Antioxidant capacity of fractions of alcoholic extracts of sugarcane leaves

Extraction of antioxidant compounds present in the top leaves of sugarcane at different stages of plant development showed that the most favorable phase for the extraction is the final stage of maturation of the plant. Flavonoid contents (5.98±0.32mg/mL) and relative antioxidant activity (AArel=257.49±16.94%) similar to the initial stage of maturation were found. Active compounds were chromatographically fractionated over silica gel and elution with solvents of increasing polarity. The antioxidant activity towards -carotene, and the sequestering activity of DPPH (1,1-diphenyl-2-picrylhydrazyl) radicals was determined. The fractions which had the highest total antioxidant capacity (TAC) were obtained from the elution of the solvent mixture: chloroform and ethanol. The results indicate that the top leaves of sugarcane are a possible source of bioactive compounds.

Avocado peel has been considered as a potential source of natural antioxidants in which phenolics are among the most important compounds. Therefore, this study aims to optimize the extraction process of phenolics using response surface methodology and evaluate the corresponding antioxidant activity. From the quadratic model, the optimal condition was determined including the ethanol concentration 54.55% (v/v), the solvent/solute ratio 71.82/1 (mL/g), temperature 53.03 oC and extraction time 99.09 min. The total phenolic content and the total antioxidant capacity at this condition with minor modifications were 26,74 ± 0,04 (mg GAE/g DW) and 188.06 ± 1.41 (mg AAE/g DW), respectively. The significant correlation between total phenolic content and total antioxidant capacity was also confirmed. Circular economy principles are based on the use of by-products from one operation as the raw materials in another. The aim of this work is to obtain extracts with high antioxidant capacity and resveratrol content for the superior capitalization of the biomass of Feteasca regala leaves obtained during vineyard horticultural operations in spring. In order to obtain a high-quality extract at an industrial level, an optimal extraction process is needed. Central composite design (CCD) was used for the experiment design, which contained three independent variables: the ratio of extraction solvent to solid matter, temperature (°C) and time (minutes). The evaluation of extracts was done by measuring the total antioxidant capacity of the extracts using photo-chemiluminescent techniques, and the resveratrol content using liquid chromatography. Process optimization was done using response surface methodology (RSM). Minitab software version 17.0 was used for the design of experiments and data analysis. Regression analysis showed that the model predicts 87.5% of the variation for resveratrol and 96% for total antioxidant capacity (TAC). The temperature had the biggest influence on the extraction yield. The optimal operational conditions for the extraction method applied had the following conditions: ratio e/m 2.92; 43.23 °C and 55.4 min. A maximum value of 34,623 µg ascorbic acid equivalent (AAE) /mL total antioxidant capacity and 182.4 µg/mL resveratrol content were obtained when the optimal extraction parameters where used. The values obtained in experiments proved that by using RSM an accurate model can be obtained for extraction of Feteasca regala leaves.

The current study was designated to explore the antioxidant and antibacterial properties of the methanolic extract of Magnolia champaca stem bark and its different fractions. Antioxidant activity was assessed using total antioxidant capacity, ferric reducing power, DPPH, hydroxyl and hydrogen peroxide scavenging assay. Antibacterial activity was evaluated against five gram positive and five gram negative bacteria using disc diffusion assay method. Among the different fractions, chloroform fraction (CHF) and ethyl acetate fraction (EAF) showed the highest antioxidant activity whereas aqueous fraction (AQF) showed lowest activity in DPPH radical scavenging assay with IC50 of 12.12, 22.41 and 55.16 μg/ml, respectively. Both of the extracts CHF and EAF also exhibited highest total antioxidant capacity, ferric reducing power and hydrogen peroxide scavenging activity with concentration dependent manner when compared to standard BHT. Moderate to potent antibacterial activity was observed against all tested organisms compared to standard azithromycin. The results from the present study revealed that the different fractions of stem bark of M. champaca specially CHF and EAF possess antioxidant and antibacterial property which support its use in traditional medicine and suggesting that the plant may be further investigated to discover its pharmacologically active natural products. Cedrus atlantica Manetti Tar was analyzed by gas chromatography coupled to mass spectrometry, it was subjected to analyzes to know their total polyphenolic and condensed tannins contents, also their Ferric-reducing antioxidant power and Total antioxidant capacity. Chemical characterization identified 88 constituents where Himachalene and α-atlantone isomers (14.51 % - 4.07 %), Calacorene (3.52 %) and ar-Turmerone 3.35 %, were the major components, the total polyphenolic content and condensed tannins contents were 57.15 ± 0.15 milligrams equivalent of gallic acid /g tar and 4.41 ± 0.05 milligrams equivalent of catechin /g tar respectively .This extract showed remarkable Ferric-reducing antioxidant power with effective concentration equal to 50 ± 0.075 mg /mL ± 0,00028 and total antioxidant capacity equal to 262.75 mg equivalents of ascorbic acid /g tar ± 14,43. The experimental results indicated that our tar has promotive antioxidant activity.

One of the richest natural sources of resveratrol - the rhizome of Reynoutria japonica in East Asia is a well-known traditional herb (Hu zhang) used in various inflammatory diseases, infections, skin diseases, scald, hyperlipidemia. Although, it has been recently included in the European Pharmacopoeia, still in Europe is an untapped resource. Some of the therapeutic effects are likely to be influenced by its antioxidant properties and this in turn is frequently associated with a high stilbene content. However, some literatures suggested that other compounds than stilbenes may add to the total antioxidant capacity. Hence, the aim of this research was to examine rhizomes of R. japonica and less studied, morphologically similar species, R. sachalinensis and R. x bohemica for their phytochemical composition and antioxidant activity and to clarify the relationship between the antioxidant activity and compounds by statistical methods. HPLC/UV/ESI-MS studies of three Reynoutria species revealed 171 compounds comprising stilbenes, carbohydrates, procyanidins, flavan-3-ols, anthraquinones, phenylpropanoids, lignin oligomers, hydroxycinnamic acids, naphthalenes and their derivatives. Our studies confirmed the presence of procyanidins with high degree of polymerization, up to decamers in the rhizomes of R. japonica and brings new data on the presence of these compounds in other Reynoutria species. A procyanidin trimer digallate was described for the first time in the studied plants. Moreover, we suggested a presence of new for these species, dianthrone glycosides and previously unrecorded phenylpropanoid disaccharide esters and hydroxycinnamic acid derivatives, mainly in R. sachalinensis. Furthemore, compounds tentatively annotated as lignin oligomers were observed for the first time in studied species. The rhizomes of all Reynoutria species exhibited strong antioxidant activity. Statistical analysis demonstrated that proanthocyanidins should be considered as important contributors to the total antioxidant capacity.

The purpose of this study is to determine the total content of phenols, flavonoids, and condensed tannins, as well as on the antioxidant activity of the extract, and their fractions were measured by 2,2-diphenyl-1-picrylhydrazyl (DPPH), ferric reducing antioxidant power (FRAP), phosphomolybdate reduction (or total antioxidant capacity), and cyclic voltammetry (CV). The hydromethanolic extract of Salvia officinalis showed the highest values of total phenolic (176 mgGAE/g of extract) and condensed tannins (162.53 mgEC/g of extract) from the Boulemane and Khenifra regions, respectively. The results showed that the best DPPH assay was found in the ethyl acetate fraction of Salvia officinalis leaves of the Boulemane region (IC50 = 0.002 mg/ml). For the ethyl acetate and butanolic fractions of Salvia officinalis leaves, those collected from different regions have a better reducing capacity (EC50 = 0.021 mg/ml, respectively). For the total antioxidant capacity, the best activity was found in the aqueous fraction of Salvia officinalis leaves of the Boulemane region (108 mgGAE/g of extract). By the cyclic voltammetry method, hydromethanolic extract of Salvia officinalis leaves from the Boulemane region showed an important result (288.8 mgGAE/g). There was a positive correlation between total phenol content (TPC), condensed tannin content (TCT), and total antioxidant capacity (TAC) (r = 0.932, r = 0.896, respectively). The main compounds that have been identified in the hydromethanolic extract of Salvia officinalis are ascorbic acid, gallic acid, 4-hydroxybenzoic acid, tannic acid, and rutin. Due to their antioxidant property, the leaf extracts from Salvia officinalis are used as natural preservative ingredients in food and/or pharmaceutical industries.

Flemingia macrophylla locally known as ‘Charchara’ in Bangladesh is a medicinal herb. Being a member of the Fabaceae family it claims a number of medicinal uses including hypoglycemic activity, neuroprotective effect and gynecological remedies. Methanolic extract of F. macrophylla was investigated to evaluate its antioxidant and general toxic properties in this study. Antioxidant potential was evaluated using total antioxidant capacity, total phenol contents, total flavonoid contents and DPPH (1,1-diphenyl-2-picrylhydrazyl) assays. Both leaf and stem extracts of F. macrophylla were found to possess significant amount of phenolics and flavonoids, expressed as gallic acid equivalents (GAE) and quercetin equivalent (QE), respectively. Interestingly total phenol content was equal for both leaf and stem extract and it was 43.8 mg GAE/g. Total flavonoid content was found to be 64.4 and 39.1 mg QE/g for leaf and stem extract, respectively. The total antioxidant capacity was expressed as ascorbic acid equivalents (AAE) and for leaf it was 5.067 mg/gm AAE and for stem 0.8167 mg/gm AAE. DPPH scavenging activity was determined by comparing with ascorbic acid. The IC50 values were 19.95, 125.89 and 707.94 ?g/ml for ascorbic acid, leaf and stem extract, respectively. The results of the present study on methanolic extracts of F. macrophylla revealed the presence of moderate antioxidant activity and extract of leaves produced better results than stem of the plant. In brine shrimp lethality bioassay, methanolic extract of F. macrophylla exhibited insignificant cytotoxicity. It has been established that antioxidants in seminal plasma play an important role in protecting the spermatozoa against oxidative stress-induced damage. This study was conducted to measure the total antioxidant capacity (TAC) of boar seminal plasma. Fifty-four ejaculates were collected from 17 mature boars of proven fertility by the gloved-hand technique. Ejaculates were collected separately in different fractions (pre-sperm, sperm-rich, and post-sperm) according to their macroscopic (color) characteristics. After centrifugation (2400g for 3 min), the sperm pellet was discarded; the supernatant was recentrifuged and filtered through a 10-μm nylon mesh filter to remove debris or clumped spermatozoa. The seminal plasma was frozen at -20°C until further use. After thawing at room temperature, seminal plasma aliquots of 5 μL were immediately assessed for total antioxidant capacity. TAC was measured using the ABTS/H2O2/HRP decoloration method (Cano A et al. 2000 Redox Report 5, 365–370) which allows differentiation between hydrophilic and lipophilic antioxidant activity capacity. TAC units were expressed as micromolar (μM) Trolox equivalents. Data were analyzed using ANOVA. Only the hydrophilic activity was measurable, with the lipophilic activity being undetected. The overall TAC of seminal samples (mean ± SEM) was 1623.7 ± 56.28 μM, ranging from 674 to 2428 μM. Different TACs were observed among males (P < 0.05) and between ejaculates of the same male (P < 0.05). Ejaculate fraction had a significant effect (P < 0.001) on the TAC levels. The post-sperm fraction had a significantly lower TAC level (1104.09 ± 57.66 μM) than the pre-sperm and sperm-rich fractions (1611.95 ± 153.68 μM and 1356.136 ± 72.47 μM, respectively, P < 0.001). In conclusion, hydrophilic antioxidant activity represented the main contribution to the TAC in boar seminal plasma, showing differences among males, between ejaculates of the same male, and also between the different ejaculate fractions. One hundred and forty-seven sorghum samples, grown in 2015 and 2017, were used to build different near infrared spectroscopic calibration models able to predict total antioxidant capacity, total phenolics, total flavonoids and condensed tannins content. Samples were separated into calibration and validation sets using a nearest neighbours algorithm. The r2pred values ranged from 0.84 (condensed tannins) to 0.95 (total phenolics), whereas the Ratio of Performance to Deviation (RPD) values ranged from 1.9 (total flavonoids) to 3.0 (total phenolics). Comparison of prediction error estimates highlighted the best models with significant differences. Model robustness was tested through a reduction of sample numbers in the calibration set; the highest robustness was found for total antioxidant capacity and total flavonoids. In addition, a partial least squares discriminant analysis model to screen the samples for their tannins level was developed and resulted in good performance; it should be useful to select tannin-free genotypes for the food industry. These models could be used for rapid screening of sorghum breeding genotypes with high antioxidant compounds.

Sargassum sp. adalah salah satu jenis rumput laut paling banyak di Indonesia dan memiliki manfaat sebagai antioksidan, obat penyakit jantung, stroke, dll. Penelitian ini bertujuan untuk menentukan kandungan antioksidan fraksi etil asetat dari ekstrak metanol Sargassum sp. Sampel diambil dari Pantai Sundak, Gunung Kidul, Yogyakarta. Metode yang digunakan adalah eksploratif diskriptif. Sampel dikeringkan dalam suhu ruangan selama 7 hari dan dimaserasi dengan pelarut metanol, lalu dievaporasi dengan rotary evaporator. Ekstrak metanol di fraksinasi menggunakan etil asetat. Pengujian aktivitas antioksidan dilakukan dengan menggunakan 2 metode, yaitu metode penangkapan radikal bebas DPPH dan total kapasitas antioksidan fosfomolibdat. Vitamin C digunakan digunakan sebagai standar penangkapan radikal bebas DPPH dan total kapasitas antioksidan fosfomolibdat. Kadar total fenolat diuji menggunakan larutan Folin-Ciocalteu dengan standar asam galat. Hasil penelitian menunjukkan bahwa kadar total fenolat fraksi etil asetat Sargassum sp. sebesar 64,42 mg GAE/g sampel. Aktivitas antioksidan dengan metode penangkapan radikal bebas DPPH memiliki nilai IC50 1.289 ppm, vitamin C memiliki IC50 sebesar 122,71 ppm, sedangkan total kapasitas antioksidan adalah 39,52 mg AAE/g sampel. Kesimpulannya, yaitu kandungan aktivitas antioksidan fraksi etil asetat dari ekstrak metanol Sargassum sp. dengan metode penangkapan radikal bebas DPPH diduga sangat lemah dan kandungan aktivitas antioksidan dengan menggunakan metode fosfomolibdat diduga tinggi.  Sargassum sp. is one of the many types of seaweed in Indonesia and has benefits as an antioxidant, a drug for heart disease, stroke, etc. This study aims to determine the antioxidant content of ethyl acetate fraction of methanol extract of Sargassum sp. Samples were taken from Sundak, Gunung Kidul, Yogyakarta. The method used is descriptive explorative. Samples were dried at room temperature for 7 days and macerated with methanol, then evaporated at a rotary evaporator. The methanol extract fractionation using ethyl acetate. Testing the antioxidant activity using two methods, the method of catching free radicals DPPH and total antioxidant capacity fosfomolibdat. Vitamin C is used as a standard arrest DPPH free radical and total antioxidant capacity fosfomolibdat. Levels of total phenolics were tested using the Folin-Ciocalteu solution with gallic acid standard. The results showed that levels of total phenolic fraction of ethyl acetate Sargassum sp. amounting to 64,42 mg GAE/g sample. The antioxidant activity with catching free radicals DPPH methods have IC50 values 1.289 ppm, vitamin C has an IC50 of 122,71 ppm, while the total antioxidant capacity was 39,52 mg AAE/g sample. In conclusion, the content of the antioxidant activity of ethyl acetate fraction of methanol extract of Sargassum sp. with catching free radicals DPPH methods allegedly extremely weak and the content of the antioxidant activity using methods fosfomolibdat predictably high.
