The effect of dynamic compressive stimulation on MG-63 cell proliferation on an auxetic PLGA scaffold was investigated. The estimated Poisson ratio of the prepared auxetic scaffold specimens was approximately (−)0.07, while the Poisson ratio estimated for conventional scaffold specimens was (+)0.12 under 10% strain compression on average. Three stimulus groups were examined: control (no stimulation), static compression, and dynamic compression. In preparation for proliferation testing, cells were seeded at 2.2 × 105 cells/80 μL on each scaffold specimen. The average proliferation rates of the static and dynamic groups were higher than those of the control group: 13.4% and 25.5% higher at culture day 1, 34.7% and 56.2% at culture day 3, and 17.5% and 43.0% at culture day 5, respectively. The static and dynamic group results at culture day 5 were significantly different (p<0.01). Moreover, proliferation rate of the dynamic stimulation group was 1.22 times higher than that of the static group (p<0.01). Conclusively, proliferation of osteoblast-like cells was enhanced through compressive stimulation, but the enhancement was maximal with dynamic compressive stimulation of auxetic scaffolds.

Objectives: This study aimed to investigate the effect of unilateral and bilateral electrical stimulation of the brain on balance in the elderly. Methods & Materials: Thirty-six elderly in Ilam City, Iran, participated in the study. In the pretest, the static balance was taken, and then the participants were randomly divided into three groups: unilateral brain stimulation, bilateral brain stimulation, and sham. The intervention took in 3 sessions. In the unilateral stimulation group, the anode electrode was positioned at the O point, and the cathode was above the left ophthalmic cavity (FP1). In the bilateral stimulation group, the anode electrode was positioned at O1, and the cathode was positioned at O2. The intensity of stimulation was 2 mA, and the duration of treatment was 15 minutes per session. In the control group, the anode and cathode electrodes were placed on the O and FP1 points, respectively, but the excitation current was stopped after 30 seconds. After the last training session, the posttest was performed. The obtained data were analyzed using 1-way ANOVA. Results: The Results showed no statistically significant difference between the three groups in the pretest phase (P<0.535). But at posttest, there was a difference between groups (P<0.002). Post-hoc test Results showed a difference between unilateral stimulation with the control group (P=0.001) and bilateral stimulation with the control group (P=0.005). But there was no significant difference between the unilateral stimulation group and the bilateral stimulation group (P=0.599). Conclusion: Unilaterally and bilaterally, cerebellar stimulation can be improved in the elderly.

Several researchers have developed finite element analysis (FEA) models on dental implants; however, few have used it to investigate the effects of nanosurface treatment on the cell-implant interface. For cell proliferation and differentiation are believed related to the physical stimulations of modification surfaces. In the present study, the cell adhesion affected by mechanical stimulation from substrate was evaluated by culturing the MG-63 osteoblast-like cells on Ti materials with different chemical composition using computer-aided programs. Three types of surfaces were made by mechanically abraded, anodized and anodized following with hydrothermal treatment on Ti substrates. The cell adhesion behavior was promoted because the physical stimulation of specific surface topography provided cells mechanical external forces. The finite element analysis (FEA) was used to investigate the deformations stresses which cells were suffered on these 3 different modifications surfaces. Results indicated that cells in the HTT group were received larger stimulation of deformations and stresses than the control group (P > 0.01). This confirmed that the superior cell proliferation and differentiation resulted from the specific roughly surfaces of HYT surface for implants.

Summary. The disbalanced feeding and the uneven distribution of micro- and macroelements in the environment leads to a trace element, in particular hypomelanosis. To accelerate the growth and preservation of young farm animals include in the diet of various biological additives and drugs, which include selenium. For stimulation of weight gain in the livestock industry, as well as for the prevention and treatment of pathological processes in addition to micro - and macrouse amino acids, primarily methionine. The aim of this work was to study the influence of composition of DAFS-25+Polizon on morpho-biochemical parameters of blood and functional state of the liver in fattening bulls of black-motley breed in the conditions of the Republic of Bashkortostan. Experiments using were conducted on bull-calves of black-motley breed of the properties in the properties age from 6 to 15 months. The first experimental group during the experiment was additionally given the composition of DAFS-25+Polizon at a dose of 2 mg/kg, the animals of the control group received a standard diet. To assess the impact of the composition DAFS-25+Polizon on metabolism cattle studied morphological and biochemical indicators of blood and conducted histological examination of the liver. It is established that the use of the composition of DAFS-25+Polizon at a dose of 2 mg/kg increases the number of erythrocytes and hemoglobin in the experimental group and reduces the amount of white blood cells. The serum content of total protein, phosphorus and calcium increases in the group of experimental animals. Microscopic examination of the liver revealed no changes in the structure of the organ and hepatocytes in the experimental group, whereas in the control group hemodynamic disorders and dystrophic changes in liver cells were observed. Thus, the use of the composition DAFS-25+Polizon at a dose of 2 mg/kg of live weight in fattening bulls black-and-white breed contributes to the increase of redox processes in the body, stimulation of metabolism, prevent the development of liver disorders of cellular mechanisms of metabolism, optimizes the structure of the liver, which generally provides higher productivity.

This study is aimed at analyzing the anticancer properties of papaya leaf extract, specifically the inhibition of cell proliferation and apoptotic induction through nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) and p53 pathways. Twenty-five mice (Mus musculus), aged 2 months and weighing 20–30 g, was injected with 0.5 mg dexamethasone for 7 days. The mice were then injected intracutaneously with 1 ml of HeLa cells (8 × 106 HeLa cells/microliter). The mice were divided into five groups (5 each): negative control (P1) (5% CMC-Na, sodium carboxymethyl cellulose), treatment II (225 mg/kg BW (body weight) papaya leaves methanol extract), treatment III (450 mg/kg BW), treatment IV (750 mg/kg BW), and treatment PV (2 mg alcohol anticancer drug). Papaya leaf extract treatments were applied for 2 weeks. Then, the tumor tissue was isolated for hematoxylin and eosin staining. Immunohistochemical imaging was used to detect Ki-67, caspase-3, NF-κB, and p53 expression. Further analysis was undertaken using the ImmunoRatio software program. The results indicated that administration of papaya leaf methanol extract significantly increased the expression of NF-κB and p53 at a dose of 450 mg/kg BW. Our results also showed that the mice treated with 450 mg of papaya leaf extract per kg of BW (P3) had the largest increase of caspase-3 expression compared to the negative control group. Papaya leaf ethanol extract decreased the cancer cell proliferation index and increased apoptosis of cancer cells in animal models of cervical cancer; it may also work to increase NF-kB expression and expression of the p53 gene.

The research aimed at investigating the effect of giving counterpressure technique to decrease the intensity of pain in the first stage of labor in the delivery room of RSUD Sawerigading Palopo. This study was conducted in a preexperimental manner with static group comparison design, in subject static group comparison design divided into two groups, where the treatment group was followed by observation and observation result compared with the observation result in the control group which only received standard treatment. Data were analyzed using T test. Where t value hit 6.971> t table 2.05, so it can be concluded that the results obtained mean that there is influence of counterpressure technique is big enough to decrease the intensity of labor pain. The method to define strain characteristics of soil under dynamic loading is proposed based on the results of experiments on dynamic compression of soils on the device for dynamic loading in laboratory conditions; the method allows solving wave problems with the statement similar to the statement of experiments. Using the proposed method, the modulus of dynamic and static compression, the modulus of unloading, the coefficient of viscosity of loess soil in the range of seismic loads are determined in accordance with elastic-visco-plastic model of soil developed by G.M.Lyakhov.

Background: Esophageal carcinoma is one of the common malignant tumors in digestive tract. BECLIN-1 is a key gene that regulates autophagy, and its abnormal expression may be related with many human tumors. However, the mechanism of BECLIN-1 in esophageal carcinoma remains unknown. Objective: In this study, we explored the effect of BECLIN-1 overexpression on tumor growth in mice with esophageal carcinoma and its mechanism. Methods: Recombined lentiviral vector containing BECLIN-1 was used to transfect human esophageal carcinoma Eca109 cells and establish stable cell line. qRT-PCR was used to detect BECLIN-1 mRNA level in the transfected Eca109 cells, CCK-8 assay was used to detect cell proliferation. Beclin-1, P62 and LC3-II protein expression levels in Eca109 cells were detected using Western blot analysis. Subcutaneous xenograft nude mice model of human esophageal carcinoma was established, and the tumor growths in Beclin-1 group, control group and empty vector group were monitored. Beclin-1 protein expression in vivo was detected by immunohistochemistry. Results: Beclin-1 mRNA and protein were overexpressed in Eca109 cells. Compared with empty vector group, the growth rate of cells transfected with BECLIN-1 decreased significantly. Compared with the control group and empty vector group, the expression level of P62 protein in beclin-1 group was significantly decreased, while the expression level of LC3-II protein was significantly increased. The tumor growth rate in nude mice of Beclin-1 group was significantly lower than that of the control group and empty vector group, and Beclin-1 protein was mainly expressed in Beclin-1 group in vivo. Conclusion: BECLIN-1 can induce autophagy in esophageal carcinoma Eca109 cells, and it can significantly inhibit the growth of esophageal carcinoma.

✓ The modulation effects of hexamethylene bisacetamide (HMBA), a differentiation-inducing agent, on growth and differentiation of cells from human malignant glioma cell line SHG-44 were studied. At cytostatic doses (2.5 mM, 5 mM, 7.5 mM, and 10 mM for 15 days), HMBA exerted a marked inhibitory effect on cell proliferation. Exposure to HMBA (5 mM and 10 mM for 12 days) also resulted in an accumulation of cells in G0/G1 phase and a decrease of cells in S phase as analyzed by flow cytometry. The reversible effects of 7.5 mM HMBA and 10 mM HMBA on cell proliferation and 10 mM HMBA on disruption of cell cycle distribution were observed when HMBA was removed from culture media on Day 6 and replaced with HMBA-free media. Colony-forming efficiency (CFE) in soft agar was remarkably decreased by HMBA (2.5 mM, 5 mM, 7.5 mM, and 10 mM for 14 days), and in 7.5 mM HMBA— and 10 mM HMBA—treated cells, the CFEs were reduced to 25% and 12.5%, respectively, of that in untreated cells. Cells treated with HMBA (5 mM and 10 mM for 15 days) remained tumorigenic in athymic nude mice, but the growth rates of the xenografts were much slower than those in the control group. The effects of HMBA on cell proliferation, cell cycle distribution, CFE, and growth of xenografts were dose dependent. A more mature phenotype was confirmed by the morphological changes from spindle shape to large polygonal stellate shape and remarkably elevated expression of glial fibrillary acidic protein in cells exposed to HMBA (5 mM, 10 mM for 15 days). Our results showed that a more differentiated phenotype with marked growth arrest was induced in SHG-44 cells by HMBA.

Loop ileostomy closure after colorectal surgery is often associated with Postoperative ileus, with an incidence between 13–20%. The aim of this study is to evaluate the efficacy and safety of preoperative stimulation of the efferent loop with probiotics prior to ileostomy closure in patients operated on for colorectal carcinoma. For this, a prospective, randomized, double-blind, controlled study is designed. All patients who underwent surgery for colorectal carcinoma with loop ileostomy were included. Randomized and divided into two groups, 34 cases and 35 controls were included in the study. Postoperative ileus, the need for nasogastric tube insertion, the time required to begin tolerating a diet, restoration of bowel function, and duration of hospital stay were evaluated. The incidence of Postoperative ileus was similar in both groups, 9/34 patients stimulated with probiotics and 10/35 in the control group (CG) with a p = 0.192. The comparative analysis showed a direct relationship between Postoperative ileus after oncological surgery and Postoperative ileus after reconstruction surgery, independently of stimulation. Postoperative ileus after closure ileostomy is independent of stimulation of the ileostomy with probiotics through the efferent loop. There seem to be a relationship between Postoperative ileus after reconstruction and the previous existence of Postoperative ileus after colorectal cancer surgery.

Background and Objectives: Chronic rhinosinusitis (CRS) is a condition that affects as much as 10.9% of the population and, along with presence of nasal polyps, is associated with significant morbidity and decreased quality of life. Studies on molecular pathways that have been activated in nasal polyp tissue are mainly based on cytokine concentration detection. Therefore, our aim is to investigate the complex appearance, relative distribution and interlinks of IL-1, IL-4, IL-6, IL-7, IL-8, IL-10, IL-12 and Ki 67 in chronic rhinosinusitis with nasal polyps (CRSwNP) affected human nasal mucosa. Materials and Methods: Samples of nasal polyps were obtained from 12 patients with previously diagnosed CRSwNP and no prior surgery. Control group consisted of samples from 17 otherwise healthy individuals with isolated nasal septum deviation. Tissues were stained for IL-1, IL-4, IL-6, IL-7, IL-8, IL-10, IL-12 and Ki67 immunohistochemically. Non-parametric statistic, Mann–Whitney U test and Spearman’s rank correlation coefficient were used. Results: All factors, except connective tissue cytokine IL-10 and proliferation marker Ki-67, had increased presence in connective tissue and decreased presence in epithelium of nasal polyps when compared to controls. Very strong and strong positive correlations between factors were observed. Conclusions: Decreased appearance of IL-1α, IL-4, IL-6, IL-7, IL-8, IL-10, IL-12 positive structures in the nasal epithelium with selective increase of IL-1α and IL-12 in nasal subepithelial connective tissue characterize the cytokine endotype with dysfunctional epithelial barrier and local stimulation of immune response in the connective tissue in case of chronic rhinosinusitis with polyps. Decrease of IL-6 in both—epithelium and connective tissue with strong correlation between it and IL-7 and IL-10 in connective tissue suggests significant stimulation of this regulatory cytokine and, possibly, the important role in pathogenesis of the development in nasal polyps. Correlations between Ki67 and cytokines indicate possible involvement of IL-4, IL-7 and IL-12 in regulation of cellular proliferation.
