Potato Dextrose Agar (PDA) is the most media to grow the mushrooms, but the price of this media is expensive. Thus we need alternative media that easy and cheap to get it. This study aims to determine the growth of Aspergillus niger in alternative Artocarpus communis and Artocarpus heterophyllus seeds media. This research was an experimental study using a completely randomized design (CRD) one factor was the type of media is PDA (M0), Artocarpus communis seed media (M1), Artocarpus heterophyllus seed media (M2) and using the test mushrooms A. niger (J1). Inoculation of A. niger used agar block method for 3 days with a temperature of 28°C. Parameter of research was colony diameter and sporulation of A. niger. Data obtained with qualitative and quantitative methods. The result of this research showed that the best growth for Aspergillus niger was after 72 hours incubation. Colony diameter continually in PDA media, Artocarpus communis media, and Artocarpus heterophyllus media is 4.7 cm, 4.3 cm, and 4.1 cm with heavy sporulation. Therefore, Artocarpus communis and Artocarpus heterophyllus seeds media can be utilized as a substitution of PDA media for the growth of mushrooms.

The efficacy of liquid smoke obtained from medang wood (Cinnamomum sp.)against Schizophyllum commune fungus was evaluated. This study aims to evaluate the antifungal properties of liquid smoke from medang against S. commune fungus. Potato Dextrose Agar (PDA) medium was used to determine the efficacy of the liquid smoke of medang wood on S. commune fungus growth. Three kinds of liquid smoke were obtained from the pyrolysis of medang wood at 370, 400, and 430°C. The efficacy of liquid smoke from medang wood for antifungal is a factorial 3 by 4 in a completely randomized design; the first factor was pyrolysis temperature of liquid smoke from medang (370, 400, and 430°C), and the second factor was the treatment of concentration of liquid smoke from medang wood (0, 0.5, 1.0, 1.5, 2.0, 2.5, and 3.0%, v/v). The results showed that pyrolysis temperature affected liquid smoke from medang, and the concentration of liquid smoke was significantly different for inhibition of fungal growth. The results indicated that the pyrolysis temperature of liquid smoke production and the concentration of liquid smoke had a significant effect on S. commune fungus growth inhibition. Medang wood liquid smoke effectively inhibited the growth of S. commune fungus about 98.57% at a concentration of 2.5% with liquid smoke pyrolysis temperature used is 430°C.Keywords: efficacy, inhibition of fungal growth, liquid smoke, medang wood, Schizophyllum commune

This study aims to determine the quality of jackfruit seed juice drinks with the addition of red ginger and to determine the level of acceptance of panelists on jackfruit seed juice drinks. Parameters observed included water content, protein content, carbohydrate content and organoleptic including color, taste, aroma and texture. This study used a Completely Randomized Design (CRD) consisting of one factor, namely the concentration of red ginger juice with four treatments, namely: 0%, 6%, 12%, 18%, each repeated three times. The results showed that jackfruit seed juice drink with the addition of 18% red ginger was the best treatment from the results of organoleptic tests. The proximate test results showed that the best treatment was 3.13% water content in 12% ginger treatment, 2.17% protein in 18% red ginger treatment and 95.61 carbohydrate in 18% red ginger treatment.

This study evaluated the synergism between several P-solubilizing fungi isolates and arbuscular mycorrhizal fungi to improve clover ( Trifolium pratense) growth in the presence of Araxá apatite. Clover was sown directly in plastic pots with 300g of sterilized washed sand, vermiculite and sepiolite 1:1:1 (v:v:v) as substrate, and grown in a controlled environment chamber. The substrate was fertilized with 3 g L-1 of Araxá apatite. A completely randomized design, in 8×2 factorial scheme (eight P-solubilizing fungi treatments with or without arbuscular mycorrhizal fungi)and four replicates were used. The P-solubilizing fungi treatments consisted of five Brazilian P-solubilizing fungi isolates (PSF 7, 9, 20, 21 and 22), two Spanish isolates ( Aspergillus niger and the yeast Yarowia lipolytica) and control (non-inoculated treatment). The greatest clover growth rate was recorded when Aspergillus niger and PSF 21 were co-inoculated with arbuscular mycorrhizal fungi. Aspergillus niger, PSF 7 and PSF 21 were the most effective isolates on increasing clover growth in the presence of arbuscular mycorrhizal fungi. Greater mycorrhizal colonization resulted in greater clover growth rate in most PSF treatments. PSF 7 was the best isolate to improve the establishment of mycorrhizal and rhizobia symbiosis.

Various problems have caused the decline in peanut production, among others the low resistance of peanut plants to various diseases. One of the obstacles in peanut production is the attack of plant diseases, such as the fungus Sclerotium rolfsii Sacc. The use of plant-based insecticides can be an alternative to disease control in peanuts. This study aimed to obtain the appropriate and effective dose of clove leaves and flower stalks in suppressing the in vitro growth of the fungus S. rolfsii Sacc causing damping off in peanuts, and to test the effectiveness of clove leaves and flower stalks on the formation of sklerotia of the fungus. This study used ‘tuni’ clove powder from Waai Village, Salahutu District. This experimental study employed a completely randomized design (CRD) with 8 treatments and 3 replications. The results showed that the growth of the fungus S. rolfsii Sacc. was inhibited by clove flower stalk powder at a dose of 1.5 g per100 mL of PDA, which was indicated by the number of sklerotia formed of 13.67% and propagule weight of 0.12 g, followed by a dose of clove leaf powder of 1.5 g per100 mL PDA, with amount of sklerotia formed of 27.33% and propagule weight of 0.17 g. These results were statistically shown on the parameters: colony diameter, Sklerotia formation time, number of Sklerotia, and weight of propagules. Keywords: Clove powder, dosage, in vitro, Sclerotium rolfsii Sacc.   ABSTRAK Berbagai masalah telah menyebabkan turunnya produksi kacang tanah, diantaranya masih rendahnya ketahanan tanaman kacang tanah terhadap berbagai penyakit. Salah satu kendala dalam produksi kacang tanah adalah serangan penyakit tanaman, seperti jamur Sclerotium rolfsii Sacc. Penggunaan insektisida nabati dapat menjadi alternatif pengendalian penyakit pada kacang tanah. Penelitian ini bertujuan untuk memperoleh dosis daun dan tangkai bunga cengkih yang tepat dan efektif dalam menekan pertumbuhan jamur S. rolfsii Sacc penyebab damping off pada kacang tanah secara in vitro, serta menguji efektifitas daun dan tangkai bunga cengkih terhadap pembentukan sklerotia jamur. Penelitian ini menggunakan bubuk cengkih tuni, dari Desa Waai, Kecamatan Salahutu. Penelitian eksperimental ini menggunakan Rancangan Acak Lengkap (RAL) dengan 8 perlakuan dan 3 ulangan. Hasil penelitian menunjukkan bahwa pertumbuhan jamur S. rolfsii Sacc. terhambat oleh bubuk tangkai bunga cengkih pada dosis 1,5 g per 100 mL PDA yang ditunjukan oleh jumlah sklerotia terbentuk 13,67% dan berat propagul 0,12 g dan diikuti oleh dosis bubuk daun cengkih 1,5 g per 100 mL PDA, dengan jumlah sklerotia 27,33% dan berat propagul 0,17 g. Hasil ini secara statistik ditunjukkan pada parameter: diameter koloni, waktu terbentuk sklerotia, jumlah sklerotia, dan berat propagul. Kata kunci: bubuk cengkih, dosis, in vitro, Sclerotium rolfsii Sacc.

Pemanfaatan sisa hasil pertanian (jerami padi) sebagai pakan dasarmenjadi hal yang strategis untuk dikembangkan guna memenuhi kebutuhan pakan berserat bagi ternak sapi potong. Mikrobia selulolitik dan bakteri asam laktat merupakan salah satu sumber inokulum yang dapat meningkatkan kualitas jerami padi sebagai pakan dasar berserat. Penelitian dilakukan dengan tujuan untuk mendapatkan sumber karbon yang tepat untuk pertumbuhan Aspergillus niger(AN) dan Lactobacillus plantarum(LP).Penelitian ini dilakukan dengan menfermentasi jerami padi menggunakan AN dan LP. Perlakuan sumberkarbon yang diberikan dua macam substrat, yaitu molasses dan dedak. Perlakuan level AN sebesar 0, 5, 10, dan 15%. Pemberian LP sebesar 10% pada setiap perlakuan. Fermentasi dilakukan selama 21 hari,sedang nilai kecernaannyadievaluasi dengan menggunakan metode in vitro Tilley andTerrydan Gas Test. Variabel yang diamati dalam penelitian ini adalah pH, asam laktat,BK, BO, PK, SK, NDF, ADF, dan TDN. Data yang diperoleh dianalisis menggunakan analisis varian rancangan acak lengkap (Completely Randomized Design/CRD) pola searah dan pola faktoril (2 x 3), bila terdapat pengaruh nyata dilanjutkan dengan uji DMRT (Duncan’s Multiple Range Test). Hasil penelitian menunjukkan penggunaan Aspergillus nigerdan Lactobacillus plantarumuntuk fermentasi jerami padi yang terbaik adalah  15% Aspergillus nige rdan  10% Lactobacillus plantarum dari bahan kering. Penggunaan substrat molases lebih baik jika dibandingkan dengan dedak, hal ini terlihat dari hasil analisis PK, SK, LK, ADF, dan hasil uji fisik jerami padi fermentasi.

The effect of some plant extracts on Deightoniella torulosa  Syd. Ellis the causative agent of black tip disease of banana.  An experiment was conducted to evaluate the effect of some plant extracts on the growth and reproduction of Deightonella torulosa. The plants  used in this experiments were Zingiber cassumunar, Acorus calamus, and Amomum cardamomum. The treatments of the factorial (3x4) experiment were arranged in a completely randomized design with three replicates. The first factor (plant extracts) consisted of  Z. cassumunar,  A. calamus, and A. cardamomum extracts. The second factor (extract concentration) consisted of  0, 100, 200, and 300 mg/l. Variables examined were  the colony diameter and conidial density of D. torulosa. The result of the experiment shows that extract of Z. cassumunar,  A. calamus, and  A. cardamomum extracts suppressed fungal colony diameter. On Z. cassumunar and A. calamus extracts, the higher the concentration the higher fungal colony diameter suppressed. The most effective suppression by the extract was at 300 mg/l.  A. calamus extract was more effective than Z. cassumunar and  A. cardamomum at 100 mg/ml. Z. cassumunar and A. calamus extracts had the same effectivity at 200 mg/l, and both was more effective compared to A. cardamomum.  Z. cassumunar  was more effective  compared to A. calamus and  A. cardamomum at 300 mg/l. The results of the experiment also showed that Z. cassumunar, A. calamus,  and  A. cardamomum suppressed  conidial production  of D. torulosa.   Z. cassumunar most effectively suppressed   conidia production at 300 mg/l.  A. calamus  extract suppressed conidial production the most effectively  at 200 and 300 mg/l. A. cardamomum  extract significantly suppressed conidia production only at 300 mg/l. Z. cassumunar and  A. calamus  extracts were more effective to suppress conidia production compared to A. cardamomum for all level of concentration except control. The effectivity of Z. cassumunar  and  A. calamus  was the same at 100 and 300 mg/l. Z. cassumunar extract was more effective  than A. calamus at 200 mg/l.

This study aims to determine the optimal time of fermentation using Aspergillus niger on the pH value, dissolved protein and levels of ash content corn straw. The design used was a Completely Randomized Design (CRD) of unidirectional pattern consisting of 4 treatments repeated 3 times namely P0 : corn straw fermented with Aspergillus niger for 0 days, P1 : corn straw fermented with Aspergillus niger for 4 days, P2 : fermented corn straw with Aspergillus niger for 8 days, P3 : corn straw fermented with Aspergillus niger for 12 days. The parameters observed were pH value, dissolved protein and ash content. The results showed that corn straw fermented with Aspergillus niger had a very significant effect (P <0.01) on the pH value, dissolved protein content and ash content. Corn straw fermentation using Aspergillus niger for 0 - 12 days has a very significant effect on the pH value, dissolved protein content and ash content (P <0.01). The average pH value of the treatment  P0: 5,00, P1: 6,13, P2: 6,51, P3: 6,84 The average value of dissolved protein content in the treatment P0: 27,09%, P1: 33,31%, P2: 24,14%, P3: 25,51%. The average value of ash content in the P0: 8,91%, P1: 11,34%, P2: 10,06%, P3: 9,22%,  As well as the optimal time achieved in fermentation for 4 days for dissolved protein content and ash content.  Keywords: Corn straw, Aspergillus niger, Fermentation, pH value, Dissolved protein content, Ash content

Putak is a local feed in west Timor, East Nusa Tenggara Province. It is obtained from soft core of palm tree which is called gebang tree (Corypha gebanga). Putak contains high carbohydrate and fiber but low in protein. As an effort to increase the ptotein level in putak a research was conducted by fermenting it with mixed-cultured of Trichoderma reesei and Aspergillus niger. A laboratorium experimental method was used and arranged factorially in Completely Randomized Design 3 x 4 x 3 namely three inoculant levels of T. reesei (T): 5,0; 7,5 and 10,0 % (w/w), and four levels of A. niger (A) 0,5; 1,0; 1,5 and 2,0% (w/w). Mixed-cultured was done by: first, T.reesei as treatment mix with putak and incubation for two days, after that mix with A.niger, incubation for two days. The best result of this study was on combination of T.reesei 7.5% level and 1,5% of A.niger were crude protein (23.62%) and soluble protein (14.92%) was highest and fiber 10,17%.

The research about the fermentation of protein-free grated coconut with Aspergillus niger to produce of crude lipase enzyme has been carried out. This study aims to determine the effect of water content on fermentation medium and the incubation time which results lipase enzyme extract with the highest activity. This study used a Completely Randomized Design (CRD) consisting of 2 factors, i.e. variations of water content in medium of 40%, 50%, 60%, and 70% and incubation times of 48, 96, 144, and 192 hours. The parameters observed were lipase enzyme activity produced in each treatment. The results showed that the highest medium water content was 70% with the lipase enzyme activity 4.33 µmol/mL.minute and the best incubation time was 96 hours with the lipase enzyme activity of 0.83 µmol/mL.minute. ABSTRAKPenelitian ini bertujuan untuk melakukan evaluasi kecernaan in vitro bahan kering dan bahan organik kulit singkong yang difermentasi menggunakan jamur Aspergillus niger dan EM-4 sehingga dapat memberikan informasi tambahan mengenai kualitas kulit siongkong yang difermentasi maupun tanpa fermentasi.  Rancangan penelitian yang digunakan adalah rancangan acak lengkap (RAL) yang terdiri atas 4 perlakuan dan 4 ulangan.  Perlakuan yang diberikan terhadap kulit singkong adalah P1 (tanpa fermentasi), P2 (fermentasi dengan Aspergillus niger), P3 (fermentasi dengan EM-4), P4 (fermentasi dengan kombinasi A. niger dan EM-4).  Evaluasi kecernaan in vitro ini dilakukan dengan menggunakan metode Tilley and Terry.  Hasil penelitian menunjukkan bahwa penambahan jamur A. niger dan EM-4 dalam proses fermentasi kulit singkong dapat meningkatkan kecernaan bahan kering dan bahan kering kulit singkong secara signifikan dimana hasil terbaik diperoleh pada penambahan kombinas A. niger dan EM-4.Kata kunci: Aspergillus niger, bahan kering, bahan organik, EM-4, kecernaan in vitroABSTRACTThe aim of this study was to evaluate the in vitro digestibility of dry matter and organic matter of fermented cassava peel using Aspergillus niger and EM-4 inoculant to provide additional information about the quality of fermented and unfermented cassava peel. The study design used was a completely randomized design (CRD) consisting of 4 treatments and 4 replications. The treatments were P1 (without fermentation), P2 (fermented with Aspergillus niger), P3 (fermented with EM-4), P4 (fermented with a combination of A. niger and EM-4). This in vitro digestibility evaluation was conducted by using the Tilley and Terry method. The results showed that the addition of A. niger fungi and EM-4 in the fermentation process of cassava peel could significantly improve the digestibility of dry matter and dry matter of cassava peel where the best results were obtained on the addition of A. niger and EM-4 combinations.Keywords: Aspergillus niger, dry matter, EM-4, in vitro digestibility, organic matter
