Cellulase adsorption onto lignin decreases the productivity of enzymatic hydrolysis of lignocellulosic biomass. Here, adsorption of enzymes onto different types of lignin was investigated, and the five major enzymes—cellobiohydrolases (CBHs), endoglucanase (Cel7B), β-glucosidase (Cel3A), xylanase (XYNIV), and mannanase (Man5A)—in a cellulase cocktail obtained from Trichoderma reesei were individually analyzed through SDS-PAGE and zymogram assay. Lignin was isolated from woody (oak and pine lignin) and herbaceous (rice straw and kenaf lignin) plants. The relative adsorption of CBHs compared to the control was in the range of 14.15–18.61%. The carbohydrate binding motif (CBM) of the CBHs contributed to higher adsorption levels in oak and kenaf lignin, compared to those in pine and rice lignin. The adsorption of endoglucanase (Cel7B) by herbaceous plant lignin was two times higher than that of woody lignin, whereas XYNIV showed the opposite pattern. β-glucosidase (Cel3A) displayed the highest and lowest adsorption ratios on rice straw and kenaf lignin, respectively. Mannanase (Man5A) was found to have the lowest adsorption ratio on pine lignin. Our results showed that the hydrophobic properties of CBM and the enzyme structures are key factors in adsorption onto lignin, whereas the properties of specific lignin types indirectly affect adsorption.

Abstract Background Agriculture by-products are considered a great potential value for utilization by ruminants as well as rabbits. They usually can be the maintenance and part of the production requirements. However, in developing countries, as well as in Egypt, animals suffer from shortage of feeds that are continuously increasing in costs. In general, biological treatments were shown to be the most effective and improved chemical composition of rice straw or corn stalks. Method This work aimed to investigate the possible ways of utilizing rice straws or corn stalks in rabbit feeding. The field work is designed to study the effect of biological treatment of Pleurotus ostreatus cultivated on rice straws and Trichoderma reesei cultivated on corn stalks and replacing clover hay by rice straws and corn stalks at levels of 0, 33, 66, and 100% either without or with microbes adding. Seventy-eight New Zealand White (NZW) rabbits aged 4–5 weeks (565 ± 13.57 g) were randomly divided into thirteen equal experimental groups. Results Untreated rice straws or biologically treated with Pleurotus ostreatus increased their contents of crude protein (CP) by 178.75 and 224.5% and nitrogen-free extract (NFE) by 6.30 and 24.53, respectively. Meanwhile, crude fiber (CF) content was reduced by 31.32 and 56.75%, and organic matter content was decreased by 2.81 and 5.51%, respectively, in comparison with the raw rice straws. Also, biological treatment of rice straws caused a decrease in values of neutral detergent fiber (NDF), acid detergent fiber (ADF), acid detergent lignin (ADL), and hemicellulose contents in comparison with either raw or treated rice straws. Furthermore, biological treatment with Trichoderma reesei realized a decrease in organic matter (OM), CF, NDF, and ADF and increased CP and ash contents in corn stalks. NFE content of corn stalks was decreased as a result of treatment without or with Trichoderma reesei experimental rations by 11.95% and 3.82% compared to raw corn stalks (CS). Biological treatments with fungi significantly (P < 0.05) improved average daily gain (ADG) and feed conversion. ADG and feed conversion were significantly (P < 0.05) improved when rabbits were fed diets containing rice straw (RS) compared to that fed CS. Rabbits fed diets replaced clover hay (CH) with 33% or 66% of RS or CS significantly increased ADG compared to control and that replaced 100% of both RS and CS containing rations. Levels of replacing had no significant effect on their dry matter intake (DMI) values. The highest improvement in feed conversion was recorded with rabbits that received diets replaced 33% of berseem hay (BH) by RS or CS, followed by that replaced 66% of BH by RS or CS. There were significantly interactions between biological treatments (T), roughage source (S), and replacement levels (L) (T × S × L) only on ADG. The best fed conversion was realized by rabbits fed diet replaced BH with 33% of RS that are treated by Pleurotus ostreatus (4.05 g DMI/g gain). Rabbits fed 33% biologically treated rice straw with Pleurotus ostreatus showed the highest economic efficiency (179%) followed by rabbits that received 33% of both rice straws treated without Pleurotus ostreatus and rabbits that received corn stalks biologically treated with Trichoderma reesei (161%). Conclusion Biological treatments of rice straws by Pleurotus ostreatus or corn stalks by Trichoderma reesei were safe, and it improves their chemical analysis and improved both daily gain and feed conversion, decreasing the costing of diet formulation which consequently decreased the price of 1-kg live body weight.

This paper proposed a scheme design for Sydney’s frontport check-in system, which completes check-in and baggage drop-off at Sydney’s Circular Quay, and transports the baggage to Sydney Kingsford Smith Airport by waterway, and provided a strengths, weaknesses, opportunities and threats (SWOT) analysis of Sydney’s frontport check-in system. Using the process method of quality management, the frontport check-in process was divided into three sub-processes: baggage consignment, baggage packing and transportation, and airport baggage handling. The eight key elements of each sub-process such as input, output, resources, and methods, etc. were discussed, the key factors influencing the cost of baggage transportation were analyzed, and the cost control measures such as adopting economic speed, reducing fuel consumption of the main engine, improving the ship loading rate, and raising loading and unloading efficiency were proposed. At the same time, two different types of baggage transportation ships and other parameters that affect the cost such as the number of berths, ships, lifting machineries, and the yard area were analyzed and calculated through calculation cases. This scheme is a beneficial addition to the existing in-town check-in system.

Purpose The purpose of this paper is to explore how an organization can combine different types of open innovations and what are the key factors that may influence the combination of different open innovations. Design/methodology/approach The basic methodology of this paper is the longitudinal inductive analysis within the conceptual framework of the open innovation proposed by Dahlander and Gann (2010). In this case study of Xiaomi Tech Inc., the open innovation combination is investigated through examining 25 new products created between August 2010 and December 2016 in terms of four general types: acquiring, sourcing, selling and revealing open innovation. Findings In practice, the combination of different types of open innovations can be realized. A firm may combine different open innovations at three levels: a single product level, a related product cluster level and a company level. In addition, different open innovations can be combined in diverse modes. The purpose of combining different types of open innovations is to overcome the disadvantages of each type and to exploit the advantages of all different types. Many factors may affect a firm’s option of how to combine open innovations. At different development stages, a firm may make and implement corresponding strategic direction based on its innovation capacity and internal resource. For a given strategy, the firm needs to create profits and manage intellectual property in the implementation of open innovations. These factors are interacted each other, rather than isolated. Originality/value The findings of this paper are helpful for better understanding how and why an organization can combine different types of open innovations. From a managerial point of view, an organization may combine different types of open innovations to leverage advantages and avoid disadvantages of each certain type of open innovation. An appropriate combination of different open innovations can effectively improve new product development.

One of the latest trends in the transport field is the increasing interest for the rejuvenation of the railway. It is considered to be a logical consequence of the gradual switch towards a more sustainable future in transports. Terminals and stations are considered to be the junction points between the various lines that constitute the railway network and can simply be described as points of arrival, departure and interchange of passengers or commodities. The most commonly used indicators that measure the level of their performance are time and cost. This study aims at exploring possible improvements that could be implemented to the infrastructure and the operation of terminals and stations in order to increase the efficiency level. Firstly, drawing upon grounded theory about rail infrastructure and terminals, a description is conducted, followed by a comparative analysis of the different types of existing terminals and stations. Secondly, the suggested improvements are presented in accordance with their time frame completion. The main contribution of this study is to illustrate the high significance of terminals, stations and yards, acknowledging them as crucial parts of the railway network, because as characterized and demonstrated in this study, their performance are key factors to the whole network performance, making the identification of their critical points and respective possible solutions, the final objective of this paper. In addition to this, emphasis is given to the need of improving and developing the existing terminal infrastructure and operations.

AbstractThe bovine whey consists of more than 200 different types of proteins, of which β-lactoglobulin, α-lactalbumin, serum albumin, immunoglobulins and lactoferrin predominate. However, their concentrations are not stable due to the existence of protein dynamics during a transition from colostrum secretion to mature milk. To evaluate the dynamics of whey proteins of Jersey cows during a colostral phase and first month of lactation and an influence of the number of lactations, 268 milk samples from 135 Jersey cows were selected through a clinical evaluation. Whey was obtained by rennet coagulation of the mammary secretion. The concentration of total proteins was determined by the biuret method and their fractions were identified by 12% dodecyl sulfate-polyacrylamide gel electrophoresis (12% SDS-PAGE). Maximum concentrations of all protein fractions were observed in the first 12 h of lactation, reducing over the course of the study. Modification of the protein predominance was also observed. The transition from colostrum secretion to milk occurred between 24 and 72 h postpartum. There was an influence of the number of lactations on the dynamics of whey proteins, indicating that multiparous cows had better immunological and nutritional quality when compared to primiparous cows.

Introduction. Whey is a by-product and an excellent source of proteins that is rather aggressive due to a large amount of organic substances it contains. The electro-activation of whey applied in the experiments is a wasteless method that allows the va-lorification of all whey components. β-lactoglobulin (β-Lg) makes up 50% of the whey proteins and 12% of the total protein content in milk. Material and methods. The recovery of β-Lg in protein-mineral concentrates (PMC) by electro-activation processing of different types of whey with different initial protein content was investigated in seven configurations. The recovery of protein fractions in the PMCs were analyzed via electrophoresis with sodium dodecyl sulfate (SDS-PAGE) and 15% non-denaturing polyacrylamide gel (PAAG).      Results. Whey electro-fractionation and the obtaining of PMCs with predetermined protein content, namely of β-Lg, were studied on three whey types, processed at different treatment regimens and in seven configurations. The proper management of electroactivation by varying the treatment regimens will allow the electro-fractionation of different types of dairy by-products. Conclusions. The maximum amount of β-Lg recovered in PMCs on electroactivation is  66-71% depending on the processed whey and on the treatment regimens. Obviously, the extraction of β-Lg from initially lower protein content shows a higher recovery degree of β-Lg. The registered temperatures allows formation of PMCs without thermal denaturation.

Abstract Background Community engagement (CE) and empowerment are required to support the sustainability and effectiveness of actions to reach Agenda 2030. There is a need to guide CE for health and well-being to take action on important societal challenges such as the growing burden of non-communicable diseases (NCDs) and health inequities. The framework proposed in this study has been designed to assist professionals, practitioners and communities to effectively engage. Methods A narrative review of existing grey literature, policy papers and models related to CE was performed. This guided the development of a systematic search strategy, performed by two researchers, which reviewed CE approaches and key influencing factors. The search strategy captured different terms used for CE. Results A total of 27 studies of different types, from around the world, were identified for inclusion into the review. The study compiled a set of widely-used theories and approaches to CE. Key factors such as governance, trust, accessibility and sociocultural contextualisation were also identified as important for the success of CE initiatives. Subsequently, the Comprehensive Community Engagement Framework (CCEF) was developed. It combines theoretical and empirical principles, proven participatory actions and key factors to produce evidence-based health and well-being outcomes across different sectors and levels of society. Conclusions This study has formed the basis of a forthcoming WHO report on CE. The CCEF enables the operationalisation of CE to guide for possible practical approaches to planning, initiating, sustaining and evaluating CE processes alongside the community. It can be used by the health sector as well as the non-health sectors to address health, well-being and broader societal challenges. Key messages The CCEF can be used to engage health and non-health stakeholders to tailor CE processes, increase impact of interventions and policies, building capacity and empowering communities. The proposed framework provides the first comprehensive guidance to conduct community engagement.

Over more than ten centuries, Maya vaults had great technical developments that allowed to achieve constructive and structural advanced systems and increasingly improved forms. One of the key factors to consider when studying its temporal evolution and the different regional variants is the stone vaults shape. This work is based on data collected during fieldwork and comparative analysis of a wide sample of vaults from different geographical zones and chronological periods, and as a result proposes a classification of the Maya stone vaults. The study of the influence of stereotomy on its form and the analysis of the use of different types of stone vaults by zones offer new data to understand the technological evolution of vaulted Maya architecture and the transfers of building knowledge.

The effect of inhibiting protein glycosylation was studied in nongrowing mycelia and protoplasts of Trichoderma reesei which secreted two endoglucanases (I and II) upon addition of sophorose. Tunicamycin (40 μg∙mL−1) inhibited incorporation of N-acetylglucosamine into secreted protein, but had no effect on secretion of total protein or endoglucanases. The secreted endoglucanases I and II exhibited relative molecular masses of 58 and 45 kilodaltons, respectively, irrespective of the presence of tunicamycin. On the other hand 2-deoxy-D-glucose inhibited the biosynthesis of extracellular as well as intracellular protein over a wide range of concentrations; at 50 μg∙mL−1, however, it inhibited the synthesis of extracellular protein more strongly. The synthesis of endoglucanases I and II was decreased accordingly under these conditions. SDS–PAGE did not reveal the secretion of endoglucanases with smaller molecular weights. When the two endoglucanases were purified and subjected to Endo H treatment or β-elimination, the former had no detectable effect, whereas the latter released all carbohydrate from the protein. Nevertheless, endoglucanases I and II contained 1.3 and 0.5 mol of glucosamine per mol enzyme, respectively. It is concluded that endoglucanases I and II from T. reesei contain mainly O-linked neutral carbohydrate, which is required for their secretion.
