BACKGROUND: Fig leaves are reported to have an effect on reducing blood glucose levels. However, the use of fresh leaves makes the effects obtained is not measurable and efficient. AIM: The purpose of this research was to determine the antidiabetic potential of ethanol extract of fig leaves and to optimize tablet dosage formulations. METHODS: Four tablet formulas were made using the wet granulation method. Formula I (FI), Formula I (FII), and Formula III (FIII) groups to give a tablet of ethanol extract of fig leaves with a dose of 40 mg, 60 mg, and 80 mg and placebo treatment group. There were eight groups of male rats strain Wistar treated as follows: Normal control, negative control, positive control (metformin tablets), basis group, placebo treatment group, F1, F2, and F3 groups. The antidiabetic activity was evaluated from a decrease in rat blood glucose levels. Previously, rats were induced first using alloxan 150 mg/kg intraperitoneally to damage β-pancreatic cells so that the rats could experience increased blood glucose levels. After giving treatment to each group for 14 days, a rat blood sample was then taken on days 9 and 14, which were then analyzed by the GOD PAP method with readings carried out using a spectrophotometer with a wavelength of 500 nm. RESULTS: The average weight (mg) of FI tablets (617.8 ± 3.21%), FII (629.35 ± 8.16%), and FIII (643.6 ± 6.21%), and placebo tablet (666.45 ± 4.36%). As for the uniformity of size, all formulas have a diameter of 0.9 ± 0.0 (cm). For the hardness values of FI (5.7 kg), FII (1.31 kg), and FIII (3.09 kg), placebo tablet (2.98%). The value of friability FI (1.42%), FII (11.8%), and FIII (0.84%), placebo tablet (1.16%). While the disintegration time of FI (11.02 min), FII (10.10 min), and FIII (17.00 min), placebo tablet (12.23%). As for the uniformity of size, all formulas have a diameter of 0.9 ± 0.0 (cm). Whereas the dissolution rate (DE45) of each formulation decreased with increasing dose of extract, FI (73.73%), FII (74.80%), and FIII (69.80%). The treatment group of the ethanol extract of fig leaves at a dose of 40 mg, ethanol extract of fig leaves at a dose of 60 mg, and ethanol extract of fig leaves at a dose of 80 mg could reduce rats’ blood glucose levels with statistically significant results (p < 0.05) compared to negative group. When it was compared to the positive group, it had significant results with a statistical value (p < 0.05). CONCLUSION: Ethanol extract of fig leaf tablets had a significant effect on lowering rats’ blood glucose levels.

Maintaining a lifestyle can reduce the incidence of DM (diabetes mellitus). DM occurs due to insulin disorders so that blood glucose levels increase, which can lead to various complications. The management of blood glucose levels by activating the insulin function can be done by using natural ingredients such as the Yakon (Smallanthus sonchifolius) plant. Yakon leaves contain phenol which can reduce blood glucose. The design of this study was experimental with a pre-posttest approach with control-group design, using male and healthy white rats (Rattus norvegicus). Rats were divided into 3 groups, treatment dose 1, treatment dose 2 and control. The rats were given a high carbohydrate diet during 9 weeks to make the rats hyperglycemic. In the treatment group, dose 1 was 150 mg/kg BW, dose 2 was 300 mg kg BW, and was given for 3 days. The results showed that the rats in the treatment group dose 1 had decreased in the average blood sugar level of 114.10 mg / dl (p 0.002) and dose 2 was 105.27 mg / dl (p 0.005). This showed that there was an effect of treatments on blood sugar levels. The comparison results showed that there was a significant difference between the dose 1, the dose 2 group and the control (Sig. (2-tailed) = 0.001 (α) = 0.05). There was no significant difference in the treatment group dose 1 and treatment dose 2 (Sig. (2-tailed) = 0.693, (α) = 0.05). Yakon leaves can be used alternative to lower to control blood glucose levels in rats receiving a high-carbohydrate diet

Gynura divaricata (GD) also known as Dewa leaf is one of the Indonesian medicinal plants which are also native to India, Thailand and China. GD is widely used as antidiabetic, antihypertensive and other diseases including several tumors. However, research on the effects of GD as antidiabetic is still not widely practiced, especially in Indonesia. The aim of this research is to investigate the effect of GD ethanolic extract on blood glucose levels of alloxan-induced diabetes mice. This research is an experimental research with pre- and post-test design using 24 male DDY strain mice. Animals experiment is divided into four groups, that are one normal group and three alloxan-induced diabetes mice group that give 1% CMC (control), 400[Formula: see text]mg/kg GD ethanolic extract and 0.65[Formula: see text]mg/kg glibenclamide, respectively. Treatment was given for 14 days. Blood glucose levels were measured before and after inducing alloxan, and after 14 days treatment. At the end of study, all of animals experiment were sacrificed for histological examination. Phytochemical analysis revealed that polyphenols, flavonoids, steroids, triterpenoids, terpenoids and and quinones were present in ethanol extract of GD. The results of this study showed significant ([Formula: see text]) decrease of blood glucose levels after GD ethanolic extract and glibenclamide treatment. The results of histological examination showed improvement in pancreas damage and significant increase in the number of beta cells in GD and glibenclamide groups. The results indicate that dewa leaf ethanolic extract has antidiabetic activity and the effectiveness is the same as glibenclamide. GD also could repair damage of pancreas caused by alloxan induction.

Abstract BackgroundOptimization of experimental conditions in streptozotocin induced diabetic model in Sprague Dawley (SD) rats to evaluate the stability of the model.MethodsMale and female SD rats were randomly divided into control group, STZ 45 group (STZ: 45 mg / kg), STZ 65 group (STZ: 65 mg / kg), STZ 85 group (STZ: 85 mg / kg), high fat diet with STZ 45 group (STZ: 45 mg / kg), high fat diet with STZ 65 group (STZ: 65 mg / kg), high fat diet with STZ 85 group (STZ: 85 mg / kg). N = 6 in each group. The changes of body weight and blood glucose were observed dynamically.ResultsThere was no significant difference in blood glucose or body weight between the STZ 45 group and the control group in both male and female rats, whether or not they were on a high-fat diet. However, there were significant differences in blood glucose between the high-dose STZ group and the control group in both male and female rats, regardless of whether the rats were on a high-fat diet or not (P < 0.05 or P < 0.01). Compared with the control group, there were significant differences in blood glucose levels (P < 0.05 or P < 0.01) and higher blood glucose levels in the male rats fed with the normal diet than that in those fed with the high-fat diet.ConclusionsIn this study, male rats fed with ordinary feed and injected STZ dose of 65 mg / kg were the most stable and ideal diabetic rat.
