To assess nephroprotective potency of methanolic extract of C. owariensis on renal histomorphology of Wistar rats during exposure to nephropathic activity of CCl4. Twenty eight (28) albino Wistar rats divided into four groups which include normal control group administered with vehicles -distilled water (1 ml/kg b.w.) and olive oil (3 ml/kg b.w.), experimental control group administered with CCl4 (3 ml/kg b.w.) twice a week, first treatment group administered with CCl4 (3 ml/kg b.w.) twice a week + methanolic extract of C. owariensis (100 mg/kg b.w.) daily and second treatment group administered with CCl4 (3 ml/kg b.w.) twice a week + methanolic extract of C. owariensis (300 mg/kg b.w.) daily for twenty eight (28) days. Phytochemical analysis of methanolic extract of C. owariensis was carried out using GC-MS. The body weight of study animals was measure at days 0, 7, 14, 21 and 28 of study. Then, kidney tissue of study animals was collected, weighed and processed for histopathological study. Tissue sections were stained using H & E, examined under microscope, photomicrographs were generated and observable histopathological changes were quantified using image-J software. Phytochemical analysis of methanolic extract of C. owariensis showed abundance of phenolic compounds which may in turn confer antioxidant property on the extract. Results of this study also showed that treatment with extract helped to reduce body and tissue weight loss that follows exposure to CCl4. Also, treatment with the extract helped to reduce significantly (p < 0.05) renal histopathological changes following exposure to CCl4. The methanolic extract of C. owariensis contains abundant phenolic compounds which confer antioxidant property that in turn mediate the nephroprotective potency of the extract against nephropathic effect of CCl4.

Abstract: Liver is the largest organ in the abdominal cavity. As the center of metabolism in the body, liver is potentially damaged by exposure of toxic substances, inter alia carbon tetrachloride (CCl4). Metabolism of carbon tetrachloride (CCl4) produces CCl3 free radicals that can damage the liver. In Indonesia, there are a lot of natural ingredients that have antioxidant properties, such as tomato. Lycopene in tomatoes contains antioxidant compounds that can prevent damages due to free radical. This study aimed to obtain liver histopathological changes of wistar rats fed with tomato juice after being induced of carbon tetrachloride (CCl4). This was an experimental study, using 10 wistar rats which were divided into 4 groups. Group I was the negative control; group II was induced with CCl4 0,05 cc/day and was terminated on day 6; group III was induced with CCl4 0,05 cc/day and was given tomato juice 3 ml/day, and terminated on day 13; group IV was induced by CCl4 0,05 cc/day, given regular pellets, and terminated on day 13. The results showed that group II had histopathological changes of the liver indicating fatty liver, meanwhile group III showed regeneration of nearly all liver cells. Conclusion: Administration of tomato juice after the induction of 3 ml carbon tetrachloride (CCl4) for 7 day showed regeneration of almost all liver cells. Keywords: histopathological changes of the liver, carbon tetrachloride, tomato juice. The purpose of this study was to evaluate effect of Coleus amboinicus (CA) stem extracts on uric acid-induced nephropathy by comparingthe levels of Macrophage CD-68 expression and concentration of serum Cystatine C (CYS C ) in Wistar rats. Twenty-four male Wistar rats(Rattus norvegicus) with a body weight (bw) of 200-250 g, were allocated into three groups, with eight animals per group. The rats in controlgroup (PO) received 0.1% carboxymethyl cellulose (CMC) solution orally The rats in group 2 (P1) were orally induced with uric acid (UA) (500mg/kg) and oxonic acid (OA) (750 mg/kg.) and the rats in group 3 (P2) received uric acid (500 mg/kg), oxonic acid (750 mg/kg), and 500 mg/kgof the CA stem extracts for 35 days. Bloods were collected for analysis of serum CYS C expression and concentration of serum creatinine andblood nitrogen urea (BUN). The rats in all groups were sacrificed for kidney tissue extractions for macrophage CD-68 identification andhistopathology analysis. The levels of CYS C concentrations were analyzed by Avidin-Horseradish Peroxidase (HRP) Sandwich-ELISA. Theresults showed that Coleus amboinicus stem extract at dose of 500 mg/kg bw can significantly reduce BUN and creatinine levels (P≤0.05), whileCys C levels were not different. In the treatment group (P2) compared with group (P1). CD-68 (ED-1) macrophage activity decreasedsignificantly (P≤0.05) in the treatment group (P2) compared to the control group and (P1). Nephrophaty induction using UA and OA causessevere kidney lesions characterized by degeneration, necrosis and inflammation of the renal tubules and glomerulus in the treatment group.

To assess the effect of methanolic leaf extract of Caladium bicolor on the histomorphology of gastrointestinal tissues of experimental animals. Twenty four Wistar rats (weighing between 175-190 g) were randomly and equally divided into four groups which include one control group (CG) and three treatment groups (TG I, TG II and TG III). The CG was administered with distilled water [2 ml/kg body weight (b.w.)] while TGs I, II and III were administered with 100 ml/kg, 200 ml/kg and 300 ml/kg (b.w.) of C. bicolor extract respectively. All administrations were done orally and once daily for a period of thirty days. The body weight of all animals was recorded at the beginning and end of study. After the period of study, gastric and small intestinal tissues of experimental animals were harvested, processed, converted to tissue blocks and sectioned. Tissue sections were stained using Haematoxylin and Eosin (H&E) technique. Thereafter, stained sections microscopically examined for observable histopathological changes within study tissues. The results of this study showed that exposure to C. bicolor extract causes significant (p < 0.05) body weight loss in TGs I-III compared to CG. In addition, prominent histopathological changes were observed in gastrointestinal tissues of experimental animals in TGs I-III including gastric mucosal surface erosion and intestinal villi degeneration compared to normal gastrointestinal histomorphology of CG animals. These histopathological changes may be associated with toxic effect of phytochemicals constituents of the extract. Therefore, its application for therapeutic purposes needs to be thoroughly re-validated or perhaps disallowed where alternative therapeutic agents with minimal toxic potential exist.

Background: Tooth extraction is the process of removing a tooth from the oral cavity potentially triggering a wound healing response in the body. As a result, many methods have been applied to improve the wound healing process, especially in wounds resulting in complications. One such method involves the application of amniotic membrane which has anti-inflammatory, anti-bacterial, anti-fibrosis, anti-scarring properties with low immunogenicity, epithelialization effects, and secretory leukocyte protease inhibitor (SLPI). It also contains collagen, various growth factors, transferrin, fibronectin, nidogen, proteoglycans, hyaluronan, and laminin. Purpose: This study aimed to determine the effects of sponge amnion on the number of VEGF expressions and new blood vessels in post-tooth extraction wounds of Wistar rats. Methods: Sponge amnion was produced by mixing freeze-dried amnion membrane from the Tissue Bank at RSUD Dr. Soetomo with 1% gelatin before freeze drying the mixture. Wistar rats were then divided into two groups. In Group 1, referred to as the control group, the post-extraction wounds of the rats received no treatment. Meanwhile, in Group 2, the treatment group, the subjects’ post-extraction wounds were treated with sponge amnion. The rats of both groups were sacrificed on day 3 to allow observation of the number of VEGF expressions and new blood vessels. A statistical analysis test, a t-test, was subsequently conducted. Results: There was a significant difference in the number of new blood vessels in the control group and that of the treatment group with a p value of 0.018 (p<0.05). There was also a significant difference in VEGF expression between the two groups with a p value of 0.000 (p <0.05). Conclusion: Sponge amnion can generate a number of VEGF expressions and new blood vessels in the post-extraction wounds of Wistar rats.

This study was initiated on 24 prepubertal Holstein x Kankrej crossbred heifers of nearly identical age (7-9 months) and body weight (130-140 kg) at University farm to evaluate the effect of high plane of nutrition on blood biochemical and minerals profile and the age at puberty. Twelve heifers were managed under routine farm feeding (control) and the rest 12 under ideal optimum feeding regime (treatment) that included extra 1 kg concentrate, 30 g min mix and ad-lib dry fodder. The body weight and ovarian ultrasonography together with blood sampling was carried out at monthly interval from 10 to 18 months of age to study the ovarian dynamics and blood biochemical changes. High plane of nutrition to growing heifers was beneficial in reducing the age of onset of puberty (by 2-3 months) compared to routine farm fed group. The mean plasma total protein and cholesterol concentrations showed a rising trend with significant variations from 10 to 16 months of age, where it got mostly stabilized indicating adult profile. The activity of enzymes GOT and GPT also rose gradually and significantly from 10 months till 14-15 months of age, and thereafter it remained more or less static till 18 months of age. The levels of both these enzymes were higher, with lower protein and cholesterol, in control than the treatment group from 15-16 months of age onwards. The mean plasma levels of both calcium and phosphorus increased gradually and significantly with advancing age till 16-17 months of age, with little higher values in supplemented than a control group. The plasma levels of zinc, iron, copper, and cobalt also showed rising trend with significant differences between 10th and 12th-14th months of age, and from 15th to 18th months of age the levels were statistically the same in all the groups with slightly higher values in the treatment group.

The aim of this study was to investigate the clinical efficacy of vanadium complexes on triglycerides (TG), total cholesterol (Chol), uric acid (UA), urea (U), and antioxidant parameters: nonenzymatic (FRAP—ferric reducing ability of plasma, and reduced glutathione—GSH) and enzymatic (glutathione peroxidase—GPx, catalase—CAT, and GPx/CAT ratio) activity in the plasma of healthy male Wistar rats. Three vanadium complexes: [VO(bpy)2]SO4⋅2H2O, [VO(4,4′Me2bpy)2]SO4⋅2H2O, and Na[VO(O2)2(bpy)]⋅8H2O are administered by gavage during 5 weeks in two different diets such as control (C) and high fatty (F) diets. Changes of biochemical and antioxidants parameters are measured in plasma. All three vanadium complexes statistically decrease the body mass growth in comparison to the control and fatty diet. In plasma GSH was statistically increased in all vanadium complexes-treated rats from control and fatty group in comparison to only control group. Calculated GPX/CAT ratio was the highest in the control group in comparison to others.
