Black pepper ( Piper nigrum L.) is a popular spice that is grown as tropical and subtropical plant throughout the world. The leaf, flower, fruit, and root are the most important elements of the plant. Asexual or vegetative propagation is becoming highly popular, although the sexual approach is still used for pepper vine cultivation. For mass production of the pepper plant, in vitro culture is also used. The bioactive components contained in them are extremely important because of their therapeutic potential against a number of diseases. They are usually classed as functional foods because, in addition to providing basic nutrition, provide physiological benefits and help to avoid chronic illness. The main component of black pepper is piperine. It has a complex phyto-chemistry includes: Volatile oil, alkaloids, and oleoresins. Because of its free-radical scavenging properties, black pepper and its active components can be prevention and control of tumor growth. Piperine, which can bind and inhibit the SARS-CoV-2 virus that causes the sickness, is present in black pepper and has antibacterial and antiviral effects. Piperine, a key alkaloid component of black pepper, it also helps to cognitive brain function, nutritional absorption, and gastrointestinal health. Black pepper is known as the “King of Spices” as well as the “King of Medicinal Agents,” since it includes a wide variety of bioactive compounds with nutraceutical and pharmacological applications. An overview of the most common applications for black pepper, along with a strong evidence is present in this review.

Erica australisL. (Ericaceae) is used in traditional medicine to treat many free-radical related ailments. In the present work, the stability and biological activity of the plant aqueous extracts submitted to anin vitrodigestive process were investigated. Chemical stability was monitored by HPLC-DAD and LC-MS/MS, while the bioactivities were evaluated through the inhibition of acetylcholinesterase (AChE) and DPPH radical scavenging activity. Both extracts, whose main components were flavonol glycosides, inhibited AChE, showing IC50values of 257.9 ± 6.2 µg/mL and 296.8 ± 8.8 µg/mL for the decoction and for the infusion, respectively. Significant radical scavenging activities were also revealed by both extracts, as denoted by the IC50values for the decoction, 6.7 ± 0.1 µg/mL, and for the infusion, 10.5 ± 0.3 µg/mL. After submission to gastric and pancreatic juices, no remarkable alterations in the composition or in the bioactivities were observed, suggesting that the extracts may pass through the gastrointestinal tract, keeping their composition and therefore their biological properties. Moreover, the bioavailability of the components of both extracts, as studied in a Caco-2 cell model, showed that compounds can permeate the membrane, which is a condition to exert their biological activities. Our results add further support to the potential ofE. australisfor its antioxidant and neuroprotective properties.

Fusarium solani f. sp. piperis (teleomorph: Nectria haematococca f. sp. piperis), causal agent of root rot and stem blight on black pepper (Piper nigrum), produces secondary metabolites with toxigenic properties, capable of inducing vein discoloration in detached leaves and wilting in transpiring microcuttings. Production of F. solani f. sp. piperis (Fsp) toxic metabolites reached a peak after 25 days of static incubation on potato sucrose broth at 25 ºC under illumination. Changes in the pH of the culture filtrate did not alter the effect of toxic metabolites. However, when the pH was changed before the medium had been autoclaved, a more intense biological response was observed, with an optimum at pH 6.0. Isolates that produced red pigments in liquid cultures were more efficient in producing biologically active culture filtrates than those which produced pink coloured or clear filtrates suggesting that these pigments could be related to toxigenic activity. Detached leaves of seven black pepper cultivars and Piper betle showed symptoms of vein discoloration after immersion in autoclaved and non-autoclaved Fsp culture filtrates indicating the thermostable nature of these toxic metabolites.

Piperine and piperidine are the two major alkaloids extracted from black pepper (Piper nigrum); piperidine is a heterocyclic moiety that has the molecular formula (CH2)5NH. Over the years, many therapeutic properties including anticancer potential of these two compounds have been observed. Piperine has therapeutic potential against cancers such as breast cancer, ovarian cancer, gastric cancer, gliomal cancer, lung cancer, oral squamous, chronic pancreatitis, prostate cancer, rectal cancer, cervical cancer, and leukemia. Whereas, piperidine acts as a potential clinical agent against cancers, such as breast cancer, prostate cancer, colon cancer, lung cancer, and ovarian cancer, when treated alone or in combination with some novel drugs. Several crucial signalling pathways essential for the establishment of cancers such as STAT-3, NF-κB, PI3k/Aκt, JNK/p38-MAPK, TGF-ß/SMAD, Smac/DIABLO, p-IκB etc., are regulated by these two phytochemicals. Both of these phytochemicals lead to inhibition of cell migration and help in cell cycle arrest to inhibit survivability of cancer cells. The current review highlights the pharmaceutical relevance of both piperine and piperidine against different types of cancers.

The cause of stunting disease of black pepper (Piper nigrum) in Indonesia has not yet been confirmed cither due o a virus or mycoplaam. However, similar disease found on black pepper plants in the Southeast Asia is caused by Piper yellow mottle virus (PYMV) which is transmitted by Planococcus citri. This expeiments was aimed o examine the cause of the stunting disease and its insect vectors. The expeiment were conducted from October 1998 to May 2001. Diseased vegetative materials of black pepper plants showing stunting disease were collected from Sukamulya, Sukabumi. The potential insect vectors were collected from black pepper plants in Bogor, IP Sukamulya (Sukabumi), Lampung and Bangka. The insects were fed on the diseased plants obtained from Lampung, Bogor and IP Sukamulya, then transferred o the healthy plants. The healthy plants were produced rom true seeds and cutings oiginated rom Bogor and IP Sukamulya. The tested plants were incubated at the green house and examined for disease development. Diseased leaf samples collected rom black pepper plant rom IP. Sukamulya and the transmitted plants were sent to the University of Minnesota, USA for Ihe virus (PYMV) by using ISEM (immunosorbent electron microscope). The result snowed that the leaves samples rom IP. Sukamulya were infected by PYMV. The morphology and size of the virus were similar to those caused stunting disease in the South East Asia. Potential insects vectors found on the diseased black <br /><br />pepper plants were two mealybugs, P. minor and Ferrisia virgata (Hemiptera; Coccoidca: PaaidbcoccidaeX aa well as an aphid Toxoptera aurantii (Hemiptera: Aphidoidea: Aphididae). The first tial on disease transmission by using P. minor, previously reared on the potato tubers, showed one out of ten tested plants produced disease sympom. The subsequent trial using P minor, bred on healthy black pepper seedlings, showed thee out often tested plants developed disease symptoms. None of the aphid transmitted plants developed Ihe disease. This study confirmed thai PYMV b the cause of stunting disease on black pepper in IP Sukamulya and Lampung and P. minor as the insect vecor of the disease.<br /><br />

This review aimed to provide a summary on the traditional uses, phytochemistry, and pharmacological activities in the cardiovascular system and cardiotoxicity of Melissa officinalis (MO), with the special emphasis on the protective mechanisms in different cardiovascular pathologies. MO is a perennial aromatic herb commonly known as lemon balm, honey balm, or bee balm, which belongs to Lamiaceae family. Active components are mainly located in the leaves or essential oil and include volatile compounds, terpenoid (monoterpenes, sesquiterpenes, triterpenes), and polyphenolic compounds [rosmarinic acid (RA), caffeic acid, protocatechuic acid, quercitrin, rhamnocitrin, luteolin]. For centuries, MO has been traditionally used as a remedy for memory, cognition, anxiety, depression, and heart palpitations. Up until now, several beneficial cardiovascular effects of MO, in the form of extracts (aqueous, alcoholic, and hydroalcoholic), essential oil, and isolated compounds, have been confirmed in preclinical animal studies, such as antiarrhythmogenic, negative chronotropic and dromotropic, hypotensive, vasorelaxant, and infarct size–reducing effects. Nonetheless, MO effects on heart palpitations are the only ones confirmed in human subjects. The main mechanisms proposed for the cardiovascular effects of this plant are antioxidant free radical–scavenging properties of MO polyphenols, amelioration of oxidative stress, anti-inflammatory effects, activation of M2 and antagonism of β1 receptors in the heart, blockage of voltage-dependent Ca2+ channels, stimulation of endothelial nitric oxide synthesis, prevention of fibrotic changes, etc. Additionally, the main active ingredient of MO-RA, per se, has shown substantial cardiovascular effects. Because of the vastness of encouraging data from animal studies, this plant, as well as the main ingredient RA, should be considered and investigated further as a tool for cardioprotection and adjuvant therapy in patients suffering from cardiovascular diseases.

Aronia fruits contain many valuable components that are beneficial to human health. However, fruits are characterized by significant variations in chemical composition dependent on the growing conditions and harvesting period. Therefore, there is a need to formulate the extracts with a precisely defined content of health-promoting substances. Aronia dry extracts (ADE) were prepared from frozen pomace applying water extraction, followed by purification and spray-drying. Subsequently, the content of anthocyanins, phenolic acids, and polyphenols was determined. The high-quality chokeberry pomace enabled obtaining extracts with anthocyanin content much higher than the typical market standards. Moreover, it was found that the antioxidant capacity of aronia extracts exceeded those found in other fruit preparations. Antioxidant and free-radical scavenging properties were evaluated using a 2,2′-diphenyl-1-picrylhydrazyl using Electron Paramagnetic Resonance (EPR) spectroscopy (DPPH-EPR) test and Oxygen Radical Absorbance Capacity (ORAC) assay. The inhibition of lipid peroxidation and the level of inflammatory markers have been also investigated using lipopolysaccharide (LPS)-stimulated RAW 264 cells. It was revealed that ADE standardized to 25% of anthocyanins depresses the level of markers of inflammation and lipid peroxidation (Interleukin 1 beta (IL-1β), tumor necrosis factor alpha (TNF-α), and malondialdehyde (MDA)) in in vitro conditions. Additionally, it was confirmed that ADE at all analyzed concentrations did not show any cytotoxic effect as demonstrated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay.

We have determined the protective effects of Thymus serpyllum (TS) extract and nanoparticle-loaded TS on hydrogen peroxide-induced cell death of mesenchymal stromal cells (MSCs) in vitro. Gas chromatography–mass spectroscopy confirmed the spectrum of active components in the extract. Out of the three different extracts, the hexane extract showed significant free radical scavenging activity. Treatment of MSCs with H2O2 (hydrogen peroxide) significantly increased intracellular cell death; however, pretreatment with TS extract and nanoparticle-loaded TS (200 μg/ml) suppressed H2O2-induced elevation of Cyt-c and MMP13 and increased the survival rates of MSCs. H2O2-induced (0.1 mM) changes in cytokines were attenuated in the extract and nanoparticles by pretreatment and cotreatment at two time points (p<0.05). H2O2 increased cell apoptosis. In contrast, treatment with nanoparticle-loaded TS suppressed the percentage of apoptosis considerably (p<0.05). Therefore, TS may be considered as a potential candidate for enhancing the effectiveness of MSC transplantation in cell therapy.

Coleonema album, a member of the South African ‘Fynbos’ biome, was evaluated for its antioxidant and free radical scavenging activity. Ethanol- and acetone-based extracts from plant material obtained from two different geographical areas were analysed. A bioassay-guided fractionation methodology was followed for screening of active compounds. The 1,1-diphenyl-2-picrylhydrazyl (DPPH)-TLC method revealed the presence of a number of antioxidants which were quantified by the DPPH-spectrophotometric assay and the oxygen radical absorbance capacity (ORAC) assay. The C. album extracts possessed significant in vitro antioxidant activity, a large portion of which appeared to be contributed by the phenolic compounds. In contrast, the reducing power of the extracts could not be correlated with the observed antioxidant activity. Identification and structural information of the active components were obtained by a combination of preparative TLC and LC-MS which revealed the presence of coumarin aglycones and glycosides. The results of this study indicate that C. album contains strong antioxidants that warrant further investigation into the relationship between the structure and activity of the active coumarin metabolites.
