Mastitis is highly prevalent infection in cattle causing cost-effective loss in dairy milk production. Escherichia coli is the most frequently isolated bacteria causing mastitis worldwide. The current study was performed to investigate the mastitis prevalence and effect of different antibiotics against pathogens causing it. In sum, 216 milk samples were collected randomly including 108 each from both cows and buffaloes. These samples were subjected to Surf Field Mastitis Test for prevalence of mastitis that found 18.50% and 23.14% in buffaloes and cows respectively. Mastitis was more prevalent both in buffaloes and cows at the age of 9-10 years that was reported in 35% and 32% respectively. The affected buffaloes showed 65% samples containg watery fluid having blood, pus and mucus whereas 36% samples of affected cows had mucus and watery fluid. Pus and blood content were found to be 12% and 16% respectively. Sub-clinical mastitis was prevalent as 56% and 55% in cows and buffaloes respectively while acute mastitis as 4% and 5% in similar fashion. In both groups Right-for part of udder was frequently affected than any other. Among commonly used four antibiotics, Norfloxacine was observed the most effective antibiotic to control mastitis as compared to Gentamycin, Penicillin and ciprofloxacin. The benefit of this study is to diagnose mastitis and use to isolate pathogens for further processing like antibiotic sensitivity and resistance. This study will help veterinarians in better selection of antibiotic to control mastitis.

Nine hundred and fifty two milk samples from clinical bovine mastitis cases, originated from 269 farms in Northern Greece, were tested. Escherichia coli was isolated in 49.3% of samples. Furthermore, Staphylococcus spp, Corynebacterium spp, Streptococcus spp, Pseudomonas spp, Proteus spp, Klebsiella spp or a combination of the above bacteria were isolated. The bacteriological result from 4.3% of samples was negative. No correlation between clinical mastitis cases and seasonal variation was observed. Resistance of E. coli isolates to tetracycline was 82-96%, to enrofloxacin 10-30%, to gentamicin 58-80%, to cephalosporins 47-75%, to sulfamethoxazole-trimethoprime 46-81%, to ampicillin 71-92% and to neomycin 83-97%. The highest resistance rate and the appearance of multi-resistant isolates of E. coli (6%), to 8 antibacterials, were recorded during the last year of the survey.

     A total of 200 samples were collected from 100 goats from more than one regions within the three governorates (Sulaimani, Erbil and Duhok); all these samples were tested by California mastitis test and these showed the following results: 103 samples (51.50%) were negative in all governorates, 28(14.00%) were trace, 35(17.50%) were (+) mark and 34(17.00%) were (++) mark. Clinical mastitis was observed on 93 out of 200 half udder of goats, in the three governorates; 37(18.5%) were acute mastitis, 27(13.5%) were sub acute mastitis and 19(9.5%) were Chronic mastitis. Examination of extracted DNA by Real Time Polymerase Chain Reaction out of 200 DNA extracted from milk samples of goats only 10 (5.00%) were positive for Mycoplasma agalactiae in the three governorates, 8(80%) from mastatic milk samples and 2 (20%) from non mastatic milk samples in result of M. agalactiae. For Mycoplasma capricolum 6 (3.00%) were positive and all were from mastatic milk.

AbstractThe study described in this research communication used phylogenetic genotyping to identify virulence genes and antimicrobial susceptibility in Escherichia coli recovered from cases of bovine mastitis. From 385 mastitic milk samples, 30 (7.8%) isolates were confirmed as E. coli. Most isolates (80%) belonged to phylo-group A. These 30 E. coli isolates were also screened for 11 different virulence genes. The majority of isolates (63%) harbored no virulence gene. Only 11 (37%) isolates tested positive for two virulence genes, either the iron uptake gene iucD in 3 (10%) isolates or the serum resistance gene traT in 2 (7%) isolates or both traT and iucD in 6 (20%) isolates. The E. coli isolates showed highest susceptibility to gentamicin, meropenem, and pipracillin. Most isolates were resistant to ampicillin, cefotaxime and streptomycin. This study suggests that mastitis causing E. coli might originate from commensal bacteria and that the presence of these virulence genes, common in extra-intestinal pathogenic E. coli (ExPEC) strains could be attributed to high genetic variability of mastitis-causing E. coli.

A cross-sectional study was carried out in and around Sebeta town dairy farms, Finfinne special zone, Ethiopia, from December 2019 to May 2020 to isolate, identify, and test antimicrobial susceptibility profile of Staphylococcus aureus from clinical mastitis. A total of 116 milk samples were purposively collected from 57 lactating cows with clinical mastitis. Isolation and identification of Staphylococcus aureus were carried out by using primary and secondary biochemical tests. Besides, Biolog was used for microbial identification systems. To know if the isolates develop resistance to antibiotics, the antimicrobial susceptibility test (ATS) was performed on Mueller-Hinton agar by the disk diffusion method. From a total of 57 lactating cows and 116 teat quarters examined, 21.05% (12/57) and 15.52% (18/116) were positive for S. aureus from clinical mastitis, respectively. From a total of 116 milk samples collected, 15.52% (18/116) Staphylococcus aureus were isolated, and from 11 farms surveyed, about 72.72% (8/11) overall farm prevalence of clinical mastitis due to S. aureus was recorded. All the 18 Staphylococcus aureus isolates were found susceptible to sulphamethoxazole/trimethoprim, erythromycin, gentamicin, ciprofloxacin, and chloramphenicol. However, high level of resistance was observed to common drugs such as penicillin (88.89%, 16/18) and tetracycline (61.11%, 11/18). The observed high level of resistance to penicillin and tetracycline also indicates the need to visit our treatment guidelines for mastitis caused by Staphylococcus aureus. Therefore, improved management and early treatment of the cases with drug of choice after the antimicrobial susceptibility test for each specific case can reduce chance of further development of resistance and are imperative to tackle clinical mastitis occurring at Sebeta and other similar farms in Ethiopia.

Background: Mastitis is one of the most prevalent complex diseases of mammals with high economic impact in dairy industry worldwide. Most of the published inland reports on bovine mastitis are mainly based on sub-clinical mastitis and some extent to clinical types. Objectives: The main objectives of this study are (a) to compare the bacterial pathogens associated with sub-clinical, clinical and chronic mastitis, and (b) to detect the antibiotic sensitivity and resistance status of the isolated bacteria from different types of mastitis. Materials and Methods: A total of 539 quarter milk samples, collected from lactating cows of Rajshahi and Mymensingh districts were subjected to standard bacterial culture and biochemical tests during the period of 2010-2011. Antibiogram test was done on bacteria isolated from sub-clinical (n = 444), acute (n = 35) and chronic (n = 60) mastitis cases include Staphylococcus spp., Streptococcus spp., Bacillus spp. and E. coli in 78.54%, 80% and 71.67% milk samples as a single and 21.46%, 20% and 28.33% as mixed infection, respectively. Results: The Staphylococcus spp. was recorded as major pathogen for all the sub-clinical (42.15%), acute (45.71%) and chronic (41.67%) mastitis cases. The right hind quarters were found significantly (p < 0.05) more affected with sub-clinical, acute and chronic types of mastitis than other three quarters in cows. The highest sensitivity (up to 100%) was recorded with gentamicin, ciprofloxacin, oxytetracycline and enrofloxacin against all the tested four organisms isolated from sub-clinical, clinical and chronic mastitis cases. Antibiotic resistance was highly prevalent, especially streptomycin (70-100%), amoxicillin (30-100%) and ampicillin (0-100%) against the four isolated bacteria of three different types of mastitis. Conclusions: It may be concluded that there is a need to establish a nationwide plan for monitoring the resistance of antibiotics and ensure the cautious use of antibiotics in the veterinary medical practices. ABSTRACT Dairy cattle in two commercial Holstein herds were randomly selected to be vaccinated twice with J5, at approximately 60 days and 28 days before the expected calving date, or to be untreated controls. Based on whether milk production changed following clinical mastitis or whether cows were culled or died within 30 days after onset, 51 mastitis cases were classified as severe or mild. J5-specific antibody responses were evaluated by enzyme-linked immunosorbent assay of all 32 severe and 19 mild cases. The amounts of J5-specific immunoglobulin M (IgM), IgG1, and IgG2 antibodies in sera from the 27 J5 vaccinates were compared with those of the 24 controls. At drying off (before J5 vaccination), all cows had similar amounts of J5-specific antibody. Immediately after calving (approximately 28 days after the second vaccination), J5 vaccinates had significantly higher production of J5-specific IgG1 and IgG2 than controls. When cows were tested following clinical mastitis, none of the three antibody classes differed significantly between the controls and the vaccinates. Vaccinates that contracted Escherichia coli mastitis had 75% less milk loss than controls. The cows that contracted clinical mastitis later in lactation, the unvaccinated controls, and those infected with E. coli had more milk loss following mastitis. The hazards of being culled for all reasons and of being culled for mastitis were significantly lower for J5 vaccinates. Vaccination with J5 was associated with protection against milk production loss and culling following clinical mastitis, and it was also significantly associated with changes in J5-specific IgM, IgG1, and IgG2 antibodies in sera of vaccinated cows.

Mastitis is the most prevalent disease of dairy animals, imparting huge economic losses to the dairy industry. There is always a dire need to monitor the prevalence of mastitis, its bacteriology, and evaluation of antimicrobial susceptibilities for mastitis control and prevention. Therefore, the objectives of this study were to investigate: (i) the prevalence of mastitis in cattle and buffaloes; (ii) identification of bacteria associated with mastitis; (iii) antimicrobial susceptibility of bacterial isolates. Milk samples (n = 1,566) from cattle (n = 1,096) and buffaloes (n = 470) were processed for detection of mastitis using the California mastitis test in the year 2018–19. A total of 633 mastitic milk samples were further processed for bacteriology and antimicrobial susceptibility testing by the disc diffusion method. Overall, the prevalence of clinical and subclinical mastitis was 17 and 57% in both species. Clinical mastitis was higher in cattle (20%) compared to buffaloes (11%), whereas subclinical was higher in buffaloes (66%) than cattle (53%). Besides, month-wise prevalence was higher in hot and humid months in both species. Staphylococci spp. (34%) were the most predominant bacterial isolates from mastitic milk, followed by Escherichia coli (19.4%), Streptococci spp. (9%), and Klebsiella spp. (8%). Most of the bacteria were susceptible to gentamicin (92%) and enrofloxacin (88%), when a panel of 16 different antimicrobials was tested. Nevertheless, most of the isolates were resistant to sulphamethoxazole (99%), lincomycin (98%), oxytetracycline (89%), ampicillin (86%), and doxycycline (85%). This study concludes a high prevalence of mastitis caused by Staphylococcal spp. in cattle and buffaloes belonging to the northwest of Pakistan, and gentamicin and enrofloxacin might be appropriate antimicrobial agents in the treatment of bovine mastitis.

Background and Aim: There is currently no published information on the prevalence and antimicrobial susceptibility patterns of commensal Escherichia coli in dogs of Grenada origin. Monitoring antimicrobial resistance helps in the empirical selection of antibiotics. This study determined the occurrence of E. coli including the O157:H7 serotype in feces of non-diarrheic dogs of Grenada origin and the antibiotic resistance pattern of the E. coli isolates. Materials and Methods: Fecal samples from 142 of the 144 (98.6%) dogs were culture positive for E. coli. Selection of up to three colonies from each of the 142 E. coli-positive samples yielded a total of 402 E. coli isolates, which were analyzed for the presence of non-sorbitol fermenting colonies, and O157-agglutination. Results: Of the 402 E. coli isolates, 30 (7.5%) were non-sorbitol fermenters. However, none of the 402 isolates gave a positive reaction (O157:H7) to the E. coli O157:H7 latex kit. Antimicrobial susceptibility tests against 12 antibiotics revealed low resistance rates to all the tested antibiotics except for tetracycline (Te) (23.4%), cephalothin (CF) (13.2%), and ampicillin (AM) (7.7%). Thirty-nine out of the 402 (9.7%), E. coli isolates were resistant to two or more antibiotics of different classes. Conclusion: This is the first report of isolation and antimicrobial susceptibilities of commensal E. coli from non-diarrheic dogs in Grenada. Some of the isolates (39/402 isolates, 9.7%) were resistant to multiple antibiotics. This study showed that presently, dogs in Grenada should not be considered a reservoir for the E. coli O157:H7 serotype and for multiple antibiotic-resistant E. coli strains. Among the 402 E. coli isolates, the resistance rate to drugs other than Te, CF, and AM was very low.
