The phosphomolybdenum method for total antioxidant activity determination showed that the hexane, dichloromethane and ethyl acetate extracts of five marine sponge species collected off misamis oriental coast-Aaptos suberitoides, Dactylospongia elegans, Stylissa massa, Haliclonasp. and an unidentified species coded as KL-05, have varying degrees of antioxidant capacity. Expressed as ascorbic acid equivalents in μg/mL of extract, the hexane extract ofDactylospongia elegans(DeH) and the ethyl acetate extract ofAaptos suberitoides(AsE) showed the highest antioxidant capacity. Although the hexane extract of KL-05 (KL-05H) has considerable antioxidant activity, the ethyl acetate extract (KL-05E) showed no antioxidant activity. The brine shrimp assay for cytotoxicity indicated high bioactivity, withHaliclonasp.,Dactylospongia elegans, Aaptos suberitoidesandStylissa massaexhibiting high % mortality and low LC50values. The antioxidant and cytotoxic activities of the marine sponges may be attributed to the zoochemicals present. All sponge species contain alkaloids, saponins, tannins, and flavonoids. Terpenoids are present only inHaliclonasp. and the cardiac glycosides, only inAaptos suberitoidesandHaliclonasp.

Peel-Off face mask is one of the cosmetics that is used to treat skin from free radicals. Peel-off face mask can minimize the effects of free radicals because it contains antioxidant. One of the antioxidant sources is red dragon fruit’s rind (Hylocereuspolyrhizus Haw.). The aim of this study is to obtain Peel-Off face mask of red dragon fruit’s rind extract that contains anthocyanin. Red dragon fruit’s rind was macerated by ethanol 96%, ethyl acetate and n-hexane added citric acid (4:1) solvents. TLC test used Chloroform: ethyl acetate: n-butanol (5:4:1) eluent and sprayed by AlCl3. Antioxidant activity of extract was tested using DPPH (1,1-Diphenyl-2-Picryhidrazyl) method. Peel-Off face mask was made using PVA, HPMC, methyl paraben, propylparaben, propylenglycol, extract, ethanol 96% and aquadest. The rendemen of ethanol 96% extract was 9,476%, ethyl acetate extract was 0,783% and n-hexane extract was 0,631%. The results of TLC test showed yellow spots on the ethyl acetate and ethanol 96% extracts which indicated that extract contained flavonoids (anthocyanin). The results of antioxidant activity test showed that IC50 value of ethanol 96% extract was 189,7422 (AAI=0,2087), ethyl acetate extract was 196,9398 (AAI=0,2011), and n-hexane extract was 385,3664 (AAI=0,1027). The result of the product evaluations showed that all the formulas complied the product requirements such as the organoleptic, homogenity, pH, dispersive power and drying time. Peel-off face mask already meet the product requirements but further research is needed to test the product stability and activity.

Abstract. Larasati F, Batubara I, Lestari Y. 2020. The presence of endophytic actinobacteria in mangosteen peel (Garcinia mangostana L.) and its antioxidant activity. Biodiversitas 21: 1488-1497. Mangosteen (Garcinia mangostana L.) is a family member of Clusiaceae which is rich in secondary metabolite compounds that can function as antioxidants. Besides being produced by its host plant, the bioactive compounds can also be produced by endophytic actinobacteria. The purpose of this study was to explore the presence of endophytic actinobacteria from mangosteen peel and determine its antioxidant activity. The actinobacteria were isolated, purified, morphologically characterized, molecularly identified, extracted with ethyl acetate and tested for antioxidant properties. The antioxidant activity was assayed using DPPH (2.2-diphenyl-1-picrylhydrazyl) and ABTS (2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) methods. The components of extracts were separated by Thin Layer Chromatography (TLC) and bioautography was done to determine the antioxidant bands. As a result, five isolates of endophytic actinobacteria in mangosteen peel showed to have difference in aerial mycelium color, substrate mycelium color, and types of spore chains. Based on 16S rRNA gene analysis, AGM3.2 isolate showed similarity with Streptomyces griseochromogenes ATCC 14511 (T) 99.06%. AGM3.1 had similarity with Streptomyces osmaniensis OU-63 (T) 98.35%. Meanwhile, AGM2.3 were similar to Streptomyces xanthophaeus NBRC B-5414 (T) 99.82%, AGM2.2 had similarity with Streptomyces xanthophaeus NBRC B-5414 (T) 98.95%. In addition, AGM2.1 has homology with Streptomyces goshikiensis NBRC 12868 (T) 99.52%. Using both DPPH and ABTS, supernatant of AGM2.1 showed the highest antioxidant activity indicated by 36.96 and 98.80 inhibition, respectively. Antioxidant capacity of ethyl acetate extract of AGM2.1 was 22.22 μg AEAC/mg extract (DPPH) and 20.34 μg AEAC/mg extract (ABTS). Meanwhile, ethyl acetate extract of mangosteen peel had antioxidant capacity by 21.17 µg AEAC/mg extract (DPPH) and 18.75 µg AEAC/mg extract (ABTS). Antioxidant bioautographic analysis of mangosteen peel ethyl acetate extract was compared with alpha mangosteen standard. The results showed that alpha mangosteen presence in the mangosteen extract with the same Rf value of 0.64 with standard. Meanwhile, actinobacterial ethyl acetate extract from AGM3.1, AGM2.3, AGM2.2, AGM2.1 each have the same Rf value with the alpha mangosteen standard. However, the spot for alpha mangosteen had dark red color, while spots of the four actinobacterial isolates showed to have blue color indicating different antioxidant compounds. The blue spot indicates the flavone, flavanone, flavonol, and isoflavone. These compounds include a subgroup of flavonoid compounds. Ethyl acetate extract AGM3.2 does not have spot compounds with the same Rf value as the alpha mangosteen standard. Study clearly shows that endophytic actinobacteria from mangosteen peel have potency as antioxidant.

This study was conducted to determine the organic constituents and to test the antioxidant activity and toxicity of gaharu fruit pulp extract (Aquilaria malaccensis Lamk.). Gaharu fruit pulp extract was obtained through Microwave Assisted Extraction (MAE) method and then extracted with a solvent with different polarity, namely ethyl acetate and n-hexane. The obtained yield of ethyl acetate extract of the gaharu fruit pulp was 40.827% w/v, whereas 0% w/v in n-hexane extract, therefore, no further test for n-hexane extract, so that the n-hexane extract was not tested for the next stage. The results of the organic constituents of the ethyl acetate extract of gaharu fruit pulp showed flavonoids, saponins, glycosides, phenols and tannins. Antioxidant activity testing was carried out using the DPPH method with vitamin C as a positive control. The results of the antioxidant activity test showed that IC50 of ethyl acetate extract of gaharu fruit pulp and vitamin C were 143.789ppm and 13.797ppm, respectively. Extracts from microwave-assisted maceration that were partitioned with ethyl acetate solvent were categorized into moderate antioxidants because the IC50 value was between 101-150ppm, while vitamin C was categorized as a strong antioxidant because the IC50 value was between 0-100ppm. Toxicity testing was also carried out on ethyl acetate extract of gaharu fruit pulp using the BSLT method. The results of the toxicity test of the ethyl acetate extract of gaharu fruit pulp showed activity with an LC50 value of 11.282ppm. Based on this research, the ethyl acetate extract of gaharu fruit pulp is considered to have an anticancer potential.

The purpose of this study is to determine the total content of phenols, flavonoids, and condensed tannins, as well as on the antioxidant activity of the extract, and their fractions were measured by 2,2-diphenyl-1-picrylhydrazyl (DPPH), ferric reducing antioxidant power (FRAP), phosphomolybdate reduction (or total antioxidant capacity), and cyclic voltammetry (CV). The hydromethanolic extract of Salvia officinalis showed the highest values of total phenolic (176 mgGAE/g of extract) and condensed tannins (162.53 mgEC/g of extract) from the Boulemane and Khenifra regions, respectively. The results showed that the best DPPH assay was found in the ethyl acetate fraction of Salvia officinalis leaves of the Boulemane region (IC50 = 0.002 mg/ml). For the ethyl acetate and butanolic fractions of Salvia officinalis leaves, those collected from different regions have a better reducing capacity (EC50 = 0.021 mg/ml, respectively). For the total antioxidant capacity, the best activity was found in the aqueous fraction of Salvia officinalis leaves of the Boulemane region (108 mgGAE/g of extract). By the cyclic voltammetry method, hydromethanolic extract of Salvia officinalis leaves from the Boulemane region showed an important result (288.8 mgGAE/g). There was a positive correlation between total phenol content (TPC), condensed tannin content (TCT), and total antioxidant capacity (TAC) (r = 0.932, r = 0.896, respectively). The main compounds that have been identified in the hydromethanolic extract of Salvia officinalis are ascorbic acid, gallic acid, 4-hydroxybenzoic acid, tannic acid, and rutin. Due to their antioxidant property, the leaf extracts from Salvia officinalis are used as natural preservative ingredients in food and/or pharmaceutical industries.

Abstract This study aimed to compare antioxidant, antibacterial and toxicity tests of various types of kesambi tree bark extract (Schleichera oleosa (Lour) Oken). The research stages included extraction of kesambi stem skin from Mancak Serang sub-district with multilevel maceration techniques using 3 solvents: Methanol, n-hexane, and ethyl acetate. The extract was then tested for antioxidant activity, antibacterial, and toxicity. The results of antioxidant testing using the DPPH method, antibacterial with agar diffusion method, and toxicity test with BSLT method, the results showed that ethyl acetate extract had better antioxidant activity (IC50 7,723 ppm.) compared to extract of methanol (IC50 7,801 ppm), and n-hexane extract (IC50 8,568 ppm). Antibacterial activity showed the ability to inhibit the growth of Streptococcus aureus compared to Escherichia coli at a concentration of 10000 ppm, and the results of the toxicity test showed that ethyl acetate extract had better activity (LC50 305,17 ppm) than n-hexane extract (LC50 374, 96 ppm) and methanol extract (LC50 431,26 ppm).Keywords: DPPH, BSLT, Agar Diffusion, Schleichera oleosa (Lour) Oken

This study aimed to investigate phytochemical screening and antioxidant activity of n–hexane, ethyl acetate and ethanol extract from lakoocha leaves. The powdered simplicia was macerated with n–hexane, ethyl acetate and ethanol 96% successively, filtered, then concentrated using rotary evaporator to obtain n–hexane extract, ethyl acetate extract and ethanol extract. Phytochemical screening and antioxidant activity was performed against these extracts. Antioxidant activity was determined by DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging method using ultraviolet-visible spectrophotometer at wavelength of 516 nm after incubated for 60 minutes in dark place. Quercetin was used as positive control. The result of phytochemical screening showed n-hexane extract contains steroid, ethyl acetate extract contain steroid, tannin, glycoside, flavonoid and saponin, whereas ethanol extract contain tannin, glycoside, flavonoid and saponin. The IC50 value of n–hexane, ethyl acetate and ethanol extract was 1062.03±1.42 ppm, 323.18±0.02 ppm and 99.23±0.07 ppm respectively, whereas for quercetin was 2.32±0.01 ppm. This study showed that ethanol extract had antioxidant activity with strong category whereas n-hexane extract and ethyl acetate extract had inactive antioxidant activity with very weak categories.       Keyword: Antioxidant Activity, DPPH, Lakoocha leaf

Sea holly leaves (Acanthus ilicifolius L.) belongs to Acanthaceae family, contain flavonoid compounds, alkaloids, an phenols. This research aimed to determine the antioxidant activity of sea holly leaf extract by free radical damping method 1,1-Diphenyl-2-Picrylhydrazil (DPPH). The extraction method multilevel maseration using n-hexane extract, ethyl acetate extract, and ethanol extract is 1,55%, 0,65% and 4,97% respectively. On each extract, the antioxidant activity was assayed by DPPH free radical inhibition method by measuring is absorbance at the maximum wavelength of 515nm using UV-VIS spectrophometer with quercetin compound as comparator. The result of antioxidant assay showed that IC50 value, ethanol extract is 34,659 μg/mL (strong antioxidant), ethyl acetate extract is 162,512 μg/mL (weak antioxidant), n-hexane extract is361,730 μg/mL (not active as antioxidant).

Bali has around 50,000 usadha palm oil which is a source of untreated herbal medicine. One of the plants that have the potential as an antioxidant and is traditionally used by the community is Dewandaru (Eugenia uniflora L.) plant. Dewandaru containing tannins, flavonoids, and anthocyanins. Dewandaru plants have antibacterial activity, antioxidants, and antidotes to free radicals.  Making extract with six different solvents, among them methanol, ethyl acetate, n-butanol, chloroform, and n-hexane. The measurements were taken of the reduction of free radical activity by observing the absorbance at the maximum wavelength with a UV-Vis spectrophotometer alternately on the six samples. Secondary metabolites contained in Dewandaru leaves, namely flavonoids, tannins, and quinones. The value obtained is IC50 methanol extract 5,857 ppm; n-butanol extract 8,893 ppm; ethyl acetate extract 15,203 ppm; n-hexane extract 162.7315 ppm; and chloroform extract 75,873 ppm. Methanol extract, n-butanol, and ethyl acetate have very strong antioxidant activity because <50 ppm; n-hexane extract has weak antioxidant activity (ranging from 150-200 ppm), and chloroform extract has strong antioxidant activity (ranging from 50-100 ppm). Major conclusions: Dewandaru leaves has the potential as a natural antioxidant in the treatment section of Usadha Bali.

Sargassum sp. adalah salah satu jenis rumput laut paling banyak di Indonesia dan memiliki manfaat sebagai antioksidan, obat penyakit jantung, stroke, dll. Penelitian ini bertujuan untuk menentukan kandungan antioksidan fraksi etil asetat dari ekstrak metanol Sargassum sp. Sampel diambil dari Pantai Sundak, Gunung Kidul, Yogyakarta. Metode yang digunakan adalah eksploratif diskriptif. Sampel dikeringkan dalam suhu ruangan selama 7 hari dan dimaserasi dengan pelarut metanol, lalu dievaporasi dengan rotary evaporator. Ekstrak metanol di fraksinasi menggunakan etil asetat. Pengujian aktivitas antioksidan dilakukan dengan menggunakan 2 metode, yaitu metode penangkapan radikal bebas DPPH dan total kapasitas antioksidan fosfomolibdat. Vitamin C digunakan digunakan sebagai standar penangkapan radikal bebas DPPH dan total kapasitas antioksidan fosfomolibdat. Kadar total fenolat diuji menggunakan larutan Folin-Ciocalteu dengan standar asam galat. Hasil penelitian menunjukkan bahwa kadar total fenolat fraksi etil asetat Sargassum sp. sebesar 64,42 mg GAE/g sampel. Aktivitas antioksidan dengan metode penangkapan radikal bebas DPPH memiliki nilai IC50 1.289 ppm, vitamin C memiliki IC50 sebesar 122,71 ppm, sedangkan total kapasitas antioksidan adalah 39,52 mg AAE/g sampel. Kesimpulannya, yaitu kandungan aktivitas antioksidan fraksi etil asetat dari ekstrak metanol Sargassum sp. dengan metode penangkapan radikal bebas DPPH diduga sangat lemah dan kandungan aktivitas antioksidan dengan menggunakan metode fosfomolibdat diduga tinggi.  Sargassum sp. is one of the many types of seaweed in Indonesia and has benefits as an antioxidant, a drug for heart disease, stroke, etc. This study aims to determine the antioxidant content of ethyl acetate fraction of methanol extract of Sargassum sp. Samples were taken from Sundak, Gunung Kidul, Yogyakarta. The method used is descriptive explorative. Samples were dried at room temperature for 7 days and macerated with methanol, then evaporated at a rotary evaporator. The methanol extract fractionation using ethyl acetate. Testing the antioxidant activity using two methods, the method of catching free radicals DPPH and total antioxidant capacity fosfomolibdat. Vitamin C is used as a standard arrest DPPH free radical and total antioxidant capacity fosfomolibdat. Levels of total phenolics were tested using the Folin-Ciocalteu solution with gallic acid standard. The results showed that levels of total phenolic fraction of ethyl acetate Sargassum sp. amounting to 64,42 mg GAE/g sample. The antioxidant activity with catching free radicals DPPH methods have IC50 values 1.289 ppm, vitamin C has an IC50 of 122,71 ppm, while the total antioxidant capacity was 39,52 mg AAE/g sample. In conclusion, the content of the antioxidant activity of ethyl acetate fraction of methanol extract of Sargassum sp. with catching free radicals DPPH methods allegedly extremely weak and the content of the antioxidant activity using methods fosfomolibdat predictably high.

Acmella uliginosa (Sw.) Cass is widely used in traditional medicine and used in the treatment of many diseases. Extraction of component in leaves of Acmella uliginosa used methanol, ethyl acetate and n-hexane solvent. The percentage yield of extract from the leaves in methanol, ethyl acetate and n-hexane were 6.50, 0.49, and 0.66, respectively. Analysis of component in various extracts of Acmella uliginosa (Sw.) Cass leaves used Gas Chromatography-Mass Spectrometry (GC-MS). The various classes of phytochemicals were identified from fractions of methanolic leaves extract by GC-MS. Qualitative analysis of phytochemical constituents in methanol, ethyl acetate and n-hexane extract were tannins, flavonoids, saponin, alkaloid and steroid. Antioxidant activity of Acmella uliginosa leaves from Indonesia was carried out by using 1,1,diphenyl-2-picryl-hydrazine (DPPH) free radical scavenging assay. The IC50 of three extract was calculated. A comparative study determined that Acmella uliginosa in ethyl acetate extract showed the highest antioxidant potential (IC50 = 28.09 µg/mL) compared to methanol extract (IC50 = 44.31 µg/mL) and n-hexane extract (181.23 µg/mL) against DPPH free radicals.
