Genetic chemical variability of the volatiles and phenolic compounds in Clinopodium serpyllifolium subsp. fruticosum (L.) Bräuchler syn. Micromeria fruticosa (L.) Druce grown in Israel

Abstract Clinopodium serpyllifolium is a perennial medicinal plant generally used as herbal tea in folk medicine. Leaves of C. serpyllifolium from the representative exemplars in the Newe Ya’ar living germplasm collection were analyzed for their polyphenolic and volatiles composition, and tested for their antioxidant activity. The oxygenated monoterpenes pulegone (10.4–50.6%), piperitenone oxide (3.2–28.6%), piperitenone (0.9–14.6%), trans-piperitone oxide (0.3–11.2%), iso-menthol (0.3–8.8%) and sesquiterpene β-caryophyllene (7.4–13.7%) were found to be the major constituents from the solvent extraction of C. serpyllifolium analyzed by gc-ms. The representative exemplars were grouped into two chemotypes: one rich in pulegone and the second rich in piperitenone oxide and piperitenone. The total polyphenolics content, determined range from 26.6 to 62.9 mg gallic acid equivalents/g dry weight (dw). The antioxidant activity ranged from (42.8–77.1 mg chlorogenic acid equivalents/g dw). hplc analysis revealed the rosmarinic acid content also showed remarkable differences in C. serpyllifolium (0.3–1.8 dry weight %). The results indicate that there is chemical diversity and also difference in the content of polyphenols of the representative exemplars. Antioxidant activity was in correlation with phenolic components.

The consumption of herbal teas has become popular in recent years due to their attractive flavors and outstanding antioxidant properties. The Five-Golden-Flowers tea is a herbal tea consisting of five famous edible flowers. The effects of microwave-assisted extraction parameters on the antioxidant activity of Five-Golden-Flowers tea were studied by single-factor experiments, and further investigated using response surface methodology. Under the optimal parameters (53.04 mL/g of solvent/material ratio, 65.52 °C, 30.89 min, and 500 W), the ferric-reducing antioxidant power, Trolox equivalent antioxidant capacity, and total phenolic content of the herbal tea were 862.90 ± 2.44 µmol Fe2+/g dry weight (DW), 474.37 ± 1.92 µmol Trolox/g DW, and 65.50 ± 1.26 mg gallic acid equivalent (GAE)/g DW, respectively. The in vivo antioxidant activity of the herbal tea was evaluated on alcohol-induced acute liver injury in mice. The herbal tea significantly decreased the levels of aspartate aminotransferase, total bilirubin, and malonaldehyde at different doses (200, 400, and 800 mg/kg); improved the levels of liver index, serum triacylglycerol, and catalase at dose of 800 mg/kg. These results indicated its role in alleviating hepatic oxidative injury. Besides, rutin, chlorogenic acid, epicatechin, gallic acid, and p-coumaric acid were identified and quantified by high performance liquid chromatography (HPLC), which could contribute to the antioxidant activity of the herbal tea.

The family Asteraceae include large number of Centaurea species which have been applied in folk  medicine. One of the family Asteraceae members is the Centaurea damascena which authentically been tested for its antibacterial activity. The aim of the study was to discuss antibacterial activities of essential oil composition and methanolic extract of the same plant aerial part leaves.  Thirty-seven components were characterized with 86% of oxygenated terpenes. The composition was dominated by 11.45% Fokienol, 8.8% thymol, 8.21% Alpha Terpineol, 7.24% Chrysanthemumic acid, 7.13% Terpinen-4-ol and 6.59% Borneol with a high degree of polymorphism in the occurrence of these compounds as compared with the different species of centaurea.. Free radical scavenging capacity of the C. damascna methanol extract was calculated by DPPH and FRAP test. DPPH radicals were scavenged with an IC50 value of 17.08 µg /ml. Antioxidant capacities obtained by the FRAP was 51.9 and expressed in mg Trolox g-1 dry weight. The total phenolic compounds of the methanol extracts of aerial parts, as estimated by Folin–Ciocalteu reagent method, was about 460 mg GAE/ g. The phenolic contents in the extracts highly correlate with their antioxidant activity, (R2 = 0.976) confirming that the antioxidant activity of this plant extracts is considerably phenolic contents-dependent.

Purpose: To evaluate the antioxidant, anti-inflammatory and anti‐arthritic effects of the methanol extract of Tamus communis roots. Methods: The total phenolic and flavonoid contents were determined using Folin–Ciocalteu’s reagent and aluminium chloride assays, respectively while the antioxidant capacity was determined using DPPH and β-carotene/linoleate tests. For anti-inflammatory activity, carrageenan-induced paw edema in rat was used. Formaldehyde-induced paw edema and adjuvant-induced arthritis in rats were used to evaluate the anti-arthritic effect of the extract. Results: The total phenolic and flavonoid contents of T. communis showed 55.2 ± 0.036 mg GAE/g dry weight and 1.191 ± 0.002 mg QEq/g dry weight, respectively while the antioxidant activity (DPPH) showed a half-maximal inhibitory concentration (IC50 of 0.128 ± 0.011 mg/mL. Using βcarotene/linoleate assay, the extract showed 88.13 ± 4.03 % inhibition. In carrageenan-induced paw edema in rat, all doses of T. communis did not show inhibition of edema. In contrast, formaldehyde induced paw edema decreased at the dose of 150 mg/kg days 2 and 4. However, doses of 300 and 600 mg/kg did not show any activity. In adjuvant-induced arthritis, all used doses caused no reduction in arthritic scores and in paw thickness except a significant decrease with the dose 150 mg/kg at the last day. Conclusion: T. communis extract exhibits high antioxidant activity related to the phenolic compound contents. However, the results of the anti-inflammatory and anti‐arthritic studies did not support its use in folk medicine in the treatment of rheumatic ailment except on low doses

Rubus matsumuranus H. Lev. & Vaniot, a famous Siberian shrub of the Rosaceae family, is used in the folk medicine of nomads (Buryats, Yakuts, Soyots, and Mongols) as a remedy for the treatment of diseases of the respiratory and hepatobiliary systems. The lack of scientific information on R. matsumuranus leaves contributed to the investigation of the metabolomic profile and biological activity of this plant. In this study, metabolites of R. matsumuranus leaves in three stages (active growth, flowering, and fruiting) were characterised using high-performance liquid chromatography with photodiode array and electrospray ionisation triple quadrupole mass spectrometric detection (HPLC-PDA-ESI-tQ-MS). In total, 63 compounds were identified, including gallic acid derivatives, hydroxycinnamates, catechins, procyanidins, flavonols, and ellagitannins. Lambertianin C (57.11 mg/g of dry weight, DW), miquelianin (39.63 mg/g DW), and kaempferol-3-O-glucuronide (31.18 mg/g DW) were the major compounds in R. matsumuranus leaves. As a result of the HPLC-PDA-based assay to determine the antioxidant activity, it was revealed that lambertianin A, sanguiin H6, lambertianin C, and sanguiin H11 were effective scavengers of free radicals (2,2-diphenyl-1-picrylhydrazyl, DPPH•) and possessed Fe2+-chelating activity. After an investigation of the phenolic content in infusions and decoctions obtained by extraction with water at different temperatures, it was revealed that a hot infusion (80 °C) is a phenolic-rich preparation of R. matsumuranus leaves. Our research suggests that R. matsumuranus leaves are a rich source of phenolic compounds with high antioxidant properties and that this could be a prospective plant for new functional products.

Tragopogon porrifolius(Asteraceae), commonly referred to as white salsify, is an edible herb used in Lebanese folk medicine to treat cancer and liver dysfunction. In this study, we investigated the antioxidant activity ofTragopogon porrifoliusmethanolic extract, bothin vitroandin vivo, in addition to its hepatoprotective and anticancer activities. Total phenolic and flavonoid contents were measured and found to be37.0±1.40 mg GAE/g and16.6±0.42 mg QE/g dry weight, respectively.In vitroantioxidant assays revealed an FRAP value of659±13.8 µmol Fe2+/g of extract and DPPH IC50value 15.2 µg/mL. In rats subjected to CCl4-induced hepatotoxicity, significant increase in CAT, SOD, and GST levels was detected. The highest dose of the extract (250 mg/kg) recorded a fold increase of 1.68 for SOD, 2.49 for GST, and 3.2 for CAT. The extract also showed substantial decrease in AST (57%), ALT (56%), and LDH (65%) levels. Additionally, the extract caused a dose-dependent decrease in cell viability and proliferation. In conclusion, the methanolic extract ofT. porrifoliusdisplayed a relatively high antioxidant activity bothin vitroandin vivoas well as hepatoprotective potential against liver toxicity in rats and anticancer effect on MDA-MB-231 and Caco-2 cells.

Verbena officinalis Linn is a traditionally known medicinal plant which is used against a number of diseases including inflammatory conditions. In this study its antioxidant activity (reducing powers, 2, 2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activities), ferric reduction activity potential (FRAP), total flavonoid concentration and antimicrobial activities of 80%, 90%, 100% methanol and chloroform extracts of V. officinalis Linn root and 90% and100% methanol leaf extracts were determined. Its antioxidant activity increases with increase in amount of extract (10% to 40%v/v). Total flavonoid content (TFC) varied from 73.32±0.002 mgQE/100g of dry weight (90% methanol) to 42.39±0.032 mgQE/100g dry weight (chloroform), 2,2-diphenyl-1-picrylhydrazyl (DPPH), radical scavenging activity (%) was varied between 87.39% (90% methanol) to 45.57% (chloroform) while Ferric reducing antioxidant power was observed between 372.93±0.04 mgAAE/100 g extract (90% methanol) to 129.41±0.026 mgAAE/100 g chloroform in the root extract. The methanolic extract of the leaf showed less antioxidant activity than the methanolic extract of the root. Crude extracts of V. officinalis root showed various degree of antimicrobial activity towards drug resistance microbial pathogens. Growth inhibition tests against bacterial pathogens demonstrated concentration dependence. Moreover, gram positive bacteria were more susceptible to V. officinalis root extract when compared to gram negative bacteria. In general V. officinalis root and leave extracts possess strong antioxidant and antimicrobial activities.

Background: The natural antioxidants have an important role in the prevention of many diseases. The aim of study is to investigate phytochemical components, antioxidant activity and acute oral toxicity of Pomegranate (Punica granatum L.) fruit peel (PFP) extract. Materials and methods: Phytochemicals of PFP were determined by qualitative chemical tests, thin layer chromatography, total polyphenol and flavonoid contents. The PFP extract was evaluated for antioxidant activity by DPPH assay and MDA assay. In vivo acute oral toxicity test was conducted using Karber-Behrens method to determine LD50. Results: Results illustrated that PFP mainly contains flavonoids, alkaloids, tannins, triterpenes, saponins, and coumarins. PFP extract exhibited the total polyphenol and flavonoid contents with 189.97 mg gallic acid equivalent/g dry weight and 9.42 mg quercetin equivalent/g dry weight, respectively. The DPPH free radical scavenging and anti-lipid peroxidation activities of PFP extract were expressed with IC50 value of 4.80 μg/mL and 0.38 μg/ mL, sequentially. Simultaneously, the Dmax (the maximum dose administered to mice that no toxicity was observed) of PFP extract was determined to be 21.28 g/kg, equivalent to 35.64 g dried herb. Conclusion: The PFP extract is relatively safe and revealed high antioxidant activity. Background: P. mucronata (Pm) comes from South America, Brazil and is characterized as “Maracujá de Restinga”. It is used in folk medicine for its soothing properties and in treating insomnia. Objective: The present study for the first time analyzed the antioxidant and cytotoxicity of the hydroalcoholic leaves extract and fractions from Pm. Method: The cytotoxicity test will be evaluated by different assays (MTT and CV) against human prostate cancer (PC3) and mouse malignant melanoma (B16F10) cell lines, and the antioxidant test by DPPH method. Results: β-Amyrin, oleanolic acid, β-sitosterol and stigmasterol were isolated of the most active, hexane fraction. These substances were tested against the tumor cell lines: β-sitosterol and stigmasterol showed the most relevant activity to PC3 in CV assay and, oleanolic acid to B16F10 by the MTT assay. In addition, it was possible to indicate that the mode of cell death for stigmasterol, presumably is apoptosis. In terms of antioxidant activity, the hydroalcoholic leaves extract presented higher activity (EC50 133.3 µg/mL) compared to the flower (EC50 152.3 µg/mL) and fruit (EC50 207.9 µg/mL) extracts. By the HPLC-MS, it was possible to identify the presence of flavones in the leaf extract (isoschaftoside, schaftoside, isovitexin, vitexin, isoorientin, orientin). Conclusions: P. mucronata hexane fraction showed promising cytotoxic effect against cancer cell lines, and stigmasterol contributes to this activity, inducing apoptosis of these cells. Furthermore, as other Passiflora species, Pm extract showed antioxidant activity and flavones are its major phenolic compounds.

Background: One of the four most incident species in mangrove is the Laguncularia racemosa, plant widely used in popular medicine against inflammation and fever. Objective: Here, L. racemosa was investigated in relation to their phytochemical profile, antioxidant activity, citotoxicity, antimicrobial and immunostimulatory effect. Method: Aqueous extract was obtained from leaves of plant, its phytochemical profile was investigated through UPLC method, the antioxidant assays performed were TAA, DPPH, ABTS, nitrite and lipid peroxidation assay. Antimicrobial assays were made using standard strains. For all biological tests were used mice splenocytes and from these cell cultures were measured cytotoxicity, proliferation index and cytokines production. Results: Laguncularia racemosa leaves showed the presence of ions, proteins, carbohydrates, vitamins and high concentration of phenolic compounds. Antioxidant activities were promoted by aqueous extract, especially in DPPH and NO assays. Extract in 6 µg/mL did not induce significant cell death, stimulated the cell proliferation and the IL-4 production. Moreover, decreases of proinflammatory cytokines IFN-γ, TNF-α and IL-6 were found. Conclusion: The presence of essential nutrients, significant antioxidant activity and immune stimulation confirm the use of this plant in folk medicine against inflammation.
