Pharmacognostical Investigation of Trapa natans L Leaves

Background: Trapa natans L., is annual aquatic plant generally kwon as Water caltrp, Water chest nut belonging to the Trapaceae or Lytraceae family. Trapa natans L is use for the treatment of wide no of diseases without proper standardization. Objective: To give the right pharmacognostical and photochemical information of the Trapanatan L leaves. In this study pharmacognostical investigation of the fresh leaves and powder drug were done to determine the macroscopical, microscopical, quantitative physicochemical and phytochemical property of the drugs. Method: Macroscopical, quantitative and qualitative microscopy, physicochemical evaluation, extractive value, florescence analysis and phytochemical analysis were done according to the WHO guideline. Result: Macroscopical analysis showed that, leaves are greenish to purplish color, rhomboidal shape; alternate, acute, margin is dentate, pinnate venation. Microscopic evaluation showed that leaf is dorsi ventral in nature, upper layer epidermis cells were covered with cuticle layer. Single layer of barrel shape cell were present bellow the upper epidermis layer. Trichomes are generally multicellular. Anomocytic stomata were observed in upper epidermis. From the experiment it was found that methanolic extract give the highest extractive value. Phytochemical analysis gives the evidence for the presence of carbohydrate, alkaloids, glycoside, steroids, flavonoids, tannin, and triterpenoids. Qualitative phytochemical analysis give the evidence for presence of high amount total phenolic content. Conclusion: Different pharmacognostical parameters assessed in this examination help to detection and standardization of Trapa natans L., leaves.

Syzygium cumini (L.) Skeels (Family: Myricaceae) commonly known as Jamun is a widely used medicinal plant in Ayurveda. In spite of its numerous medicinal attributes, no published work is available on pharmacognostic characterization, physicochemical analysis of its leaves. The measures taken for pharmacognostic characterization were macroscopy, microscopy, powder microscopy, physicochemical evaluation, fluorescence analysis, and preliminary phytochemical screening. The microscopic evaluation showed useful characters for the recognition of leaf of Syzygium cumini (L.) Skeels . Transverse section, showed presence of single layer of wavy epidermal cells with striated cuticle single layer Palisade beneath upper epidermis in lamina region (contains compact elongated cells), Spheraphide in lamina region, collenchyma below the upper epidermis and above the lower epidermis in midrib region, Xylem and phloem in center, Sclerenchyma in between vascular bundle and collenchyma in midrib region. Wavy epidermal cell in the upper epidermis, anisocytic stomata in the lower epidermis, Bordered pitted reticulate vessels, Calcium oxalate crystal sheath in the lamina, fragment of fibers and Spiral vessels were observed in powder microscopy. Physicochemical studies showed total ash (3.1%), acid insoluble ash (0.7%), alcohol soluble extractive values (10.96 %), and water-soluble extractive values (12.32 %). Phytochemical analysis revealed the presence of sugar, lipid, Glycoside, saponins, phenols, flavonoids, tannins, tri-terpenoids and steroids. The study forms the first report on pharmacognostic characters and a physicochemical parameter which could be useful for identification and authentication of the plant.

<em>Cassia auriculata</em> L. (CAL) popularly identified as Tanner’s Cassia and <em>Cassia fistula</em> L. (CFL) is generally known as Golden Shower. Both plants belong to the Family, Fabaceae. These plants are used in skin disease, as hepatoprotective, as anticancer agent and as antioxidant agent. The intention of current article is to put forward the comparative pharmacognostical analysis of <em>Cassia auriculata</em> and <em>Cassia fistula</em> roots in terms of macroscopic evaluation, microscopic evaluation, physicochemical evaluation, extractive values and phytochemical analysis. Thin Layer Chromatography study was carried out for CAL and CFL and data pertaining to the above cited evaluations were recorded for both, CAL and CFL roots. The present study may help in differentiating among these species and these pharmacognostic parameters may serve as a tool for identification, authentication and standardization of CAL and CFL.

This study aimed to carry out morphoanatomical, histochemical and phytochemical analysis of leaves and stems of Tetrapterys multiglandulosa Cav. (Malpighiaceae), a native plant responsible for abortion and sudden death in beef cattle. Plant specimens underwent anatomical study, in which leaves and stems were dried and ground with aqueous and ethanol extracts subjected to histochemical and phytochemical analysis. Anatomical observation of vegetative organs detected the presence of simple columnar cells in the adaxial epidermis, palisade mesophyll, parenchyma with a layer of cells and paracitic type stomata. Stem cross-sections presented circular section, single layer (uniseriate) epidermis with thick cuticle and Malpighi’s trichomes, the phelogen was in differentiation. The cortex showed angular collenchyma, parenchyma with exogenous channels and druse shaped calcium oxalate crystals. Phytochemical tests of aqueous and ethanol extracts of leaves and stems indicated the presence of phenolic compounds, tannins, flavonoids, saponins, alkaloids and cardiotonic heterosides. Potentially phytotoxic secondary metabolites evidenced in the leaves and stems, including saponins and cardiotonic heterosides were correlated with clinical signs observed in intoxicated animals.

Sideritis niveotomentosa Hub. -Mor. is a local endemic species belonging to the Lamiaceae family. In this study, GC/MS analysis, total antioxidant capacity and anticancer effects of different extracts obtained from S. niveotomentosa were investigated comparatively. Total phenolic contents of extracts were determined by the Folin–Ciocalteu method, total flavonoid contents by aluminum chloride method, and also the free radical scavenging activities of the extracts by DPPH (2,2-diphenyl-1-picryl-hydrazyl-hydrate) assay. The cytotoxic effect of the extracts was studied via MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium Bromide) assay on DLD1, HL60 and ARH77 cell lines. Pro-apoptotic gene expression levels were also tested in the most sensitive cell line ARH77 by Real-Time PCR. The expression levels of 4 pro-apoptotic genes, APAF, BAX, CASP3, and HRK were found to be upregulated in ARH77 cells that were treated extracts. Results showed that methanolic extracts contain more phenolic content than acetone extracts, consistent with DPPH results. As a result, Sideritis niveotomentosa extracts, especially methanolic extracts, are rich in phenolic content and have a strong radical scavenging effect. In addition, the extracts showed selective effects on cell lines. This study is pioneering in terms of future studies, and the findings provide hope for future experimentation.

The phytochemical analysis of Vinca minor, V. herbacea, V. major, and V. major var. variegata leaf extracts showed species-dependent antioxidant, antibacterial, and cytotoxic effects correlated with the identified phytoconstituents. Vincamine was present in V. minor, V. major, and V. major var. variegata, while V. minor had the richest alkaloid content, followed by V. herbacea. V. major var. variegata was richest in flavonoids and the highest total phenolic content was found in V. herbacea which also had elevated levels of rutin. Consequently, V. herbacea had the highest antioxidant activity followed by V. major var. variegata. Whereas, the lowest one was of V. major. The V. minor extract showed the most efficient inhibitory effect against both Staphylococcus aureus and E. coli. On the other hand, V. herbacea had a good anti-bacterial potential only against S. aureus, which was most affected at morphological levels, as indicated by scanning electron microscopy. The Vinca extracts acted in a dose-depended manner against HaCaT keratinocytes and A375 melanoma cells and moreover, with effects on the ultrastructure, nitric oxide concentration, and lactate dehydrogenase release. Therefore, the Vinca species could be exploited further for the development of alternative treatments in bacterial infections or as anticancer adjuvants.

Propolis is one of the natural products produced by kelulut bees and is still not widely used. The type of stingless bee that is the prima donna in the community is Heterotrigona itama. This study aims to determine the phytochemical content of the n-hexane fraction of Heterotrigona itama bee propolis collected from Kutai Kartanegara, East Kalimantan. The n-hexane fraction was obtained from the methanol extract of H. itama propolis by the liquid-liquid partition method. After obtaining the n-hexane fraction, the research continued with a qualitative phytochemical test to identify the compound and determine total phenolic. Antibacterial activity was determined by the agar well diffusion method with a serial concentration in Escherichia coli bacteria. Qualitative phytochemical analysis in the form of color changes showed that the n-hexane fraction of H. itama propolis contained flavonoids, alkaloids, saponins, and tannins. Based on the results, the total phenolic content of the n-hexane fraction sample was 490 mgGAE/100 g. It caused the n-hexane fraction to have lower phenolic content than the methanol extract (792 mg GAE100 g). Furthermore, this result indicated that the non-polar fraction was not substantial enough to extracted phenolic compounds. It correlated to the antibacterial activity of the n-hexane fraction, which was very weak (2  mm ± 1.5) at  200µg/mL concentration.

Objective: In this study, the leaf powder of Aegle marmelos (L.) Correa. was subjected to preliminary phytochemical and in vitro antioxidant analysis. Methods: The freshly prepared plant leaf extract was subjected to preliminary phytochemical screening, which revealed the presence of alkaloids, tannins, saponins, flavonoids, glycosides, phenolic compounds, terpenoids, and steroids. Reducing power, superoxide (SO) anion radical, nitric oxide (NO) radical, and hydroxyl radical scavenging assays were carried out to evaluate the antioxidant potential of the methanolic leaf extract of this plant. The amounts of total phenolic and flavonoid compounds were also determined. Results: This study has revealed that the A. marmelos (L.) Correa. leaf extract showed considerably high amounts of most of the phytochemicals, total antioxidant capacity, total phenolic, and flavonoid content. The study also indicated that the A. marmelos (L.) Correa. showed comparatively good scavenging activity, i.e., inhibition of hydroxyl radical, NO and SO anion scavenging and reducing power activities when compared with the respective standards. Conclusion: The leaf powder of A. marmelos (L.) Correa. can be used as easily accessible source of natural antioxidant and as a possible food supplement or in pharmaceutical industry.

This study deals with the Pharmacognostical physicochemical and phytochemical evaluation of the whole plant of Piper longum L. Pharmacognostical studies include macroscopic, microscopic and physicochemical evaluation. Physicochemical constants showed the values of total ash (10.8%), acid insoluble ash (1.3%), water soluble ash (3.2%), sulphated ash (12%), nitrated ash (1.85%), hot extraction (11.2%), water soluble extractive (4.8%), ethanol soluble extractive (12.4%), ether soluble extractive (5.6%), loss on drying (29%), swelling index (0.33) and mucilage content (0.8%) w/w. The preliminary phytochemical screening of the alcoholic extract showed presents of alkaloids, phenolic compounds, tannins, mucilage, starch, steroid and flavonoids. This study helps to development of different standard parameters of the plant which help the plant identification and authentication. This study was aimed to identify the potential use of active compounds extracted from Staurogyne sp as an antibacterial agent to control disease-causing bacteria in fish. Staurogyne sp, an aquatic plant collected from Bantimurung, South Sulawesi were subjected to allelochemical compound. Plant extracts were tested to five bacteria including four types of bacterial pathogen, such as Aeromonas hydrophilla, Edwardsiella ictaluri, Streptococcus agalactiae, and Flavobacterium columnare, and one bacterial sensing quoroum, Chromobacterium violaceum. Based on phytochemical analysis, Staurogyne’s extracts derived from both stem and root contained flavonoids, phenols, and antioxidants compounds with the concentration of 0.018 mgQE/g, 0.3471 mgGAE/g, and  1004,391 IC50 µg/mL, respectively. In addition, Staurogyne plant extracts derived from both their stem and root has also produced both saponins and glycosides compounds. Plant extracts derived from Staurogyne’s leaves revealed three major chemical compounds such as flavonoids (0.77 mgQE/g), phenol (0.0629 mgGAE/g), and glycosides (+). However, no antioxidants and saponin compounds were detected.  Applying plant extracts as an antibacterial on five disease-causing bacteria causing diseases in aquaculture, such as Aeromonas Hydrophilla, Edwardsiella ictaluri, Streptococcus agalactiae, Flavobacterium columnare, and Chromobacterium violaceum bacterials revealed that at concentration of 0.1 g the Staurogyne sp extracts did not influence all the bacterials growth. However, by application of plant extract at concentration of 0.2 g on the growth media,three out of 5 tested bacterials (Aeromonas hydrophilla, Edwardsiella ictaluri, and Streptococcus agalactiae) showed intermediate inhibition responses on bacterial growth. The growth of remaining two bacterials pathogen, Flavobacterium columnare and Chromobacterium violaceum at 0.2 g plant extract of Staurogyne sp was not affected. This study revealed that the use of active compound derived from Staurogyne sp would be potential to be used in inhibiting disease-causing bacterial in fish in future. However, optimum concentration of the plant extracts, in particular on the inhibition of the growth of disease-causing bacteria in fish is still needed to adjust.Key words: Antibacterial, aquatic plant, Staurogyne sp, fish disease

Background: Herbal medicine mostly contains wide range of chemical compounds responsible for medicinal therapeutic use. Costus woodsonii is commonly called as Red Button Ginger and synonyms of the botanical name are Costus spiralis, Alpinia spiralis and Costus pisonis. In Malay, it is known as Setawar Halia Merah. In Chinese, it is known as Hong Bi Qiao Jiang. Objective: This research was conducted to study the pharmacognostical, phytochemical, antioxidant and antimicrobial activity of C. woodsonii leaf extracts. Method: Macroscopy, microscopy, phytochemical analysis, thin layer chromatography, antioxidant activity and antimicrobial activity of C. woodsonii leaf were carried out. Total flavonoids were estimated in the leaf extract. The total phenolic content of C. woodsonii leaf was determined using Folin-Ciocalteu reagent. The antioxidant activity of leaf extract of C. woodsonii was determined by performing DPPH radical scavenging. The microbial activity was determined by Well diffusion test, MIC (Minimum Inhibitory Concentration) test and MBC (Minimum Bactericidal Concentration) test. Result and Discussion: C. woodsonii belongs to costaceae with elliptical green leaves. Till now are no extensive studies on C. woodsonii. Preliminary phytochemical analysis revealed the presence of flavonoid, steroid, fat, phenol, tannin and mucilage in leaf extract. Physicochemical studies further revealed the ash value of leaf as 8.7%. Among the three extractions, alcohol extractive values showed the highest as 13%. Loss on drying at 105 degree Celsius in leaf was found to be 12.67%. The plant extract showed total phenolic content of 7.941 mg GAE/g at concentration of 5µg/ml. As for flavonoids content, plant extract showed 21.7 mg RE/g at concentration of 200µg/ml and 43.4 mg RE/g at concentration of 400µg/ml. For antioxidant activity, the plant extract showed weak antioxidant activity in DPPH scavenging activity assay. For antimicrobial test, the leaf extract of C. woodsonii showed weak antimicrobial activity. Conclusion: From this study, it can conclude that C. woodsonii leaf extract possess weak antioxidant activity and weak antimicrobial activity which need to be further validated by using more antioxidant assays and antimicrobial tests.
