ABSTRACTIn Vitro mutation using colchicine on 2 month of self-pollinated protocorm like bodies of Dendrobium sylvanum var. flava was conducted to determine the effects of concentration and immersion duration in colchicine on proliferation of PLBs, and to identify of ploidy variants based on stomatal variable. Research was arranged using factorial completely randomized design with three factors in three replications. The first factor was concentration of colchicine, consisted of five concentrations (0.02; 0.04, 0.06; 0.08; dan 0.1%). The second factor was duration of immersion in the colchicine, consisted of four durations (1; 24; 48; 72 hours). The third factor was proliferation medium consisted of two concentrations of BAP (1; 0.5 mg L-1). Analysis of variance showed the significant effect of colchicine treatment on percentage of survived explants. LD50 in media 1 mg L-1 BAP was obtained at a colchicine concentration of 0.069% with duration immersion of 58.19 hours. On Media 0.5 mg L-1 BAP, LD50 was obtained at colchicine concentration of 0.054% with duration immersion of 47.63 hours. Percentage of solid polyploid mutant of Dendrobium sylvanum var. flava can not be determined on MV2 generation because the stomata leaf showed chimeras based on the chloroplast number in cell guard and stomata size.Keywords: colchicines, Dendrobium sylvanum, mutation, number of chloroplast stomatal density.ABSTRAKMutasi dengan kolkisin pada PLBs hasil selfing Dendrobium sylvanum var. flava umur 2 bulan dilakukan secara In Vitro dengan tujuan mempelajari pengaruh konsentrasi kolkisin, durasi perendaman dalam kolkisin, media proliferasi terhadap pertumbuhan PLBs Dendrobium sylvanum var. flava serta mengidentifikasi variasi ploidi berdasarkan variabel stomata. Penelitian disusun dalam Rancangan Acak Lengkap Faktorial 3 Faktor dengan 3 ulangan. Faktor pertama ialah konsentrasi kolkisin yang terdiri atas 5 taraf (0.02; 0.04, 0.06; 0.08; dan 0.1%). Faktor kedua ialah durasi perendaman yang terdiri atas 4 taraf (1; 24; 48; 72 jam). Faktor ketiga ialah media proliferasi media V&W yang ditambah BAP terdiri atas 2 taraf (1; 0.5 mg L-1). Hasil analisis ragam menunjukkan pengaruh kolkisin yang nyata terhadap variabel persentase hidup. LD50 pada media 1 mg L-1 BAP diperoleh pada konsentrasi 0.069 % dengan durasi perendaman 58.19 jam. Pada media 0.5 mg L-1 BAP, LD50 diperoleh pada konsentrasi 0.054 % dengan durasi perendaman 47.63 jam. Persentase mutan poliploid pada MV2 Dendrobium sylvanum var. flava ini belum dapat ditentukan karena stomata daunnya masih kimera berdasarkan karakter jumlah kloroplas sel penjaga dan ukuran stomata.Kata kunci: Dendrobium sylvanum, jumlah kloroplas, kerapatan stomata, kolkisin, mutasi.

ABSTRAK  Penelitian bertujuan untuk mengetahui pengaruh pemberian bubuk daun Moringa oleifera (MO) terhadap kuantitas dan kualitas semen sapi peranakan ongole (PO). Metode penelitian menggunakan percobaan lapang dengan rancangan acak lengkap dengan tiga perlakuan pemberian MO sebesar 0 kg/ekor/hari (P1); 0,05 kg/ekor/hari (P2) dan 0,1 kg/ekor/hari (P3). Analsis data menggunakan one way analysis of variance. Masing-masing perlakuan terdiri atas empat ekor dengan tiga periode pengamatan sebagai ulangan, yaitu periode pertama dan ketiga tanpa MO, dan periode kedua diberikan MO pada P2 dan P3. Hasil penelitian menunjukan bahwa volume dan pH semen perlakuan P1, P2 dan P3 tidak berbeda semua periode pengamatan. Motilitas massa spermatozoa P2 dan P3 periode kedua dan ketiga menunjukan nilai +++ berbeda nyata (P<0,05) dibandingkan dengan P1 (++); motilitas individu  perlakuan P1 menunjukkan nilai terendah 60,56±2,94 % (P<0,05) dibandingkan P2 dan P3 pada periode kedua dan ketiga (75,50±2,29 dan 72,50±2,34%). Konsentrasi spermatozoa P2 dan P3 periode kedua dan ketiga menunjukkan rata-rata  >1.350 juta/ml berbeda nyata (P<0,05) daripada P1 (876±152juta/ml), sedangkan total spermatozoa motil menunjukkan nilai tertinggi 5.647±829 juta/ml pada P3 periode kedua.Viabilitas spermatozoa P2 dan P3 menunjukkan nilai >88-89 % beda nyata (P<0,05) daripada P1 (<84 %) pada periode kedua dan ketiga, sedangkan nilai abnormalitas spermatozoa yang terbaik adalah P2 sebesar 4,30% pada periode kedua dan P3 sebesar 5,33% pada periode ketiga. Disimpulkan pemberian bubuk daun MO dengan dosis 0,1 kg/ekor/hari dapat memperbaiki kuantitas dan kualitas semen (viabilitas dan total spermatozoa motil) sapi pejantan PO.Kata kunci:  kuantitas dan kualitas semen, Moringa oleifera, sapi pejantan ABSTRACTThe research aimed to determine the effect of Moringa oleifera (MO) leaf powder on the quantity and quality of semen of ongole crossbreed. The research method used was a field experiment with a completely randomized design with three treatments giving MO of 0 kg/head/day (P1); 0.05 kg/head/day (P2) and 0.1 kg/head/day (P3). Each treatment consisted of four heads with three periods of observation as replications, namely the first and third periods without MO, and the second period was given MO on P2 and P3. Data analysis used a one-way analysis of variance. The results showed that the volume and pH of semen treated P1, P2, and P3 didn’t different in all observation periods. The second and third period spermatozoa mass motility of P2 and P3 showed that the value of +++ was significantly different (P<0.05) compared to P1 (++); Individual motility of P1 treatment showed the lowest value 60,56±2,94 % (P<0.05) compared to P2 and P3 in the second and third periods (75,50±2,29 and 72,50±2,34%). The second and third periods of P2 and P3 spermatozoa concentrations showed an average of >1,350 million/cc significantly different (P<0.05) than P1 (876±152 million/cc), while the total motile spermatozoa showed the highest value of 5,647±829 million/cc in the second period P3. The spermatozoa viability of P2 and P3 showed values >88-89% significantly different (P<0.05) than P1 (<84%) in the second and third periods, while the best spermatozoa abnormalities were P2 at 4.30% in the second period and P3 at 5.33% in the third period. It was concluded that the giving of MO leaf powder at a dose of 0,1 kg/head/day could improve the quantity and quality of semen (viability and total motile spermatozoa) of Ongole Crossbreed bulls. ABSTRACTThis study aims to determine the digestibility of dry matter and organic matter level waste silage mixed vegetables and Gliricidia leaves were tested in vitro. This study used a completely randomized design with 3 treatments and 3 replications. Each of these treatments is R0 (Gliricidia leaves 100%), R1 (Gliricidia leaves 70% + 30% silage vegetable waste), and R2 (Gliricidia leaves 40% + 60% silage vegetable waste). Data were analyzed using analysis of variance (ANOVA) and further testing using the test Honestly Significant Difference (HSD). The results of ANOVA showed that the mixture of vegetable waste silage was highly significant (p <0.05) on dry matter digestibility and significantly (p <0.05) on the digestibility of organic matter. It can be concluded that the mixed of vegetable waste silage and Gliricidia leaves can improved digestibility of dry matter and organic matter, treatment of 40% and 60% Gliricidia leaves plus waste vegetable produce silage dry matter digestibility and percentage of organic matter is best (72,24% and 68,19%).Keyword: Silage vegetable waste, gliricidia leaves, dry and organic matter digestibilityABSTRAKPenelitian ini bertujuan untuk mengetahui kecenaan bahan kering dan bahan organik tingkat campuran silase sampah sayur dan daun gamal yang diuji secara in vitro. Penelitian ini menggunakan Rancangan Acak Lengkap dengan 3 perlakuan dan 3 ulangan. Masing-masing perlakuan tersebut ialah R0 (daun gamal 100%), R1 (daun gamal 70% + silase sampah sayur 30%), dan R2 (daun gamal 40% + silase sampah sayur 60%). Data yang diperoleh dianalisis dengan menggunakan analisis sidik ragam (ANOVA) dan uji lanjut menggunakan uji Beda Nyata Jujur (BNJ). Hasil analisis sidik ragam menunjukkan bahwa campuran silase sampah sayur berpengaruh sangat nyata (p<0,05) terhadap kecernaan bahan kering dan berpengaruh nyata (p<0,05) terhadap kecernaan bahan organik. Dapat disimpulkan bahwa perlakuan campuran silase sampah sayur dan daun gamal dapat meningkatkan kecernaan bahan kering dan bahan organik, perlakuan 40% daun gamal dan 60% silase sampah sayur menghasilkan persentase kecernaan bahan kering dan bahan organik yang terbaik yaitu (72,24% dan 68,19%).Kata kunci : Silase sampah sayur, daun gamal, kecernaan bahan kering, dan bahan organik.

The aims of the research are to prove the anthelmintic effects of Ocimum sanctum Linn. leaves ethanol extract on Raillietina echinobothrida in vitro and to know the value of LC50 and LC90of Ocimum sanctum Linn. leaves ethanol extract. The research used completely randomized design. The research had five treatments and each treatment was done in four replications. The research used 5 Raillietina echinobothrida adult worms for each treatment and all replications. The observation and the recording were done at the first hour until the sixth hour of the research. The Raillietina echinobothrida adult worms were noted dead if there was no movement when they were disturbed by anatomic tweezer, the death of Raillietina echinobothrida were confirmed when they dipped mildly in warm water (50°C) and they showed no movements. The acquired data was analyzed using ANOVA and continued with LSD (5%). The results were Ocimum santum Linn. ethanol extract has the anthelmintic activity in 1.25%, 2.5%, 5% and 10% concentrations. The LC50 and LC90 was analyzed using probit analysis. The results of LC50 calculation were 7.8% at the third hour, 6% at the forth hour, 3.2% at the fifth hour, 2.5% at the sixth hour and the results of LC90 were 6.5% at the fifth hour, 5.2% at the sixth hour. The higher concentration of the extract, the higher anthelmintic activity worked.

In vitro dendrobium orchid plantlet growth is highly determined by the cytokinin and auxin concentration. The aim of this research was to study the effect of 6-Benzylaminopurine (BAP) and Naphthaeeneacetic Acid (NAA) on growth of Dendrobium orchid plantlet. The research was arranged in completely randomized design (CRD) with two treatments. The first treatment was the concentration of BAP (A1 : 0 ppm, A2 : 1 ppm, A3 : 2 ppm) and the second treatment was the concentration of NAA (B0 : 0 ppm, B1 : 0,25 ppm, B2 : 0,50 ppm) with 4 replications. The observed variables were number of buds, plantlet height, number of leaves, and length of leaves. The data obtained were analyzed by Analysis of Variance and followed by Least Significance Different (LSD). The result showed that treatment of BAP and NAA only significantly affected the number of buds variable. The combination of BAP 1 ppm and NAA 0,25 ppm was the best treatment for increasing number of buds of dendrobium orchid plantlet.

The present study was conducted to polyploidy possibility induction of two Iranian endemic mint (Mentha mozaffarianii) ecotypes. For this purpose, three experiments were done. The first experiment was factorial, based on completely randomized design with three factors and three replications that rhizomes were used for treatment. The first factor including different colchicine concentrations (0, 0.025, 0.05, 0.1 and 0.2% that 0 as control). The second factor including two Mentha ecotypes (Ecotype A: Kamarej region and Ecotype B: Pirmohlat region) and the third factors consist of two soaking time (6 h and 12 h). In second experiment, apical meristem was treated. The factorial experiment based on randomized completely design with two factors and five replications. The factors including different colchicine concentrations (0, 0.035, 0.07, 0.15, 0.3 and 0.7%) and two ecotypes. In the third experiment, colchicine as combined with irrigation was applied. For this purpose, a factorial experiment in randomized complete design with two factors (colchicine concentrations: 0, 0.025, 0.012, 0.006% and two mint ecotypes) and three replications was conducted. At the end of all experiments, survival rate and tetraploidy percentage (by morphological change, stomata traits, flow cytometry and chromosome counts) were measured. The results showed that different treatment had significant effects on survival percent on all experiments and increasing of colchicine concentration caused decreasing plants survival. On the other hand, tetraploidy changes only in the first experiments were observed. Only in 0.025% colchicine treatment with 6 h soaking time on ecotype A, 12.66% tetraploidy was identified. Totally, it is seems that Mentha mozaffarianiihardly response to colchicine for tetraploidy induction.

Shallot (Allium ascalonicum L.) has been known as traditional medicine. Shallot which has same genus with garlic(Allium sativum L.) contains allicin that is also found in garlic and has been suspected has fungicidal activity toCandida albicans. It is supported by several researches. Therefore, shallot is suspected has antifungal activity too.The aim of this research was to know antifungal activity of shallot’s water extortion againsts Candida albicans invitro. This was a laboratory experimental research which used completely randomized design, with diffusion method.Shallot’s water extortion was devided into three concentrations, there were 50%, 100% and 200%. Ketoconazole 2%was positive control and aquadest was negative control. The result of this research based on analysis of varians(Anova), there was significant difference between several treatments and was confirmed with Duncan New MultipleRange Test (DNMRT) p<0,05, there was significant difference between 100% shallot’s water extortion with othertreatments, but there was no significant difference between 50% shallot’s water extortion with 200% shallot’s. Theconclusion was shallot’s water extortion had antifungal activity againsts Candida albicans with the best concentration100%, but it was lower than ketoconazole 2%.

Dendrobium anosmum is one of natural orchids in Indonesia. Optimization of medium composition for orchid propagation through in vitro culture is necessary to enhance propagule multiplication capabilities and quality. This study was aimed to study the influence of concentration of coconut water in culture medium on in vitro growth and development of D. anosmum orchid species and to determine the optimal coconut water concentration in culture media.  The experiment were arranged in a Completely Randomized Design with four treatments and eight replications. The treatments consisted of the addition of coconut water with concentrations: 0 ml•l -1 (control), 50 ml•l-1, 100 ml•l-1 and 150 ml•l-1. The results showed that addition of coconut water in culture medium gave different effect on shoot growth and multiplication of D. anosmum orchids.  Coconut water concentration of 100 ml•l-1 was the best concentration for growth and multiplication of D. anosmum orchids, based on both shoots and roots growth, plantlet height and wet weight.

<em>Inhibition of Pseudomonas fluorescens isolates the origin of tomato, soybean and corn against Ralstonia solanacearum tested using a completely randomized design (CRD).        P. fluorescens growth was measured at King's B medium by way of suspension antagonist put on filter paper of 0.5 cm in diameter. Inhibition of P. fluorescens is done by placing the antagonist suspension of 0.5 cm diameter filter paper on NA media that has been deployed R. solanacearum. The results showed that: (a) the growth of P. fluorescens origin of tomato, soybean and corn on King's B media were not significantly different, (b) the inhibition of P. fluorescens isolates against R. solanacearum not significantly different and, (c) the inhibition of isolates P. fluorescens origin of tomato, soybean and corn against R. solanacearum in vitro relatively strong</em>

The objective of this study was to evaluate the efficacy of the active ingredient abamectin, either in isolation or in combination with fungicides and insecticides formulated for the industrial seed treatment, on the population of Meloidogyne javanica, tested under greenhouse and in vitro conditions. In both tests, the combination of the following treatments was assessed: abamectin; thiamethoxam; fludioxonil + metalaxyl-M + thiabendazole. Water was used as control for the in vitro assay, whereas under greenhouse conditions, controls were inoculated and not inoculated with M. javanica. The tests were conducted in a completely randomized design with six (in vitro) and ten (greenhouse) replicates. For in vitro studies the effect on hatching, motility, and mortality of juveniles of M. javanica was evaluated. Under greenhouse conditions, the soybean ‘BRS 133’ seeds were treated, and at 15, 30, and 60 days after inoculation (DAI) with M. javanica, plant measurements were recorded. The penetration of second stage juveniles (J2) was also evaluated at 15 DAI. At 30 DAI, galls, egg masses, nematodes/g of root, and final population were evaluated. At 60 DAI, the final population of nematodes in the roots was quantified. The treatments containing abamectin were the most effective in diminishing the hatching of juveniles. All treatments had an effect on nematode motility when compared to the control, and in the treatment containing only abamectin, total juvenile mortality was observed. In greenhouse conditions, at 15 DAI, the treated soybean plants had the highest root mass and shoot length, differing statistically from the inoculated control. All treatments reduced the number of nematodes per gram of root, differing from the control. At 30 DAI, treatment efficiency was observed in reducing the final population of M. javanica, particularly the treatments using abamectin, and abamectin + thiamethoxam + fludioxonil + metalaxyl-M + thiabendazole. However, at 60 DAI, the effect of the treatments on the population of M. javanica did not persist.

The purpose of this research is to determine the concentration of young coconut water that is appropriate for the growth of orchid plantlets (Dendrobium sp.) In vitro. This study used an experimental method, descriptive data analysis to compare several different concentrations of coconut water. The design of this study uses a completely randomized design (CRD). The treatments consist of 0% coconut water concentration (as a control), 15%, 30% and 60%. Each concentration was carried out 5 replications and each repetition consisted of 5 Dendrobium sp plantlets in each culture bottle conducted for 40 HST, for observing the root length carried out for 50 HST. The highest number of shoots and leaves were produced at the same concentration, namely 150 ml / L coconut water treatment (15% concentration) with an average of 2.8 shoots and the average number of leaves 10.8 leaves. The average number of roots and the longest root length was produced at a concentration of 600 ml / L coconut water (60% concentration) with an average of 6 roots, and the longest root length was 0.5 cm.Keywords: Young coconut water, (Cocos nucifera L.), Dendrobium sp., in vitro, growth.ABSTRAKTujuan penelitian ini ialah menentukan konsentrasi air kelapa muda yang tepat untuk pertumbuhan planlet anggrek (Dendrobium sp.) secara in vitro. Penelitian ini menggunakan metode eksperimen, analisis data secara deskriptif untuk membandingan beberapa konsentrasi air kelapa yang berbeda. Rancangan penelitian ini menggunakan Rancangan Acak Lengkap (RAL). Perlakukan terdiri dari konsentrasi air kelapa 0 % (sebagai kontrol), 15% , 30% dan 60%. Masing-masing konsentrasi dilakukan 5 kali ulangan dan setiap ulangan terdiri dari 5 planlet Dendrobium sp dalam setiap botol kultur yang dilakukan selama 40 HST, untuk pengamatan panjang akar dilakukan selama 50 HST. Jumlah tunas dan jumlah daun terbanyak dihasilkan pada konsentrasi yang sama, yaitu perlakuan air kelapa 150 ml/L (konsentrasi 15%)  dengan rata-rata jumlah tunas terbanyak 2,8 tunas dan rata-rata jumlah daun terbanyak 10,8 helai daun. Rata-rata jumlah akar terbanyak dan panjang akar terpanjang dihasilkan pada konsentrasi air kelapa 600 ml/L (Konsentrasi 60%) dengan rata-rata jumlah akar terbanyak sebanyak 6 akar, dan rata-rata panjang akar terpanjang 0,5 cm.Kata kunci : Air kelapa Muda (Cocos nucifera L.), Dendrobium sp., in vitro, pertumbuhan
