Long W, Ansari A, Seecharran D. 2017. The effect of urea on epigeic earthworm species (Eisenia foetida). Cell Biol Dev 1: 46-50. The use of chemical fertilizers in intensive agriculture has no doubt increased crop production but it has resulted in adverse effect on soil properties over a long period of time. The effects are clearly seen in the soil environment including the soil organisms living in that ecosystem. This study was conducted to determine effects of Urea on epigeic earthworm species (Eisenia foetida) in clay soil. A total of five doses of Urea were used i.e 50 mg/kg, 100 mg/kg, 150 mg/kg, 200 mg/kg and 250mg. They were labelled as T1, T2, T3, T4, and T5, respectively. A control group was also set up and the treatment groups were replicated three times. The results indicated that the addition of urea affected the soil chemical properties in treatments inoculated with earthworms (Eisenia foetida). The adult earthworm population was 70% in control when compared to different treatment dosage of Urea 50 mg/kg (T1)-85%, 100 mg/kg (T2)-66.67%, 150 mg/kg (T3)- 68.34%, 200 mg/kg (T4)- 86.67% and 250 mg/kg (T5)- 51.67%. The percentage mortality minimum in treatment T4 and T1 whereas it was maximum in treatment T5 (48%). 250 mg/kg was the most toxic dose of Urea in the soil inoculated with earthworms. The results were significant at p=0.05.

Three synthetic growth-promoting substances were administered to cauliflower plants (Brassica oleracea) to determine their effect on flower weight and flower growth. A Youden square experimental design was employed to set up treatment groups for all possible combinations of three substances at four concentration levels and three frequencies of application, and an untreated control group. The substances employed were 3-indolylbutyric acid (IBA), 2,3,5-tri-iodobenzoic acid (TIBA), and β-naphthoxyacetic acid (NOXA); concentrations used were 0.01, 0.2, 4.0, and 80.0 parts per million; and frequencies adopted were 2, 3, and 6 times in a 12-day period. Final flower weights were analyzed by an analysis of variance which showed that the treatments differed significantly (P <.01). Treatments with NOXA and IBA produced flowers weighing significantly more than those of the control group. Flower growth was characterized by a rapid initial rise, which subsequently levelled off in the control group. For treated groups, the initial rise continued throughout the flowering period. The results suggest that the use of certain synthetic growth-promoting substances may be feasible commercially for enhanced production of cauliflower crops.

Carbon nanotubes (CNTs) are promising candidates for various applications including biomedical purposes. Owing to their remarkable physical, mechanical, electrical and chemical properties, CNTs have become an area of intense research and industrial activity in recent years. Therefore, toxicity and risk assessment studies are becoming increasingly important. The present study was designed to assess the effects of hydroxyl-functionalized multiwalled CNTs (OH-f MWCNTs) on sperm health and testes of adult Wistar rats. Animals were treated with different doses of OH-f MWCNTs (0.4, 2.0 and 10.0 mg/kg) along with a control group receiving only vehicle. Assessments after 15 alternate intraperitoneal doses revealed dose-related adverse effects on many endpoints tested. Results of the study showed significant impairment of sperm health at 2.0 and 10.0 mg/kg. Histology of testes demonstrated degeneration of germinal epithelium and loss of germ cells in the treatment groups. The exposure resulted in increased oxidative stress in testes in a dose-dependent manner. The findings of the study demonstrate that CNTs are potentially harmful for male reproductive health.

Background: The god’s crown fruits have properties as antioxidants and anti-inflammatory. Toxic doses of paracetamol can injure the liver through toxic metabolite bonds with cytoplasmic proteins that cause free radicals to form. The aim of this research is to know the effect of the crown of gods extracts on paracetamol hepatotoxicity. Method: A total of 30 of Webster swiss mice with a weight of ± 20 grams, age 3 months were randomly assigned to five groups, negative control, positive control, treatment 1,2 and 3. A 70% ethanol extract of god’s crown fruit given in doses 60mg, 120mg and 240mg per kgBB mice. The extract was administered for 14 days in the treatment groups, then on day 15 paracetamol ware administered in a given dose 300mg/kgBB for 1 day for the positive control group and treatment groups. On day 16, the serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels were examined from the orbital sinuses and animals terminated liver tissues taken and immediately fixed in 10% buffer formalin for histological examination. Results and Discussion: The 70% ethanol extract of the god’s crown fruits decreased blood serum levels of AST and ALT, and these results were supported by histopathologic scores of the liver in which histopathologic scores were improved with the increasing doses (p < 0.05). The secondary metabolite contents of the god’s crown fruit extract served as an antioxidant and anti-inflammatory, protecting hepatic injury from the toxic metabolite of paracetamol. Conclusion: A 70% ethanol extract of god’s crown fruit (Phaleria macrocarpa) have hepatoprotective properties that effectively prevent hepatic injury due to paracetamol toxic dose.

The study objective was to evaluate targeted reprogramming of peripheral blood mononuclear cells in systemic juvenile idiopathic arthritis (sJIA) modeled in 6-month-old male Wistar rats by co-administration of complete Freund’s adjuvant and lipopolysaccharide. Methods. On day 40 of the experiment, rats were divided into three groups: control, doxycycline, and dexamethasone groups. Blood samples were collected on days 0, 41, and 55. Peripheral blood mononuclear cells were isolated gravimetrically and stained for markers and cytokines. Monocytes (CD3-CD4+) and T-helpers (CD3+CD4+) were differentiated as target cells. IL-4 was considered a marker for the pro-M2 phenotype since IL-4 can activate M2 macrophage polarization; IFN-g was considered a marker for the pro-M1 phenotype. Time-related changes in the intracellular expression of IL-4 and IFN-g were studied using flow cytometry. Data were analyzed using nonparametric statistical tests (Mann-Whitney-Wilcoxon) in the R environment for statistical computing. Results and conclusions. Monocytes (like macrophages) underwent reprogramming during the development of modeled sJIA disease. In monocytes of doxycycline and dexamethasone treatment groups, pro-M2 effects were observed earlier (day 41) than in the control group. Pro-M1 effects were observed in monocytes of doxycycline and dexamethasone groups on day 55, as compared with the control group. Characteristic time-related changes of intracellular cytokine expression were described for different groups. Importantly, the differently directed phenotype polarization was observed in sJIA and treatment groups for both monocytes and T-helpers.

Background: The activation of Nrf2/HO-1 pathway has been shown to protect against cisplatin- induced nephrotoxicity by reducing oxidative stress. Berberine (Ber), an isoquinoline alkaloid, has demonstrated antioxidant, anti-inflammatory and anti-apoptotic activities in various experimental models. Aim: To check the effect of Ber on cisplatin-induced nephrotoxicity and to explore the involved mechanism. Methods: Adult male Wistar rats were divided into 6 groups: Normal, cisplatin-control, treatment groups and per se group. Normal saline and Ber (20, 40 and 80 mg/kg; p.o.) was administered to rats for 10 days. A single intraperitoneal injection of cisplatin (8 mg/kg) was injected on 7th day to induced nephrotoxicity. On 10th day, rats were sacrificed, the kidney was removed and stored for the estimation of various parameters. Results: As compared to cisplatin-control group, Ber pretreatment improved renal function system and preserved renal architecture. It also diminished oxidative stress by upregulating the expression of Nrf2/HO-1 proteins. In addition, Ber attenuated the cisplatin mediated inflammation and apoptosis. Furthermore, it also reduced the phosphorylation of p38/JNK and PARP/Beclin-1 expression in the kidney. Conclusion: Ber attenuated renal injury by activating Nrf2/HO-1 and inhibiting JNK/p38MAPKs/ PARP/Beclin-1 expression which prevented oxidative stress, inflammation, apoptosis and autophagy in renal tissue.

The present study was conducted to investigate the efficacy of Ovsynch and Ovsynch plus protocol in postpartum (day 60) Sahiwal cows (n=18). Animals were randomly divided into three equal groups, viz., Ovsynch group Ovsynch plus group and Control group. Animals of group I (n = 6) were treated with traditional Ovsynch protocol. The animals (n = 6) of group II were treated with Ovsynch plus protocol which consisted of an initial intramuscular injection of eCG (Folligon) @ 250 IU on day 60 postpartum followed 3 days later by GPG (Ovsynch) protocol. In group-III Control, no treatment was given to animals (n = 6). Treated animals were inseminated at a fixed time between 14 and 20 hrs after second GnRH injection, irrespective of estrus detection. Blood samples were collected from each animal on days 50 and 60 postpartum to determine the status of cyclicity in animals based on serum concentrations of progesterone (P4). A third blood sample was collected on the day of prostaglandin treatment to determine the response of first GnRH injection. Four animals each were cyclic, and two were acyclic in both treatment groups. Four animals each responded to first GnRH treatment in both treatment groups. Similarly, two animals each got conceived giving conception rate of 50% (2/4) in each treatment. In the control group, one out of 6 animals got conceived yielding 16.66 % conception rate (1/6) during the study period. It may be thus concluded that Ovsynch and Ovsynch plus protocol may be used during the early postpartum period to improve the reproductive efficiency in postpartum Sahiwal cows.
