Abstract Chicken meat is a source of animal protein that is very popular with all people, especially in Indonesia. The meat also contains nutrients that are good for the body. However, broiler meat also has a weakness because it is a food that is easily damaged; most of the damage is caused by poor handling, thus providing opportunities for growth microbes. Broiler meat is easily contaminated by various microorganisms that are in the environment. This study aimed to determine the level of microbial contamination in broiler chicken meat in the Ambon city market. The study used a random sampling technique and used 12 samples from the market in Ambon city. Parameters of microbiologic status observed total plate count and E. coli. Data were described descriptively. The result showed that total plate count (TPC) and E-coli was not more than the standard. The microbial contamination of broiler chicken meat markets in Ambon city was not in excess with the National Standardization Agency (NSA, 2009). Thus, broiler meat is safe for consumption.

Big Eye Tuna (Thunnus obesus Lowe, 1839) is one of the fish species that can increase sources of animal protein and has high economic value in the world of trade because it is the second-largest export commodity after shrimp. The purpose of this study was to test the content of microbial and formalin contamination in the flesh of T. obesus fish from the Fish Auction Place (TPI) and Mobile Fish Trader (PIK) in Panimbang Village, Pandeglang, Banten. The research was carried out at the Regional Technical Implementation Unit (UPTD) Testing and Application of Quality of Fishery Products, Department of Marine Affairs, and Fisheries of Banten Province. This research is a descriptive laboratory study with purposive sampling. Twelve samples of T. obesus fish obtained from TPI (6 fishes) and PIK (6 fishes) were taken for 25 g of meat. The tested for microbial contamination content with Total Plate Count (TPC) using Butterfield's phosphate (BFP) media, and Plate Count Agar (PCA), Coliform-Test, and E. coli-Test using Lauryl Tryptose Broth (LTB). Brilliant Green Lactose Bile (BGLB), EC Broth and Levine's Eosin Methylene Blue (LEMB), and Formaldehyde-Test using  Formaldehyde-Test Kits. The results showed that the flesh of T. obesus fish contained microbial contamination with the average values ​​of TPC, Coliform MPN, and E. coli MPN, respectively, namely 1.6 103 colony/g, 15.2 MPN/g and < 3 MPN/g ( TPI), and 1,7103 colony/g, 61.3 MPN/g and < 3 MPN/g (PIK). Therefore, fish in TPI and PIK are safe for consumption as stipulated in SNI.

Abstract This study aims to determine the effect of the microbiological quality of broiler chicken meat that is marinated using pineapple extracts with different storage times at refrigerator temperature (180C). The stages of this research consisted of 2 stages, namely the first stage of making pineapple extract from fresh pineapples and the second stage was the marination process in which the broiler chicken meat samples were marinated using pineapple extract with a concentration of 30%. The experimental design used in this study was a completely randomized design (CRD) with one treatment factor (0 days, 3 days, 6 days, 9 days and 12 days) with each treatment repeated 4 times, in order to obtain 5 x experimental units. 4 = 20 experimental units. The microbiological analysis observed was the inhibition zone analysis and Total Plate Count (TPC). Giving marination with pineapple extract to the storage time of chicken meat has a significant effect on the inhibition zone. The highest zone of inhibition was 3.23 mm (for 6 days) while the lowest zone of inhibition was 2.21 mm (for 0 days). Provision of pineapple extract marination on the storage time of broiler chicken has a significant effect on the TPC. The highest TPC was 2.29 (for 12 days) while the lowest TPC was 0.30 (for 0 days). Keywords: Broiler chicken; Marination; Microbiological quality; Pineapple extract.   Abstrak Penelitian ini bertujuan untuk mengetahui pengaruh kualitas mikrobiologi daging ayam broiler yang dimarinasi menggunakan ekstrak buah nenas dengan lama penyimpanan yang berbeda pada suhu refrigerator (180C). Tahapan penelitian ini terdiri dari 2 tahapan yaitu tahap pertama pembuatan ekstrak buah nenas yang berasal dari buah nenas segar dan tahap kedua adalah proses marinasi dimana sampel daging ayam broiler dimarinasi dengan menggunakan ekstrak buah nenas dengan konsentrasi 30%. Rancangan percobaan yang digunakan dalam penelitian ini adalah Rancangan Acak Lengkap (RAL) dengan satu faktor perlakuan (0 hari, 3 hari, 6 hari, 9 hari dan 12 hari) dengan masing-masing perlakuan diulang sebanyak 4 kali, sehingga diperoleh unit percobaan 5 x 4 = 20 unit percobaan. Analisis mikrobiologi yang diamati adalah analisis zona hambat dan Total Plate Count (TPC). Pemberian marinasi dengan ektrak buah nenas terhadap lama penyimpanan daging ayam berpengaruh nyata terhadap zona hambat. Zona hambat tertinggi 3,23 mm (selama 6 hari) sedangkan zona hambat terendah 2,21 mm (selama 0 hari). Pemberian marinasi ekstrak buah nenas terhadap lama penyimpanan daging ayam broiler berpengaruh nyata terhadap TPC. TPC tertinggi 2,29 (selama 12 hari) sedangkan TPC terendah 0,30 (selama 0 hari). Vegetable salad ingredients (lettuce, tomatoes, broccoli, and cauliflower) purchased from three grocery-store deli operations were analyzed for total plate count, coliforms, yeasts, and molds. The temperature of the vegetable ingredients was measured at the time of purchase and the pH was measured on all samples within one-half hour after purchase. In the second phase, fresh broccoli was processed into florets, inoculated with E. coli ATCC 23742, and subjected to three washing treatments. The temperature of the salad ingredients ranged from 5.1°C to 18.9°C. The pH ranges for the vegetables were broccoli, 5.46 to 6.39; cauliflower, 5.82 to 6.65; lettuce, 4.92 to 6.38; and tomatoes, 3.30 to 4.47. The total aerobic count for the vegetables ranged from 5.51 to 6.63 log CFU/g. Coliforms on the vegetables ranged from 4.89 to 6.30 log CFU/g. Yeasts and molds were found on all vegetables. The results of the study indicate that the temperature conditions and pH ranges for the broccoli, cauliflower, and lettuce could support microbial growth. The pH range of the tomatoes was below 4.6, but if contaminated and added to low acid foods, the tomatoes may also act as a vehicle for microbial contamination. When a chlorine wash solution was used, it slightly reduced the aerobic microbial load on previously inoculated broccoli and reduced the coliform population of the broccoli by approximately one log unit.

Chicken meat is one of the foodstuffs that have high nutritional value because it contains carbohydrates, proteins, fats, minerals and other substances that are beneficial to the body. Chicken meat is very high risk in terms of bacterial contamination, this study aims to determine the treatment of soaking native chicken meat in soursop leaf infusion on chemical and microbiological quality. This method uses a completely randomized design (CRD). The data obtained were analyzed statistically using Analysis Of Variance (ANOVA). If there is a significant significant difference between treatments, the test is continued with Duncan Multiple Range (DMRT). Parameters observed in this study include phenol test, total plate count test, and bacterial inhibition zone test. The results showed the chemical and microbiological quality of free-range chicken meat by soaking soursop leaf infusion for one hour and different storage times, namely the difference was not significant (P<0.1) in the phenol test, the difference was very significant (P<0.01) in the total test. plate count, and non-significant (P>0.1) in the bacterial inhibition zone test. The results showed a very significant difference in the total plate count test due to contamination during research or storage.

Escherichia coli (E. coli) is a gram-negative bacteria that can reduce meat quality and cause diarrhea if the amount is too much. This study investigates the presence of E. coli contamination in chicken meat sold in traditional markets in Medan. This research was conducted using the purposive sampling method and collected samples from 30% of each nine traditional markets with 32 samples. The traditional markets in Medan, namely Petisah, Central, Sei Kambing, Kampung Lalang, Kwala Bekala, Helvetia, Kampung Baru, Kemiri and Padang Bulan. Research was conducted by using the CFU / ml method with chromocult media. Samples were analyzed in the laboratory of the Regional Veterinary Disease Investigation Center Region 1 Medan. The results showed 32 samples of chicken meat had exceeded the maximum limit of microbial contamination. The highest total microbial average was in the Kampung Lalang market, 3.49 Log CFU / g, and the lowest was in the Kemiri market of 2.79 Log CFU / g. Based on the research results, it was found that the meat in the traditional markets of Medan was all contaminated with E. coli and had passed the maximum limit of bacterial contamination.

Chicken meat is one of the foods favored by the public because it has high nutritional content, delicious taste, affordable prices, and a soft fiber texture that makes it easy to digest. The high nutritional content can make chicken meat a good medium for the growth and development of microorganisms.The research was conducted to determine the amount of bacterial contamination in broiler meat sold in several markets in Ambon city. This study used an experimental method, namely the analysis of microbial contamination in the famous chicken meat sample in Ambon City. The research was started by taking samples of chicken breasts and thighs from various markets in Ambon city. The sample used was chicken meat that had been stored for 2 days. Furthermore, the bacterial isolation stage is carried out. The results showed that the microbial contamination in the sample of chicken breasts and thighs was still below the maximum limit (less than 1 x 106 cfu/g) determined by the National Standardization Agency. Broiler chicken meat in Ambon City is safe for consumption by the public.

Sumedang tofu is one of favorite foods for Indonesian society. But many sellers or producers are not aware to the food safety of sumedang tofu. The aims of this study were to analyze the microbial, formalin, and lead (Pb) contents in ready-to-eat sumedang tofu which were sold in traffic jams area in Cicurug, Ciawi, and Cisarua. The analysis were carried out by Total Plate Count (TPC) testing for microbial analysis, potassium permanganate reaction (KMnO4) testing for formaldehyde analysis, and atomic absorption spectrophotometry (AAS) method for lead content analysis in the samples. The results obtained for the microbial analysis were sumedang tofu that were sold in the traffic jam areas of Cicurug, Ciawi, and Cisarua contained contaminant above the treshold (1.4 x 105 colonies / gram to 2.2 x 105 colonies / gram of microbes). All of the samples of sumedang tofu were positive containing formaldehyde. Otherwise, there were no lead (Pb) content in all samples regarding to AAS analysis. This study concluded that the ready-to-eat sumedang tofu those were sold in traffic jam area in the Cicurug, Ciawi, and Cisarua were not suitable for consumption because it contained exceed microbial contamination and formalin which are harmful for human health.Keywords : formalin, microbes, sumedang tofu, lead, traffic jam

Yogurt merupakan minuman susu yang difermentasi dengan menggunakan bakteri asam laktat Lactobacillus bulgaricus dan Streptococcus thermophyllus sebagai starter. Tujuan penelitian untuk mengetahui kemampuan tertinggi dari yogurt berbasis air kelapa dalam menghambat pertumbuhan bakteri pathogen (Salmonella, Staphylococcus, Klebsiela pneumonia dan E.coli) secara in vitro. Penelitian ini menggunakan rancangan acak lengkap (RAL) dengan empat perlakuan dan empat ulangan. Keempat perlakuan tersebut: Ko: susu skim 12%+air; K1: susu skim 12%+air kelapa gading; K2: susu skim 12%+kelapa gading bulan; K3: susu skim 12%+air kelapa hijau. Peubah yang diamati adalah aktivitas antimikroba yogurt terhadap bakteri pathogen (Salmonellatyphi, Staphylococcus, Klebsiella pneumonia dan E.coli), total plate count (TPC), total bakteri asam laktat dan E,coli. Hasil penelitian menunjukkan bahwa aktivitas antimikroba tertinggi diperoleh pada yogurt berbasis air kelapa bulan terhadap bakeri Salmonella typhi dan Staphylococcus masing-masing sebesar 0,21 dan 0,26 cm dengan total bakteri asam laktat1,1×107 cfu/mL sedangkan untuk Klebsiella pneumonia dan E.coli diperoleh pada yogurt berbasis air kelapa gading sebesar 0,38 dan 0,64 mm dengan total bakteri asam laktat 3,1×106 cfu/mL.Dari hasil penelitian dapat disimpulkan bahwa yogurt berbasis air kelapa gading, bulan, hijau dan tanpa air kelapa memiliki aktivitas antimikroba berspektrum luas (menghambat bakteri gram positif dan negatif).

Dry cocoa bean quality is also determined by its microbe contamination level. Steaming process for dried cocoa beans as a pretreatment process was selected because of less effect on organic compound inside the dried cocoa bean. This experiment aim was to study microbial contamination level of cocoa beans using steaming process, determining its microbial population and evaluate its chemical changes. Experiment was carried out in Postharvest Laboratory of Indonesian Coffee and Cocoa Research Institute. Cocoa beans for the experiment were lots collected from four farms in Jayapura, Papua with different microbial contamination level for each lot. Results of this experiment showed that optimum steaming process was 15 minutes at 100 O C with 10 minutes preheating time. Microbial analysis result of the four lots after complete steaming process by total plate count method showed the same result (<3.0 x 103 cfu). Most of the decrease in microbial contaminant appeared in the plate was 73.5% of Staphylococcus aureusand 0.058% of Penicilliumsp. Bean acidity (pH) after steaming increased (4.76 to 4.80) and free fatty acid increased (1.81% to 1.96%) while carbohydrate content decreased (17.5% to 15.9%) and as well as protein content (12.6% into 11.7%).
