Lung adenocarcinoma (LUAD) is one of the most common malignancies, and there is still a lack of effective biomarkers for early detection and prognostic prediction. Here, we comprehensively analyze the characteristics of. an RNA sequencing data set of LUAD samples. In total, 395 long non-coding RNAs (lncRNAs), 89 microRNAs (miRNAs), and 872 mRNAs associated with c-Myc were identified, which were differentially expressed between tumor and normal tissues. The most relevant pathway was found to be WT1-AS–miR-200a-3p–IGF2BP2 according to the rules of competitive endogenous RNA (ceRNA) regulation. WT1-AS and IGF2BP2 expression were positively correlated and increased in LUAD samples, while miR-200a-3p had relatively low expression. The high expression of WT1-AS and IGF2BP2 was associated with poor prognosis in LUAD patients, while low expression of miR-200a-3p predicted reduced survival (p < 0.05). The analysis of the multi-gene regulation model indicated that the WT1-AS (downregulation)–miR-200a-3p (upregulation)–IGF2BP2 (downregulation) pattern significantly improved the survival of LUAD patients. Finally, reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting were detected in LUAD cells, and the results are consistent with the bioinformatics analysis. In summary, the WT1-AS/IGF2BP2 axis is a potential prognostic biomarker in LUAD and is expected to become an effective target for diagnosis and treatment.

Intelligence is innate, but grit is something everyone can develop. Grit not only enables students to stick to their goals, but also to persevere even when they fail. Career adaptability is an important concept in vocational education of college students, which is a person engaged in some work, must have a certain physical and psychological quality. Base on the self-regulation theory, this study investigated the relationship between grit and career adaptability of Chinese college student based on the self-regulatory processes. We surveyed 839 Chinese college students and tested a self-regulation model. As expected, grit was related to greater career adaptability via greater career exploration and decision self-efficacy, positive affect, and goal commitment. These findings not only broaden the theoretical framework for the effect of grit on career adaptability, but also open up a new horizon for improving college students’ career adaptability in practice.

AbstractThe epithelial-mesenchymal transition (EMT) is a cellular process critical for wound healing, cancer metastasis and embryonic development. Recent efforts have identified the role of hybrid epithelial/mesenchymal states, having both epithelial and mesehncymal traits, in enabling cancer metastasis and resistance to various therapies. Also, previous work has suggested that NRF2 can act as phenotypic stability factor to help stablize such hybrid states. Here, we incorporate a phenomenological epigenetic feedback effect into our previous computational model for EMT signaling. We show that this type of feedback can stabilize the hybrid state as compared to the fully mesenchymal phenotype if NRF2 can influence SNAIL at an epigenetic level, as this link makes transitions out of hybrid state more difficult. However, epigenetic regulation on other NRF2-related links do not significantly change the EMT dynamics. Finally, we considered possible cell division effects in our epigenetic regulation model, and our results indicate that the degree of epigenetic inheritance does not appear to be a critical factor for the hybrid E/M state stabilizing behavior of NRF2.

Milk fat is the most important and energy-rich substance in milk and plays an important role in the metabolism of nutrients during human growth and development. It is mainly used in the production of butter and yogurt. Milk fat not only affects the flavor and nutritional value of milk, but also is the main target trait of ruminant breeding. There are many key genes involve in ruminant milk fat synthesis, including ACSS2, FASN, ACACA, CD36, ACSL, SLC27A, FABP3, SCD, GPAM, AGPAT, LPIN, DGAT1, PLIN2, XDH, and BTN1A1. Taking the de novo synthesis of fatty acids (FA) and intaking of long-chain fatty acids (LCFA) in blood to the end of lipid droplet secretion as the mainline, this manuscript elucidates the complex regulation model of key genes in mammary epithelial cells (MECs) in ruminant milk fat synthesis, and constructs the whole regulatory network of milk fat synthesis, to provide valuable theoretical basis and research ideas for the study of milk fat regulation mechanism of ruminants.

Abstract Objective: To build an intelligent regulation model of oxytocin (OT) for women having a vaginal birth after cesarean section (VBAC) and further explore the intelligent and precise regulation of the scheme of the OT medication at delivery. Methods: A cross-sectional study design was used by collecting the data of the VBAC parturitions who delivered in the Obstetrics Department of the First Affiliated Hospital of Wenzhou Medical University from January 2014 to May 2020 and used the OT during the labor process. The multiple linear regression was used to analyze the modeling variables from the electronic medical records and the variables such as fetal heart rate (FHR) and uterine contraction (UC) frequency extracted from the cardiotocography. The OT drip speed predictive model was established based on the XGBoost algorithm and was compared using the logistic regression model and traditional decision tree. The data set was divided into the training set and test set at the 8:2 ratio, and the predictive performance of the model was evaluated for the accuracy, precision, recall rate, and F1 score. Results: A total of 1005 records with oxytocin regulation were included from 124 parturitions involving the VBAC. The XGBoost model performed the best prediction. Through the five fold crossover operation, the accuracy was 0.82, precision was 0.84, the recall rate was 0.80, and the F1 value was 0.82. Among them, the contraction duration, uterine pressure, FHR, UC frequency, and interval time from the previous cesarean section were the variables with great contribution to the modeling. Conclusion: This study constructed an OT regulation model based on the XGBoost, which recognized that the real-time intelligent regulation of the prenatal OT, with fast response speed, high model accuracy, and strong extrapolation had positive significance to obstetric clinical nursing.

Farmers in the trade chain for agricultural commodities, including corn, are in the weakest position compared to other parties. As a result, the trading system often does not take sides or even tends to harm farmers from year to year. Therefore, state intervention is needed in the context of evaluation which will lead to the reformulation of regulations regarding the maize trade system. Farmers are certainly an important element in realizing food security in this country. Therefore, this research was conducted to produce a regulatory model regarding the corn trade system which has unequivocal support for the protection of farmers. The purpose of this research is to identify the advantages and disadvantages of the current regulatory model, both by the central government and by local governments. To the corn trade system and find a model of corn trading system regulation based on protection for farmers. The research method used in this research is qualitative research and doctrinal legal research with several approaches, namely: statutory approach, analytical approach, and case approach with data collected through literature study and combined with empirical research (field research). This research is very important and useful for stakeholders (especially for farmers, entrepreneurs, and the government) as well as legislators in formulating policies on the trading system of agricultural commodities, especially corn which can ensure that farmers get serious attention and protection from the state.

Comfort can be considered as subjective feeling, which could be affected by the external ambient, by the physical activity, and by clothing. Considering the human body heat transfer system, it mainly depends on various parameters including clothing materials, external and internal environment, etc. The purpose of the current paper is to study and establish a quantitative relationship between one of the clothing parameters, ease allowance (air gap values) and the heat transfer through the human body to clothing materials and then to the environment. The study considered clothing which is integrated with the 3D ease allowance from the anthropometric and morphological data. Such incorporating of the clothing’s 3D ease control was essential to properly manage the air space between the body and the proposed clothing thermal regulation model. In the context of thermal comfort, a clothing system consisting of the human body, an ease allowance under clothing, a layer of textile materials, and a peripheral layer adjacent to the textile material was used. For the complete system, the heat transfer from the skin to the environment, which is influenced by thermoregulation of the human body, air gap, tissue, and environmental conditions were also considered. To model and predict the heat transfer between the human body and the temperature of skin and clothes, a 3D adaptive garment which could be adjusted with ease allowance was used. In the paper, a thermoregulatory model was developed and proposed to predict the temperature and heat within clothing material, skin, and air space. Based on the result, in general the main difference in the temperature of clothing and skin from segment to segment is due to the uneven distribution of air layers under the clothing.
