The botanical family Solanaceae has many species producing compounds with insecticidal properties, e.g. nicotine and capsaicin, which are used for pest management in agriculture. This fact provides perspectives to identify insecticidal compounds in Brazilian native species of Solanaceae. In this study, we performed a screening with 25 ethanolic extracts from 17 Solanaceae species in order to evaluate their bioactivity against the Mexican bean weevil, Zabrotes subfasciatus(Coleoptera: Chrysomelidae: Bruchinae). The bioactivity of Solanaceae ethanolic extracts (2500 mg kg-1) was tested with residual contact bioassays. Adults ofZ. subfasciatus were exposed to treated bean grains, and adult mortality, oviposition, F1progeny and damages on grains were quantified. Most of the ethanolic extracts from Solanaceae reduced the number of eggs per sample, the egg-adult viability, the F1progeny and the damages on bean grains promoted byZ. subfasciatus, but none of them interfered on its sex ratio. Ethanolic extract from leaves of Solanum lycocarpumA. St.-Hil promoted the most promissory effects on Z. subfasciatus. This ethanolic extracts can be a suitable alternative to control Z. subfasciatus in stored beans, mainly for small farmers and organic farmers.

The aim of this work was to assess the antifungal and antioxidant activity of essential oils and ethanolic extracts from distilled solid by-products from aromatic plants (Artemisia dracunculus, Hyssopus officinalis, Lavandula stoechas, Origanum vulgare and Satureja montana) against 14 fungi strains isolated from sheep cheese and identified at species level using DNA barcoding based on β-tubulin sequence analysis. In addition, capacity of fungi to produce ochratoxin A, patulin, cyclopiazonic acid and sterigmatocystin was analyzed. Of the isolates, 85.7% belonged to Penicillium (P. commune/biforme, P. crustosum) and 14.3% to Aspergillus (A. puulaauensis and A. jensenii), the first time that these Aspergillus species have been found in sheep’s cheese. All P. commune isolates were producers of cyclopiazonic acid, and the two Aspergillus strains produced sterigmatocystin, but the others did not produce any tested mycotoxin. Among the essential oils tested, oregano, savory and tarragon had a significant antifungal activity against all the isolated strains, but no ethanolic extract showed antifungal activity. By contrast, ethanolic extracts showed great potential as antioxidants. The identification of new molds in cheese will help the dairy industry to know more about those molds affecting the sector, and the use of aromatic plants in the control of fungal spoilage could be a suitable alternative to chemical preservatives used in the agri-food industry.

AbstractMexican bean weevil, Zabrotes subfasciatus Boheman, is a significant pest of stored beans (Phaseolus vulgaris L.) that causes considerable loss in the quality and quantity of bean grain in Africa. In the past two to three decades, researchers have attempted to identify botanicals with better insecticidal potential against storage pests. However, there is a lack of information on the potency of botanical combinations, their toxicology, and optimal application rates. This study was conducted to examine the bioefficacy of unitary and binary botanical combinations of Jatropha curcas (L.), Datura stramonium (L.), Chenopodium ambrosioides (L.), Schinus molle (L.), and Azadirachta indica (A. Juss) against Z. subfasciatus at 1% and 2% w/w dosages. The insecticidal activities of the botanicals were measured based on their effect on adult mortality of Z. subfasciatus and progeny reduction, grain weight loss, and weevil perforation index. Among botanical admixtures, binary combinations of C. ambrosioides + D. stramonium, J. curcas + C. ambrosioides, and S. molle + C. ambrosioides were the most potent. They resulted in the highest mortality of Z. subfasciatus, low weevil perforation index or none, and grain weight loss at the most moderate test dosage (1% w/w). Treatment with C. ambrosioides resulted in the highest efficacy when applied individually. We failed to observe any notable difference in toxicity between dosage levels among the binary botanical combinations. However, the effectiveness of unitary formulations improved with the increase in dosage rate. Application of botanicals did not adversely affect bean seed germination. Our study suggests that botanicals in binary mixtures may enhance the potency of constituent components for effective control of Z. subfasciatus.

Mastitis is responsible for both damage to animal health and economic losses to the industry. To identify effective treatments for this disease, products extracted from a variety of plants with antimicrobial potential have gained attention. The present study aimed to assess the in vitro antibacterial potential of the ethanolic extract of two plant species from the Caatinga biome against bacteria isolated from small ruminants with subclinical mastitis. The leaves of Mimosa tenuiflora and Commiphora leptophloeos were dried and processed to obtain crude ethanolic extracts and their phenolic composition was evaluated. In total, 33 Staphylococcus spp. isolates from the bacterial collection of the Laboratory of Food Quality Control of IF SERTÃO-PE were used for evaluation of biofilm production. Furthermore, an antimicrobial susceptibility test was conducted using the minimal bactericidal concentration (MBC) method against the two ethanolic extracts. The toxic potential was measured through a toxicity test with Artemia salina. The quantification of the phenolic compounds revealed that the ethanolic extracts of M. tenuiflora and C. leptophloeos possessed higher amounts of myricetin (43.2 and 294.9 mg in 10 g, respectively) in relation to the other compounds. A 39.4% positivity rate was observed in the nuc gene investigation. The biofilm production analysis revealed that 96.9% of the isolates produced biofilm, evidencing the evolution the microorganisms regarding the development of resistance mechanisms. The MBC results showed an inhibition range between 195.30 and 3125.00 µg mL-1 and between 781.20 and 6250.00 µg mL-1 for the extracts of M. tenuiflora and C. leptophloeos, respectively. The M. tenuiflora extract showed the highest activity, suppressing 100% of the bacterial isolates (n=26), whereas the extract of C. leptophloeos showed an inhibition percentage of 69.23%. The crude ethanolic extract (EEB) of M. tenuiflora was found to be toxic, presenting a DL50 of 118.356 µg mL-1. In contrast, the EEB of C. leptophloeos was found to be non-toxic (DL50 = 1527.430 µg mL-1). In conclusion, both native Caatinga species presented antibacterial activity and myricetin was the major compound. These findings highlight the need for further studies regarding the identification of anti-mastitis products from natural extracts.

AbstractApplication of 5-fluorouracil (5-FU) in cholangiocarcinoma (CCA) is limited by adverse side effects and chemoresistance. Therefore, the combination therapy of 5-FU with other substances, especially natural products may provide a new strategy for CCA treatment. The aim of this study was to evaluate the combination effects of 5-FU and two ethanolic extracts of Thai noni juice (TNJ) products on CCA cell lines and nude mice xenografts. The results of antiproliferative assay showed the combination treatment of 5-FU and each TNJ ethanolic extract exerted more cytotoxicity on CCA cells than either single agent treatment. Synergistic effects of drug combinations can enable the dose reduction of 5-FU. The mechanism underlying a combination treatment was apoptosis induction through an activation of p53 and Bax proteins. In the nude mouse xenograft model, combination treatments of 5-FU with each TNJ ethanolic extract suppressed the growth of CCA cells implanted mice more than single agent treatments with no effects on mouse body weight, kidney, and spleen. Moreover, low doses of TNJ ethanolic extracts reduced the hepatotoxicity of 5-FU in nude mice. Taken together, these data suggested that the ethanolic extracts of TNJ products can enhance the anti-CCA effect and reduce toxicity of 5-FU.

Betel leaf ethanolic extract dechlorophyllized using sedimentation (BLEE-SED) exhibited enhanced antioxidant and antibacterial activities. BLEE-SED could extend the shelf-life of Nile tilapia fillets during refrigerated storage (4 °C) up to 9 days.

Uncaria tomentosa (Willd.) DC is a woody climber species originating from South and Central America that has been used in the therapy of asthma, rheumatism, hypertension, and blood purification. Our previous study showed that U. tomentosa extracts altered human erythrocyte shape, which could be due to incorporation of the compounds contained in extracts into the erythrocyte membrane. The aim of the present study was to determine how the compounds contained in U. tomentosa extracts incorporate into the human erythrocyte membrane. The study has assessed the effect of aqueous and ethanolic extracts from leaves and bark of U. tomentosa on the osmotic resistance of the human erythrocyte, the viscosity of erythrocyte interior, and the fluidity of erythrocyte plasma membrane. Human erythrocytes were incubated with the studied extracts in the concentrations of 100, 250, and 500 µg/mL for 2, 5, and 24 h. All extracts tested caused a decrease in erythrocyte membrane fluidity and increased erythrocyte osmotic sensitivity. The ethanolic extracts from the bark and leaves increased viscosity of the erythrocytes. The largest changes in the studied parameters were observed in the cells incubated with bark ethanolic extract. We consider that the compounds from U. tomentosa extracts mainly build into the outer, hydrophilic monolayer of the erythrocyte membrane, thus protecting the erythrocytes against the adverse effects of oxidative stress.

ABSTRAK. Tujuan penelitian ini untuk mengetahui motilitas Ascaridia galli dewasa dalam ekstrak etanol biji Veitchia merrillii. Ekstrak etanol V. merrillii dianalisis fitokimia. Sebanyak 16 ekor cacing A. galli dewasa dibagi kedalam empat kelompok. Cacing pada kelompok pertama adalah kelompok tanpa perlakuan. Cacing pada kelompok kedua diberi 0,6 mg/ml levamisole. Cacing pada masing-masing kelompok ketiga dan keempat diberi 50 dan 100 mg/ml crude ekstrak biji V. merrillii. Motilitas A. galli ditentukan dalam skor persentase setelah 12, 24, 36 jam dengan menggunakan kriteria: 3 (badan bergerak), 2 (hanya sebagian badan bergerak), 1 (tidak bergerak tetapi masih hidup), 0 (mati). Hasil fitokimia V. merrillii mengandung alkaloids, saponins, tannins, flavonoids, terpenoids. Ekstrak biji V. merrillii dosis 100 mg/ml secara in vitro dapat mempersingkat selama 12 jam waktu motilitas cacing A. galli dewasa. Penelitian ini mengindikasikan potensi anthelmintik berbasis herbal untuk pengendalian A. galli.  (Motility of Ascaridia galli adult worms in vitro in ethanolic extracts of Nuts Veitchia merrillii) ABSTRACT. The purpose of this research was to know the motility of Ascaridia galli adult worms in aqueous ethanolic extracts of nuts Veitchia merrillii. The ethanolic extract of the V. merrillii was analyzed. Amount of sixteen head A. galli adult worms were divided into four groups. The first group, worms were left as un-treated normal controls. The second group, worms were treated with concentrations of 0,6 mg/ml levamisole. The third and fourth group, worms were treated with crude aqueous ethanolic extract of 50 and 100 mg/ml concentrations nuts of the V. merrillii, respectively. Motility of A. galli were determined after 12, 24, 36 hour by mean of persentage scored using the following criteria: 3 (moving whole body), 2 (moving only parts of the body), 1 (immobile but alive), and 0 (died). The result of phytochemical V. merrillii contains alkaloids, saponins, flavonoids, tannins, and terpenoids. V. merrillii nuts extract concentrations of 100 mg/ml in vitro can shorten the time motility A. galli adult worms for 12 hours. The study indicated the potential for developing herbal-based anthelmintics to control A. galli.

Catunaregam spinosa leaves have been ethnopharmacologically accounted for acquiring various pharmacological properties. The present study was undertaken to evaluate anti-inflammatory, analgesic and antipyretic potential of leaves of C. spinosa.The ethanolic extract was selected for this purpose based on phytochemical screening. Inflammation was inhibited at the dose of 200 mg/kg with percent inhibition of inflammation 32.06, 37.28 and 43.16 %, respectively, at 1, 3 and 5 h, while in egg albumin model % inhibition was found to be 47.81%. There was no significant analgesic activity seen in acetic acid induced writhing response method while significant effects were observed in the doses of 25 and 100 mg/kg on hot plate test. No antipyretic activity was shown by ethanolic extracts(25, 100 and 200 mg/kg) against Brewer’s yeast induced pyrexia in rats. This study aimed to understand Strychnosnuxvomica and Holarrhena pubescens Stem bark extract action towards M3 receptor in controlling blood glucose levels. Strychnos nux vomica  and Holarrhena pubescens are both alkaloidal drugs can help in controlling Hyperglycemic level. This will be useful in the formulation of a new herbal drug molecule for treating diabetes. Chloroform and ethanolic extracts of selected alkaloidal plants were extracted using the soxhlet apparatus and obtained quotes were tested for acute toxicity studies and carried out anti-diabetic action on Wister albino rats for 21 days. Results obtained from Blood glucose levels and histopathological study of test groups are compared with blood glucose levels of standard group, and highly significant action was identified by the chloroform extract of Strychnos nux vomica and Holarrhena pubescens group. Moderate anti-diabetic action was observed remaining two groups of ethanolic extracts. Strychnos nux vomica and Holarrhena pubescens ethanolic extract groups are acting on M3 receptors and controlling Hyperglycemic levels.
