Purpose: To determine the antidiabetic effect of methanol extract of Plectranthus lanuginosus leaves in streptozotocin-induced hyperglycemic (HGD) rats. Methods: P. lanuginosus leaves were collected from Saad Medhas, Al Baha, Kingdom of Saudi Arabia. After defatting with n-hexane, they were extracted in vacuo at 40 oC with 75 % methanol. Streptozotocin (50 mgkg−1, i.p.) was used to induce hyperglycemia (diabetes) in the rats. The HGD rats received either standard drug (glibenclamide, 10 mgkg−1, p.o.) or Plectranthus lanuginosus leaf methanol leaf extract (PLLM) at doses of 200 and 400 mgkg−1/day, p.o. for 21 consecutive days. Blood samples were taken from the rat tails 2 h after dosing, and at 7-day intervals (i.e., 0, 7th, 14th and 21st days). The blood samples were used for measurement of fasting blood glucose (FBS), using a glucometer. On the 21st day, the rats were sacrificed via cardiac puncture. The activities of liver marker enzymes (SGPT and SGOT), and serum lipid profile (cholesterol, triglycerides, HDL and LDL) were determined using a hemolyzer. Results: Streptozotocin treatment produced significant hyperglycemia in the rats (348.9 ± 5.6) when compared to control (79.2 ± 1.3). However, PLLM (200 and 400 mg kg−1) produced significant and dose-dependent anti-diabetic (166.4 ± 5.6 and 123.86 ± 6.8 respectively) and antihyperlipidemic effects in HGD rats, at levels similar to those produced by the standard drug, glibenclamide (120.6 ± 6.4). Conclusion: P. lanuginosus leaf extract possesses pronounced anti-diabetic and anti-hyperlipidemic properties which may be due to the presence of phenolic and flavonoid constituents in the plant. Therefore, the plant extract can be further developed for the management of diabetes

Aims: Ocimum gratissimum is a medicinal plant that has been traditionally used in the management of many diseases including diabetes mellitus. The aim of this research was to evaluate hypoglycemic, hypolipidemic and antioxidant activities of Ocimum gratissimum leaf extracts on diabetic rats. Study Design: Mention the design of the study here. Place and Duration of Study: Department of Biochemistry, Faculty of Life Science, Kebbi State University of Science and Technology Aliero, Kebbi State, Nigeria. Between february 2021 and June 2021. Methodology: The phytochemical screening was carried out using standard procedures. The extract was administered orally (100, 200 and 400 mg/kg body weight, for 21 days) to alloxan-induced diabetic rats. Hypoglycemic effects, change in body weight, lipid profile and antioxidant activities of diabetic rats treated with the extract were assessed and compared with normal, diabetic control and standard drug treated rats. Histological examination of the pancreas during 21 days of treatment was also carried out. Results: The extract produced a significant reduction in fasting blood glucose level in alloxan-induced diabetic rats. Significant differences were also observed in body weights, serum lipid profile and levels of antioxidant vitamins and enzymes of Ocimum gratissimum methanol extract treated diabetic rats, when compared with diabetic, normal and standard drug treated rats. Histopathological studies of the pancreas showed comparable regeneration of the cells by extract which were earlier necrosed by alloxan. Methanol leaf extract of Ocimum gratissimum exhibit significant hypoglycemic, hypolipidemic and antioxidant activities in alloxan-induced diabetes in rats. The extract could be further processed towards the management of diabetes mellitus. Conclusion: Ocimum gratissimum extract showed a promising good hypoglycemic effect, the extracts also exhibit hypolipidemic and antioxidant activities on diabetic rats. There was regeneration of pancreatic islets of Langerhans. Therefore at acute dose the extracts can serve as an alternative in the management of diabetes mellitus.

Aim: To evaluate the nephroprotective property of Cnidoscolus chayamansa aqueous leaf extract in diabetic rats. Study Design: Rats were randomly divided into five groups with group 1 as the normal control. Diabetic was induced in groups 2-4. Group 2 was used as the test control while groups 3 and 4 were treated with different concentrations of the leaf extract; group 5 was treated with the standard drug, glipizide. Place and Duration of Study: Departments of Biochemistry and Anatomy, University of Jos, Nigeria, between August to November, 2019. Methodology: Fourty (40) male albino Wistar rats were grouped into five groups. The rats were treated for fourteen days and then sacrificed by decapitation after anaesthesia. Blood was collected for biochemical parameters; kidney was excised and stored in formaldehyde until required for histopathological study. Serum urea, creatinine, uric acid, sodium, potassium, chloride and bicarbonate were determined using appropriate methods. Results: The test control had a significant (P < .05) decrease in the concentrations of Na+, Cl- and HCO3- ions; significant (P < .05) increases in serum K+ ion, urea, uric acid and creatinine. Treatments of the test groups with the different doses of the leaf extract and the standard drug increased the concentration of Na+ ion which was not significantly (P < .05) different from the test control. On the other hand, the concentration of Cl- and HCO- ions  were significantly (P < .05) increased; the concentrations of K+, urea, uric acid and creatinine were significantly (P < .05) decreased. The histochemistry of the kidneys revealed that the injury brought about under diabetic condition was ameliorated with the treatments with the low and high doses of the leaf extract, and the standard drug. Conclusion: The results show that the aqueous leaf extract has nephroprotective property.

Diabetes is a chronic disease which characterized by hyperglycemia (elevated or abnormally high blood sugar levels) and other metabolic disturbances, including metabolism of lipids and haemostasis. Caesalpinia pulcherrima has previously showed strong anti-diabetic and hepatoprotective potential. The present research work was to investigate the anti-diabetic activity and hepatoprotective activity Caesalpinia pulcherrima in streptozotocin-induced (STZ) diabetic rats. The dose-dependent effects of 45days oral treatment with methanol extract of plant (200 and 300mg/kg) of CPAE on body weight, blood glucose level, total protein, albumin, liver marker enzymes and carbohydrate metabolizing enzymes were evaluated in STZ-induced diabetic rats. Oral administration methanolic extract of Caesalpinia pulcherrima of showed significant restoration of the body weight and decrease in the blood glucose level, liver marker enzymes (ALT, AST ALP) and carbohydrate metabolizing enzymes were observed in diabetic rats. These results suggest that fruit extract of Caesalpinia pulcherrima has valuable anti-diabetic activity in STZ-induced diabetic rats which is comparable to the standard drug metformin and hence might be of use in the management of diabetes.

Abstract Background Curcuma amada is the most popular traditional medicine in India for the treatment of diabetes. The present study aimed to focus the antidiabetic and antioxidative activity of C. amada through the analysis of biochemical and genomic levels in a dose-dependent manner in streptozotocin-induced male adult rat. Method Streptozotocin-induced diabetic rats were administered orally with hydro-methanolic extract of C. amada at the dose of 10, 20, 40 and 80 mg/100 g body weight of rats for 28 days. The antidiabetic and antioxidative efficacy of the extract on glycemic, enzymatic, genomic and histological sensors along with toxicity study was investigated. Results The result showed a significant antidiabetic and antioxidative effect of the extract at dose-dependent manner. The significant recovery of fasting blood glucose level, serum insulin, activity of carbohydrate metabolic enzymes and antioxidative enzymes in extract-treated diabetic group as compared to untreated diabetic group were noted. After the extract treatment, the size of pancreatic islet and cell population densities were significantly increased. Activities of glutamate oxaloacetate transaminase and glutamate pyruvate transaminase in liver were significantly recovered along with the correction of Bax and Bcl-2 gene expression in hepatic tissue after the extract treatment in diabetic rats in respect to untreated diabetic group. Out of all the doses, the significant effects were noted at the dose of 20 mg/100 g body weight which has been considered as threshold dose in the concern. Conclusion It may be concluded that the significant and corrective effect in most of the sensors was noted at the minimum dose of 20 mg/100 g body weight of hydro-methanolic extract of C. amada without producing any toxicity.

The study was designed to evaluate the glucose and cholesterol lowering effect of the aqueous extract of Centella asiatica leaf using the alloxan-induced diabetic rats and compared the activity with diabetic control and antidiabetic drug (Glibenclamide). Leaf extract (50 mg/kg) of C. asiatica and Glibenclamide were administered to normal and experimental diabetic rats for the duration of 10 days. In the alloxan-induced diabetic rat model, C. asiatica extract (50 mg/kg) significantly (p < 0.05) lowered the fasting blood glucose level as well as the total cholesterol level. Serum insulin levels were not stimulated in the animals treated with the extract. In addition, changes in body weight, serum lipid profiles and liver glycogen levels assessed in the extract treated diabetic rats were compared with diabetic control and normal animals. Significant results (p < 0.05) were observed in the estimated parameters. Surprisingly, body weight was increased significantly (p < 0.05) in the C. asiatica treated diabetic group. Introduction: Euphorbia antiquorum is a fleshy shrub belonging to the family Euphorbiaceae. It is widely distributed in the hotter parts of India. It is used as a substitute for the Ayurvedic drug ‘Snuhee’ whose accepted botanical source is Euphorbia neriifolia Linn. The leaf, stem, latex and root of Snuhee are used in treatment of diseases and conditions like, diabetes, coryza, and oedema. Objective: The present study is aimed to investigate the renal protective effect of  E. antiquorum alcoholic extracts of the stem   on alloxan (150mg/kg) induced diabetic Wistar rats. Methods: All animals were treated for 21 days and sacrificed on the last day. Unwanted placebo effect in case of the control group was blocked by executing appropriate vehicle treatment and same animal handling pattern. Extract and standard drug doses given orally were suspended in 0.9% normal saline. Blood samples were drawn from retro orbital sinus using Diethyl-ether anesthesia at weekly intervals for 3 weeks. Blood samples were labeled in such a way that analysis team was kept blind about groups and nature of the study design. Body weight measurement and blood sampling were done on 1st, 7th, 14th and 21st day of the study. Conclusion: 21 days daily treatment with a test drug of ethanolic extract of stems of Euphorbia antiquorum significantly reduced the elevated Blood glucose level, Urinary Albumin, urine creatinine, serum creatinine in alloxan induced diabetic rats. Background: Beluntas (Pluchea indica L.) is an Indonesian plant that grows wild and is used as traditional medicine. Beluntas leaves are reported to contain phenolic, and flavonoid is a part of phenolic compounds. Phenolic compounds other than flavonoids include 1,3,4,5-tetra-O-caffeoylquinic acid, 3,4,5-tri-O-caffeoylquinic acid, chlorogenic acid, and ferulic acid. Flavonoid compounds in beluntas leaves are quercetin, apigenin, luteolin and chrysoeriol. Methanol solvents are reported to be able to extract higher polyphenol and flavonoid contents than other solvents. Objective: The purpose of this study is to determine the effect of 20%, 50% and 75% solvent concentration of methanol on total phenol and total flavonoid levels of beluntas leaf extract. Methods: In this study, the extraction process was carried out by maceration, the determination of total phenol content was carried out by the Follin-Ciocalteu method. Determination of total flavonoid levels was carried out by the Colorimetric method. Data on phenol and flavonoid levels were analyzed statistically. Results: The yield of beluntas leaf extract in this study was respectively from the lowest methanol concentration of 24.094% w/w, 31.126% w/w, 24.838% w/w. The value of total phenol levels increased with increasing methanol concentration, namely 124.84 mg GAE/g, 138.3 mg GAE/g, and 147.91 mg GAE/g. The highest total flavonoid value in 75% methanol extract is 69.72 mg QE/g, followed by 20% methanol extract at 46.29 mg QE/g and the lowest is found in 50% methanol extract at 32.80 mg QE/g. The results of statistical analysis using the Kruskal-Wallis test showed that there were significant differences (p<0.05) in the value of total phenol levels and total flavonoids of the three extracts. Conclusion: The difference in the concentration of methanol solvents affects the value of total phenol levels and total flavonoids with the highest value produced by a 75% methanol solvent. Aim: Acute toxicity and protective effect of ethanol leaf extract of Rhus longipes Engl. against Paracetamol induced oxidative stress was investigated. The LD50 of the leaf extract was determined using up and down technique and the effect of 1/10th and 1/20th/ LD50 of the extract on antioxidants enzymes and non-enzymes were assessed in the serum and isolated liver of normal and Paracetamol intoxicated rats. Data obtained were analyzed by one-way analysis of variance (ANOVA) and Dunnett’s t-test was used as the test of significance. Values were considered significant at P value < 0.05. The results obtained indicated that LD50 of Rhus longipes Engl. leaf extract is greater than 5000 mg/kg /body weight. A significant (p<0.05) increase was observed in the level of hepatic (H) TBARs (81.97%), Catalase (38.42%) and serum (S) TBARs (164.44%) and catalase (64.72%) respectively but, a significant (P<0.05) decrease in hepatic activities of SOD, GPX, GR, vitamin C and E in paracetamol treated groups when compared with the serum and normal control group respectively. The extracts (250 and 500 mg/kg/body/weight) and the standard silymarin significantly (p<0.05) restored the derange antioxidants parameters to near normal in dose dependent manners. The activities of the extract at the highest concentration (500 mg/kg/b.wt) compared favourably with the standard drug. The results suggested that the leaf extract of Rhus longipes Engl. contain bioactive compounds which could protect against toxicity induced oxidative stress. The results of this study can be used as a basis for further investigations in the search for the bioactive principle.

Diabetes mellitus is an endocrine and metabolic disorder of humans and animals characterized by hyperglycemia and low blood insulin levels or insensitivity of target organs to insulin and it’s a major health problem affecting patient’s quality of life due to its many complications. Infertility is one of the major secondary complications in diabetes. Although numerous drugs have been used for intervention studies on diabetes-induced infertility worldwide, there are currently no treatments for diabetes associated infertility in humans. This study was performed to investigate the effects of Annona muricata ethanolic leaf extract (AMELE) on fertility of male diabetic rats and levels of blood glucose. Twenty male Wistar rats (150-200g) were randomly distributed into 4 groups (n=5) treated thus: CTRL (control), DNT, DT1 and DT2 (diabetic, single intraperitoneal injection, streptozotocin, 60 mg/kg). Group DT1 and DT2 received AMELE orally at 100 mg/kg and 200 mg/kg respectively daily for fourteen days. Data were analysed using ANOVA at α0.05. The animals were sacrificed after 2 weeks via thiopental injection and testicular weights were recorded. Fasting blood glucose was determined using a digital glucometer. Sperm count, motility, viability and morphology were assessed microscopically. Testes were histologically evaluated. The results showed that oral administration of AMELE at 100 mg/kg and 200 mg/kg to diabetic male rats for fourteen days significantly decreased blood glucose level and also ameliorated diabetes-induced decreases in sperm functions in streptozotocin-induced diabetic male rats.

The methanol extract of Acacia ferruginea (Family- Mimosaceae) was studied for possible antidiabetic activity on normoglycaemic, OGTT and alloxan induced diabetic rats at doses of 100, 200, and 400 mg/kg, p.o. The acute toxicity studies were carried out on Swiss albino mice to determine the LD50 values. The experiments were performed as per OECD guidelines. The results of the normoglycaemic, OGTT and hyperglycaemic studies revealed that the extract exhibited reduction in blood glucose concentration in a dose dependant manner as compared to the standard drug metformin (250 mg/kg, p.o.). The acute toxicity studies revealed no signs of mortality in animals treated with a single dose of 2000 mg/kg body weight. Preliminary phytochemical studies of the methanol extract revealed presence of alkaloids, steroids, triterpenoids, saponins, flavonoids, tannins and phenolic compounds, carbohydrates, gums and mucilages, proteins and amino acids. The present study justifies the use of the plant for treating diabetes as suggested in folklore remedies.
