The study was conducted with the primary objective to establish the influence of melatonin on adrenocortical functions to ameliorate thermal stress in goats. Endocrine secretions and several other blood biochemical parameters reflecting the animals adrenocortical stress response were determined over a one-week period after goats had been exposed to40C∘and 60%RH for 10 days. The study was conducted for a period of 17 days in psychrometric chamber. The animals served as self-controls prior to start of the experiment. Blood samples were drawn on day 10 to establish effect of thermal stress. Chemical adrenalectomy was achieved using metyrapone followed by exogenous melatonin treatment.40C∘of thermal stress which is quite normal in tropical zone significantly(P≤.05)influenced all parameters except plasma insulin. Metyrapone treatment significantly(P≤.05)affected plasma levels of glucose, total protein, total cholesterol, cortisol, and aldosterone. Metyrapone aggravated thermal stress by decreasing cortisol level in goats. Melatonin treatment at 11:00 AM significantly(P≤.05)influenced plasma levels of glucose, total protein, total cholesterol, cortisol, aldosterone and insulin. Metyrapone treatment aggravated thermal stress although administration of melatonin could ameliorate the condition. This establishes the role of melatonin in relieving thermal stress in goats.

Scandium (Sc) is an element with many industrial applications, but relatively little is known about its physiological and/or toxicological effects, and very little data are available concerning the role of Sc in chronic renal failure (CRF). This paper examines the changes in plasma levels of Sc in predialysis patients with CRF and the relationship with blood parameters. The participants in this trial were 48 patients with CRF in predialysis and 53 healthy controls. Erythrocyte, haemoglobin, and haematocrit counts in blood were determined, and levels of creatinine, urea, uric acid, albumin, total protein and Sc were measured in plasma. The glomerular filtration rate (GFR) was calculated using the Cockcroft-Gault index. The CRF patients were found to have higher plasma levels of creatinine, urea, uric acid, albumin, total protein, and Sc and a lower GFR than that the controls. Scandium in plasma was positively correlated with creatinine and plasma urea and negatively correlated with GFR, haemoglobin, and haematocrit and was associated with the risk of lower levels of erythrocytes, haemoglobin, and haematocrit. CRF was associated with increases in the circulating levels of scandium.

PURPOSE: To compare some major hematological and biochemical blood parameters of 3 newly introduced poultry breeds – Australorp, Marans and Araucana, during different stages of their egg-laying period. METHODS: Five blood parameters were investigated: hematocrit, hemoglobin, glucose, total protein and total cholesterol. A hundred blood samples were tested, collected at three laying stages – beginning, middle and end. RESULTS: Statistically significant differences in the hemoglobin levels by sexes were established: in Marans breed (males – between 8.38 and 13.45; females – between 7.30 and 9.02 g*dl-1) and Araucana (males – between 9.17 and 13.65; females – between 9.08 and 9.40 g*dl-1). The hematocrit levels in the males (from 23.16% in Marans to 40.88% in Australorp) were significantly higher than the levels in the females (from 23.72% in Marans to 30.48% in Australorp). The blood glucose test of the cocks (from 32.07 in Marans to 40.88 mmol*l-1 in Australorp) showed higher levels compared to the female (from 23.72 in Marans to 30.48 mmol*l-1 in Australorp). The total protein content in the layers’ blood (between 5.97 in Araucana and 8.73 g*dl-1 in Marans) was higher compared to cocks. The total cholesterol levels were, as follows: males – from 2.05 in Araucana to 3.49 mmol*l-1 in Australorp; females – from 1.92 in Australorp to 6.57 3.49 mmol*l-1 in Araucana. CONCLUSIONS: The variations in the investigated parameters in the blood are a result of the simultaneous effect of several factors - breed genetic differences, differences in egg-laying effort and degree of adaptation.

Background: The industrial hub of Punjab, Ludhiana is polluted with heavy metals. The dairy animals experience stress due to pollution of heavy metals as well as the hot summer season. The stressors lead to disturbance in metabolic profile and affect milk production. The present study aimed at evaluation of detrimental effect of environmental heavy metal pollution on the metabolic profile of buffaloes during summer and winter seasons in Ludhiana district of Punjab, India. A total of 100 buffaloes were randomly selected from heavy metal exposed (n=60) and control area (n=40) during summer and winter seasons. Methods: The location of the experimental area surveyed on the basis of levels of heavy metals. The dairy farms are located in and around Ludhiana, Punjab were identified with levels of heavy metal above the permissible limits (FSSAI, 2010) viz. Chromium-0.05 µg /ml; Nickel-0.02 µg /ml; Arsenic-0.05 µg /ml and Lead -0.05µg /ml. The plasma samples were analyzed for metabolic profile. The water levels of heavy metals were below the permissible limits; therefore, it was taken as uncontaminated or control area. Result: Buffaloes of heavy metal exposed areas exhibited significantly (P less than 0.05) higher levels of glucose, total cholesterol, triglycerides, BUN, creatinine, AST, ALT, GGT, ALP and CK levels as compared to control area. A significant (P less than 0.05) decrease was observed in plasma total protein, albumin, A:G ratio, BUN: creatinine ratio in the buffaloes of exposed area as compared to the control area. However, significantly (P less than 0.05) higher levels of plasma glucose, BUN, creatinine, AST, ALT, GGT, ALP and CK levels were observed during summer as compared to winter season in both exposed and control areas. Significantly (P less than 0.05) decreased levels of plasma total cholesterol, triglycerides, total protein, albumin and BUN: Creatinine ratio were observed in the buffaloes during summer as compared to winter in both exposed and control areas. The Cr, Ni, As and Pb levels showed highly significant (P less than 0.01) positive correlation with plasma levels of glucose, total cholesterol, creatinine, ALT, AST, CK and GGT. Highly significant positive relationship (P less than 0.01) was observed among plasma levels of glucose, total cholesterol, creatinine, BUN, ALT, AST, ALP, CK and GGT whereas, plasma BUN had highly significant (P less than 0.01) negative correlation with plasma total protein and A:G ratio considering both the areas together. Plasma levels of total protein showed significant (P less than 0.01) negative correlation with all other metabolic profile constituents of both control and heavy metal exposed areas. Thus it may be concluded that the metabolic disturbance in heavy metal exposed buffaloes may have been mediated by heavy metal pollution.

SummaryInfusion of desamino-d-arginine vasopressin (DDAVP) results in an increase in plasma plasminogen activator activity. Whether this increase results in the generation of plasmin in vivo has never been established.A novel sensitive radioimmunoassay (RIA) for the measurement of the complex between plasmin and its main inhibitor α2 antiplasmin (PAP complex) was developed using monoclonal antibodies preferentially reacting with complexed and inactivated α2-antiplasmin and monoclonal antibodies against plasmin. The assay was validated in healthy volunteers and in patients with an activated fibrinolytic system.Infusion of DDAVP in a randomized placebo controlled crossover study resulted in all volunteers in a 6.6-fold increase in PAP complex, which was maximal between 15 and 30 min after the start of the infusion. Hereafter, plasma levels of PAP complex decreased with an apparent half-life of disappearance of about 120 min. Infusion of DDAVP did not induce generation of thrombin, as measured by plasma levels of prothrombin fragment F1+2 and thrombin-antithrombin III (TAT) complex.We conclude that the increase in plasminogen activator activity upon the infusion of DDAVP results in the in vivo generation of plasmin, in the absence of coagulation activation. Studying the DDAVP induced increase in PAP complex of patients with thromboembolic disease and a defective plasminogen activator response upon DDAVP may provide more insight into the role of the fibrinolytic system in the pathogenesis of thrombosis.

Abstract. Basal plasma levels for adrenalin (A), noradrenalin (NA), l-triiodothyronine (T3), and l-thyroxine (T4) were determined in rats with a chronically inserted catheter. The experiments described in this report were started 3 days after the surgical procedure when T3 and T4 levels had returned to normal. Basal levels for the catecholamines were reached already 4 h after the operation. The T3/T4 ratio in plasma was significantly increased after 3, 7, and 14 days in rats kept at 4°C and the same holds for the iodide in the 24-h urine after 7 and 14 days at 4°C. The venous NA plasma concentration was increased 6- to 12-fold during the same period of exposure to cold, whereas the A concentration remained at the basal level. During infusion of NA at 23°C the T3/T4 ratio in plasma was significantly increased after 7 days compared to pair-fed controls, and the same holds for the iodide excretion in the 24-h urine. This paper presents further evidence for a role of the sympathetic nervous system on T4 metabolism in rats at resting conditions.

During the last few years, a significant number of studies have attempted to clarify the underlying mechanisms that lead to the presentation of atrial fibrillation (AF). Inflammation is a key component of the pathophysiological processes that lead to the development of AF; the amplification of inflammatory pathways triggers AF, and, in tandem, AF increases the inflammatory state. Indeed, the plasma levels of several inflammatory biomarkers are elevated in patients with AF. In addition, the levels of specific inflammatory biomarkers may provide information regarding to the AF duration. Several small studies have assessed the role of anti-inflammatory treatment in atrial fibrillation but the results have been contradictory. Large-scale studies are needed to evaluate the role of inflammation in AF and whether anti-inflammatory medications should be routinely administered to patients with AF.

This study was initiated on 24 prepubertal Holstein x Kankrej crossbred heifers of nearly identical age (7-9 months) and body weight (130-140 kg) at University farm to evaluate the effect of high plane of nutrition on blood biochemical and minerals profile and the age at puberty. Twelve heifers were managed under routine farm feeding (control) and the rest 12 under ideal optimum feeding regime (treatment) that included extra 1 kg concentrate, 30 g min mix and ad-lib dry fodder. The body weight and ovarian ultrasonography together with blood sampling was carried out at monthly interval from 10 to 18 months of age to study the ovarian dynamics and blood biochemical changes. High plane of nutrition to growing heifers was beneficial in reducing the age of onset of puberty (by 2-3 months) compared to routine farm fed group. The mean plasma total protein and cholesterol concentrations showed a rising trend with significant variations from 10 to 16 months of age, where it got mostly stabilized indicating adult profile. The activity of enzymes GOT and GPT also rose gradually and significantly from 10 months till 14-15 months of age, and thereafter it remained more or less static till 18 months of age. The levels of both these enzymes were higher, with lower protein and cholesterol, in control than the treatment group from 15-16 months of age onwards. The mean plasma levels of both calcium and phosphorus increased gradually and significantly with advancing age till 16-17 months of age, with little higher values in supplemented than a control group. The plasma levels of zinc, iron, copper, and cobalt also showed rising trend with significant differences between 10th and 12th-14th months of age, and from 15th to 18th months of age the levels were statistically the same in all the groups with slightly higher values in the treatment group.

Introduction: Patients’ decisions to undergo major surgery such as orthognathic treatment are not just about how the decision is made but what influences the decision. Objectives: The primary objective of the study was to identify the key processes involved in patients’ experience of decision making for orthognathic treatment. Methods: This study reports some of the findings of a larger grounded theory study. Data were collected through face-to-face interviews of patients who were seen for orthognathic treatment at a teaching hospital in the United Kingdom. Twenty-two participants were recruited (age range 18–66 y), of whom 12 (male = 2, female = 10) were 6 to 8 wk postsurgery, 6 (male = 2, female = 4) were in the decision-making stage, and 4 (male = 0, female = 4) were 1 to 2 y postsurgery. Additional data were also collected from online blogs and forums on jaw surgery. The data analysis stages of grounded theory methodology were undertaken, including open and selective coding. Results: The study identified the central role of dental care professionals (DCPs) in several underlying processes associated with decision making, including legitimating, mediating, scheduling, projecting, and supporting patients’ decisions. Six categories were related to key aspects of decision making. These were awareness about their underlying dentofacial problems and treatment options available, the information available about the treatment, the temporality of when surgery would be undertaken, the motivations and expectation of patients, social support, and fear of the surgery, hospitalization, and potentially disliking their new face. Conclusion: The decision-making process for orthognathic treatment is complex, multifactorial, and heavily influenced by the role of DCPs in patient care. Understanding the magnitude of this role will enable DCPs to more clearly participate in improving patients’ decision-making process. The findings of this study can inform future quantitative studies. Knowledge Transfer Statement: The results of this study can be used both for informing clinical practice around enabling decision making for orthognathic treatment and also for designing future research. The findings can better inform clinicians about the importance of their role in the patients’ decision-making process for orthognathic treatment and the means to improve the patient experience. It is suggested that further research could be conducted to measure some of the key constructs identified within our grounded theory and assess how these change during the treatment process.

Abstract Background Our understanding of the relationship between plasma and cerebrospinal fluid (CSF) remains limited, which poses an obstacle to the identification of blood-based markers of neuroinflammatory disorders. To better understand the relationship between peripheral and central nervous system (CNS) markers of inflammation before and after surgery, we aimed to examine whether surgery compromises the blood-brain barrier (BBB), evaluate postoperative changes in inflammatory markers, and assess the correlations between plasma and CSF levels of inflammation. Methods We examined the Role of Inflammation after Surgery for Elders (RISE) study of adults aged ≥ 65 who underwent elective hip or knee surgery under spinal anesthesia who had plasma and CSF samples collected at baseline and postoperative 1 month (PO1MO) (n = 29). Plasma and CSF levels of three inflammatory markers previously identified as increasing after surgery were measured using enzyme-linked immunosorbent assay: interleukin-6 (IL-6), C-reactive protein (CRP), and chitinase 3-like protein (also known as YKL-40). The integrity of the BBB was computed as the ratio of CSF/plasma albumin levels (Qalb). Mean Qalb and levels of inflammation were compared between baseline and PO1MO. Spearman correlation coefficients were used to determine the correlation between biofluids. Results Mean Qalb did not change between baseline and PO1MO. Mean plasma and CSF levels of CRP and plasma levels of YKL-40 and IL-6 were higher on PO1MO relative to baseline, with a disproportionally higher increase in CRP CSF levels relative to plasma levels (CRP tripled in CSF vs. increased 10% in plasma). Significant plasma-CSF correlations for CRP (baseline r = 0.70 and PO1MO r = 0.89, p < .01 for both) and IL-6 (PO1MO r = 0.48, p < .01) were observed, with higher correlations on PO1MO compared with baseline. Conclusions In this elective surgical sample of older adults, BBB integrity was similar between baseline and PO1MO, plasma-CSF correlations were observed for CRP and IL-6, plasma levels of all three markers (CRP, IL-6, and YKL-40) increased from PREOP to PO1MO, and CSF levels of only CRP increased between the two time points. Our identification of potential promising plasma markers of inflammation in the CNS may facilitate the early identification of patients at greatest risk for neuroinflammation and its associated adverse cognitive outcomes.
