CONDITION OF MORPHOLOGICAL CHANGES IN THE KIDNEYS ON THE 1ST DAY UNDER POLYTRAUMA-Histological examination of kidney tissue of first experimental group on the 1st day of the experiment, we observed that the cortical layer of the kidney glomerular layer was presented. Glomeruli were expanded, full- blooded but perivascular edema was observed only in animals with severe trauma combined with burn and wound scalp. On the other injured animals of the experimental group excretory tubules majority were expanded, but their lumen was weakly visualized. In the loop of henle and straight tubules we observed epithelial edema, which dramatically narrowed their lumen. In the experimental group 3 animals in the distal tubules occurred hidropichna dystrophy protein, cytoplasmic eosinophilia and little in the damaged epithelium with basal membrane
Ключові слова: тяжка травма, опік, скальпована рана, нирки.
Ключевые слова: тяжелая травма, ожог, скальпированная рана, почки.
Key words: severe trauma, burns scalp wound, kidneys.

Abstract Background and Aims It is believed that septicemia and septicopyemia can lead to functional failure of many organs, including the kidneys. In this regard, it is important to study the morphological features of ultrastructural changes in kidney tissue. Method Structural changes in renal tissue of 30 patients who died as a result of sepsis were investigated by electron-microscopy. The burial was carried out at an early stage (1-6 hours after death). The sections obtained for ultrasonic examination were examined on “James 100 S ” electron microscope after contrasting with uranyl acetate and lead citrate solution. Results An ultrastructural study of the cells of the cortical and medial layers of the kidneys revealed that there are changes in extracellular contacts and cell fragmentation as a result of lysis of the cytoplasmic membrane of cells. The integrity of the basal membrane of epithelial cells of the proximal and distal tubules are preserved. However, the basal layer did not have a homogeneous structure, intersected and transparent in certain areas. Numerous vacuoles, lipid supplements and fragments of lysed intracellular proteins in the cytoplasm have been identified in the cytoplasm of cells. Most of the nucleus was in the collapse phase. The lumen of tubules is narrow and most of the microvilli of the brush border are destructed. It was revealed that in sepsis renal failure and severe destructive changes and the formation of necrosis sites in the structural elements of the nephron as a result of bacterial toxins were mainly observed in areas where bacteria were localized. Conclusion Abnormalities of podocytes, of endothelial cells, disruption of the basal membrane layers as a result of abnormal damage of organelles especially mitochondria, changes of proximal and distal tubules may be caused by severe renal failure due to sepsis.

During follicular development in the mammalian ovary, several functional changes occur in the granulosa cells in response to steroid hormones and gonadotropins (1,2). In particular, marked changes in the content of membrane-associated receptors for the gonadotropins have been observed (1).We report here scanning electron microscope observations of morphological changes that occur on the granulosa cell surface in response to the administration of estradiol, human follicle stimulating hormone (hFSH), and human chorionic gonadotropin (hCG).Immature female rats that were hypophysectcmized on day 24 of age were treated in the following manner. Group 1: control groups were injected once a day with 0.1 ml phosphate buffered saline (PBS) for 3 days; group 2: estradiol (1.5 mg/0.2 ml propylene glycol) once a day for 3 days; group 3: estradiol for 3 days followed by 2 days of hFSH (1 μg/0.1 ml) twice daily, group 4: same as in group 3; group 5: same as in group 3 with a final injection of hCG (5 IU/0.1 ml) on the fifth day.

The article presents data on the long-term effect (nine months) of a silicon compound supplied with drinking water – nonahydrate sodium metasilicate (10 mg/l in terms of silicon), on CD68-positive macrophages in the liver and spleen of laboratory rats. Changes in the morphological characteristics of this cell population were found. There was a decrease in the average cell area (in the liver of the control group of rats, the average macrophage area was 179.23±5.94 microns2, and in the group receiving silicon with drinking water – 117.04±3.35 microns2; in the spleen-136.02±3.93 microns2 and 103.44±2.8 microns2, respectively). Macrophages in the liver preparations of the experimental group of rats had a fewer processes and a darker cytoplasmic membrane. The number of macrophages in the liver per unit area was comparable, for the control group of rats it was 18.78±1.24, and for the rats that received with water with the addition of silicon – 19.41±0.75 cells. CD68+ macrophages of the red splenic pulp in laboratory rats that received silicon also underwent the following morphological changes: they were located in a denser way and had fewer processes, while the number of macrophages per unit area was 73.7±2.3 for the control group, 91.6±5.0-for the experimental group, respectively. The distance between them did not change. There was a change in the intensity of CD68 expression on the surface of the cytoplasmic membrane and in the cytoplasm of liver and spleen macrophages. These changes can be interpreted as the adaptive ability of liver and spleen macrophages to silicon introduced with drinking water. Given the heterogeneity of the macrophage population in the liver and spleen, further studies using markers for different subpopulations of macrophages are needed to clarify their role in the response of tissues to silicon supplied with drinking water.

Plasma leakage in inflammation results from intercellular gaps that form in the endothelium of venules. These gaps and related morphological changes in endothelial cells are not readily seen by light microscopy. In this study we sought to visualize such changes by using the selective binding properties of plant lectins. Acute inflammation was induced in the trachea of pathogen-free F344 rats by injecting substance P intravenously, and 1, 3, or 10 min later the vasculature was perfused with fixative followed by a biotinylated lectin. Lectin binding was localized by avidinbiotin complex-peroxidase histochemistry and viewed in tracheal whole mounts by differential-interference contrast microscopy. The binding patterns of the 20 lectins tested fell into 4 groups. Most of the lectins either bound uniformly to the endothelium of normal and inflamed venules (group 1, e.g., Lycopersicon esculentum lectin) or bound weakly or not at all to venules (group 2, e.g., Maackia amurensis I lectin). The uniform binding of group 1 lectins not only revealed the overall vascular architecture but also made visible intercellular gaps and fingerlike processes at endothelial cell borders in inflamed venules. In postcapillary venules after substance P, the fingerlike processes were present along an average of 32% of the endothelial cell perimeter at 1 min, 25% at 3 min, and 7% at 10 min, compared with a baseline value of 2%. A third group of lectins (group 3, e.g., concanavalin A) bound selectively to focal patches of inflamed venules but bound weakly to normal venules. The fourth group (group 4, e.g., Ricinus communis I lectin) bound preferentially to focal patches in inflamed venules and also bound uniformly to normal venules. The focal binding of group 3 and 4 lectins coincided with sites of plasma leakage marked by extravasation of the particulate tracer monastral blue and was associated with subendothelial components of the vessel wall. We conclude that selected lectins reveal novel features of focal sites of plasma leakage, endothelial gaps, and fingerlike processes at endothelial cell borders in inflamed venules.

The aim of our study was to investigate if green tea and bilberry have protective effect on gentamicin-induced kidney damage, when applied together, and to make a connection between their effects. GM group of rats received only gentamicin, GT group received green tea only, B group received only bilberry, whereas control (C) group received saline only. GT+GM group received green tea together with gentamicin, and B+GM group received bilberry together with gentamicin. Biochemical analysis showed significantly increased urea and creatinine levels in GM group when compared to groups that also received bilberry or green tea. Histological analysis showed complete disruption of glomerular basal membrane as well as basal membranes of both proximal and distal tubules in GM group. These destructive effects were significantly milder and limited only to proximal tubules when bilberry or green tea was applied simultaneously with gentamicin. Both green tea and bilberry protective effect on gentamicin-induced nephrotoxicity is manifested because of their strong antioxidant activity. Since they are strong antioxidants, widely distributed in nature, they can offer available and inexpensive adjuvant therapy in Gram-negative infections, which can relieve gentamicin nephrotoxicity, but will not affect its bactericidal effect.

In order to reach better production and financial results of pork production different technological ways of pig fattening are researched. The purpose of this paper is to show separate results of polyenzyme application and the results of the use of heat-treated cereals in pig fattening as well as the results of their combined application. Pigs fattened on a diet with polyenzyme preparation and heat-treated cereals, either separately or combined, were characterised by an insignificantly higher growth rate and their feed intake per kilogram of weight gain was low compared to pigs fed standard mixtures (control group). In statistical terms, the relative portion of muscle tissue in pork carcass was significantly lower in experimental group 3 fed the mixture with heat-treated cereals than in the other groups. The absolute portion of muscle tissue in pork carcass was statistically more important in experimental group 2 (45.15 kg) receiving the mixture of enzyme additive and heat-treated cereals, and the least important in experimental group 3 (40.92 kg) fed heat-treated cereals. During pig fattening the income for experimental groups was higher than for the control group because of better production results. Average values covering variable expenses show the increase only in the third experimental group in the second fattening period compared to the control group, but these differences were of no statistical importance.

Acute kidney injury is a frequent disorder that can be mimicked by the application of different nephrotoxic agents, including carbon tetrachloride (CCl4), where kidney injury marker-1 (KIM-1) has been recognized as a highly specific marker. Melatonin is one of the most powerful natural antioxidants and has numerous beneficial properties. We evaluated the nephroprotective potential of 2 melatonin treatment regimens (pre- and post-intoxication) in a CCl4-induced acute kidney injury model based on the standard serum parameters, kidney tissue antioxidative capacity, KIM-1 levels, and kidney tissue morphological changes. The two treatment regimens were found to preserve kidney function, as judged from the evaluated standard serum parameters. Only when administered after the intoxication, melatonin preserved total kidney antioxidant capacity; pre-treatment melatonin only preserved reduced glutathione levels. An increase in tissue KIM-1 level was found to be prevented by both treatment regimens, which correlated with the morphological changes seen in the kidney tissues of animals treated with melatonin and CCl4. The findings of our study are in agreement with the known actions of melatonin in relieving kidney tissue oxidative burden, but also contribute to the understanding of its action by preventing an increase in KIM-1.

Ethanol has chronobiological effects, which are associated with inhibition of melatonin synthesis and secretion and disruption of the sleep-wake cycle. Ethanol is known to cause sleep fragmentation due to frequent awakenings, prolong wakefulness, and reduce the duration of the slow sleep phase. At the same time, changes in the morphology of the pineal gland under chronic exposure to ethanol remain poorly studied. Of particular interest are changes in the basic morphometric parameters of pinealocytes, because they are a marker of the functional state of the pineal gland. The aim of the study was to morphometrically study the features of morphological changes in rat pinealocytes in the physiological norm and in chronic ethanol intoxication. To achieve this goal, we used 20 laboratory male rats: a control group and an experimental group. The control group was under standard vivarium conditions. For the experimental group, alcohol intoxication was modeled by injecting a 40% ethanol solution at the rate of 12 mg/kg of body weight intragastric 4 times a day. The morphometric parameters of pinealocytes were studied on day 30 from the start of the study. According to the results of morphometric measurements, a significant increase in the parameters of light pinealocytes and a decrease in the parameters of dark cells were established. It was determined that the average values of the cytoplasm area of light pinealocytes increase by 54.55% (p<0.05), the area of the nucleus increases by 61.32% (p<0.05), and the area of the nucleolus by 32.84% (p<0.05) compared with the control group. The area of the cytoplasm of dark pinealocytes decreases by 27.2% (p<0.05), and the area of the nucleus by 37.33% (p<0.05). Changes in the ratio of light and dark pinealocytes were also noted. An increase in the number of active light cells by 8.17% was found. The detected morphometric changes indicate high functional activity of cells, which has a compensatory nature in response to apoptosis of pinealocytes.

The research aimed to identify presence and severity of contralateral kidney tissue changes while opposite kidney inflammation progresses. Our own model was used for disease evoking in rats. The observation period lasted 10 days. Starting from the 3rd day, signs of hematogenous dissemination of infection/inflammation into contralateral kidney were registered. Starting from the 5th day urinogenous dissemination process was observed. By the 3rd day of study a cortical hyperemia in contralateral kidney was seen, which possibly was of compensatory origin. Based on research results, a primarily bilateral urinary tract inflammation/infection can be suspected.
