To evaluate the effects of dietary fiber from corn husks on hyperlipidemia, the hyperlipidemia model of mice was induced by trans-fatty acids diet,and the mice were randomly divided into the blank control group, the high fat control group(TFA group) and the low, middle and high dose group (2.5,5.0,10.0 G / kg) of dietary fiber from corn husks. Except the blank control group, the mice were fed with high fat diet. The body mass, serum total cholesterol, triglyceride, low density lipoprotein(LDL) and high density lipoprotein(HDL) were measured. The intake of trans-fatty acids resulted in a significant increase in the body weight, serum total cholesterol, triglyceride and LDL(P<0.01), HDL level was significantly lower(P<0.01). Corn husks dietary fiber intervention can reduce the body weight of mice, the serum total cholesterol, triglyceride, LDL concentration and increase HDL concentration. The difference between the high-dose group and the TFA group was significant(P<0.01), and the intervention effect was best in the group with high dose of corn husks dietary fiber. Corn husks dietary fiber can effectively reduce blood lipid in hyperlipidemia mode mice.

Our purpose in this study was to investigate the effect of acupuncture therapy on body weight and on levels of the serum total cholesterol, triglyceride, high-density lipoprotein (HDL) cholesterol and low-density lipoprotein (LDL) cholesterol in obese women. Fifty-five women were studied in three groups as follows: (1) control group ( n = 12; mean age = 43.3 ± 4.3, and mean body mass index { BMI } = 32.2 ± 3.4); (2) electroacupuncture (EA) ( n = 22; mean age = 39.8 ± 5.3, and BMI = 34.8 ± 3.3); and (3) diet restriction ( n = 21; mean age = 42.7 ± 3.9, and BMI = 34.9 ± 3.3). EA was performed using the ear points, Sanjiao (Hungry) and Shen Men (Stomach), and the body points, LI 4, LI 11, St 25, St 36, St 44 and Liv 3, once daily, for 30 minutes, for 20 days, whereas patients on diet restriction had a 1425 Kcal diet program, that consisted of 1425 Kcal daily for 20 days. There was a 4.8% weight reduction in patients with EA application, whereas patients on diet restriction had a 2.5% weight reduction. There were significant decreases in total cholesterol and triglyceride levels in EA and diet groups compared with the control group ( p < 0.05 in both cases). Furthermore, there was a decrease in LDL levels in the EA group compared with the control group ( p < 0.05). No significant changes could be found in HDL levels among the three groups. Our results suggest that EA application in obese women may decrease the serum total cholesterol, triglyceride, and LDL cholesterol levels by increasing the serum beta endorphin level. This lipolytic effect of EA may also reduce the morbidity of obesity by mobilizing the energy stores that result in weight reduction.

An experimental study was done to observe the hypocholesterolaemic effect of wheatgrass (Triticum aestivum) juice on experimentally induced hypercholesterolaemic rats, especially on serum low density lipoprotein (LDL) level, which was carried out in the Department of Pharmacology & Therapeutics, Dhaka Medical College, Dhaka, between July, 2009 and June, 2010. A total number of 48 healthy adult male Long Evans rats (Norwegian strain) were used for the present study. 6 rats were taken in each group of total 8 groups treated differently with 1 control group. Rats of the first 5 groups were sacrificed at 29th day and their serum total cholesterol levels were measured. The rest of the rats were sacrificed at 57th day and their serum total cholesterol levels were measured. The mean serum LDL of 10ml/kg grass juice fed normal rats was decreased by 11.64% than that of the laboratory diet fed control group (P<0.01). Serum LDL of the group of rats treated with 10ml/kg grass juice and1% cholesterol diet were decreased by 36.59% than that of the hypercholesterolaemic control group (P<0.001). Serum LDL of the hypercholesterolaemic rats treated with 10ml/kg grass juice for 28 days were decreased by 29.10%, and in hypercholesterolaemic rats treated with 20ml/kg grass juice for 28 days were decreased by 39.46% (P<0.001). Reduction of LDL as evident on hypercholesterolaemic rats treated with 20ml/kg grass juice, which was similar to hypercholesterolaemic rats treated with 0.14mg/kg Atorvastatin for 28 days. OBJECTIVE: The aim of the present study was to investigate the lipid profiles of the hepatic and adipose tissues of Wistar rats treated for 21 days with a diet high in saturated fat (high saturated fat, n=6) or high in hydrogenated fat, that is, having 50% partially hydrogenated vegetable oil in its composition (high hydrogenated fat, n=6), and compare them to those of a control group (control group, n=6). METHODS: Adipose tissue and total hepatic fat were higher in the saturated fat group than in the hydrogenated fat group. Hepatic lipid peroxidation was greatest in the saturated fat group, with consequent lower hepatic vitamin E and A levels. In contrast, serum vitamin A was highest in the saturated fat group. Analysis of hepatic lipid fractions found more cholesterol and less high density lipoprotein-cholesterol in the hydrogenated fat group. The hydrogenated fat group had the highest levels of triacylglycerols, followed by the saturated fat group. RESULTS: Significant amounts of trans fatty acids were detected in the hepatic and adipose tissues of the hydrogenated fat group. Among the identified fatty acids, 18:1n9 had a higher positive association with hepatic cholesterol and triacylglycerols, and a higher negative association with high density lipoprotein-cholesterol. Partially hydrogenated vegetable oil promotes greater accumulation of cholesterol and triacylglycerols in the liver than saturated fats. CONCLUSION: Trans fatty acids were incorporated into hepatocytes and adipocytes in a highly efficient manner.

AbstractObesity and high cholesterolaemia are major health problems in industrialized countries. The effects of the antiobesity drug orlistat at 0.3 g kg-1 and amidated alginate at 40 g kg-1 on serum and hepatic cholesterol and the faecal output of fat and sterols were compared in female rats. Rats were fed diets containing cholesterol and palm fat at 10 and 70 g kg-1, respectively. Palm fat was provided by coconut meal. Amidated alginate (the octadecylamide of alginic acid) is a sorbent of lipids, and orlistat (tetrahydrolipstatin) is an inhibitor of pancreatic lipase. Both agents significantly increased the faecal loss of fat, orlistat, however, did not significantly decrease serum total cholesterol and its effect on hepatic cholesterol was less pronounced. Amidated alginate at 40 g kg-1 significantly decreased serum total cholesterol, LDL cholesterol, hepatic cholesterol, and hepatic lipids, and increased the faecal output of fat and coprostanol (a metabolite of cholesterol). Both orlistat and amidated alginate modified the fatty acid profile in excreted lipids. The concentration of saturated fatty acids decreased and the concentration of unsaturated fatty acids increased. Despite different modes of action, orlistat and amidated alginate were equally efficient in the removing dietary fat from the body. Amidated alginate, however, was more active in the control of serum and hepatic lipid metabolism.

This study investigated the effects of berberine on amelioration of hyperglycemia and hyperlipidemia and the mechanism involved in high glucose and high fat diet-induced diabetic hamsters. Golden hamsters fed with high glucose and high fat diet were medicated with metformin, simvastatin, and low or high dose of berberine (50 and 100 mg·kg−1) for 6 weeks. The results showed that the body weights were significantly lower in berberine-treated groups than control group. Histological analyses revealed that the treatment of berberine inhibited hepatic fat accumulation. Berberine significantly reduced plasma total cholesterol, triglyceride, free fatty acid, low density lipoprotein cholesterol, malondialdehyde, thiobarbituric acid-reactive substance, and 8-isoprostane level but significantly increased plasma superoxide dismutase activity. Glucose and insulin levels were significantly reduced in metformin and berberine-treated groups. Glucose tolerance tests documented that berberine-treated mice were more glucose tolerant. Berberine treatment increased expression of skeletal muscle glucose transporter 4 mRNA and significantly decreased liver low density lipoprotein receptor mRNA expression. The study suggested that berberine was effective in lowering blood glucose and lipids levels, reducing the body weight, and alleviating the oxidative stress in diabetic hamsters, which might be beneficial in reducing the cardiovascular risk factors in diabetes.

Background:: Lung cancer remains the most common cause of cancer-related deaths in China and worldwide. Traditional surgery and chemotherapy do not offer an effective cure although gene therapy may be a promising future alter-native. Kallistatin (Kal) is an endogenous inhibitor of angiogenesis and tumorigenesis. Recombinant adeno-associated virus (rAAV) is considered the most promising vector for gene therapy of many diseases due to persistent and long-term transgen-ic expression. Objective:: The aim of this study was to investigate whether rAAV9-Kal inhibited NCI-H446 subcutaneous xenograft tumor growth in mice. Method:: The subcutaneous xenograft mode were induced by subcutaneous injection of 2×106 H446 cells into the dorsal skin of BALB/c nude mice. The mice were administered with ssrAAV9-Kal (single-stranded rAAV9) or dsrAAV9-Kal (double-stranded rAAV9)by intraperitoneal injection (I.P.). Tumor microvessel density (MVD) was examined by anti-CD34 stain-ing to evaluate tumor angiogenesis. Results:: Compared with the PBS (blank control) group, tumor growth in the high-dose ssrAAV9-Kal group was inhibited by 40% by day 49, and the MVD of tumor tissues was significantly decreased. Conclusion:: The results indicate that this therapeutic strategy is a promising approach for clinical cancer therapy and impli-cate rAAV9-Kal as a candidate for gene therapy of lung cancer.

The objective of this study was to evaluate short-term variations of trans fatty acids (TFA) in plasma lipoproteins and ruminal fermentation parameters of non-lactating cows subjected to ruminal pulses of vegetable oils. Three non-lactating, non-pregnant Holstein cows, each with a ruminal cannula, were arranged in a 3 × 3 Latin square design with three-day pulsing periods and four-day washout intervals between treatments. Cows were treated with single ruminal pulses of: (1) control (skimmed milk (SM); 500 mL); (2) soybean oil (SO; 250 g/d in 500 mL of SM) and (3) partially-hydrogenated vegetable oil (PHVO; 250 g/d in 500 mL of SM). Time changes after infusion in TFA contents were only observed for plasma C18:1 trans-4, trans-5 and trans-12, and high-density lipoprotein fraction C18:1 trans-9. After ruminal pulses, concentration of acetate decreased linearly; molar concentrations of propionate and valerate increased linearly; molar concentrations of butyrate and isovalerate changed quadratically and were greater at 1 h than at other times. There was an accumulation of several C18:1 TFA in plasma and lipoproteins, especially on the third day of pulsing. Overall, naturally occurring C18:1 TFA isomers (produced during ruminal biohydrogenation of SO) and preformed TFA (supplied by PHVO) elicited differential TFA partitioning and transport in plasma and lipoproteins.

The effects of atorvastatin and carboxymethylated β-glucan (CMG) on the lipoprotein-cholesterol (LP-C) and lipoprotein-triglyceride (LP-TG) fractions and subfractions at the early stage of murine hyperlipidemia, and its pleiotropic anti-inflammatory effects, were studied. Atorvastatin and CMG were administered in ICR male mice with acute lipemia induced with a single injection of poloxamer 407 (P-407). A novel small-angle X-ray scattering method for the determination of fractional and subfractional composition of LP-C and LP-TG was used. In P-407-treated animals, there was a drastic increase of total cholesterol and especially TG. Atorvastatin decreased both the total cholesterol and TG, but not to control levels. CMG primarily decreased TG and was not as potent as atorvastatin. P-407 increased atherogenic LDL-C (IDL-C and LDL1–3-C subfractions) and very low-density lipoprotein-C (VLDL-C) (VLDL1–2-C and VLDL3–5-C subfractions) fractions, with an increase of the total anti-atherogenic HDL-C fraction (HDL2-C subfraction). Atorvastatin treatment of lipemia was followed by a decrease in the total LP-C, total LDL-C (LDL1–3-C subfraction), and the LDL1–3-TG subfraction. Additionally, atorvastatin treatment resulted in an increase in the serum matrix metalloproteases activity both in control and P-407-treated mice. In general, high-dose atorvastatin therapy exerts its lipid-lowering and pleiotropic effects in the early stages of acute lipemia induced in mice by treatment with P-407.

Abstract Moringa oleifera (MO) is a widely used as the nutritious and non-traditional feed supplementation containing kinds of bioactive substances. However, the enhancement effect of Moringa oleifera leaf Polysaccharide (MOLP) as a feed additive in broilers growth performance and immunity remains unclear. In this study, MOLP was obtained by water extraction and alcohol precipitation method, then purified with Trichloroacetic acid (TCA) assay. Chickens were randomly divided into 4 groups, to receive different doses of MOLP (0, 0.1, 0.2, 0.4g/kg) in feed for 3 weeks. The body weight gain (BWG) and feed consumption were recorded for feed conversion ratio (FCR) and average daily feed intake (ADFI) calculation. Broiler chickens were sacrificed and sampled on day 14, 21, 28 (D 14, D 21, and D 28) respectively. Serological indicators, including total protein (TP), albumin (ALB), globulin (GLO), and creatinine (CREA) were detected. ELISA kits were applied for detecting the levels of immunoglobulin A (IgA), immunoglobulin G (IgG), interleukin-2 (IL-2), and tumor necrosis factor (TNF-α). From D 21 to D 28, the results showed that middle dose of MOLP significantly increased BWG and ADFI as well as liver and bursa indexes when compared with the control group. In addition, TP and GLO were also increased (P<0.05). All MOLP treatments enhanced the serum concentrations of IgG and IL-2 (P<0.01). Furthermore, results of quantitative RT-PCR showed that high dose of MOLP treatment significantly increased (P<0.001) the mRNA expression levels of IL-2 and TNF-α of chickens relative to the control group. In conclusion, the results showed that MOLP supplementation contributed to improve growth performance and immune response in broiler chickens, and MOLP could be considered as a promising feed additive.
