effect against all tested cancer and normal cells to be obtained. Anti-cancer activity of the plant extract of S. spinosum remains under-investigated, so this research describes how the three major compounds identified in the ethyl acetate extract can exert a significant dose dependent in vitro cytotoxicity.
