A national survey was conducted of the microbiological quality of three dry ingredients used in beverages and one frozen non-dairy topping obtained at retail markets. Geometric mean aerobic plate counts (APCs) of units examined at 35°C were as follows: 1,313 units of cocoa powder, 6,600 CFU/g; 1,552 units of dry instant chocolate drink mix, 290 CFU/g; 1,559 units of dry non-dairy coffee creamer, 37 CFU/g; and 1,532 units of frozen non-dairy topping, 34 CFU/g. At 30°C, the geometric mean APC was 34 CFU/g for frozen nondairy topping. Geometric means for most probable number determinations of coliform bacteria and Escherichia coli were <3/g for the four products. Geometric mean values for Staphylococcus aureus in three of the products were <10/g; no S. aureus was found in cocoa powder. Geometric mean values for yeasts and molds in dry instant chocolate drink mix and dry nondairy coffee creamer were 8 and 6 CFU/g, respectively.

OBJECTIVE: To evaluate the microbiological quality of treated and untreated water samples came from urban and rural communities and to examine the relationship between coliforms occurrence and average water temperature, and a comparison of the rainfall levels. METHODS: A sample of 3,073 untreated and treated (chlorinated) water from taps (1,594), reservoir used to store treated water (1,033), spring water (96) and private well (350) collected for routine testing between 1996 and 1999 was analyzed by the multiple dilution tube methods used to detect the most probable number of total and fecal coliforms. These samples were obtained in the region of Maringá, state of Paraná, Brazil. RESULTS: The highest numbers water samples contaminated by TC (83%) and FC (48%) were found in the untreated water. TC and FC in samples taken from reservoirs used to store treated water was higher than that from taps midway along distribution lines. Among the treated water samples examined, coliform bacteria were found in 171 of the 1,033 sampling reservoirs. CONCLUSIONS: Insufficient treatment or regrowth is suggested by the observation that more than 17% of these treated potable water contained coliform. TC and FC positive samples appear to be similar and seasonally influenced in treated water. Two different periods must be considered for the occurrence of both TC and FC positive samples: (i) a warm-weather period (September-March) with high percentage of contaminated samples; and (ii) cold-weather period (April-August) were they are lower. Both TC and TF positive samples declined with the decreased of water temperature.

Abstrak Minuman Teh Poci merupakan produk minuman yang berkembang pesat dan banyak dijual oleh pedagang kaki lima di Padang terutama di Pasar Raya kota Padang. Penjualan Teh Poci oleh pedagang kaki lima di Pasar Raya tidak mengikuti sistem franchise yang mempunyai standar kebersihan penyajian menyebabkan proses pembuatan Teh Pociyang  dijual di Pasar Raya tidak terjamin kebersihan penyajianya. Tujuan penelitian ini adalah menguji kualitas mikrobiologis  minuman Teh Poci yang dijual pedagang kaki lima di Pasar Raya Padang. Minuman yang diperiksa adalah minuman Teh Poci yang dijual di sepanjang jalan Pasar Raya dengan mengambil 13 sampel. Penelitian dilakukan dari bulan November 2011- September 2012 dengan menggunakan metode Most Probable Number (MPN) yang terdiri dari tes penduga dan tes konfirmasi. Dari 13 sampel minuman yang diperiksa seluruh sampel tersebutmengandung bakteri Coliform. Faktor-faktor yang dapat mempengaruhi adalah kebersihan penjual minuman dalam menutup kemasan minuman, kurangnya higiene perseorangan, lokasi penjualan di tengah pasar, es batu yang tidak bersih, dan alat-alat yang digunakan. Kesimpulan penelitian ini ialah minuman Teh Poci yang dijual di Pasar Raya Padang tidak memenuhi standar kualitas mikrobiologis air minum yang sudah ditetapkan pada Peraturan Menteri Kesehatan tahun No. 492 tahun 2010.Kata kunci: pedagang kaki lima, teh Poci, higieneAbstract Teh Poci has become popular nowadays. There is a significant movement of Teh Poci street sellers in Padang especially in Pasar Raya Padang. Teh Poci street sellers in Pasar Raya do not buy the franchise system which has the appropriate standard for food serving. The Teh Poci stands are also located in the middle of traditional market which cause the serving to the society do not fulfill the sanitary standard.One of the indicators of water contamination is by measuring the amount of the coliform bacteria presents in water.  The objective of this study was to do themicrobiological quality test on Teh Poci in Pasar Raya Padang. The subject of this test was thirteen Teh Poci sold around Pasar Raya Street. The research occured from November 2011 until September 2012 using the Most Probable Number method, consists of two phases presumtive and confirmative.The result of the test showed that all thirteensamples were positive which means all the samples did not fulfill the microbiological quality of drinks published by the Indonesian Health Ministry.Keywords:  street sellers, Teh Poci, higiene

The microbial quality of raw fillets of aquacultured catfish, salmon, tilapia, and trout was evaluated. A total of 272 fillets from nine local and nine Internet retail markets were tested. Mean values were 5.7 log CFU/g for total aerobic mesophiles, 6.3 log CFU/g for psychrotrophs, and 1.9 log most probable number (MPN) per gram for coliforms. Differences in these microbial levels between the two kinds of markets and among the four types of fish were not significant (P > 0.05), except that Internet trout fillets had about 0.8-log higher aerobic mesophiles than did trout fillets purchased locally. Although Escherichia coli was detected in 1.4, 1.5, and 5.9% of trout, salmon, and tilapia, respectively, no sample had ≥1.0 log MPN/g. However, E. coli was found in 13.2% of catfish, with an average of 1.7 log MPN/g. About 27% of all fillets had Listeria spp., and a positive correlation between the prevalence of Listeria spp. and Listeria monocytogenes was observed. Internet fillets had a higher prevalence of both Listeria spp. and L. monocytogenes than did those fillets purchased locally. L. monocytogenes was present in 23.5% of catfish but in only 5.7, 10.3, and 10.6% of trout, tilapia, and salmon, respectively. Salmonella and E. coli O157 were not found in any sample. A follow-up investigation using catfish operation as a model revealed that gut waste exposed during evisceration is a potential source of coliforms and Listeria spp.

Water is the main source for life and also the most severe substance caused by pollution. The mandatory parameters for determining microbiological quality of drinking water are total non-fecal Coliform bacteria and Coliform fecal (Escherichia coli). Coliform bacteria are a group of microorganisms commonly used as indicators, where these bacteria can be a signal to determine whether a water source has been contaminated by bacteria or not, while fecal Coliform bacteria are indicator bacteria polluting pathogenic bacteria originating from human feces and warm-blooded animals (mammals) . The water inspection method in this study uses the MPN (Most Probable Number) method which consists of 3 tests, namely, the presumption test, the affirmation test, and the reinforcement test. The results showed that of 15 drinking water samples 8 samples were tested positive for Coliform bacteria with the highest total bacterial value of sample number 1, 15 (210/100 ml), while 7 other samples were negative. From 8 positive Coliform samples only 1 sample was stated to be negative fecal Coliform bacteria and 7 other samples were positive for Coliform fecal bacteria with the highest total bacterial value of sample number 1 (210/100 ml).

The consumption of lamb meat in Brazil has increased in the last years but little information about the microbiological quality of this product is available. To evaluate the hygienic-sanitary conditions of lamb carcasses, the quantification of microorganism populations indicators (mesophiles and psychrotrophs; total and thermotolerant coliforms; Escherichia coli; moulds and yeasts) and the pathogenic microorganisms indentification (Salmonella sp. and Listeria spp.) were performed. A total of 60 lamb carcasses were sampled from one abattoir in São Paulo. Swab samples were collected from three points (forequarter, back and hindquarter) on the muscle surface after carcasses final washing. Statistical analysis consisted of descriptive evaluation of the results whose counts were grouped by intervals of microorganism populations. Counts ranged from 1.0 x 10¹ to 8.0 x 10(4) colony-forming unit cm-2 (CFU cm-2) for mesophiles; 1.0 x 10(0) to 4.4 x 10(4)CFU cm-2 for psychrotrophs; < 1.0 x 10(0) to 4.4 x 10(4)CFU cm-2 for moulds and yeasts; < 0.3 to > 32.0 most probable number/cm² (MPN cm-2) for total and thermotolerant coliforms and Escherichia coli. Salmonella sp. and Listeria spp. were not found in any of the carcasses. Most carcasses presented low counts for all microorganisms. Overall results may be explained by the small size of the industry where the study was taken. Results suggest that good microbiological quality lamb meat is possible to be obtained, but improvement in hygienic-sanitary conditions is still required.

The microbiological quality of 150 units of raw ground beef obtained from a local retail store was determined. The range of aerobic plate counts was from 6.9 × 104 to 8.3 × 107/g. By using the most probable number method 96.7% of the 150 units were positive for coliforms, 94.7% for Escherichia coli and 61.3% for Staphylococcus aureus. By the plate methods, 99.3% of the units were positive for fecal streptococci and 56% were positive for Clostridium perfringens. No salmonellae were isolated. Aerobic and anaerobic organisms were isolated and identified. E. coli was the most frequently isolated aerobe followed by organisms in the Klebsiella-Enterobacter group. Among the anaerobic isolates, C. perfringens was the organism most frequently encountered.

<p>Monitoring the microbiology of honey is necessary to ensure its final quality. The microbial contamination is a danger to public health, a major concern for the surveillance agency. The objective of this study was to evaluate the microbiological quality of honey produced in the cerrado region, Mato Grosso State. The apiary was installed in the town of Conquista D'Oeste city, a total of 30 boxes of honey covered with asbestos tile and exposed to the sun. The honey was collected in November of 2008. The physical and chemical analysis were carried out: moisture, total soluble solids (TSS) and pH. Microbiological analysis were: mesophilic bacteria facultative aerobic, yeast and mold, Staphylococcus, the most probable number (MPN) of coliforms at 35&ordm;C and 45&ordm;C, and the presence of Salmonella sp. The methods used followed the directions of the Normative Instruction no. 62, August 23, 2003 of Minist&eacute;rio da Agricultura, Pecu&aacute;ria e Abastecimento - MAPA. The pH value resulted in 3.80, SST at 81.5 and humidity at 17 %. Microbiological analysis showed no presence of Salmonella, Staphylococcus and yeast. The count of facultative aerobic bacteria was 5.0 x 101, the two groups of coliforms in NMP was &lt;3.0, and 3.0 x 101 mold. The presence of refill drinking water in the urban community especially Badung Regency is favored as an alternative of water supply because it cost relatively cheaper compared to water in pacage. The number of samples in this study was 45 outlets with refill drinking water as its study material. The bacteriological examination of the material of study was done on included most probable number (MPN) of coliform and Eschericia coli. The result of refill drinking water investigation bacteriological examination revealed 88.9 % outlets were considered qualified, 11.1 % of them were unqualified because they contained coliform bacteria and one stall had Eschericia coli. The observation hygiene 15.6 % unqualified and sanitation outlets examination 17.8 % unqualified. Correlation test showed no significant relationship between raw materials, equipment, processing, hygiene and sanitation autlets with bacteriological quality of drinking water

ABSTRACT Cronobacter is a bacterial genus that includes seven species, and the species Cronobacter sakazakii is most related to meningitis and septicemia in infants associated with powdered infant formula (PIF). The objectives of this study were to evaluate the presence of C. sakazakii and to determine the microbiological quality of PIF for infant consumption. To do this, a total of 128 PIF samples were analyzed in four brands and countries (Chile, Mexico, Holland, and Brazil), considering three types of PIF: premature (PIF1), infant (PIF2), and follow-up (PIF3). Aerobic plate counts (APC) and Enterobacteriaceae (ENT) were assessed in accordance with Chilean official standards. The outer membrane protein A (ompA) gene was amplified to detect Cronobacter spp. and the fusA gene was amplified to identify C. sakazakii by using the PubMLST Web site and BLAST (NCBI). The antibiotic resistance profile was performed according to the Clinical and Laboratory Standards Institute standards. The pathogen was quantified by the most probable number (MPN). The results showed that APC median values for PIF1, PIF2, and PIF3 were 3.2, 4.9, and 4.8 log CFU g−1, respectively. The APC were higher in PIF2 (P &lt; 0.01) from Holland (P &lt; 0.01) in the commercial brand 4 (P &lt; 0.01). The ENT median values in PIF1, PIF2, and PIF3 were 1.8, 1.5, and 1.7 log CFU g−1, respectively. Five strains of C. sakazakii and one strain of Cronobacter malonaticus were identified as having values between 0.023 and 2.3 MPN/g. All strains (100%) harbored the ompA, plasminogen activator (cpa), and hemolysin (hly) virulence genes. To conclude, C. sakazakii was found in four PIF samples from four Chilean products and one from Mexico, which is distributed throughout America. C. sakazakii strains exhibit virulence factors and resistance to ampicillin, thus posing a risk when PIFs are consumed by infants. HIGHLIGHTS

This study investigated the efficacy of a POU water treatment system featuring sari-cloth filtration and/or Moringa oleifera coagulation as pre-treatments for solar disinfection (SODIS). Surface water from a peri-urban slum in Chennai, India, was treated and analysed for turbidity, organic content via chemical oxygen demand (COD) and microbiological quality via most probable number (MPN) enumeration of total coliforms. Pre-treatment with both moringa coagulation and sari-cloth filtration significantly improved the turbidity of raw water compared to no pre-treatment controls (P = 0.0002). Optimal moringa coagulation did not outperform sari-cloth filtration (P = 0.06), but combining optimal moringa coagulation with sari-cloth filtration significantly outperformed either pre-treatment independently with respect to turbidity (P = 0.016 and P = 0.0001, respectively). The addition of moringa was found to increase COD in treated water, with greater doses of moringa resulting in higher COD levels (P = 0.04). Increased organics may have encouraged the re-growth of coliform bacteria that was observed in those jars receiving moringa coagulant such that, with respect to MPN, those jars which were subject to optimal moringa coagulation did not outperform those undergoing sari-cloth filtration alone (P = 0.41). Sari-cloth filtration is recommended as a pre-treatment for SODIS whereas moringa is not, as further investigation on the relationship between organics and bacterial re-growth is necessary.
