AbstractThe objective of this study was to investigate the effect of diets containing different levels of dried tuber of Jerusalem artichoke, Helianthus tuberosus, on skatole levels in back fat and on the CYP2E1 mRNA expression in the liver of commercial crossbred pigs. A total of 23 uncastrated male pigs from 10 litters of a commercial crossbred population of Large White × (Landrace × Large White), were used in this study. Boars were randomly divided into four different dietary treatment groups - a control group (K1; 5 boars; without supplementation of Jerusalem artichoke,) and three experimental groups (6 boars each) that were fed with the diet containing different levels of dried Jerusalem artichoke (K2 – 4.1%; K3 – 8.2%; K4 – 12.2%) for 14 days before slaughter. Significant effects of diet on skatole levels were observed between the control group and the experimental groups (P = 0.0078). The lowest level of skatole was in the K3 group with 8.2% of Jerusalem artichoke. As for CYP2E1, a negative correlation was observed between the levels of skatole and CYP2E1 mRNA expression. Significant effect (P = 0.0055) was found in all experimental groups compared to the K1 group, and most pronounced in the K2 and K3 groups. The supplementation with Jerusalem artichoke resulted in lower level of skatole and higher CYP2E1 mRNA expression. The results suggest that affecting the expression of CYP2E1 by feed supplements could be an option to effectively reduce the levels of skatole in adipose tissue of entire male pigs.

This study aimed to obtain a comprehensive look at the influence of castration on mRNA expression of the genes CYP2E1, CYP1A2, CYP2A19, HSD3B, SULT2A1 and SULT1A1 and their correlation with boar taint compounds (androstenone, skatole and indole) and Improvac-specific antibodies in a Czech commercial hybrid (Large White × Landrace (sow) × Duroc (boar)). Pigs were divided into groups of entire male pigs (NC), pigs castrated surgically (SC), pigs immunologically castrated and slaughtered 8 weeks (IM8) or 15 weeks (IM15) after the second dose of Improvac, and gilts (GI). Hepatic mRNA expression, measured by quantitative real-time polymerase chain reaction, differed significantly between the control group (entire male pigs) and all groups of interest for CYP2E1, CYP1A2 and CYP2A19. The mRNA level of the HSD3B gene differed significantly between the control group and the IM8, IM15 and GI groups. SULT1A1 gene expression was significantly different between the control group and the SC, IM8 and GI. In the case of SULT2A1, a significant difference was observed only between the control group and IM8 pigs. For all genes and treatment groups described above, expression was increased relative to the control. Significant differences for Improvac-specific antibodies between IM8 and IM15 groups were observed, indicating decrease of antibodies over time. Moreover, negative correlations between androstenone and mRNA levels of CYP2A19, CYP2E1 and SULT1A1 suggest that gene expression is suppressed.

This study was carried out to determine the influence of different levels of alfalfa meal and the addition of enzyme on performance, carcass and intestinal parameters in growing quails. Japanese quails (n=240), 1 days of age, were divided into 6 dietary treatment groups and the experiment lasted for 5 weeks. In each experiment group there were 4 replicates, and in each replicates there were 10 chicks. Six diets, arranged a factorial design with three alfalfa meal levels (0, 2.5 and 5.0 %) and two levels of added enzyme (0.0 and 1.0 g/kg) were used. The treatments did not effect on the body weight, feed intake and feed conversion ratio. However, relative liver, leg+back, intestine weights and crypt depth were significantly increased, and relative heart and breast weights and villus width decreased by dietary alfalfa meal levels. The addition of enzyme to diets containing alfalfa meal did not affect the performance and carcass parameters, but reduced the intestinal histomorphology parameters in quails. According to the results obtained from the experiment, it can be said that the use of alfalfa meal in growing quail diets did not affect the performance, but the use of alfalfa meal at the level of 2.5% in the diet promotes intestinal development.

The present study aims to distinguish the effect of tocotrienol on an important gastric protective factor, prostaglandin E2(PGE2), in stress-induced gastric injury. Twenty-eight Wistar rats were divided into four groups of seven rats each. Two control groups were fed commercial rat diet, and two treatment groups were fed the same diet but with additional dose of omeprazole (20 mg/kg) or tocotrienol (60 mg/kg). After 28 days, rats from one control group and both treated groups were subjected to water-immersion restraint stress for 3.5 hours once. The rats were then sacrificed, their stomach isolated and gastric juice collected, lesions examined, and gastric PGE2content and cyclooxygenase (COX) mRNA expression were determined. Both the regimes significantly attenuated the total lesion area in the stomach compared to the control. Gastric acidity, which was increased in stress, was significantly reduced in rats supplemented with omeprazole and tocotrienol. The PGE2content was also significantly higher in the rats given tocotrienol supplementation compared to the control followed by an increase in COX-1 mRNA expression. We conclude that tocotrienol supplementation protected rat gastric mucosa against stress-induced lesions possibly by reducing gastric acidity and preserving gastric PGE2by increasing COX-1 mRNA.

Abstract The objective of this study was to investigate the effects of diets supplemented with sodium stearoyl-2-lactylate (SSL), polyglycerol fatty acid ester (PGFE), and combined emulsifiers (0.02% SSL and 0.08% PGFE) on growth performance, nutrient digestibility, and plasma lipid profiles in weaned piglets and to further evaluate the possible effects of feeding exogenous emulsifiers on digestive enzyme activities and liver bile acid (BA) metabolism. Twenty-eight barrows (age at 35 d, Duroc × Landrace × Yorkshire) with an initial BW of 10.13 ± 0.16 kg were randomly assigned to 4 dietary treatment groups (7 pigs/treatment). Dietary treatment groups included the following: 1) basal diet (Control, CTR); 2) basal diet with 0.1% SSL (SSL); 3) basal diet with 0.1% PGFE (PGFE); and 4) basal diet with 0.08% PGFE+0.02% SSL (PG-SL). SSL diet increased ADG and ADFI of piglets during day 0 to 17 (P < 0.05) compared with the CTR treatment. Piglets fed emulsifier diets experienced a significant improvement in the digestibility of nutrients (DM, CP, ether extract, energy, calcium, and phosphorus) during the first 17 d (P < 0.05). The level of low-density lipoprotein cholesterol (LDL-C) was lower in the PGFE and PG-SL treatment groups than in the CTR treatment group (P < 0.05). Feeding emulsifier diets increased the lipase activity of the pancreas when compared with the CTR diet (P < 0.05). Moreover, the emulsifier diets significantly increased the mRNA expression of FXR (P < 0.05) and decreased the mRNA expression of CYP27A1 (P < 0.05) in the liver. In conclusion, the addition of emulsifiers improved nutrient digestibility and increased the mRNA expression of FXR BA receptors while inhibiting the mRNA expression of BA biosynthesis by CYP27A1 in weanling piglets.

SUMMARYThe carcass characteristics of young adult female pigs (gilts) slaughtered after weaning their first litters have been investigated using three groups of 19 Landrace × (Landrace×Large White) gilts. In two treatment groups gilts were mated at their pubertal oestrus and suckled their piglets for 5 to 11 and 35 to 42 days respectively. These gilts were slaughtered on average 10 days after weaning. A third control group consisted of unmated gilts slaughtered at 118 kg live weight.There was no significant difference in the killing-out percentage for the three groups. The carcasses of the gilts which farrowed contained significantly less fat (P<0·001) than those of the unmated controls. The carcasses of the farrowed gilts had significantly lighter middle sections (P<0·001) and significantly heavier shoulders (P<0·001) than the controls.Despite the variations in tissue yield and distribution, the yield of prime joints for the gilts which had farrowed was only 0·62 percentage units less than that of the controls.No commercially significant variations between groups in meat quality were observed.

Abstract Objective: To observe the expression levels of serum high mobility group box 1 (HMGB1) , nicotinic acetylcholine receptor α7 (α7 nAChR) and nuclear factor kappa-B mRNA(NF-κB mRNA) in colon tissue after electroacupuncture (EA) on Shangjuxu, Zusanli, Xiajuxu,Yanglingquan and Chengjin points, by comparison of the different effects we could thus explore the relative specificity of Shangjuxu on treating its corresponding viscera’ s disease. Methods: Seventy SD rats were randomly assigned to seven groups labeled by blank control (A), Model (B), Shang Ju Xu (C), Zu San Li (D), Xia Ju Xu (E), Yang Ling Quan (F), Cheng Jin (G), ten (half males and half females) in each group. Except for the blank control group, animals in other groups were modeled to ulcerative colitis (UC) by lavation of 2-4-6 trinitro-benzene-sulfonic acid (TNBS)/ethanol solution. After successful modeling animals in the five treatment groups were respectively treated with electroaccupuncture at corresponding acupoint for 10 consecutive days. Then histological changes in the colon were observed by light microscopy. ELISA method was used to check models’serum HMGB1 content, and α7 nAChR in the colon were detected by western blot , RT-PCR was used to detecte NF-κB mRNA. Results: ① Compared with the model group, the tissue injury changes in 5 treatment groups improved to a certain extent, and the colon tissue NF-κB mRNA expression of them decreasd signifcantly(P<0.01),as well, the α7 nAChR expression of Shang Ju Xu group, Zu San Li group and Xia Ju Xu group increased signifcantly (P<0.01 or P<0.05), and their serum HMGB1 content made obvious decrease(P<0.01 or P<0.05), the α7 nAChR expression in Yang Ling Quan group was also increased significantly(P<0.01). ② Compared with Shang Ju Xu group, the tissue injury changes of groups including Xia Ju Xu,Yang Ling Quan and Cheng Jin are much severe under light microscope, and the α7 nAChR expression of them decreased with significance(P<0.01),both serum HMGB1 content and colon tissue NF-κB mRNA expression increased significantly as well(P<0.01),the serum HMGB1 content of Zu San Li group also increased significantly(P<0.01). Conclusion: ① Electroacupuncture on all five different acupoints can improve the UC through regulating HMGB1, α7 nAChR and NF-κB mRNA, and effectively inhibiting the immune response, reducing inflammatory response of colon and improving pathological changes of colonic mucosa. ②The effect of treating UC inflammation in rats when electroacupuncture at Shang Ju Xu was better than Zu San Li, Xiajuxu, Yang Ling Quan and Cheng Jin, which demonstrated that Shangjuxu treating UC has some relative specificity.

The present study was conducted to investigate the effect of supplementation of different levels of dried Tamarind pulp on meat cholesterol, carcass yield and dressing percentage of broiler chicken. 300 healthy day old commercial broiler chicks of Vencob’ strain were selected and divided into five groups of 60 chicks in each group, three replicates of 20 birds in each group. The chicks of control group (Group I) were fed with standard broiler diet without dried Tamarind pulp, whereas dried Tamarind pulp was supplemented @250 gm, 500 gm, 1000 gm and 1500 gm per 100 kg of broiler ration respectively to groups -II, III, IV and V.The meat cholesterol was found to be significantly lower in treatments groups than in control group. However, carcass yield and dressing percentage of treatment groups were significantly higher than control Group.The carcass yield and dressing percentage were increased and meat cholesterol level was decreased on supplementation of dried Tamarind pulp in broiler rations.

The present study was undertaken to evaluate the efficacy of aspirin as anti-heat stressor. Broiler chicks were reared on a standard diet up to 14 days of age. Thereafter, the chicks were randomly distributed into three dietary treatment groups viz., T1 (Control group: Standard diet), T2 (Standard diet with aspirin@250 mg/kg) and T3 (Standard diet with aspirin@500 mg/kg). Each treatment was having five replicates of eight birds per replicate. Experiment was carried out during hot-dry (April-May, 30.0±0.70 to 37.0±1.40C, Rh, %: 58.05±1.32 to 70.11±0.82) summer. Feed intake and live weight gain was comparable among the treatments but FCR, protein and energy efficiency improved significantly (P less than 0.001) on aspirin supplementation. The relative yield of thymus and bursa at 4th and 6th week of age improved significantly (P less than 0.05). The percentage of protein, aspartate transaminase (AST) and alanine transaminase (ALT) increased significantly (P less than 0.001) due to aspirin supplementation at 4th as well as 6th week of age. While H:L ratio, serum corticosteron and serum cholesterol (only at 42nd day of age) decreased significantly (P less than 0.001) in aspirin supplemented groups at 28th as well as 42nd day of age. Supplementation of aspirin at 500 mg/kg reduced relative expression of HSP70 in jejunum tissues during 28th or 42nd day of age. It can be concluded that aspirin supplementation 500 mg/kg in diet of heat stressed broilers improved performance as well as welfare..

Chronic renal failure (CRF) is a serious disease related to increasing incidence and prevalence as well as decline in quality of life. Bu-Shen-Huo-Xue formula (BSHX), one of traditional herbal formulations, has been clinically employed to treat CRF for decades, but the mechanisms involved have not been investigated. In the present study, we investigated the effects of BSHX on some closely related parameters in 5/6 nephrectomy CRF rats. Rats with CRF were divided into five groups, namely, one control group, one enalapril group, and three BSHX treatment groups (0.25, 0.5, and 1 g/kg·d). The rats subjected to sham operation were used as a normal control. After eight weeks of treatment, BSHX significantly decreased the levels of Scr and BUN, downregulated the mRNA expression levels of TGF-β1, CTGF, NF-κB, TNF-α, and OPN, upregulated the mRNA expression of PPARγ, and reducedin situexpression of fibronectin and laminins. Histological findings also showed significant amelioration of the damaged renal tissue. BSHX protects 5/6 nephrectomy rats against chronic renal failure probably via regulating the expression of TNF-α, NF-κB, TGF-β1, CTGF, PPARγ, OPN, fibronectin, and laminins and is useful for therapy of CRF.

Mycotoxins are toxic secondary metabolites produced by different strains of fungi, such as aspergillus, fusarium, and penicillium that can contaminate feed ingredients or the entire feed of poultry and animals. Mycotoxins can cause many serious complications to both humans and animals due to carcinogenic, mutagenic, and immunosuppressive disorders. Therefore, the present experiment aims to investigate the effect of broiler chickens’ diets supplemented with different levels of nanosilica (NS) as an adsorbent agent of mycotoxins on their growth performance and hepatic histopathology. Detectable levels of toxins were present in the feed before feeding, and all levels of mycotoxins were above the normal limit. A total of 180 one-day-old male Arbor Acres broiler chickens were allocated randomly to six treatment groups with three replicates per group, including ten chickens per replicate. The experiment lasted for five weeks, and dietary treatments included control diet and diets with four levels of nanosilica as 0.05%, 0.10%, 0.15%, and 0.20% as well as 0.50% bentonite (fixfin® Dry) diet. Bodyweight, body weight gain, average daily feed intake, and feed conversion ratio were measured weekly. At the end of the fifth week, six chickens per treatment were sacrificed to investigate the effects of NS and bentonite on carcass characteristics and hepatic histopathology. The results showed that providing broiler chickens’ diets with an adsorbent agent, such as NS or bentonite, can reduce the side effects of mycotoxins and enhance their growth performance. The best record was achieved with NS at 0.20%, compared with the control group and other dietary treatment groups. Accordingly, 0.20% of NS could be used in broiler chickens’ diets to minimize the harmful effects of mycotoxins.
